Proteins arc the major molecules performing life activities, and their spatial and temporal expression profiles in organisms are very important for understanding their accurate functions. Phospholipid hydroperoxide glutathione pcroxidase (PHGPx) is a unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes and plays a vital role in defense against mernhrane peroxidation damage. The protein expression profiles of rice PHGPx (OsPHGPx) were investigated in different rice tissues and under various stress treatments by using Western blot analysis. The results showed that in mature rice plants, OsPHGPx was mainly distributed in leaves, especially flag leaves, and in rice seedlings, OsPHGPx was detected in shoots and leaves. Moreover, OsPHGPx expression in rice seedlings could be markedly induced by H2O2 and NaCI, but weakly influenced by several plant hormones. Time- and dose-dependent effects were observed in both H2O2 and NaCI treatments, and the strongest induction was observed when rice seedlings were treated with 0.5 mmol/L H2O2 for 12 h or 500 mmol/L NaCI for 24 h. Additionally, dimethylthiourea, a H2O2 trap, inhibited H2O2-enhanced expression of OsPHGPx, but did not impair the enhanced effect of NaCI, implying that NaCl-induced OsPHGPx expression was not mediated by H2O2. These results will contribute greatly to further study the exact physiological function of OsPHGPx in rice.

Clinical Medicine of China 2011;27(8):833-835
Objective To investigate the expression and significance of VEGF and NF-κB/p65 in the two subtypes of pleomorphic adenoma. Methods Immunohistochemistry was applied to detect the expression of VEGF and NF-κB/p65 in pleomorphic adenoma of 60 patients including 31 cases of cell-rich and 29 cases of stroma-rich as well as normal salivary gland tissues of 30 cases from adjacent tumor. Results VEGF positive staining was mainly found in tumor epithelia, while NF-κB/p65 positive was detected in gland alveolus cell and ductal epithelia. The Mean optical density( MA ) values of VEGF were 955.67 ± 305.79,149. 13 ± 60. 85 and 53.46 ± 9. 66, respectively, in cell-rich adenoma, stroma-rich adenoma and normal control. The difference in VEGF expression between the groups was significant (Ps < 0. 05 ) . The MA values of NF-κB/p65 were 529. 80 ± 164. 81,43.40 ±5.46 and 6. 84 ± 1.91 ,respectively,in three groups mentioned above. The difference in NF-κB/p65 expression between the groups was significant ( Ps < 0. 05 ). In pleomorphic adenoma, the expression level of NF-κB/p65 was positively correlated with VEGF. Conclusion ( 1 ) The angiogenesis and proliferation potential of carcinomas increased with the cell component in pleomorphic adenoma. Stroma-poor adenoma is more frequently subjected to malignant transformation than stroma-rich adenoma. (2) NF-κB/p65 may have effects on angiogenesis by activating VEGF. Detecting the expression of VEGF and NF-κB/p65 may be helpful to predict the biological behavior of pleomorphic adenoma and prognosis of the patients, which couldprovide useful information for future targeted therapy of pleomorphic adenoma.

Objective:To investigate the efficacy and characteristics of subarachnoid xenogenetic adrenal medullary transplants on neuropathic pain. Method: Eighty rats were assigned to 4 different groups before their right sciatic nerves were tied according to the method described by Bennet. Each rat received subarachnoidly xenogenetic medullary pieces or isolated chromaffin cells according to the groups. The basal pain thresholds to thermal and electrical stimuli were determined before nerve ligations, and the analgesiometric tests were repeated at weekly intervals following transplantation for 10 weeks. Behavioral indications of spontaneous pain were recorded concomitantly. The effects of naloxone, phentolamine and fentanyl on the antinociception of chromaffin cells were evaluated. Result:The pain thresholds to noxious thermal and electrical stimuli increased after transplantation of medullary pieces or isolated chromaffin cells subarachnoidly. The Behavioral indications of spontaneous pain and hyperalgesia to thermal and electrial were also eliminated after the transplantation. These antinociceptive effects can be reversed by naloxone and phentolamine. Conclusion:Transplanting xenogenetic chromaffin cells into subarachnoid space can reduce neuropathic pain effectively, and this antinociception is conducted via adrenoreceptors and opiate receptors.

Aim To investigate the role of phospho-lipase C epsilon 1 ( PLCE1 ) in modulating the apoptot-ic mechanism in esophageal cancer ( Eca ) cells. Methods Eca cell lines, OE33 and CP-C cells were cultured to assess the expression of PLCE1 . siRNA suppress expression of PLCE1 . The expression of PLCE1 and p53 was evaluated by quantitative real time PCR and Western blot. Methylation analyses of p53 were performed by bisulfite conversion of genomic DNA. Apoptosis was assessed by flow cytometry. Results OE33 and CP-C cells expressed high levels of PLCE1 . Knockdown of PLCE1 markedly increased the expression and hypomethylation of p53 , and in-creased the frequency of apoptosis. Conclusion PLCE1 suppresses apoptosis of Eca cells via promoting p53 promoter methylation and inhibiting expression of p53 .

Objective To clarity the role of VEGF and its receptors in laryngeal squamous cell carcinoma development and metastasis. Methods The protein and mRNA expression of VEGF and its receptors (Fit-1 and KDR) were examined by Western-blotting and semiquantitative reverse transcriptase polymerase chain reaction analysis in laryngeal squamouse cell carcinoma (20 specimens) and the adjacent non-neoplastic laryngeal tissues (18 specimens). Results The protein and mRNA expression levels of VEGF and its receptors(Fit-1 and KDR) were more abundant in laryngeal squamous cell carcinoma than that in the adjacent non-neoplastic laryngeal tissues (ordedy 4.32±2.21, 2.00±0.91. 1.20±0.55, 0.29~0.31. 2.50±1.69, 0.85±0.28. P<0.01). The expression of VEGF and its receptors(Fit-1 and KDR) in laryngeal squamous cell carcinoma was significantly higher in lymph node positive group than that in lymph node negative group (P<0.01 or P<0.05). Positive correlation was observed between VEGF and KDR in the laryngeal squamouse cell carcinoma and adjacent non-neoplastic laryngeal tissues (P<0.01). There was no correlation between VEGF and Fit-1 expression(P>0.05). Conclusion The intense expression of VEGF, Fit-1 and KDR in laryngeal squamous cell carcinoma provides strong evidence linking VEGF, Fit-1 and KDR expression to the angiogenesis associated with laryngeal squamous cell carcinoma. The positive correlation between VEGF expression and KDR expression clearly demonstrates that KDR is the main signaling receptor for VEGF in laryngeal squamouse cell carcinoma. The VEGF/KDR system plays a functional role in the progression of laryngeal squamous cell carcinoma.

Objective:To investigate the protective role of heme oxygenase-1 and its reaction product, carbon monoxide against acute liver injury induced by carbon tetrachloride in rats. Methods: Thirty male Sprague-Dawley rats were randomly divided into six groups with five in each. The control group received a single dose of corn oil injection.Carbon tetrachloride was injected intraperitoneally(i.p) to establish acute liver injury models in rats.Hemin(50 ?mol/kg) was administered i.p.12 hours before CCl_4 treatment, with an aim to induce HO-1 protein expression in the liver of rats.Carbon monoxide was injected i.p.12 hours prior to CCl_4 injection, resulting in about 8%-12% carboxyhemoglobin concentration in vivo. The expression of HO-1 in the liver of hemin-treated rats was determined by western blot method at different time points. At 24 h after carbon tetrachloride administration, all rats were sacrificed to collect blood samples for the examination of ALT,AST levels and to remove liver tissues for analysis of MDA concentration,SOD activity and caspase-3 activity as well as TNF-a contents. In addition, histopathological changes were investigated and hepatocyte apoptosis was detected by TUNEL method. Results: The administration of carbon tetrachloride to rats caused a marked hepatic damage, characterized by significant elevation of serum ALT,AST levels(2 136.3?163.4 U,1 422.7?221.7 U) and liver MDA con-tent(5.28?0.93 ?mol/g),caspase-3 activitiy(optical density value(4.69)?1.02) and TNF-? level(256.3?27.3 ng/L) combined with a remarkable reduction in liver SOD activity(45.9?14.8 U/mg) as compared with the control rats.Histopathological observations revealed severe damage in the liver and prominent hepatocyte apoptosis took place in CCl_4treated rats. However, pretreatment with hemin could induce high expression of HO-1 protein and exert potent protective effects against liver injury,as demonstrated by a significant decrease in ALT,AST levels(287.1?24.3 U,246.2?21.7 U) and MDA concentration(3.27?1.34 ?mol/g),reduction in caspase-3 activity(optical density value 2.49?1.47) and TNF-? level(132.6?19.5 ng/L),as compared with the CCl_4-treated rats. Moreover, hepatocyte apoptosis and liver injury were both attenuated remarkably in the liver of rats pretreated with hemin. In contrast to hemin administration, single injection of exogenous CO produced the same protective effects,as indicated by the remarkable reduction of ALT,AST levels and caspase-3 activity and TNF-a levels. Conclusion: The above results suggest that HO-1/CO system has a potent protective effect on acute liver injury induced by carbon tetrachloride in rats.Induction of HO-1 expression and low concentration of CO can inhibit the progress of hepatic damage, which might be due to the alleviation of lipid peroxidation and reduction of caspase-3 activity or inhibition of TNF-? level.
