1.Relationship between coronary heart disease and polymorphisms of endothelial nitric oxide synthase gene G894T in Han Nationality

Objective:To explore the relationship between polymorphism of endothelial nitric oxide synthase(eNOS)gene G894T and coronary heart disease(CAD)in Han Nationlity.Methods:Polymerase chain reaction-restricted fragments length polymorphism(PCR-RFLP)was used to determine the genotype of the eNOS gene G894T in 103 patients with CAD and 104 controls.Results:There was significant difference in frequencies of allele in G894T polymorphism of eNOS gene between CAD and control groups(P=0.029),but there was no significant difference in genotype distribution between two groups.Logistic regression analysis revealed that hypertension,diabete mellitus and smoking history were independent risk factors for the incidence of CAD,but T allele was not.Conclusion:The frequency of eNOS gene G894T mutation in the CAD group is higher than that in the control group and T allele may be related to with the development of CAD.

3.Relation of body mass index to vitamin D, PTH, and bone turnover markers levels among women in Shanghai area

Objective To examine the relationship of body mass index (BMI) with levels of vitamin D,parathyroid hormone (PTH),and bone turnover markers among women in Shanghai area.Methods Altogether 810 Chinese women aged 45 year and older were enrolled to study the associations by multiple linear regression analyses.Bone mineral density was measured by a dual energy X-ray absorptiometry (Lunar,Prodigy,GE),Serum 25-hydroxyvitamin D [25 (OH) D],PTH,C-telopeptide of type Ⅰ collagen (CTX),osteocalcin (BGP),and bonespecific alkaline phosphatase (BSALP) were measured.Results (1) 25 (OH) D,BGP,BSALP,and CTX levels were significantly lower in obesity group than in control group ; PTH level was significantly higher in obesity group than in control group.(2) In multiple linear regression analyses,BMI was significantly associated with lower 25 (OH) D (β =-0.017,P =0.006),BGP (β =-0.077,P =0.019),and higher PTH (β =0.025,P =0.000).Conclusions (1) There were siginificant associations between higher BMI and lower 25 (OH) D among women in Shanghai.(2) Serum osteocalcin was associated not only with bone metabolism but also with energy metabolism.
4.Comparative study of antidepressant combined with improved Morita therapy in the treatment of somatoform disorder

Objective To explore the effects of antidepressant combined with improved Morita therapy in the treatment of somatoform disorder. Methods 78 patients with somatoform disorder were randomly divided into study group (medication combined with improved Morita therapy,39 cases) and control group (medication,39 cases). Both groups were assessed with symptom checklist 90 (SCL-90), Eysenck personality questionnaire ( EPQ )and global assessment scale ( GAS ) before and after treatment. Results After treatment, the factor scores in SCL-90 of the study group were (1. 14±0.41),(1.81 ±0.44),(1.27 ±0.41),(1.20±0.54),(1.49 ±0. 66 ), ( 1.25 ± 0. 42 ), ( 1. 09 ± 0.48), ( 1.07 ± 0. 42 ), ( 1.16 ± 0. 42 ), and that of the control group were ( 1. 46±0.44) ,(2.19±0.50),(1.56±0.40),(1.54±0.56),(1.85 ±0.48),(1.47±0.44),(1.33±0.54),( 1.38 ± 0. 58 ), ( 1.38 ± 0. 45 ), there were significant differences ( P < 0. 05, P < 0. 01 ). EPQ-E and EPQ-N of the study group were 7.02 ±4.06 and 12.7 ±3.89 respectively ,EPQ-E and EPQ-N of the control group were 14.8± 4.3 and 8.97 ± 4.32 respectively, there were significant differences ( P < 0.05 ). The obvious effective rates of two groups were 82. 05% and 56.41% (x2 =4.875, P<0. 05 ). Conclusion The effects of antidepressant combined with improved Morita therapy is much better in the treatment of somatoform disorder.
5.Association between the efficacy of alendronate and serum level of vitamin D in patients with osteoporosis

Objective To examine the association between vitamin D and alendronate response,and to investigate the proper vitamin D levels for the efficacy of alendronate treatment of osteoporosis.Methods In this retrospective study,559 post-menopausal women with osteoporosis were devided into two groups:non-responders and responders,based on the European Study of Forsteo (EUROFORS).Demographic and clinical data including mean 25-hydroxy vitamin D (25-OHD) levels between dual-energy X-ray absorptiometry (DEXA) scans were obtained.Mean 25-OHD levels were compared between responders and non-responders and multiple logistic regression analysis was performed to identify factors associated with non-response.Results There were 437 (75.5%) responsers to alendronate therapy and 142 (24.5%) non-responders.Responders with a mean serum 25-OHD level of (59.96 ±12.56) nmol/L,obtained a more favorable result than non-responders,whose serum 25-OHD level was (47.50 ±9.92) nmol/L (P<0.01).25-OHD level was significantly associated with response.Conclusion Patients with a mean 25-OHD ≥50 nmol/L had a substantially greater likelihood of maintaining bisphosphonate response.
6.An advance standard curve method in fluorescence real-time PCR

0bjective To establish a standard curve method with more accuracy employed in fluorescence real-time PCR(RT-PCR)as a alternation of the general method.Methods β-actin and KLK11 plasmid DNA for quantitative standard curve were constructed in our study,and Plasmids of β-actin was employed as a internal control.After serial dilution these plasmid were used as DNA standard to obtained slope.Expression of these two genes in malignant prostate cancer cell line LNCAP were tested by real-time PCR,and we analyzed the RT-PCR results with two different methods and compared their accuracy.Results Thestandards curves made from these linear DNA standards showed good linearity (R2=0.991 and 0.992 for β-actin and KLK11 standards graphs),but also displayed a discrepancy in their PCR efficiency(β-actin 123% and KLK11 99%).There were different results after two different stand curve analytical method:the expression of KLK11 mRNA in LNCAP was downregulated in general standard curve method.In the new analytical method,howerer,KLK11 upregnlated for 4.46-fold.And there was a significant difference between aplification efficiency of targt gene and internal control gene(t=4.829,P<0.05).Conclusions Compared with general standard curve method,the new advanced standard curve method described here avoids an error which considers there is identical amplification efficiency between target gene and internal reference gene.It is considered to be a more correct analytical method in fluorescence real-time PCR.

Objective To identify PDZ domain containing proteins interacting with PTEN and its characterization with NHERF-1 by proteomic analysis. Methods The interactions between PTEN and PDZ domain containing proteins were screened with PDZ protein array, and the novel one was then identified with GST pull-down and co-immunoprecipitation assay. Results Using a PDZ protein array, we proved PTEN binding with NHERF-1. The interaction of PTEN and NHERF-1 was further characterized by GST pull down assay, and we demonstrated that PTEN associated with NHERF-1 via the binding of PTEN carboxyl-terminal with the PDZ domain 1 (PDZ1) of NHERF-1. The last four amino acids (I-T-K-V) of the PTEN were the key determinants of this interaction as mutation of any of the four amino acids to alanine resulted in markedly reducing association of PTEN with NHERF-1. In addition, the full-length of PTEN robustly associated with NHERF-1 was also determined by co-immunoprecipitation experiment in cos-7 cells. Conclusion PTEN/NHERF-1 association was mediated via the binding of PTEN carboxyl-terminal]with the PDZ1 of NHERF-1, and the last four amino acids of the PTEN carboxyl-terminal were important for PTEN/NHERF-1 interaction.
9.Comparison of epidemiological situation in chronic kidney disease between urban and rural areas in Guangxi province

ObjectiveTo investigate and compare the prevalence,awareness and risk factors of chronic kidney disease (CKD) between urban and rural population in Guangxi province in order to provide information for prevention and treatment of CKD.MethodsBy a stratified multistage random sampling method,18 to 74 years old residents in Guangxi province were surveyed. They wereevaluatedbyquestionnaire,urinaryalbumin/creatinineratio, hematuria (microscopic examination of centrifuged urine sediment),kidney B-mode ultrasound,and abnormal results were reviewed 3 months later.Estimated glomerular filtration rate(eGFR) was calculated with the simplified MDRD equation modified by a Chinese coefficient.The risk factors associated with CKD were also investigated.Results There were no significant differences between urban and rural residents in the prevalence of albuminuria (5.22% vs 5.47%) and hematuria (1.07% vs 1.11%)(all P>0.05).The prevalence of renal lithiasis in rural residents was significantly higher than that in the town(10.54% vs 6.95%)(P<0.05).The decreased renal function between urban and rural residents(3.87% vs 4.04%,P>0.05) had no significant difference.The prevalence of CKD was 9.58% in urban and 9.42% in rural(P>0.05).The prevalence of albuminuria according to the age distribution was different between urban and rural,which increased along with the age in urban but showed two peaks(30-40 years old and 60-74 years old) in rural.Based on logistic regression analysis,the risk factors for albuminuria were diabetes,hyperuricemia,the history of cardiovascular disease,chronic tonsillitis and HBsAg positive.The risk factors for kidney function decline were age,hyperuricemia,hypertension,diabetes,renal lithiasis and history of cardiovascular disease.The awareness rate of CKD in urban was significantly higher than that in rural (14.45% vs 6.27%,P<0.05).Conclusions The prevalenceof CKD has no significant difference between urban and rural in Guangxi province.The awareness rate of CKD in urban is significantly higher than that in rural.It is needed to enhance the prevention and treatment of CKD in rural.
10.Study on therapeutic effect of auricular acupuncture combined with drug in patients with frequent ventricular premature beat

Objective To compare the efficacies between auricular acupuncture combined with drugs and drug alone in the treatment of frequent ventricular premature beats(FVPB).Methods A total of 51 pa-tients with FVPB admitted to this hospital from January to December 2022 were selected as the study sub-jects,including 27 patients in the treatment group and 24 cases in the control group.The treatment group was given auricular acupuncture combined with oral metoprolol tartrate tablets,the heart and ear points and adre-nal ear points were selected,the two ears were treated alternately and the needles were replaced after 3 days.The control group was only given oral metoprolol tartrate tablets,and the adhesive tape of the same color and material were pasted at the same auricular acupoints.The number of premature beats,the TCM symptom score and clinical efficacy on 1 day before treatment and on 1,6 days after treatment were compared between the two groups.Results The number of premature beats on 1 day after treatment in the treatment group was 733(427,1 637)times,which in the control group was 2 244(1 023,2 808)times.On 6 days after treatment,the number of premature beats in the treatment group was 182(116,321)times,which in the control group was 1 025(678,1 127)times.On 6 days after treatment,the number of premature beats in the treatment group was averagely reduced by 765.49 times compared with the control group(95%CI:-976.41 to-554.56,P<0.001).On 1,6 days after treatment,the TCM symptom scores in the treatment group all were lower than those in the control group,and the difference was statistically significant(P<0.05).The effective rate of the treatment group was 92.59%,which was higher than 83.33%in the control group,and the differ-ence was statistically significant(P<0.05).Conclusion Compared with drug therapy alone,auricular acu-puncture combined with drug therapy for treating FVPB could achieve better clinical efficacy.
