1.A Comparative Study of CISA, WAIS-RC in the Violent Adolescents with Lower Intelligence

Object: To compare validity of CISA and WAIS-RC in a special group, violent adolescents with low intelligence.Method:Thirty-eight adolescents with violent delinquency were screened out by CRT (combined Raven test) with IQ lower than 85. CISA (Chinese intelligence scale for adult) and WAIS-RC (Wechsler adult intelligence scale-revised Chinese version) were applied to them in different sequence.Result:CISA IQ and WAIS-RC IQ were very close to each other. The adolescents were in borderline intelligence with more impairment in verbal IQ than in performance IQ.Conclusion:CISA and WAIS-RC have same validity in identifying intelligent impairment of violent adolescents.
2.Correlation Analysis of 113 Patients with Drug-induced Liver Disease

OBJECTIVE: To analyze 113 patients with drug-induced liver disease(DILD) so as to clinically validate the international generally-accepted diagnostic criteria.METHODS: 113 DILD cases were reviewed and categorized according to international generally-accepted diagnostic criteria and then graded separately by Maria clinical scale and DDW-J scale to analyze the correlations.RESULTS: 113 out of 822 patients with liver diseases were DILD(13.7%),among whom 17 were of hepatocellular type(15.0%),92 cholestasis type(81.5%) and 4 mixed type(3.5%).Application of Maria clinical scale and DDW-J scale led to different number of DILD cases diagnosed.DILD were mostly induced by antihyperlipidemic drugs and immunosuppressants followed by anticoagulant drugs.CONCLUSION: DDW-J scale,which is more close to clinical diagnostic criteria,contributed to the standardization of the diagnostic criteria of DILD,yet it has its limitations as well.
3.QT-interval and its dispersion in type 2 diabetic and non-diabetic patients with post-myocardial infarction and effects of different hypoglycemic drugs on QT-interval dispersion

Objective To compare the QTc and QTcd between type 2 diabetic and non-diabetic patients with post-myocardial infarction (post-MI) ,and to compare the QTcd in type 2 diabetic patients with post-MI treated with insulin,sulfonylurea,mefformin,or diet alone. Methods We measured the QTc and QTcd through simultaneous 12-lead Electrocardiogram in 138 post-MI patients,including 70 type 2 diabetic (of which,23 were assigned to re-ceive insulin,20 glipizide,16 mefformin,11 diet control) and 68 non-diabetic patients. Result Compared with post-MI patients without diabetes,those with type 2 diabetes had significantly higher QTc [(377.2±24.3) ms vs (342.9±27.5)ms,t=7.79,P<0.01] and QTcd [(48.8±19.7)ms vs (40.3±26.6)ms,t=2.14,P<0.05]. There were no significant difference between the mefformin group and the diet control group (P>0.05). The QTc and QTcd in the insulin group were significantly higher than those in the other three group s(P<0.05),and the QTc and QTcd in the glipizide group were higher than those in the mefformin group or diet control group(P<0.05,and P<0.01,respectively). Conclusion Type 2 diabetes is associated with an additional increase in the QTcd in post-MI patients,suggesting higher mortality risk in post-MI patients with type 2 diabetes. Insulin and glipizide may in-crease the QTc and QTcd in post-MI patients with diabetes. These effects were more significant in the insulin therapy group.
4.Analysis on Resistance Status of 279 Strains of Mycobacterium tuberculosis

Objective The purpose of the study was to understand the resistance status of to anti-tuberculosis drug in Yunnan province, and to provide evidence for tuberculosis (TB) control. Methods The proportional method was used in drug sensitivity test according to the guidelines recommending by WHO. Results Of 301 mycobacterial isolates, 279 (92.7%) strains were complex (MTBC) . The total drug resistance rate was 31.5% (88/279),the initial drug resistance rate of was 27.3%, and the acquired drug resistance rate was 44.3%. The drug resistance rates to INH,RFP,EMB and SM were 19.7%, 10.8%, 12.9%and 19.0%, respectively. The multi-drug resistance (MDR) rate was 9.3%. Conclusion The drug resistant rate of is still high in Yunnan province,and TB control is still a long way to go. Early detection and regular treatment should be done for MDR patients. Drug resistance monitoring as an important part of TB control programs should be regular observation and research.

Objective To study the expression of matrix metalloproteinase (MMP) and the tissue inhibitor of matrix metalloproteinase (TIMP) in hepatocellular carcinoma (HCC) and its relation with the prognosis of patient. Methods MMP 1,MMP 2,MMP 9 and TIMP 2 in human HCC tissue and HCC cell line were measured and quantified with immunohistochemistry, Northern blot hybridization and image analysis. Results MMP 1,MMP 2 and MMP 9 were localized in the cytoplasm of tumor cells. Objective To explore the relationship between ERG4 gene overexpression and azole resistance in clinical Candida albicans strains. Methods The National Committee for Clinical Laboratory Standards (NCCLS)M27-A2 broth microdilution method was conducted to evaluate antifungal susceptibility of 34 clinical Candida albicans isolates in vitro. Total RNA was extracted from these Candida albicans strains and transcribed into cDNA. Real-time fluorescence-based quantitative PCR was performed to determine the mRNA expression of ERG4 gene. Statistical analysis was carried out by a two-sample t-test. Results The expression level of ERG4 mRNA was significantly higher in fluconazole-resistant than in -sensitive Candida albicans strains (4.20 ± 2.56 vs. 1.72 ± 1.33, t = 3.99, P < 0.05), higher in itraconazole-resistant than in -sensitive Candida albicans strains (3.60 ± 2.47 vs. 1.66 ± 1.61, t = 3.71, P < 0.05), and higher in voriconazole-resistant than in -sensitive Candida albicans strains (3.99 ± 2.72 vs. 2.07 ± 1.58, t = 2.91, P <0.05). Further more, increased ERG4 mRNA expression was also observed in isolates cross-resistant to all the three azole antifungal agents compared with those susceptible to all of them (4.49 ± 2.73 vs. 1.69 ± 1.82, t = 3.81, P < 0.05). Conclusions The overexpression of ERG4 gene may be associated with cross resistance to fluconazole, itraconazole and voriconazole in clinical Candida albicans strains, but its exact role is expected to be investigated through downregulation of the ERG4 gene.

Objective To investigate the risk factors for postmenopausal osteoporosis in Shanghai.Methods Five hundred postmenopausal community-dwelling women aged 45-80 years were recruited. The case-control study was performed from June 2008 to September 2008.A total number of 250 women with postmenopausal osteoporosis identified with their bone mineral density (BMD) were selected as case group, and 250 non-osteoporosis women were selected as control group. BMD was measured by dual energy X-ray absorption (DEXA). Results Univariate logistic regression analysis showed that age, eduction level, occupation, years since menopause, BMI, use of calcium,historyofnon-violencefracture,fall,diabetesmellitus,chronicgastricdiseases, gastrointestinal resection and diarrhea were related to osteoporosis.Multiple logistic regression showed that age, years since menopause and nutritional status were the risk factors for osteoporosis. ConclusionsThe occurrence of osteoporosis is related with many factors in postmenopausal women in Shanghai, and women with early menopause, low BMI and older age should pay more attention to the prevention and treatment of osteoporosis.
9.Expression of p66Shc in heart failure patients and its relationship with oxidative stress

Objective To investigate the expression of cardiac muscle protein p66Shc in patients with congestive heart failure and its relationship with oxidative stress.Methods From October 2016 to March 2017, eighty patients with acute heart failure treated in the cardiology department of Shanghai Pudong Hospital were collected in this study,including 22 cases of grade II,26 cases of grade III,32 cases of grade IV,and 56 healthy subjects were selected in the control group.SOD activity,MDA content and p66Shc mRNA expression of the acute stage of heart failure,the recovery stage of heart failure and the control group were measured.The changes of SOD,MDA and p66Shc in patients with heart failure and the control group were analyzed.The expression of p66Shc,along with MDA and SOD were analyzed.Results There were significant differences in the SOD,MDA and p66Shc mRNA of patients in the acute stage of heart failure,the recovery stage and the control group were (SOD:(65.67±8.31)kU/L vs.(75.77±7.56)kU/L vs.(90.03±7.76)kU/L,MDA:(10.22±1.56)μmol/L vs.(8.34 ± 1.75)μmol/L vs.(5.21 ± 2.16)μmol/L,p66Shc mRNA:(5.79 ± 1.71)vs.(3.95 ± 1.24) vs.1.87±0.93))(F=2.903,2.762,2.696,P<0.05).With the aggravation of heart failure,the level of SOD decreased significantly(NYHA grade Ⅱ:(67.54 ± 7.29)kU/L;grade Ⅲ:(61.62 ± 6.96)kU/L;gradeⅣ:(57.94±6.75)kU/L,F=2.872,P<0.05)and the levels of MDA(NYHA grade Ⅱ:(9.32±1.18)μmol/L;grade Ⅲ:(10.95 ± 1.26)μmol/L;grade Ⅳ:(12.13 ± 1.07)μmol/L,F=2.703,P<0.05)and p66Shc (NYHA grade Ⅱ:(4.92 ± 0.96);grade Ⅲ:(5.66 ± 1.17);grade Ⅳ:(6.28 ± 1.15),F=2.681,P<0.05) increased significantly.The expression of p66Shc and mRNA was positively correlated with MDA in acute attack (r=0.722,P<0.05),and negatively correlated with SOD(r=-0.670,P<0.05).Conclusion P66shc is closely related to oxidative stress in heart failure,and p66Shc may be involved in the process of heart failure oxidative stress,which can provide a basis for the new treatment of heart failure.
10. Platelet derived growth factor-BB regulates phenotype transformation of pulmonary artery smooth muscle cells via SIRT3 affecting glycolytic pathway

Objective:
To investigate whether platelet-derived growth factor-BB (PDGF-BB) can regulate phenotypic transformation of pulmonary artery smooth muscle cells (PASMCs) via SIRT3 affecting glycolytic pathway.
Methods:
The PASMCs were isolated from Sprague Dawley rats. PASMCs were divided into 3 groups by using 2-deoxyglucose (2-DG), an inhibitor of the glycolytic pathway: normal control group, PDGF-BB group(30 ng/ml) and PDGF-BB (30 ng/ml)+2-DG (10 mmol/L) group. In lentivirus-mediated overexpression assay, cells were divided into control group, PDGF-BB group(30 ng/ml), PDGF-BB+deacetylase sirtuin-3 (SIRT3) overexpression group and PDGF-BB+empty vector group. The expression levels of phenotype related index such as α-smooth muscle actin (α-SMA), smooth muscle myosin heavy chain (SM-MHC), calponin, vimentin were detected by qRT-PCR and Western blot. Meanwhile, the expression of α-SMA was detected by cellular immunofluorescence staining. EDU staining was used to detect the proliferation of PASMCs. The expression of SIRT3 was detected by Western blot. The expressions of glucose transporter 1 and aerobic glycolytic enzymes were detected by qRT-PCR and Western blot in lentivirus-mediated overexpression assay.
