Objective To investigate the expression of neurotrophin- 3 (NT- 3) gene in Schwann cells (SCs) of rat sciatic nerve introduced by adenovir al vector in vivo. Methods Recombinant adenoviral vector for NT- 3 was propagat ed in 293 packaging cells and was titered by tissue culture infections dose 50 ( TCID50) method. Ad- NT- 3 was injected intraneurally into the rat sciatic nerv e following axotomy of the sciatic nerve which was repaired after the immediate injury. Immunohistochemical staining was employed to determine the expression of NT- 3 in SCs in rat sciatic nerve, and the expressive intensity difference was measured with LEICA M550 imagine analysis system on the tissue slides. Results Two days after injection of NT- 3 into the rat nerve, positive stain in the SCs were apparent in the vicinity of anastomosis. There was significant increase of the amount of NT- 3 expression 7 days following injection of Ad- NT- 3 (vs. 2 days group P0.05). AIM: Hydroxymethylglutaryl CoA (HMG-CoA) reductase inhibitors, such as simvastatin, have been shown to reduce atherosclerotic cardiovascular morbidity and mortality by mechanisms unrelated to its lipid-lowering effect. Several studies have shown that simvastatin induces apoptosis in a varieties of cell lines including vascular smooth muscle cells (VSMC). The aim of this study was to investigate the signal pathways involved in apoptosis induced by simvastatin. METHODS: Cultured VSMC were treated with simvastatin. Calpain activity was determined by measuring Ca 2+ ionophore-specific calpain substrate (suc-LLVY-AMC), caspase-3 activation was detected by Western blot, and apoptotic changes were distinguished by annexin Ⅴ binding and DNA laddering. Cell smears were made from fresh mammary tissue during operation. There are 32 samples with definite pathological diagnosis. DNA content was measured in interphase nuclei of single epithelial cells of lesions of the human breast by a microspectrophotometric technique. The normal lymphocytes from 2 cases of normal blood samples and from 4 cases of mammalopathic tissue were used as a reference for the DNA value corresponding to a diploid number of chromosomes. The normal lymphocyte had mean DNA value in 11.2?0.2 A.U. and 10.9?1.3 A.U. The rest of the measurements showed: 12.1?1.3 A.U. in the, epithelial nuclei of 10 cases of neighbouring mammalopathic tissue; 22.6?1.0 A.U. in 9 cases of fibrcma of the breast; 31.9?2.3 A.U. in 8 cases of carcinomas of the breast. The cell nuclei of carcinomas showed abnormal DNA values and the multiploid cells were dominant. It's histograms showed a shift to right and either a double-peak or multiploid-peak distribution. The DNA contents of the benign lesion cells were significantly lower than that of cancer cells, but higher than that of normal cells. The peak of their histogram showed they are between 2C-4C.

AIM: To observe the effect of simvastatin on the proliferation of vascular smooth muscle cells(VSMCs) induced by serum and growth factor PDGF-BB and the effect of simvastatin on the expression of PTEN,a important regulator of G 1/S cell cycle transition. METHODS: The DNA synthesis was determined by -TdR incorporation, cell cycle was examined with flow cytometry, the protein level of PTEN was measured by Western blot method. RESULTS: (1)Simvastatin inhibited -TdR incorporation in a dose dependent manner. (2) Flow cytometric DNA analysis revealed that simvastatin induced significantly enhancement of G 0/G 1 phase and decrease in S phase VSMCs.(3)Simvastatin increased protein level of PTEN and mevalonate, a metabolite of HMG-COA, reversed the effect of simvastatin on PTEN protein expression. CONCLUSION: Simvastatin may inhibit proliferation of VSMCs and retarded cell cycle in G 0/G 1 phase by increasing PTEN expression through inhibiting synthesis of mevalonate.

Objective To compare the QTc and QTcd between type 2 diabetic and non-diabetic patients with post-myocardial infarction (post-MI) ,and to compare the QTcd in type 2 diabetic patients with post-MI treated with insulin,sulfonylurea,mefformin,or diet alone. Methods We measured the QTc and QTcd through simultaneous 12-lead Electrocardiogram in 138 post-MI patients,including 70 type 2 diabetic (of which,23 were assigned to re-ceive insulin,20 glipizide,16 mefformin,11 diet control) and 68 non-diabetic patients. Result Compared with post-MI patients without diabetes,those with type 2 diabetes had significantly higher QTc [(377.2±24.3) ms vs (342.9±27.5)ms,t=7.79,P<0.01] and QTcd [(48.8±19.7)ms vs (40.3±26.6)ms,t=2.14,P<0.05]. There were no significant difference between the mefformin group and the diet control group (P>0.05). The QTc and QTcd in the insulin group were significantly higher than those in the other three group s(P<0.05),and the QTc and QTcd in the glipizide group were higher than those in the mefformin group or diet control group(P<0.05,and P<0.01,respectively). Conclusion Type 2 diabetes is associated with an additional increase in the QTcd in post-MI patients,suggesting higher mortality risk in post-MI patients with type 2 diabetes. Insulin and glipizide may in-crease the QTc and QTcd in post-MI patients with diabetes. These effects were more significant in the insulin therapy group.

OBJECTIVE To validate the anticancer effect of Angong Niuhuang pill (AGNH) and pinpoint associated molecular mechanisms using human cancer cells.METHODS Human MGC-803 gastric carcinoma and human BEL-7402 hepatocarcinoma cells were incubated with AGNH 9,30,90,300 and 900 mg·L-1 for 24,48and 72 h,respectively.Cell viability was detected with 3-(4,5-dimethylthiazol-2-yl) -5-( 3-carboxymethoxyphe-nyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay and colony formation assay.Apoptosis was measured with flow cytometry and Hoechst 33258/PI staining.Change in mitochondrial membrane potential (△qψ) was detected by spectrofluorophotometer.RESULTS AGNH inhibited MGC-803 cell proliferation ( for 48 h,r =0.996,P =0.002; for72 h,r=0.756,P=0.024 ) and BEL-7402 cells (for 48 h,r =0.732,P=0.030; for72 h,r=0.702,P =0.037) in a concentration-dependent manner,as showed by MTS assay.AGNH inhibited colony formation on MGC-803 cells (r =0.914,P =0.011 ) and BEL-7402 cells (r =0.871,P =0.024) in a concentration-dependent manner for 24 h.Hoechst 33258/PI staining and flow cytometry assay showed that AGNH 900 mg·L-1 for 24 h induced apoptosis of MGC-803 and BEL-7402 cells,and the apoptosis rate was 27.2％ and 19.7％,respectively.Compared with normal control group,AGNH 900 mg·L-1 for 3 min decreased the mitochondrial membrane potential of MGC-803 and BEL-7402 cells to 15.9％ and 15.0％ of control group.CONCLUSION AGNH inhibits proliferation of human cancer cells.Apoptosis and depolarization of mitochondrial transmembrane potential are probablly its mechanism.

Objective:To explore the association between dopamine-β-hydroxylase (DβH), norepinephrine transporter (NET) gene polymorphisms and panic disorder(PD).Methods:The structured clinical interview for the diagnostic and statistical manual of mental disorders fourth edition (DSM-Ⅳ) axis Ⅰ disorders was administered by trained clinical psychiatrist, 139 patients with PD(PD group) and 196 healthy controls(control group) were enrolled in the study.Single nucleotide polymorphism(SNP) genotyping was performed using an improved multiplex ligation detection reaction technique.SPSS 16.0 and PLINK softwares were used to compare the allele frequency and genotype distribution.Results:(1)Compared with control group, PD group carried more G allele(76.3% vs 68.4%) and fewer A allele(23.7% vs 31.6%) in NET rs5569, and the difference was significant(χ 2=4.986, OR=0.67, 95% CI: 0.47-0.95, P<0.05). However, the correlation was no longer significant after adjusting for Bonferroni’s multiple testing( P>0.05). (2)The additive model of NET rs5569 showed a association with PD ( OR=0.68, 95% CI: 0.48-0.96, P<0.05). And the recessive model of DβH rs1611114 showed a association with PD( OR=0.42, 95% CI: 0.18-0.96, P<0.05). However, these correlations were no longer significant after adjusting for Bonferroni's multiple testing( P>0.05). (3)No matter allele or genotype, there were no significant differences in DβH (rs129882, rs1611114, rs1611115) and NET (rs2242446, rs28386840) gene polymorphisms between panic disorder group and control group(all P>0.05). Conclusion:The present study indicates that there is no significant association of DβH and NET gene polymorphisms with PD.

Objective To assess the clinical application values of HC visual laryngoscope in emergency tracheal intubation for severe craniocerebral trauma patients with basilar fracture.Methods Retrospective analysis was used to review 60 patients of severe traumatic brain injury with basilar fracture in emergency resuscitation room who needed the hospital emergency tracheal intubation.There were 30 patients operated with visual HC laryngoscope (the observation group), and the other 30 patients underwent common direct laryngoscope (the control group).The index of the degree of exposure,intubation time,success rate of intubation were compared between the two groups.ResultsThere were 28 cases (93.33%) of Cormark-Lehane grading Ⅰ to Ⅱ in observation group,which was significantly higher than 20 cases (66.67%) in the control group(P< 0.05).The average intubation time in observation group was (34.3±6.7) seconds,and it was shorter than (44.5±5.7) seconds in the control group (P<0.05).In observation group,26 cases (86.67%) were of one-time successful intubation,which was higher than 19 cases (63.33%) in the control group,and the difference was statistically significant(P<0.05).ConclusionHC visual laryngoscope in emergency tracheal intubation for severe craniocerebral trauma patients with basilar fracture can improve the success rate of intubation,shorten the intubation time,reduce adverse reactions,and it is worthy of clinical application.

OBJECTIVE To investigate the correlated resistant genes encoding ?-lactamases,aminoglycoside and genetic marker of integron and transposon in multi-resistant Pseudomonas aeruginosa(MRPA) isolated from clinical specimens and study their relationship by phylogenetic analysis.METHODS Twenty-one resistant genes,two integron-Ⅰ genes and one transposon genetic gene were analyzed by PCR and verified by DNA sequencing.Multi-resistant genes cluster analysis was performed by UPGMA.RESULTS The positive rates of CARB,oprD2,aac(3)-Ⅱ,aac(6′)-Ⅱ,ant(2″)-Ⅰ,intⅠ1,qacE△1-sul1 and merA in 20 strains of MRPA were 15%,100%,70%,15%,15%,85%,85% and 85%,respectively,and other genes were negative.It was classfied to three subgroup,by the multi-resistant genes cluster analysis.CONCLUSIONS MRPA isolated from clinical specimens has carried many resistant genes.The deficiency rate of oprD2 gene is very high.The positive rate of genetic mark genes about integron and transposon is very high.It may be the main multi-resistant mechanism of MRPA.Multi-resistant genes cluster analysis shows that there is clone transmission in MRPA and it can induce nosocomial infection prevalence.

Objective To assess the prevalence and severity of burnout in medical staffs,and investigate associated demographic,perceived social support and job burnout.Methods Totally 618 medical staffs form Sichuan province people's hospital were selected by the systematic sampling method,and were tested by general state questionnaire,job burnout questionnaire and perceived social support scale.Results ①The prevalence of job burnout was 75.6％,including mild job burnout (36.1％),moderate (32.2％),severe (7.3％).②The scores of emotional exhaustion in the female medical staffs(20.50±8.35) was higher than the male medical staffs (19.07±7.76,P＜0.05).Divorced medical staffs had higher scores(18.28±7.13) than that of single and married medical staffs (15.05±5.00) in low personal accomplishment (P＜ 0.01).The medical personnel of different ages were statistically significant difference in total job burnout scores,emotional exhaustion and depersonalisation (F=4.475,3.061,3.942,all P＜0.05).The medical personnel of different post,professional title and working years were statistically significant difference in total job burnout scores and emotional exhaustion (F=6.920,13.968; 3.506,3.391;3.190,3.300,all P＜ 0.05).The medical personnel of different educational attainment were statistically significant difference in total job burnout scores,emotional exhaustion and low personal accomplishment(F=6.218,2.837,2.850,all P＜0.05).)Correlation analysis found that emotional exhaustion,depersonalisation,low personal accomplishment and total job burnout respectively with family support,friend support,and other support and total social support was significantly negative correlation (r=-0.126～-0.403,P＜0.01).④Regression analysis showed that education,family and friends support were protective factors for total job burnout(OR=-0.248,95％ CI =0.625-0.975 ; OR=-0.084,95％ CI =0.862-0.980 ; OR=-0.091,95％ CI =0.860-0.968 ; all P＜0.05).Conclusion The prevalence of job burnout is high in medical staffs.The medical staffs of different sex,age,title,jobs,education,working years,marital status are showed the differences job burnout scores,which is closely related to the perceived social support.
