1.Study on the β-catenin regulating osteogenesis of human periodontal ligament stem cells under mechanical tension

Objective To investigate the effect of β-catenin on the osteogenic differentiation of human periodontal ligament stem cells (PDLSCs) under mechanical tension.Methods PDLSCs were seperately cultured in vitro from the periodontium attached at freshly extracted teeth.β-catenin-targeting agonist or inhibitor was applied to the PDLSCs to upregulate or downregulate the expression of activity of β-catenin protein.The expression of β-catenin protein and the osteogenetic related markers(ALP,BMP2,Runx 2) under the mechanical tension with different period(0,6,12,24 h) were assessed with Western blot in the PDLSCs irritated with agonist or inhibitor.Results Compared with the PDLSCs without mechanical tension,the expression of osteogenesis related marker proteins,ALP and Runx 2,in PDLSCs were enhanced under the circular mechanial tensile stress (P < 0.05).In the PDLSCs irritated with WAY-262611,the β-catenin-targeting agonist,the expression of osteogenetic related markers ALP in the PDLSCs was improved during the first 12 h period of mechnical tensile load,which was significantly higher than that in the DMSO group.Conclusion Wnt/β-catenin signaling pathway plays an important role in the early osteogenesis differentiation of PDLSCs under cyclic tension.The β-catenin promotes osteogenic differentiation of PDLSCs under mechanical tension.
2.Effect of vancomycin different dose on blood concentration and kidney function in the elderly

Objective:To discuss the influence of different vancomycin administration dosage in elderly patient's through blood concentrations and renal function.Methods:The clinical data about 113 elderly patients treated from January 2016 to December 2018 in our hospital was reviewed. After therapy, the through blood concentrations, administration dosage and renal function changes were detected to analyze their relationship.Results:The peak plasma concentration of 113 elderly patients was significantly higher than that of the young and middle-aged group ( P<0.05). Plasma concentrations in 185 cases were detected, including 35 cases with through blood concentrations<10 mg/L, 71 cases at 10-20 mg/L and 79 cases at >20 mg/L. There were 15 cases with 0.5 g/d of vancomycin, 96 cases at 1.0 g/d, 54 cases at 1.5 g/d and 20 cases at 2.0 g/d. The through blood concentrations among patients with different daily doses were significantly different ( P<0.05); at 1.0g/d, the ratio with through blood concentrations at 10-20 mg/L was the highest; serum creatinine (Cr) and blood urea nitrogen (BUN) contents in patients with through blood concentrations >20 mg/L were higher than those at <10 mg/L and 10-20 mg/L ( P<0.05); before treatment, Cr and BUN contents among groups were not significantly different ( P>0.05); among 113 elderly patients, the results indicated 21 renal function impairment cases and 92 normal renal function cases. The daily doses and through blood concentrations in renal function impairment group were higher than normal group ( P<0.05); BUN contents in renal function impairment group were higher than normal group while endogenous creatinine clearance rate was lower than normal group ( P<0.05); before and after treatment, BUN contents in groups were not significantly different ( P>0.05); after treatment, endogenous creatinine clearance rate for two groups was lower than before ( P<0.05); the results indicated that renal function was negatively correlated with vancomycin administration dosage and positively correlated with through blood concentrations ( P<0.05). Conclusions:During therapy, the renal function shall be closely monitored. According to patient's conditions and through blood concentrations, the vancomycin administration dosage shall be adjusted. In particular, the defined daily doses shall be reduced in renal function impairment patients, which through blood concentrations are kept at 10-20 mg/L.
3.Relationship between defense style and family environment among male prisoners of different terms of imprisonment

ObjectiveTo explore the psychological defense styles and family environment among male prisoners of different terms of imprisonment, and to analyse the relationship between their psychological defense styles and family environment. MethodsA total of 200 male prisoners in a prison in Guangdong were randomly selected from April to June 2015, and they were classified into less than 5 years of imprisonment group (n=108) and 5 years or more imprisonment group (n=92). Their defense style and family environment were evaluated by Defense Style Questionnaire (DSQ) and Family Environment Scale-Chinese Version (FES-CV). Pearson correlation analysis was used to test the correlation between each scale score. ResultsThe factor scores of immature defense mechanism and intermediate defense mechanism of DSQ in the group with less than 5 years of imprisonment were lower than those in the group with 5 years or more imprisonment, with statistically significant differences (t=4.198, 1.137, P<0.01). The scores of FES-CV family intimacy, emotional expression and organizational factors in less than 5 years of imprisonment group were higher than those in 5 years or more imprisonment group (t=3.122, 2.993, 2.203, P<0.01), and the scores of contradiction factors were lower than those in 5 years or more imprisonment group (t=-3.682, P<0.01). The scores of DSQ immature, intermediate defense mechanism and concealment factor of male prisoners were negatively correlated with the scores of FES-CV family intimacy and emotional expression factor (r=-0.428~-0.172, P<0.05 or 0.01), and the scores of 4 factors in DSQ were all positively correlated with the scores of contradiction factor in FES-CV (r=0.175~0.384, P<0.05 or 0.01). ConclusionCompared with male prisoners with less than 5 years of imprisonment, those with 5 years or more are prone to adopt immature and intermediate defense mechanisms, and their family environments are characterized by apparent contradiction and a lack of family intimacy, emotional expression and organization. Furthermore, the defects of psychological defense mechanism of prisoners are related to their family environment.
