1.The genetic features of pan-drug resistant pseudomonas aeruginosa

Objective To investigate the genetic features of pan-drag resistant pseudomonas aeruginosa. Methods The susceptibilities to 14 antibacterial agents were detected in pseudomonas aerations by K-B paper diffusing method. A strain of pan-drug resistant pseudomonas aeruginosa was selected randomly, and was used to amplify genes for β-Lactam antibiotic resistance (TEM, SHV, CTX-M-1 group,CTX-M-2 group, CTX-M-9 group, OXA-1 group, OXA-2 group, OXA-10 group, PER, GES, VEB,CARB, LCR, BEL, DHA, IMP, VIM, SPM, GIM, SIM and oprD2), aminoglycosidase drug resistance genes (including aminoglycosides modification enzymes gent,16S rRNA methylase gene) and genetic markers of plasmid (traA and traF), integrons (tnpA, tnpU and merA), transposons Ⅰ (int Ⅰ 1 ) by PCR,and the sequences of above genes were analyzed. Results The pseudomonas aeruginosa was resistant to all 14 antibacterial agents, and the following genes were positive in PCR: TEM-1 and CARB-3 types of genes for β-Lactam antibiotic resistance (deletion of oprD2), aac (6')-Ⅱ and ant (2")-Ⅰ of aminoglycosidase resistance genes, transposons Ⅰ (int Ⅰ 1 ), and the merit of integrons. Conclusions Pseudomonas aeruginosa shows high resistance to most antibacterial agents, which should draw attention in clinic.
2.Enzyme Production and Drug Resistance of Pseudomonas aeruginosa

OBJECTIVE To investigate the 16S rRNA methylases gene and AMEs of 30 strains multi-resistant Pseudomonas aeruginosa isolated from clinic work in our hospital.METHODS Collected 50 P.aeruginosa strains from two hospitals(30 strains of multi-resistant P.aeruginosa from Yantai,Shandong,20 strains of pan-drug resistant P.aeruginosa from Jiangsu),and analyzed 6 kinds of 16S rRNA methylases(armA,rmtA,rmtB,rmtC,rmtD and npmA) and 6 kinds of AMEs gene aac(3)-Ⅰ,aac(3)-Ⅱ,aac(6′)-Ⅰb,aac(6′)-Ⅱ,aac(3″)-Ⅰ,and aac(2″)-Ⅰ by PCR and sequence analysis.RESULTS Among 30 strains of multi-resistant P.aeruginosa from Yantai,the positive rate of 4 kinds of genes aac(3)-Ⅱ,aac(6′)-Ⅱ,aac(3″)-Ⅰ,and aac(2″)-Ⅰ was 20%(6/30),46.7%(14/30),36.7%(11/30) and 3.3%(1/30).The other 8 kinds genes were all negative.The total positive rate of AMEs gene was 46.7%(14/30).In 20 strains of pan-drug resistant P.aeruginosa,the positive rate of 6 kinds of genes rmtB,aac(3)-Ⅱ,aac(6′)-Ⅰb,aac(6′)-Ⅱ,aac(3″)-Ⅰ,and aac(2″)-Ⅰ was 55%(11/20),60%(12/20),35%(7/20),60%(12/20),50%(10/20) and 45%(9/20).The other 6 kinds genes were all negative.The total positive rate of AMEs gene was 95%(19/20).CONCLUSIONS It is the first report that 16S rRNA methylases gene is existed in P.aeruginosa;there is very high positive rate of AMEs genotypes in P.aeruginosa;there are differences in gene existing among two hospitals.
3.Distribution and Resistance of Coagulase-negative Staphylococcus

OBJECTIVE To provide laboratory evidence for the prevention and control of coagulase negative staphylococcus(CNS),and study the distribution and drug resistance of CNS in our hospital.METHODS CNS of inpatients from Oct 2005 to Dec 2007 was isolated and identified with ATB Expression microbe identification and drug sensitivity system.RESULTS A total of 354 strains of CNS were isolated,from the main samples of secretion,sputum,blood and cerebrospinal fluld.The isolation rate from departments of pediatrics,ICU,orthopedics and neurology were 9.90%,9.30%,9.00% and 5.60%,respectively.CONCLUSIONS CNS is playing a significant role in nosocomial infection.The drug resistance of CNS is very serious.To pevent nosocomial infection,it is critically important to monitor the antimicriobial resistance of CNS and use autibiotics more rationally.
4.Influence of Clinical Pharmacy Service on Community Hypertension and Coronary Heart Disease Patients with Chronic Diseases

Objective: To evaluate the effects of clinical pharmacists on the community hypertension and coronary heart disease management.Methods: The clinical pharmacy service management program for community chronic disease patients was designed, and the medication files of the patients with hypertension and coronary heart disease were established, and then the medication safety cognition and chronic disease medication of the patients were investigated.Pharmaceutical interventions were given twice for the irrational drug use and living habits and the views of pharmacy were put forward.The improvement of medication compliance, pharmaceutical opinion acceptance and blood pressure etc was evaluated, and then the drug cost and the cost of health care products were analyzed and evaluated.Results: The medication and hypertension compliance increased, the patients were satisfied with the clinical pharmacy service, and the acceptance rate of pharmaceutical intervention opinions was improved significantly.The total cost of medication was without significant change, and the cost of health care products decreased significantly.Conclusion: It is an effective way to expand the impact of clinical pharmacy performed by clinical pharmacists on chronic disease medication management in communities, which can improve the rationality and safety of drug use in the patients with chronic diseases and has positive effects in promoting the management of community chronic diseases.
5.Study on the β-catenin regulating osteogenesis of human periodontal ligament stem cells under mechanical tension

Objective To investigate the effect of β-catenin on the osteogenic differentiation of human periodontal ligament stem cells (PDLSCs) under mechanical tension.Methods PDLSCs were seperately cultured in vitro from the periodontium attached at freshly extracted teeth.β-catenin-targeting agonist or inhibitor was applied to the PDLSCs to upregulate or downregulate the expression of activity of β-catenin protein.The expression of β-catenin protein and the osteogenetic related markers(ALP,BMP2,Runx 2) under the mechanical tension with different period(0,6,12,24 h) were assessed with Western blot in the PDLSCs irritated with agonist or inhibitor.Results Compared with the PDLSCs without mechanical tension,the expression of osteogenesis related marker proteins,ALP and Runx 2,in PDLSCs were enhanced under the circular mechanial tensile stress (P < 0.05).In the PDLSCs irritated with WAY-262611,the β-catenin-targeting agonist,the expression of osteogenetic related markers ALP in the PDLSCs was improved during the first 12 h period of mechnical tensile load,which was significantly higher than that in the DMSO group.Conclusion Wnt/β-catenin signaling pathway plays an important role in the early osteogenesis differentiation of PDLSCs under cyclic tension.The β-catenin promotes osteogenic differentiation of PDLSCs under mechanical tension.
6.INFLUENCE OF ARTESUNATE ON INFECTIVITY OF GAMETOCYTES OF PLASMODIUM FALCIPARUM

A study was carried out in south of Vietnam 15 Patients with Plasmodium falciparum gametocytes and asexual parasites were distributed into groups A, B and C. Artesunate was given orally at a total dose of 600 mg for 5 days in group A, 200 mg for 2 days in group B and intravenously at a total dose of 360 mg for 5 days in group C respectively. Gametocytes count was done before medication and daily after medication. Meanwhile, Anopheles dirus as a vector was employed to study the infectivity of gametocyte, The result showed that the mean gametocyte clearance time in three groups were respectively 15.4?5.0, 20.6?4.8 and 20.3?4.0 days. The mosquitoes were not infected from the blood in 2, 5 and 5 of 5 patients respectively on days 7, 14 and 21 in group A; 1 and 5 of 5 patients on day 14 and 21 in group B; 2 of 5 patients and 3 of 3 patients on days 14 and 21 in group C. It indicates that artesunate has remarkable effect on Plasmodium falciparum gametocytemia and its infectivity to mosquitoes.
7.Chemical constituents of roots of Boehmeria nivea.

OBJECTIVETo investigate the chemical constituents from roots of Boehmeria nivea.
METHODThe constituents were isolated by repeated column chromatography and preparative liquid chromatography; and their structures were elucidated by chemical properties and spectroscopic analyses.
RESULTSeven compounds were isolated and their structures were identified as tormentic acid (1), hederagenin (2), maslinic acid (3), 2alpha-hydroxyursolic acid (4), trans-p-hydroxycinamic acid (5), 2,4,4'-trihydroxychalcone (6), rutin (7).
CONCLUSIONCompounds 1-6 were obtained from this genus for the first time.

8.Antidepressant activities of flavonoids from Glycyrrhiza uralensis and its neurogenesis protective effect in rats.

Adult rats chronic unpredictable stress model of depression (CUS) was adopted to elucidate the antidepressant pharmacological activity and related neurogenesis protective effect of the total flavonoids extract (licorice flavonoids, LF) from the Glycyrrhiza uralensis Fisch. cultivated locally in Ningxia. The rats were exposed to 9 kinds of unpredictable sequence of stressors and were given flavonoids (300 mg x kg(-1), 100 mg x kg(-1) and 30 mg x kg(-1)) for 28 days. The antidepressant effect was elucidated by open field test, forced swimming test and tail suspension test. The level of serum corticosterone was detected by radioimmunoassay. 5'-Bromo-2'-deoxyuridine (BrdU) labeling experiments was employed to study the neurogenesis protective activities. The flavonoids can increase the sum of line crosses and number of rears, and decrease the number of fecal boli produced in the open field test of the CUS rats. Also the flavonoids can decrease the immobility time in forced swim test as well as in the tail suspension test. In addition, the flavonoids (300 mg x kg(-1)) can decrease the serum corticosterone level of the CUS rats, and increase the number of the new born BrdU positive progenitor cells at the subgranular zone (SGZ) of dentate gyrus (DG) region in hippocampus. The results demonstrated that the total flavonoids extract from the cultivated Glycyrrhiza uralensis Fisch. could produce the anti-depressive effect on chronic unpredictable stress of depression model rats and its mechanism may be associated with its neurogenesis protective effect.
9.Expression of porcine interferon-gamma and its safe antiviral assay.

In order to ensure the biosafety of the IFN-gamma antiviral activity assay, we used a replication-deficient VSV carrying GFP as an interferon sensitive indicator virus (VSVdeltaG*G). The antiviral activities of porcine IFN-gamma expressed in Escherichia coli and in baculovirus on MDBK cells were assessed. The results showed that the antiviral activity of porcine IFN-gamma expressed in baculovirus could reach 10(5) IU/mL, while the porcine IFN-gamma expressed in E. coli showed some antiviral activity (32 IU/mL) after refolding. The results of the VSVdeltaG*G-based antiviral assay were almost identical to that of the VSV*GFP-based assay, suggesting it is highly feasible to use VSVdeltaG*G as a substitute for VSV*GFP, making assays for IFN-gamma antiviral activity safer and more accurate.

In the paper, it is introduced professor's recognition on the pathogenesis and professor's experience in the treatment of the motor impairment of the trunk after stroke. Professorbelieves that the motor impairment of the trunk after stroke is the essential factor affecting the rehabilitation in stroke. The motor impairment of the trunk after stroke results from brain marrow damage and spiritual impairment. Hence, regaining the consciousness and promoting the circulation of the governor vessel are the basic principles of the treatment, named regulating the mind and controlling, benefitingand warming, tonifying the kidney and filling up the essence, and promoting the circulation of the governor vessel. Those four therapeutic methods are equally important. Acupuncture, moxibusiton and herbal medicine are applied in combination in the treatment. Additionally, the psychotherapy and rehabilitation are the accessory therapies.
