1.Therapeutic effect of erythropoietin gene-modified bone marrow mesenchymal stem cell transplantation on rat cerebral infarction

BACKGROUND:Previous studies have shown that erythropoietin can protect neurons and promote nerve regeneration. OBJECTIVE:To explore the therapeutic effect of erythropoietin gene-modified bone marrow mesenchymal stem celltransplantation via caudal vein on rat cerebral infarction. METHODS:Western blot assay was used to identify the expression of exogenous erythropoietin in bone marrow mesenchymal stem cells. A model of middle cerebral artery occlusion was established in Wistar rats using thread method. And then, model rats were randomly divided into model group (PBS injection via the caudal vein), transplantation group (transplantation of bone marrow mesenchymal stem cellsuspension), erythropoietin group (transplantation of erythropoietin-transfected bone marrow mesenchymal stem cellsuspension). Neurologic function was assessed at 3 days, 1, 2, 3, 4 weeks after celltransplantation. Four weeks after transplantation, the rats were decapitated after anesthesia to take brain tissues for RT-PCR detection of Bcl-2/Bax gene expression. cellapoptosis was measured by TUNEL. Hematoxylin-eosin staining and fluorescence microscopy were employed to observe the survival and distribution of PKH26-labeled bone marrow mesenchymal stem cells. RESULTS AND CONCLUSION:Western blot results showed that erythropoietin-transfected bone marrow mesenchymal stem cells could express the erythropoietin in vitro. At 1, 2, 3, 4 weeks after transplantation, the neurological defect scores in the transplantation group and erythropoietin group were significantly lower than those in the model group (P<0.05, P<0.01). The expression of bcl-2 gene in the infarct region was significantly higher in the erythropoietin group than the transplantation and model groups (P<0.05), but the expression of bax was significantly decreased (P<0.05). In the erythropoietin group, the number of apoptotic cells was reduced, and the number of PKH26 positive cells was increased as compared with the other two groups (P<0.05). These findings indicate that the transplantation of erythropoietin-modified bone marrow mesenchymal stem cells via caudal vein can significantly improve the neurological function in the rats with cerebral infarction.
2.Effect of bone marrow mesenchymal stem cells combined with Danhong injection on expression of GAP-43 and Bcl-2 after cerebral infarction

BACKGROUND:Danhong injection, scavenging free radicals and inhibiting lipid peroxidation, can improve microenvironment injury after cerebral infarction. OBJECTIVE:To explore the influence of bone marrow mesenchymal stem cel s combined with Danhong injection on expression of GAP-43 and Bcl-2 after cerebral infarction in rats. METHODS:Sixty Wistar rats were selected to prepare models of cerebral infarction by middle cerebral artery occlusion and then randomly divided into control group, bone marrow mesenchymal stem cel group, and combination group. Control group received tail vein injection of PBS. Bone marrow mesenchymal stem cel group received tail vein injection of 2.5×109/L bone marrow mesenchymal stem cel suspension. Combination group received injection of 2.5× 109/L bone marrow mesenchymal stem cel suspension+2 mL/kg Danhong injection, for 5 consecutive days, once a day. RESULTS AND CONCLUSION:There were no significant differences in the neurological dysfunction scores among the three groups at 24 hour and 3 days after implantation (P>0.05). The neurological dysfunction scores in the ombination group were significantly lower than those in the bone marrow mesenchymal stem cel group and control group at 1 and 2 weeks after transplantation (P<0.05). In the combination group, GAP-43 and Bcl-2 expression was significantly higher than the bone marrow mesenchymal stem cel group and control group (P<0.05). Bone marrow mesenchymal stem cel transplantation combined with Danhong injection can significantly promote the local expression of GAP-43 and Bcl-2 after cerebral infarction, and has obvious inhibitory effects on cel apoptosis in rats with cerebral infarction.

OBJECTIVE: To investigate the effects of deproteinised calf blood injection(DCBI) on blood glucose and lipid metabolism and its ameliorating effect on insulin resistance(IR) in model rats with type 2 diabetes mellitus(T2DM) complicating cerebral infarction.METHODS: Rats were fed with high fat and high glucose diet and injected with streptozotocin(STZ) vial caudal vein to duplicate T2DM model,followed by duplication of middle cerebral artery occlusion(MCAO) rat model.Then the rats were assigned to model group,sham-operated group,DCBI low and high dosage group(30,60 mg? kg-1?d-1 intraperitoneally),and rosiglitazone control group;meanwhile,another normal group was established.After receiving the corresponding drugs for 5 weeks for each group,fasting blood glucose(FBG),serum insulin(FINS) and sugar tolerance(OGTT) were determined,and levels of insulin resistance index(IRI),insulin sensitivity index(ISI),total cholesterol(TC),triglyeride(TG),high-density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),and free fatty acid(NEFA) levels were computed.RESULTS: As compared with model group,in DCBI-treated model rats with T2DM complicating cerebral infarction,FBG were significantly decreased,sugar tolerance increased significantly(P0.05);IR was ameliorated(P
4.Efficacy of traditional Chinese medicine in the treatment of intrahepatic cholestasis of pregnancy

Objective To discuss the efficancy of traditional Chinese medicine in treatment of intrahepatic cholestasis of pregnancy .Methods A total of 60 patients who were admitted to our hospital for severe intrahepatic cholestasis of pregnancy(ICP) from January 2009 to December 2012 were randomly divided into two group using random number table .Patients in study group (n=30) took traditional Chinese medicine three times a day plusUDCA twice a day orally .Patients in control Group (n=30)were on‐ly given UDCA twice a day orally .Conventional treatment and care for ICP were given to both groups .Biochemical index (TBA , ALT ,and AST)level ,pregnancy outcome and Caspase‐3 of the two groups were recorded and compared and compared .Results The expressing of Caspase‐3 in placental cytotrophoblast ,syncytiotrophoblast .Decidual cells and interstitial cells of study group were (10 .58 ± 2 .62)% ,(15 .8 ± 4 .02)% ,(10 .17 ± 3 .58)% ,(14 .23 ± 4 .39)% ,respectively ,which were significantly lower than those in control group(P<0 .05) ,which were (20 .46 ± 8 .68)% ,(33 .89 ± 7 .05)% ,(20 .56 ± 4 .23)% ,(28 .29 ± 5 .58)% .TBA ,ALT、AST in two groups were declined ,and ctedy group was better than in control group(P<0 .05) .The index of liver function were all lower in study group than that of control group(P<0 .05) .Incidence rate of neonatal asphyxia was lower in study group than that of con‐trol group (P<0 .05) .Conclusion Traditonal Chinese medicine can reduce the expression of Caspase‐3 in study group .It is safe and eflfective in the treatment of ICP .
5.Effects of Radix inulae in Guanxi Suhe Pill on cardiomyocytes injury induced by H_2O_2

AIM: To observe the change of Guanxi Suhe Pill(GXSH) in cardiomyocytes suffering from hydrogen peroxide-induced injury due to replacement of Aristolochia debilis sieb.et Zucc by Radix inulae. METHODS: Myocardial cells from neonatal SD rats were cultured in dulbecco's modified eagle's medium(DMEM).The primary culture cardiomyocytes were injured by hydrogen peroxide(1 mmol/L) for 4 h,and serum containing GXSH was added to the medium at the same time.The creatine kinase(CK) activity,lactate dehydrogenase(LDH) activity and malondialdehyde(MDA) content in culture medium were determined.Intracellular superoxide dismutase(SOD) activity,the activity of Na~+-K~+-ATPase and Ca~(2+)-Mg~(2+)-ATPase in the cardiomyocytes were also observed. RESULTS: GXSH containing Aristolochia debilis sieb.et Zucc,GXSH containing Radix inulae,and GXSH without above both herbs significantly inhibited the release of LDH and CK,decreased the content of MDA and increased SOD activity,increased the volume of Na~+-K~+-ATPase and Ca~(2+)-Mg~(2+)-ATPase of hydrogen peroxide-injured cardiomyocytes.The effects of TG and BG were stronger significantly than those of QG on some parameters. CONCLUSION: GXSH all has a remarkable protective effect on cardiomyocytes suffering from hydrogen peroxide-induced injury without and with Aristolochia debilis sieb.et Zucc or taking the place of the GXSH containing Radix inulae do not influence its protective effects on cardiomyocytes suffereing from hydrogen peroxide-induced injury.
6.Comparative pharmacokinetics of active component in Guanxin Suhe Pills containing Radix Inulae in place of Radix Aristolochiae

Objective To study the pharmacokinetics of the active component in series of Guanxin Suhe Pills including the prescription containing Radix Aristolochiae(QG),the prescription containing Radix Inulae(TG),and the prescription without any of them(BG) in Beagle dogs and to compare the pharmacokinetics among the series of prescription.Methods HPLC Method was developed to determine acid in plasma of Beagle dogs.Blood plasma was collected after series of Guanxin Suhe Pills were ig administrated to Beagle dogs 0—8 h later.The concentration of cinnamic acid in the plasma was determined and DAS 2.1 software was used in calculation of compartment model and pharmacokinetic parameters.Results The standard curve was linear from 0.031 25 to 32 mg/L in plasma of Beagle dogs,the lowest detectable limit was 31.25 ?g/L,the recovery rate of the method was over 90 % with satisfactory relative standard deviations(RSDs) of intra-day and inter-day both lower than 5 %.The metabolism of cinnamic acid in plasma of Beagle dogs after medication of series in QG,TG,and BG all fitted in a first order absorption of two-compartment model.The peak concentration and the area under curve in TG and BG groups had the higher tendency than those in the QG group,but there was no significant difference among the three groups.Conclusion The series of Guanxin Suhe Pills all have a similar pharmacokinetic process.Taking away the Radix Aristolochiae from the previous prescription that including the Radix Aristolochiae or taking the place of the Radix Aristolochiae with Radix Inulae does not influence the pharmacokinetic process of the main active component.

8.Arthroscopic treatment of ulnar impaction syndrome

Objective To investigate ulnar impaction syndrome imaging characteristics and wrist arthroscopy in the diagnosis and treatment effects.Methods From September 2006 to April 2014,41 patients with ulnar impaction syndrome were treated.Of all 41 patients,28 were male and 13 female with an average age of 34.5±3.9 years old (ranging from 15 to 52 years old).The preoperative values of wrist range of motion (ROM) were 44.17°±6.22° at volar flexion,42.72°±3.37° at dorsal extension,17.45°±4.11° at radial deviation,19.12°±3.70° at ulnar deviation,66.81°±4.28° at pronation,and 70.27°±5.30° at supination.Arthroscopic exploration was conducted in all patients,aiming to observe the ulnar variance,wrist osteonecrosis,articular cartilage damage and triangular fibrocartilage complex tear.Arthroscopic synovectomy were performed in all cases;33 cases of articular cartilage injury were treated with arthroscopic chondroplasty;16 cases of osteonecrosis were treated with arthroscopic drilling decompression;30 cases with tear of TFC were treated with arthroscopic repair;26 cases with ulna positive mutation were treated with arthroscopic distal ulna resection.Patients were followed at 3,6,and 12 months after operation.At 12 months follow-up,the values of wrist range of motion was evaluated for volar flexion,dorsal extension,radial deviation,ulnar deviation,pronation,and supination.GreenO'Brien method was used to evaluate the function of wrist joint.Results Primary healing was presented in all patients without neurovascular injury and joint infection.All patients were followed up 12-36 months (mean,17.5 months).At 12 months followup,the values of ROM compared with that preoperative increased with significant difference,49.13° ±3.47° at volar flexion,47.13°±2.39° at dorsal extension,19.33°±3.29° at radial deviation,25.17°±4.89° at ulnar deviation,75.73°±7.66° at pronation,and 77.22°±4.89° at supination.According to Green-O'Brien standard,the results were excellent in 21 case,good in 17 cases and fair in 3 case,with 93% excellent and good rate.Conclusion Wrist arthroscopy dose not only contribute to the diagnosis in ulnar impingement syndrome but also can be used in treating inflammatory synovium and cartilage lesions,bone drilling decompression,TFC repair and arthroscopic distal ulna resection with improvement in the range of motion of the wrist joint and function.
9.Investigation on Occupational Hazards and Electric Welding Operation

Objective To study occupational hazards effects of electric welding,and evaluate protective measurements for different occupational hazards. Methods During 2008,occupational health services for 11 factories with shield carbon-dioxide arc welding work were provided,including surveillance of the work environment and surveillance of workers’health. Surveys of workers’health through various types of health examinations were carried out. Four hundred and thirty-three welding workers of 11 factories were included. Results The concentrations of 17 welding fume samples and 6 manganese dioxide samples were over the released limits of national standard,the maximum was 5.5,1.6 times of the limit respectively. The exceeding standard rate. of welding fume had positive correlation with the density of welding equipment(r= 0.72,P=0.01) and negative correlation with the ventilation area(r =-0.63,P =0.04) .The examination results showed that nobody suffered occupational diseases or occupational contraindications. Conclusion For the environmental risk factors existed in the electric welding operation site,more attention should be paid to the potentially occupational adverse effects of electric welding.
10.The effects of different methods of getting out of bed for the first time in patients receiving lumbar disc herniation operations

Objective To compare the effects of different methods of getting out of bed for the first time in patients receiving lumbar disc herniation operations.Methods Totally 60 patients receiving lumbar disc herniation operations were divided into the experimental group and the control group with 30 patients in each group.The patients in the experimental group got out of bed with prone position.The patients in the control group got out of bed with lateral position.The pain scores and degrees of comfort were recorded.Results The experimental group scored 5.4±1.3 in pain degree,and the control group scored 6.1±1.8,t=0.37,P > 0.05,the difference was not significantly different.The experimental group scored 85.7±10.9 in comfort degree,and the control group scored 70.6±11.5,t=4.21,P < 0.05,the difference was significantly different.Conclusions The method of getting out of bed with prone position can reduce negative emotions of patients receiving lumbar disc herniation operations and improve degrees of comfort.
