ObjectiveTo explore the effect of atopic status on interleukin-4 (IL-4),interferon-γ (IFN-γ) and interleukin-5 (IL-5) levels in serum of children wih mycoplasma pneumoniae (MP) infection,and to investigate the relation between these cytokines and disease severity.MethodsNinety-five children with MP infection were enrolled in our study,which were divided into atopic group and non-atopic group,mild group and severe group,normal-TIgE group and high-TlgE group.Meanwhile,38 normal children were enrolled as normal control group.The serum levels of IL-4,IFN-γ and IL-5 were detected by ELISA method.Results(1) Thelevels of serum IL-4 (pg/ml),IFN-γ (pg/ml) and IL-5 (pg/ml) in MP infection group[2.82(2.12,7.13 ),39.70(18.82,181.42),16.12( 10.35,28.07 ) ] were higher than those in normal control group [ 1.17(1.05,1.60),13.25(8.77,22.56),9.72(6.11,12.39) ] ( P＜0.05 ).(2) Serum IL-5 levels in atopic group [ 15.55 (10.35,22.61 ) ],severe group [ 15.98 ( 11.95,26.58 ) ] and high-TlgE group [ 16.55( 10.35,23.65 ) ] were much higher than those in normal control group( P＜0.05 ).Rather than the serum IL-5 levels of atopic group,mild group and normal TIgE group showed no significant difference compared with the normal control group( P＞0.05 ).(3) No significant differences of serum IL-4,IFN-γ,andIL-4/IFN-γ were found between atopic group and non-atopic group,mild group and severe group,normalTIgE group and high-TIgE ( P＞0.05 ).ConclusionIL-4,IFN-γand IL-5 are involved in the pathogenesis of MP infection,furthermore,atopic status of the host may lead to the increase of serum IL-5,which may enhance the progress of the disease.

Mycoplasma pneumoniae(MP) is one of the most important pathogens for respiratory tract infections in older children and adults. And it also has a close relationship with the development, acute exacerbation and persistence of chronic difficult-to-control symptoms of asthma.But so far,the mechanisms inducing or causing asthma remain unclear.Possible mechanisms include direct damage of bronchial mucosa, IgE-mediatad immune response, a variety of cytokines and inflammatory mediators releasing and the propensity of host.

Objective:To study the inhibitory effect of tumor-selective replication-competent adenovirus-mediated human endostatin(CX-hE)on transplanted ovarian cancer(OV-90) in nude mice.Methods: BALB/c nude mice were inoculated subcutaneously with OV-90 cells to establish the animal model bearing human ovarian cancer.Eighteen nude mice bearing cancer were divided into 3 groups to receive intratumoral injection of PBS,CX-hE or Ad-hE,1/2 d,5 times.Then their livers were harvested for pathologic examination.Another 15 nude mice were divided into 3 groups to receive single intratumoral injection of CX-hE,Ad-hE or ONYX-015.Venous blood was collected on day 1,3 and 7 after injection for hEndo measurement by ELISA.The tumors were harvested for pathologic examination and immunohistochemical staining.Results: The tumors grew more slowly in CX-hE group and their sizes were markedly smaller than those of Ad-hE group(P

Objective To screen the liver cell proteins interacting with the novel protein encoded by the 2.2 kb singly spliced variant of hepatitis B virus genome. Methods The splicing-specific gene TPss generated by the 2.2 kb singly spliced variant of HBV genome was amplified by PCR and cloned into the bait vector pGBKT7. After exclusion of self-activatian capacities of TPss protein, a two-hybrid library screening was performed using a pre-transformed human liver cDNA library to screen the liver cell proteins interacting with TPss. Mammalian two-hybrid assay was also done to further confirm the interactions between the bait and prey proteins in Huh7 and HepG2 hepatacytes. Results TPss gene with the size of 336 bp was successfully amplified and cloned, and the TPss protein expressed well in AHI09 yeast cells. Four liver cell proteins interacting with TPss, i. e. , cathepsin B, epoxide hydrolase 1, cathepsin D and fibrinogen gamma chain, were screened by yeast two-hybrid assay and further confirmed by mammalian two-hybrid assay. Conclusion TPss could interact with liver cell proteins.

Objective The changes of thyroid hormone and cyclic nucleotides levels in rats with spleen-kidney deficiency syndrome were observed to evaluate the preventive and therapeutic effect of Yougui Wan(YW) for spleen-kidney deficiency syndrome,and to explore the relationship between the spleen and the kidney at microscopic level.Methods Male SD rats were randomly divided into normal control group,model group,and YW group.Rat models of spleen-kidney deficiency syndrome were established by gavage firstly with rhubarb(4g for each rat) decoction and then with hydrocortisone(25 mg?kg-1?d-1,im).The model group and YW group were treated with distilled water and YW(5.8 g?kg-1?d-1) respectively.The general status of rats was observed,and the plasma cyclic adenosine monophosphate(cAMP) and cyclic guanosine monophosphate(cGMP) levels as well as serum thyroid hormones(T3,T4 and rT3) and thyroid-stimulating hormone(TSH) contents were determined in all rats by radioimmunoassay(RIA).Results Compared with the normal control group,the serum contents of T3,T4,rT3 were decreased significantly(P

【Objective】To observe the morphologic changes of organs in mice with spleen deficiency and the effect of Qiangji Jianli Oral Liquid(QJOL).【Methods】Forty healthy male NIH mice were randomized into 4 groups:A(normal),B(model),C(treated with QJOL)and D(treated with Si Junzi Decoction).Except group A,the mice in other 3 groups received intraperitoneal injection of reserpine to induce spleen deficiency.Meanwhile,groups C and D received QJOL and Si Junzi Decoction in the dosage of 26?g?kg~(-1)?d~(-1) by gastric gavage respectively,and groups A and B received the same volume of distilled water for 21 days.Twelve hours after last administration,the body weight and pathological changes of liver,kidney,spleen and thymus were examined.【Results】In group A,the body weight decreased,and liver,kidney,spleen and thymus gland showed different degrees of damage.QJOL and Si Junzi Decoction could increase the body weight and promote the repair of the organs.【Conclusion】Different degrees of damage can be found in liver,kidney,spleen and thymus gland of mice with spleen deficiency,and spleen-strengthening medicinal herbs can promote their repair,indicating that the theory of spleen controlling the muscles,being the solitude organ and localized the center to cultivate the surroundings organs like the earth,which is described in Huangdi's Canon of Medicine,has the morphologic evidence.
