1.Effects of annexin Ⅱ antisense vector on the growth of transplanted tumor in nude mice

Objective To analyze the effects of annexin Ⅱ antisense expression vector on the growth of the transplanted tumor in nude mice. Methods The SPC-A-1 cells (parental group) and SPC-A-1-annexin Ⅱ cells (antisense group) were inoculated subcutaneously in nude mice respectively. The forming time, volume, and weight of tumor were measured. The tumor cells and the surrounding tissues were observed microscopically. Results The forming time of tumor in the antisense group was later than that in the parental group. The volume and weight of tumor in the antisense group were lower than those in the parental group. Furthermore, invasion of tumor in the surrounding tissues was found in 2 cases in the parental group, but not in the antisense group. Conclusion The annexin Ⅱ gene may be important in promoting tumor cell growth and invading the surrounding tissues.
2.Effects of annexin Ⅱ antisense vector on the growth of lung cancer cells

Objective To explore the effects of antisense vector of annexinⅡ gene on the growth of SPC A 1, a human lung cancer cell line. Methods The total RNA was isolated from human lung cancer cell line SPC A 1 and the target DNA fragments were amplified by RT PCR. The antisense expression vector was constructed by double restriction endonuclease cleavage directional clone method. Annexin Ⅱ antisense expression vector was introduced into SPC A 1 cells by liposome transfection reagent. The expression of annexin Ⅱ mRNA was analyzed by semi quantitative RT PCR. The effects of antisense vector of annexinⅡ gene on the growth of SPC A 1 were observed. Results The antisense vector of annexinⅡ gene was constructed and introduced into SPC A 1 cells successfully. Semi quantitative RT PCR showed that the annexin Ⅱ mRNA expression reduced by about two thirds in the transfected cells as compared with that in the untransfected cells. Compared with the untransfected cells, transfected cells decreased significantly in cell growth, clone formation efficiency in plating and DNA synthesis. Cell cycle was blocked in G 0 G 1 phase. Conclusion Annexin Ⅱ could promote the growth of lung cancer cells and may be helpful for the development of lung cancer.
3.Two kinds of decompression and implant internal fixation for the treatment of cervical spinal stenosis:C5 nerve root palsy and stability

BACKGROUND:Some scholars suggest that the nerve root palsy after cervical spinal stenosis treated with
decompression and implant internal fixation is related with the cervical stability and cervical lordosis, but there is controversial.
OBJECTIVE:To explore the C 5 nerve root palsy and stability after cervical spinal stenosis treated with posterior laminectomy lateral mass fixation and single-door decompression laminoplasty.
METHODS:Twenty-nine cervical spinal stenosis patients were selected and treated with posterior
decompression and implant internal fixation. Posterior laminectomy lateral mass fixation for the treatment of
cervical spinal stenosis:C3-6 lateral mass and C7 pedicel screw internal fixation was performed and caused rough surface on the facet joint;the unstable segment was confirmed according to the preoperative anteraposterior
plain film and dynamic radiographs combined with MRI and CT images, and then the corresponding segments were treated with lateral mass internal fixation, single-door decompression laminoplasty and laminoplasty.
RESULTS AND CONCLUSION:Al the 29 cervical spinal stenosis patients were fol owed-up for 8 months to 2.3 years. Among them, 14 cases were treated with posterior laminectomy lateral mass fixation, two cases had nerve root palsy in the early stage after implantation, three cases had incomplete paralysis after long-term symptom recurrence and treated with second surgery of scar remove and decompression;15 cases were treated with single-door decompression
laminoplasty, and one case had C 5 never root palsy and shoulder abduction dysfunctionafter treatment, no preoperative symptom recurrence. The nerve root palsy wil restored in 6 weeks for shortest and 9 months for longest. As the limitation of the case number, it is not clear whether there were significant differences in the correlation between C 5 nerve root
palsy and segmental stability, cervical lordosis, spinal decompression degree and the range for spinal cord shift, as wel as the nerve root palsy degree and the cervical spinal stenosis recurrence caused by forward scar between two
treatment methods, so accumulation observation of the cases and clinical experience are needed.

5.Neural Stem Cells Combined with Olfactory Ensheathing Cells Improve the Recovery of Motor Function in Rats Subjected to Spinal Cord Transected Injury

Objective To explore the engrafted effect of NSCs combined with OECs on the motor function repair of hindlimbs in rats subjected to spinal cord transected injury.Methods The NSCs and OECs were isolated from the Transgenic Green Fluorescent Protein(GFP) embryo mouse' hippocamp and neonatal mouse' olfactory bulb.Rats were divided into NSCs group,OECs group and co-transplantation group as well as control group.The recovery of posterior limb motor function in rats subjected to spinal cord transected injurywas evaluated by the BBB locomotion score respectively in each weekend among eight weeks.This was followed by a histochemiscal observation to know the engrafted cells survival and migration in the spinal cord of host under the fluorescence microscope.Results The NSCs and OECs cells survived and migrated in the spinal cord of transplantation groups:the motor functional improvement of hindlimb in rats was seen in all cell transplanted groups;NSCs group,OECs group and co-transplantation group demonstrated a significant increase than that of control group at 3 and 4 weeks post operation(P 0.05).Conclusions The engrafted NSCs,OECs and co-transplantation can improve partly the motor function recovery of hindlimb in rats at 3 to 4 weeks following cord transection;The OECs group and co-transplantation have better effects than that of NSCs group,but there is no obvious difference between the OECs group and co-transplantation group at observed time point in this investigation.
6.Changes of sTim-3/HMGB1 and Spot forming cells of TGF-β secreting from peripheral blood mononuclear cell in patients infected with brucella

Objective:To investigate the changes of sTim-3,HMGB1 and TGF-β in the brucellosis patients and to analyse the relationship between the changes of these molecules and brucella infection. Methods:28 cases of brucellosis patient untreated and 28 healthy control cases in the age and gender matched with brucellosis cases were collected. The serum levels of sTim-3 and HMGB1 were detected by ELISA,and the levels of Spot forming cells secreting TGF-β were measured by ELISPOT in patients and healthy control group. Results: Compared with healthy controls, sTim-3/HMGB1 expression levels and Spot forming cells secreting TGF-β were significantly increased in the brucellosis patients ( P<0. 01 ) . The changes of Spot forming cells secreting TGF-β were positively correlated with the levels of HMGB1 (P<0. 05). Conclusion:The serum levels of sTim-3/HMGB1 and Spot forming cells of secreting TGF-β from peripheral blood mononuclear cell are significantly increased in the brucellosis patients. Those molecules may be involved in the process of brucella infection and may play a significant role in the immune escape of patients infected with brucella.
7.Comparison of various sedative regimens during TACE for hepatocellular carcinoma

Objective To investigate the efficacy and safety of using various sedative regimens during transcatheter arterial chemoembolization (TACE) for hepatocellular carcinoma (HCC).Methods 156 HCC patients were randomly divided into four groups.The control group (n =30):intramuscular injection of 10 ml saline;The diazepam group (n =42):intramuscular injection of 10 mg of diazepam;The promethazine group (n =42):intramuscular injection of promethazine 25 mg;and The combined group (n =42):intramuscular injection of 10 mg of diazepam and promethazine 25 mg.Results The blood pressure and heart rate of the control group was significantly higher than the other three groups,while the combined group was significantly lower than the diazepam and the promethazine groups.The sedative rating:Grade 0 of the four groups were:30,11,18,0 patients respectively;Grade 1 of the four groups were:0,21,15,24 patients respectively;Grade 2 of the four groups were:0,10,9,18 patients respectively;No patients were in grades 3 and 4.The anxiety score of the control group was significantly higher than the other three groups.There was no significant difference in complications among the four groups.Conclusion Diazepam and/or promethazine could be used effectively and safely during TACE,and they could reduce stress response of HCC patients during TACE,and improved tolerance of TACE.
8.Problems about performance of the essential medicine system based on the Smith-Model

To study performance problems found in China's essential medicine system.The SmithModel of system implementation was called into play in a systematic collation and analysis for the ideal policies,system implementation agencies,target groups and policy environment in its performance,along with relationships among the four factors.The system is found with a number of loopholes as the system itself is highly idealized,its system objectives set inappropriately,problems found with the four factors,and tension and conflicts among these factors.Given these problems found in its performance,it is inappropriate to make drastic changes to avoid instability of the system at its early stage.Government departments in question are advised to comprehensively analyze the four factors and their relationships then taking effective measures to deal with them and the tension,conflict among them.This can ensure effectively implementation of the essential medicine system.
9.The clinical application of endovascular treatment for subclavian steal syndrome

Objective To investigate the efficacy and safety of endovascular treatment for subclavian steal syndrome (SSS). Methods During the period from April 2004 to Feb. 2013 at authors’ hospital, a total of 12 patients with SSS, including subclavian artery stenosis (n = 7) and occlusion (n = 5), underwent endovascular treatment. Percutaneous transluminal angioplasty (PTA) was carried out in 4 cases and stent implantation in 8 cases. The clinical data were retrospectively analyzed. Results Technical success rate was 100%(12/12). The diseased side/healthy side blood pressure ratio increased from preoperative (0.46 ± 0.28) to postoperative (0.89 ± 0.32), and the difference was statistically significant (P < 0.05). After the treatment, the clinical symptoms were improved in different degrees in all patients. During the follow - up period lasting for (16.7 ± 7.9) months, restenosis occurred in one case with left SSS thirteen months after PTA, and the condition was improved after stenting angioplasty. Another patient with severe subclavian artery stenosis developed peripheral micro- vascular thrombosis of the diseased limb in 23 days after PTA, which was relieved after medication to improve microcirculation and vasodilation. No severe complications such as stroke occurred. Conclusion For subclavian artery steal syndrome, endovascular management is a minimally invasive, safe and effective treatment. It can significantly relieve the clinical symptoms due to vertebral- basilar artery insufficiency caused by subclavian steal syndrome, and improve limb weakness, low body temperature, etc. Therefore, this technique should be recommended in clinical practice.
10.Research progress in mechanism of pyroptosis mediated by inflammasomes and its targeted drugs after traumatic brain injury

After traumatic brain injury(TBI), as an immune signal complex in cytoplasm, the assembly and activation of inflammasomes can induce pyroptosis, trigger extensive inflammations, aggravate brain tissue damage, lead to delayed cell death and progressive neurodegeneration, and cause neurological dysfunction, which plays a key role in the development and prognosis of the disease. After TBI, the mechanism of pyroptosis mediated by inflammasomes and its targeted treatments have become the focus of recent researches. The authors review the research progress in inflammasomes-mediated pyroptosis in terms of related signal pathways, their activation and regulation and application of targeted drugs after TBI, so as to provide references for clinical treatments.
