1.EFFECT OF ELEMENTAL SELENIUM AT NANO SIZE (Nano-Se) WITH LOWER TOXICITY ON THE ANTI-TUMOR EFFECT OF CISPLATIN

Objective: To investigate the effect of Nano-Se with lower toxicity on the anti-tumor effect of cisplatin. Method: For toxicity comparison, mice were orally administered Nano-Se and sodium selenite to observe death, body weight loss and liver injury. As to the protective effect of Nano-Se on the toxicity of cisplatin, mice with and without Nano-Se supplementation were injected cisplatin to observe death and body weight loss. For evaluation of whether Nano-Se influence the anti-tumor effect of cisplatin, Lewis lung carcinoma was transplanted into mice, and the survivor and tumor volume were recorded to compare the effect of Nano-Se supplementation on cisplatin. Results: The toxicity of Nano-Se is significantly lower than sodium selenite. Nano-Se can decrease toxicity of cisplatin without compromising its anti-tumor effect. Conclusion: Nano-Se with lower toxicity can improve the therapeutic effect of cisplatin.

To evaluate the effect of delayed treatment with monoclonal antibody to tumor necrosis factor-alpha (TNF-? MAb) on systemic hemodynamics and multiple organ dysfunction following prolonged hemorrhagic shock and resuscitation. Method: Adult male Sprague-Dawley rats were subjected to prolonged hemorrhagic shock (MAP of 4.00-4.66 kPa for 180 rain)followed by resuscitation over 50 min. Background Gene encoding optineurin (OPTN) is a causative gene for glaucoma and amyotrophic lateral sclerosis,with a more expression in retina.Our previous study isolated OPTN-interacting proteins and identified that the gene encode the basic leucine zipper (bZIP) transcription factor neural retina leucine (NRL) zipper,a causative gene for retinitis pigmentosa,and further study demonstrated the interaction between OPTN and NRL proteins in nuclei of cultured HeLaS3 cells.Objective This study was to determine the protein binding site of OPTN necessary for NRL binding.Methods A deletion series of OPTN-expression plasmids were constructed and co-expressed with hemagglutinin (HA)-tagged NRL in HeLaS3 cells,respectively.The cytoplasmic and nuclear fractions were used to perform co-immunoprecipitate (CoIP) and Western blot with anti-tag antibodies.Results In the nuclear fractions of cells transfected with the del1 st,del2nd or del3rd plasmid,a band of coimmunoprecipitated HA-labelled NRL (HA-NRL) was detected.However,the del4th plasmid did not produce a band.The NRL band was not found in cytoplasmic fractions from transfected cells with any of the deletion plasmids or with the whole-length OPTN plasmid.Conclusions The protein binding site of OPTN necessary for NRL binding is determined.This result demonstrates the binding of Flag-OPTN and HA-NRL in HeLaS3 cells.A series of partial-deletion OPTN plasmids demonstrated that the tail region (423-577 amino acids) of OPTN was necessary for binding with NRL.
4.Detection of hepatitis B virus YMDD variants using a matrix-assisted laser desorption time of flight mass spectrometric analysis

Objective To investigate the value of detecting hepatitis B virus (HBV) YMDD variants by matrix-assisted laser de-sorption time of flight mass spectrometry (MALDI-TOF MS). Methods The assay is based on polymerase chain reaction (PCR) amplifica-tion and mass measurement of oligonucleotides containing sites of mutation of the YMDD motif. Result The MALDI-TOF MS-based genoty-ping assay was sufficiently sensitive to detect as few as 100 copies of HBV genome per milliliter of serum, and this method had superior spe-cificity for determining mixtures of wild-type and variant viruses. When sera of 40 patients were analyzed, the MALDI-TOF MS-based assay correctly identified known viral variants and additional viral quasi-species not detected by previous methods, as well as their'relative abun-dance. Conclusion The sensitivity, specificity and amenability to high-throughput analysis make MALDI-TOF MS-based assay suitable for mass screening of HBV infected patients who are receiving lamivudine.
5.Comparison of Fixation Effects of Three Different Complex Fixatives on Mouse Lens Tissue

Objective To compare the effects of three kinds of fixative solutions on paraffin section of mouse lens tissue and optimize the fixing?method of paraffin section in mouse lens tissue. Methods Three kinds of conventional fixatives were selected for the test ,including the conven?tional Davison’s solution,modified Davison’s solution and 10%neutral buffered formalin. Mice eyeball tissues were fixed with three different fixa?tives,embedded,sliced and then stained with HE method. The paraffin slices were observed under the light microscope. Results The structures of lens and retina fixed in conventional Davison ’s fixative solutions were clear and intact ,and the cells were arranged regularly and compactly. There was no eyeball distortion,contraction and retinal detachment in the eyeballs fixed in modified and conventional Davison’s fixative solution. However,the ones fixed in 10%neutral buffered formalin showed eyeball distortion and contraction,space and spherules. Conclusion The mice lens slides made from tissues fixed by conventional Davison ’s fixative solution are better than fixed by modified Davison ’s fixative solution and the 10%neutral buffered formalin fixed ones.
6.Function of IL-17 in viral myocarditis mice

Objective:To study the effect of IL-17 on viral myocarditis in mice. Methods:We randomly selected 20 wild type BALB/c mice as the control group,20 IL-17A-/- mice for IL-17A-/- group,20 IL-17A-/- mice for IL-17 group. Each mouse had intrap-eritoneal injection of Coxsackie virus B3 (Coxsackie virus B3,CVB3) to construct VMC model. The serum levels of IL-17 were detected by ELISA method,and Th17 cells were detected by flow cytometry. After 3 days of treatment with CVB3,100 μg IgG antibody was injected intraperitoneally in control group and L-17A-/- group,and 100 μg IL-17mAb was injected with IL-17 group. Mice myocardium was collected at 3rd,7th and 14th days,respectively,after CVB3 treatment. Pathological examination was carried out on the myocardial sections of mice and stained with H&E. Virus titer in mouse myocardium was examined. The contents of TNF-α,IL-6,IL-23 and IL-17 in myocardial tissue were detected by enzyme linked immunosorbent assay. Results:We successfully constructed the model of mice with viral myocarditis. The levels of Th17 and IL-17 in serum of IL-17 group were significantly lower than those in control group and IL-17-/-group. On the fourteenth day after CVB3 treatment,the degree of myocardial tissue damage in the control group was significantly higher than that in the IL-17A-/- group and the IL-17 group(P<0. 05),the degree of myocardial injury in IL-17 mice was higher than that in the IL-17A-/- group. The virus titer of the control group was higher than that of IL-17A-/-group and IL-17 group,and the control group was increased with the increase of CVB3 treatment phase (P< 0. 05). After the IL-17mAb was injected,the virus titer of IL-17 group was higher than that of IL-17A-/- group. The levels of IL-17, IL-23, IL-6 and TNF-α in IL-17-/- group and IL-17 group were significantly lower than those in control group. The levels of IL-17,IL-23,IL-6 and TNF-αin IL-17 group were significantly higher than those in IL-17-/-group (P<0. 05). Conclusion:IL-17 is an important inflammatory factor in viral myocarditis,and IL-17 deletion can protect myocardium from viral myocarditis in mice.
7.Clinical Study on Treatment of Calcanodynia with a Needle-Knife

Purpose: To investigate the clinical effect of miniature needle-knives in treatment of calcanodynia.Methods: Of 96 calcanodynia patients, 64 were treated with a needle-knife and 32, by local block therapy. The curative effects were evaluated. Results: The effective rate was 96.8% in the needle-knife group and the 78.1%in the local blockage group. There was a significant difference in curative effect between the two groups (P<0.01). Conclusion: Needle-knife lysis has a marked effect on calcanodynia.
8.Application of three-dimensional computed tomography reconstruction in costal cartilage fracture

To explore the clinical application value of 3-dimensional and multiplane reconstruction with MSCT in diagnosing costal cartilage trauma. Totally 19 cases with costal cartilage trauma underwent MSCT 5 mm scanning and 0.625 mm reconstruction, and then went through three-dimensional and multiplane reconstruction. The ac-quired data were transmitted to the computer workstation through the network, and then three-reconstruction was per-formed with the software on AW4.3 platform. There were all 32 costal cartilage fractures in the 19 patients in-volving 15 cases with rib fracture, which included 2 cases and 3 fractures at the chondrosternal junction, 13 cases and 25 fractures in the middle of the costal cartilage, 4 cases and 4 fractures at the junction between costal cartilage and rib. Three-dimensional reconstruction with spiral CT could display clearly the location and number of costal cartilage frac-tures. Three-dimensional and multiplane reconstruction shows clearly the fracture and displacement of the costal cartilage, and the combination of MRP, MIR and VR may contribute to the diagnosis and clinical planning of the costal cartilage fracture.
9.STUDY ON BIOAVAILABILITY AND TOXICITY OF SELENIUM FROM METHYLSELENOCYSTEINE

Objective:With selenomethionine as reference,we investigated the bioavailability and toxicity of methylselenocysteine. Method:Selenium retention,glutathione peroxidase and thioredoxin reductase activities were tested to compare bioavailability. Acute toxicity and short-term selenium-tolerance were studied to compare toxicity. Results:Selenomethionine and methylselenocysteine were comparable in ability of increasing selenoenzymes. Selenomethionine caused much higher selenium retention,especially at higher levels and lower toxicity indicated by acute toxicity and short-term selenium-tolerance. Conclusion:Compared with selenomethionine,methylselenocysteine has equal efficacy on selenoenzymes with the characterization of less retention,but higher toxicity.
10.Indwelling Venous Catheter-related Infection in Artificial Liver Support System:Clinical Analysis of 61 Patients

OBJECTIVE To analyze clinical and etiologic characteristics of indwelling venous catheter-related infection(CRI) in artificial liver support system(ALSS)-treated patients and explore the measures of prevention and treatment.METHODS Bacterial culture and drug sensitivity test were performed in cusp of catheters after being pulled out and the peripheral blood in liver failure patients.RESULTS Sixty four strains were isolated including 56 Gram-positive strains,2 Gram-negative strains and 6 fungi ones,respectively.The most frequent organism was Staphylococcus epidermidis which had high sensitivity to vancomycin.CONCLUSIONS The most effective measure is removing catheters in time,and vancomycin is the most optimal agent for CRI.
