Patients with ataxia-telangiectasia ( A-T) syndrome were charachaterized by profound hypersensitivity to ionizing radiation in clinic. Many studies have shown that this hypersensitivity possibly attributed to ATM gene whose critical compartment was ATM kinase. So inhibitors of the ATM kinase such as caffeine, pentoxifylline, methyl xanthines and 7-hydroxystaurosporine (UCN-01) were developed and have achieved a few encouraging results in basic and clinical stuides.
3.Ultrasound guided foam sclerosis for the treatment of lower extremity incompetent perforating veins

Objective To discuss the curative effect and clinical significance of sclerosing foam agent in the treatment of chronic venous diseases (CVD) with incompetent perforating veins.Methods 65 cases of C4,C5,C6 CVD (a total of 84 limbs) underwent foam sclerotherapy closure for incompetent perforating vein.Patients were followed-up for 3 months,ultrasonography was used to observe the closure of perforating veins and the curative effect.Results 7 patients (9 limbs) were lost to follow-up,all with grade C4.In the 66 (C4,C5) limbs,pigmentation significantly subsided in 45,partial regression in 9,no change in 2.In the 9 limbs with grade C6,complete healing of ulcer in 5,partially healed in 3.Ultrasound observed satisfactory perforating vein closure.Conclusion Foam sclerotherapy closure for incompetent perforating vein has satisfactory effect,with good short-term curative result.
4.Human umbilical cord blood plasma can replace fetal bovine serum for primary culture, proliferation and cryopreservation of umbilical cord mesenchymal stem cells

BACKGROUND:Fetal bovine serum based media used for expanding and cryopreserving human mesenchymal stem cells raise safety concerns in the clinical setting. OBJECTIVE:To investigate the feasibility of human umbilical cord blood plasma as a replacement for fetal bovine serum in culture and cryopreservation of human mesenchymal stem cells derived from umbilical cord. METHODS:Umbilical cord blood units were suitable for this research if they fulfil ed the donor selection criteria of the Guangzhou Cord Blood Bank strictly. Cord blood plasma was ready to use after col ected from the plasma reduction during the suitable cord blood units processing and pooling. Umbilical cord mesenchymal stem cells were harvested from the umbilical cord tissue of health ful-term newborns after delivery by enzyme digestion and were cultured in the presence of Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing either fetal bovine serum or pooled cord blood plasma. Morphology, proliferation, immunophenotype detected by flow cytometry and differentiation toward adipogenic and osteogenic lineages were utilized for investigating the effect of media on umbilical cord mesenchymal stem cells after 3-5 passages. Then cells were cryopreserved in media containing 10%dimethyl sulfoxide, 20%fetal bovine serum or 20%pooled cord blood plasma for at least 6 months. Viability, adhesion, proliferation, immunophenotype and osteogenic differentiation of the cells were assessed after thawing. RESULTS AND CONCLUSION:The morphology (spindle-shaped and plastic-adherent), phenotype and differentiation potential (osteogenic and adipogenic) were almost indistinguishable between cells cultured in fetal bovine serum or cord blood plasma medium, while cells grown in cord blood plasma medium demonstrated significantly higher proliferation rates than those in medium containing fetal bovine serum. After thawing, the cells maintained their adherence to the culture surface and differentiation potential to osteoblasts, but cells from cord blood plasma cryopreservation medium showed significantly better plastic attachment and produced greater cellnumbers than fetal bovine serum for the first three post-thaw passages. The results demonstrate that cord blood plasma can sever as an effective substitute to fetal bovine serum for growth, maintenance and differentiation of umbilical cord mesenchymal stem cells, and thus it wil be a safe choice for clinical-scale production of human mesenchymal stem cells.
5.Screening, identification and culture optimization of a newly isolated aromatic nitrilase-producing bacterium--Pseudomonas putida CGMCC3830.

Microbial nitrilases have attracted increasing attention in nitrile hydrolysis for carboxylic acid production in recent years. A bacterium with nitrilase activity was isolated and identified as Pseudomonas putida CGMCC3830 based on its morphology, physiological and biochemical characteristics, as well as 16S rRNA gene sequence. The nitrilase production was optimized by varying culture conditions using the one-factor-at-a-time method and response surface methodology. Glycerol 13.54 g/L, tryptone 11.59 g/L, yeast extract 5.21 g/L, KH2PO4 1 g/L, NaCl 1 g/L, urea 1 g/L, initial pH 6.0 and culture temperature 30 degrees C were proved to be the optimal culture conditions. It resulted in the maximal nitrilase production of 36.12 U/mL from 2.02 U/mL. Investigations on substrate specificity demonstrate P. putida nitrilase preferentially hydrolyze aromatic nitriles. When applied in nicotinic acid synthesis, 2 mg/mL P. putida cells completely hydrolyzed 20.8 g/L 3-cyanopyridine into nicotinic acid in 90 min. The results indicated P. putida CGMCC3830 displayed potential for industrial production of nicotinic acid.

6.Therapeutic effects and quality of life in 37 glioma patients with postoperative intensity-modulated radiotherapy

Objective To evaluate treatment outcomes and quality of life (QOL) in glioma patients treated with postoperative intensity-modulated radiotherapy (IMRT), and to explore the possible clinical factors of affecting QOL. Methods From 2007 to 2009, 37 patients with low or high grade glioma were analyzed retrospectively. All patients were operated by tumor resection below microscopy. IMRT began at 2-4 week postoperstion with 2.0 Gy/fractior, 5 fractions/week and to shrink portal and to add dose to 50-60 Gy/25-30 fractions after 40-50 Gy. The gross tumor volume (GTV) was defined as preoperation T2WI MRI high sign area and postoperation tumor cavity for low grade glioma, and with preoperation T1WI MRI enhanced abnormity area and postoperation tumor cavity for high grade glioma. The clinical target volume ( CTV ) was defined as GTV with a margin of 1.5 cm for low grade glioma and a margin of 2.5 cm for high grade ghoma, the planning target volume (PTV) with CTV plus 0.4 cm margin for setup errors according to the European Organization for Research and Treatment of Cancer ( EORTC ).The treatment outcomes and QOL were assessed. Results The half-year and one-year survival rates for all the patients were 100% and 79.2%, respectively. The median progression-free survival time was 10 months. The main side-responses after postoperative IMRT were fatigue and mild memory decline or cognitive disabilities, which were radiation dose-dependent. Conclusions Postoperative IMRT is an effective and safe modality of therapy for glioma patients.
7.Application of duct endoscopy in the early diagnosis of breast cancer with nipple discharge

Purpose:To evaluate the significance of duct endoscopy on the early diagnosis of breast cancer with nipple discharge. Methods:From Oct. 1997 to Aug.1999,11 cases of breast cancer were found in 393 patients who underwent duct endocopy for nipple discharge. These 11 cases were followed up, and their endoscopic presentation were reported. Results:The presentation of ductal cancer under duct endoscopy was characterized by irregularity of the duct wall and the lumen, such as patch like elevation and aberrant bleeding. However,the presentation of a ductal neoplasm had no particularity. The diagnostic sensibility and specificity of duct endoscopy on breast cancer was 82%(9/11) and 98.7%, respectively. All patients received sugical biopsy. 5 non palpable breast cancers, who received surgical biopsy were located by ductoscopy and diagnosed as DCIS with/without micro invasion. Conclusions:Through duct endoscopy, breast cancer can be diagnosed early due to the finding of intraductal change on duct wall and lumin. We can also succeed in locating the malignant lesion and carrying out biopsy directly by using duct endoscopy. The highly suspecious lesion found by duct endoscopy should be further confirmed by histological diagnosis.
8.Analysis of the relationship between the cell cycle related factors and carcinogenesis of breast cells and biological characteristics of breast cancer cells

Purpose:To clarify the different expression patterns of the cyclins and P21 between the normal breast epithelium cells and breast cancer cells of different degrees of malignancy.Methods:Using Western Blot, we detected the protein expression of cyclin D1, E, and p21 in the normal breast epithelium cells AG11132A, ER positive MCF 7 breast cancer cells and ER negative MDA MB 231 breast cancer cells.Results:(1) P21 protein was highly expressed and cyclin E lowly expressed in the normal breast epithelium cells. Compared with the normal breast epithelium cells, Cyclin E were highly expressed in breast cancer cells of MCF 7 and MDA MB 231 cells. Abnormal low molecular weight cyclin E was co expressed with normal cyclin E in both MCF 7 and MDA MB 231 breast cancer cells, but not in normal breast epithelium cells AG11132A. (2) In breast cancer cells, compared with the ER positive MCF 7 cells the ER negative MDA MB 231 cells expressed a high level of cyclin E protein but low p21.Conclusions:(1) There are many forms of differences both quantitive and qualitative between normal cells and cancer cells, as well as between relatively low degree and high degree invasive malignant cancer cells. (2) Cyclin E and P21 may be potential indicators that reflect the malignancy of breast cancer cells, but they should be analyzed as a whole in order to get more accurate prognostic information. [
9.Advances in the research of the effects of tea polyphenols in cancer prevention and treatment

The main component of the tea is tea polyphenols, which can inhibit the processes of carcinogenesis and development of a variety of human cancers by diverse mechanisms. Further research may provide us with more information about the anticancer activities of these promising compounds and facilitate the usage in cancer prevention and treatment.
10.Clinical Study on Treatment of Calcanodynia with a Needle-Knife

Purpose: To investigate the clinical effect of miniature needle-knives in treatment of calcanodynia.Methods: Of 96 calcanodynia patients, 64 were treated with a needle-knife and 32, by local block therapy. The curative effects were evaluated. Results: The effective rate was 96.8% in the needle-knife group and the 78.1%in the local blockage group. There was a significant difference in curative effect between the two groups (P<0.01). Conclusion: Needle-knife lysis has a marked effect on calcanodynia.
