Acta Universitatis Medicinalis Anhui 2015;(5):612-615
Objective To study the correlation between alopecia areata and the susceptibility genes identified in our previous study with Han Chinese population. Methods The study performed an independent replication study using 736 cases and 1 840 controls. DNA was extracted from peripheral blood leukocytes by salting out with saturat-ed NaCl solution according to standard methods. The evidence for association had been obtained from former gene study. Then a fine-mapping study and genotyped the locus with an additional 17 SNPs were performed. Data were analyzed with the use of Plink 1. 07 software. Results Only one SNP achieved nominal significance, rs3087243 (P = 0. 041, OR = 1. 18, 95% CI = 1. 01 ~ 1. 38). The other 16 genes (TLR1, DMBT1, CHIT1, GBP4, CIITA, IL31RA, CD96, INPPL1, MASP2, IL-13, KIAA0350, PTPN22, SPATA5, TRAF1 / C5, IL1A, IL2R) failed. A further stratification analysis of alopecia areata was adopted, including the analysis of family history, the age of on-set and the severity of alopecia areata. The stratification analysis revealed that the age of onset > 20 years achieved nominal significance P < 0. 05 for one SNP, rs2416808 (P = 0. 018, OR = 1. 35, 95% CI = 1. 05 ~ 1. 74), where-as, the other results were of no statistical significance. Conclusion The results indicate that 17 SNPs may not be associated with AA in Han Chinese population. Further study should be performed in a larger Han Chinese sam-ples.

This study is to investigate the effects of sophoridine on NF-kappaB signaling pathway in kidney tissue of endotoxemia mice and the mechanism involved. BALB/c mice were challenged with lipopolysaccharide (LPS) caudal vein injection, then sophoridine was administered by intraperitoneal injection. Totally 50 mice were divided into 5 groups: control group, LPS model group, sophoridine treatment 12 mg x kg(-1) group, 6 mg x kg(-1) group and 3 mg x kg(-1) group. All animals were sacrificed at 6 hours after treatment. Kidney and blood samples were harvested. IKKbeta mRNA and TNF-alpha mRNA expression of renal tissue was measured by the reverse transcription polymerase chain reaction (RT-PCR), and phosphorylation IKKbeta protein (pIKKbeta) was detected by immunohistochemistry. NF-kappaB P65 protein expression and distribution of renal tissue were observed by Western blotting and immunofluorescence laser confocal microscopy. Serum TNF-alpha level was detected by radioimmunoassay. The results showed that the sophoridine significantly reduced the expression of IKKbeta mRNA and pIKKbeta protein, and inhibited the expression of NF-kappaB P65 protein and decreased the entry nuclear rate of NF-kappaB P65 in the renal tissue of endotoxemia mice. Thereby the renal TNF-alpha mRNA expression and serum TNF-alpha level were significantly reduced. These results suggest that sophoridine could inhibit inflammatory reaction induced by LPS through inhibiting activation of NF-kappaB signaling pathway.

Objective To investigate the expression of substance P ( SP) and neurokinin-1 recep-tor (NK1R) in patients with breast cancer and to further understand the correlations of them with the clinico-pathological features and the prognosis of breast cancer. Methods SP levels in serum samples and superna-tants of breast cell culture were measured by ELISA.The expression of total NK1R, full-length NK1R (NK1R-FL) and truncted NK1R (NK1R-Tr) in 82 patients with breast cancer and 30 patients with breast hyperplasia were detected by using immunohistochemistry and Western blot. Results The levels of SP in patients with breast cancer were higher than those in patients with breast hyperplasia and healthy subjects ( P<0.05) .The expression of NK1R-FL was inhibited, while the expression of NK1R-Tr was significantly en-hanced in breast cancer tissues as compared with those in normal and hyperplastic breast tissues (P<0.01). Compared with patients with ductal carcinoma in situ ( DCIS ) , patients with invasive ductal carcinoma (IDC) showed a decreased expression of NK1R-FL, but an increased expression of NK1R-Tr (P<0.05). The stages of breast cancer classified by using the TNM system showed a negative correlation with NK1R-FL expression, but positive correlations with the expression of NK1R-Tr and SP (P<0.01).Moreover, the en-hanced expression of NK1R-Tr and SP and suppressed expression of NK1R-FL were positively related to lymph node metastasis and high expression of vascular endothelial growth factor ( VEGF) , human epidermal growth factor receptor 2 ( HER2) and Ki-67, but negatively related to the expression of estrogen receptor (ER) and progesterone receptor (PR) in breast cancer tissues (P<0.05).The expression of NK1R-FL and NK1R-Tr showed no significant correlation with age, histological grades and the expression of P53 in breast cancer tissues (P>0.05).Conclusion The invasion and metastasis of breast cancer showed a negative cor-relation with the expression of NK1R-FL, but a positive correlation with the expression of NK1R-Tr and SP.

Objective To explore the effect of ultra-early hyperbaric oxygenation (HBO) on bone calcium, biomechanical properties and bone collagen of femur in rats with complete spinal cord transaction. Methods A total of 75 Sprague-Dawley rats were randomly divided in-to sham group (n=15), model group (n=20) and HBO group (n=40). HBO group was divided into three hours group (HBO1 group, n=20) and twelve hours group (HBO2 group, n=20). All groups underwent laminectomy at T10, while the model group, HBO1 group and HBO2 group underwent complete spinal cord transection at the same level. Three hours and twelve hours after surgery, HBO1 group and HBO2 group received HBO, respectively, for three courses with ten days in a course. After treatment, the femoral biomechanical properties, bone calcium and hydroxyproline (Hyp) were determined. The morphology of bone trabecula and the bone collagen was observed with HE stain-ing and Masson triad color staining, respectively. Results After treatment, compared with the sham group, the femoral biomechanical proper-ties, the content of bone calcium and Hyp decreased in the model group (P<0.05);compared with the model group and HBO2 group, they in-creased in HBO1 group (P<0.05). The number of bone trabecula and the bone collagen decreased, and derangement and sparseness were ob-served in the model group;however, the changes were substantially mild in HBO1 group. Conclusion Ultra-early HBO could increase the content of bone calcium and Hyp of femur, improve the morphology of the femur bone collagen, and improve the femoral biomechanical properties in rats with complete spinal cord transection.
