1.Construction of shRNA targeting AGR2 gene and effect of AGR2 on bio-logical function of nasopharyngeal carcinoma cells

AIM: To construct the shRNA targeting anterior gradient protein 2 (AGR2) gene for exploring the effect of AGR2 on the biological behavior of nasopharyngeal carcinoma (NPC) cells.METHODS: The expression of AGR2 at mRNA and protein levels in NPC cell lines 6-10B and 5-8F was detected by real-time PCR and Western blot.The pSR-GFP/Neo-AGR2-shRNA expression vector targeting AGR2 was constructed.Based on the interference targeting AGR2, the cell migration and motility were determined by Transwell migration and motility assays.RESULTS: The expression of AGR2 was increased in NPC cell line 5-8F compared with NPC cell line 6-10B (P <0.05).When the AGR2 expression in 5-8F cells was interfered, the cell migration, invasion and tumorigenicity were weakened.CONCLUSION: The expres-sion of AGR2 is up-regulated in NPC cell line 5-8F.pSR-GFP/Neo-CLU-shRNA successfully inhibits the expression of AGR2 in NPC cell line 5-8F.AGR2 inhibits the migration, invasion and tumorigenicity of 5-8F cells in vivo.
2.Depression in systemic lupus erythematosus

Objective To investigate the level of depression in patients with systemic lupus erythematosus (SLE).Methods Fifty six in patients and out patients with SLE were investigated with questionnaire of self rating depressive scale (SDS).Results Of these patients 41% had clinical features of depression.Tiredness,hebetude,hyposexuality,despair,irritability,depression,thinking difficulty,insommia,uneasiness and vacancy were more common.Most of these patients are manual worker,poorly educated ( P 0 05).Conclusion Patients with SLE have symptoms of depression to a certain extent.Doctors should pay close attention to their mentality and emotion in diagnosis and treatment of the prtients with SLE.
3.Efficacy of pregabalin in patients with painful radiation-induced brachial plexus injury

Objective To evaluate the efficacy of pregabalin on painful radiation-induced brachial plexus injury.Methods A total of forty-five patients with painful radiation-induced brachial plexus injury were assigned to treatment group with pregabalin 150-600 mg/d combined with conventional therapy, and to control group with conventional treatment for 12 weeks.The visual analogue scale (VAS) and neuropathic pain symptom inventory (NPSI) were evaluated.The response rate (proportion of subjects with at least a 30% to 50% reduction in visual analogue scale) was evaluated as the primary endpoint.Results The response rate of the pregabalin group(36.4%) was significantly higher than that of the control group (9.1%) (x2 =4.98, P < 0.05).Although hospital anxiety and depression level, clinical global impression of change, and clinical global impression of change (CGIC) did not differ significantly between pregabalin group and control group (P > 0.05), improvements in neuropathic pain symptom inventory (NPSI), patient global impression of change (PGIC) and the sleep interference score(t =-6.62,-5.65,-6.16, P < 0.05)suggested some utility in the management of radiation-induced brachial plexus.Conclusions Pregabalin treatment combined with conventional treatment exerts efficacy on patients with painful radiation-induced brachial plexus and patients are well tolerated.
4.Efficacy of donepezil in patients with cognitive dysfunction caused by radiation-induced encephalopathy

Objective To evaluate the efficacy and safety of donepezil in the treatment of cognitive dysfunction caused by radiation-induced encephalopathy.Methods A total of fifty-five patients with radiation-induced cognitive impairment were divided into treatment group with extra donepezil 5-10 mg/d combined with conventional therapy and control group with conventional treatment for 16 weeks.The cognitive function was assessed according to Montreal cognitive assessment (MoCA) and mini-mental state examination (MMSE) before and 16 weeks after treatment.Results After 16 weeks of treatment,the patients in treatment group displayed significantly greater improvement in cognitive function.In treatment group,the scores of patients after donepezil therapy in MoCA and MMSE were obviously higher than the control group (t =5.40,3.88,P < 0.01).The scores in the visual space and executive function,naming,attention,abstract thinking,delayed memories also had improved,which suggested the statistically significant difference(t=-3.55,-3.08,-3.21,-2.58,-3.65,P<0.05).The scores of control group unchanged accordingly.Conclusions Donepezil combined with conventional treatment was signally effective in the therapy of cognitive dysfunction caused by radiation-induced encephalopathy.
5.Aberrant expression and mutation of FOXQ1 in colorectal carcinoma and adenoma

Objective To investigate the clinical pathological association of FOXQ1 with colorectal carcinoma (CRC) and colorectal adenoma (CRA). Colorectal mucosa specimens were collected between Jane 2007 and June 2009 in the First hospital of Yunnan prorince, and consisted of CRC (n=76), CRA (n=50) and normal (n=48,≥8 cm apart from cancer) tissues. Methods The expression of FOXQ1 in colorectal mucosa with was detected using immunohistochemistry (SP method). PCR amplification and direct DNA sequencing were used to identify FOXQ1 gene mutations in 23 CRC, 22 CRA and 18 normal specimens. Results There was no expression of FOXQ1 in normal specimens. Aberrant expression of FOXQ1 in either CRC or CRA specimens was significantly higher than in normal colorectal mucosa tissue (76.3% vs 0.0%, 26.0% vs 0.0% respectively, P<0.01).Aberrant expression rates of FOXQ1 was significantly higher in CRC than that in CRA (76.3% vs 26.0%, P<0.01). Expression level of FOXQ1 was increase gradually along with the pathological process of colorectal adenoma-carcinoma sequence. In CRC, the aberrant expression of FOXQ1 was not only distributed in nuclear of the tumor cells, but also found in the cytoplasm and the extracellular matrix. For CRC patients, the aberrant expression rates of FOXQ1 in extracellular matrix in Dukes C/D was significantly higher than that in Dukes A/B (61.5% vs 3.3%, 61.5% vs 5.0% respectively,P<0.01);The aberrant expression of FOXQ1 in extracellular matrix in patient with lymph node metastasis was significantly higher than those without lymph node metastasis (61.5 % vs 4.0 %, P<0.01). FOXQ1 gene mutation was only identified in one out of 23 CRC specimens, but not found in 22CRA nor 18 normal colorectal mucosa samples. Conclusions Aberrant expression of FOXQ1 may play an important role in the carcinogenesis of CRC and FOXQ1 gene may be involved in the cancer erosion and lymph node metastasis. FOXQ1 mutation is not the primary cause of aberrant FOXQ1 expression in CRC.
6.Mechanism of miR-3126-5p targeting LASP1 to inhibit the proliferation and migration of colorectal cancer cells

Objective:To explore the effect of microRNA (miRNA)-3126-5p on the proliferation and migration of colorectal cancer cells by inhibiting the expression of LIM and SH3 protein 1 ( LASP1). Methods:Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect the expression levels of miR-3126-5p in colorectal cancer cell lines (HT-29, HCT116, LoVo, SW480) and normal intestinal mucosal epithelial cells (HIEC). The cell line with the lowest expression level was selected as the experimental object. The experiment was divided into 2 groups: the negative control group (transfected with miR-NC) and the miR-3126-5p group (transfected with miR-3126-5p). Cells of each group were collected 48h after transfection. qRT-PCR method was used to detect the expression level of miR-3126-5p in each group. The MTS method and the scratch healing experiment were used to detect the proliferation level and migration ability of the cells in each group. The bioinformatics software microRNA.org and the dual-luciferase reporter gene experiment were used to predict and verify the target genes of miR-3126-5p, respectively. qRT-PCR and Western blot were used to detect the expression levels of target genes in each group of cells. Measurement data were expressed as mean±standard deviation ( Mean± SD), t test was used for comparison between two groups, and one-way analysis of variance was used for comparison between multiple groups. Results:Compared with normal intestinal mucosal epithelial cells (HIEC), the expression level of colorectal cancer cell line miR-3126-5p was significantly reduced ( P<0.05), and the cell line with the lowest expression level was HCT116 cells ( P<0.01). The expression of miR-3126-5p in HCT116 cells in the negative control group and miR-3126-5p group were (1.05±0.16) and (7.91±1.26) respectively, and the difference was statistically significant ( t=5.40, P<0.01). Compared with the negative control group, the proliferation ability of HCT116 cells in the miR-3126-5p group was significantly reduced ( t=4.52, P<0.05), and the migration ability was significantly reduced ( P<0.01). microRNA.org shows that miR-3126-5p has complementary binding sites with LIM and SH3 protein 1 ( LASP1) gene mRNA. miR-3126-5p can target LASP1 mRNA ( P<0.01). Compared with the negative control group, the expression of LASP1 gene in HCT116 cells of the miR-3126-5p group was significantly reduced ( t=4.56, P<0.01). Conclusion:The expression of miR-3126-5p in colorectal cancer cell lines is low, and miR-3126-5p can reduce the proliferation and migration ability of colorectal cancer HCT116 cells by inhibiting the expression of the target gene LASP1.
7.Molecular mechanism of lncRNA HAGLR regulating the proliferation and migration of gastric cancer cells by targeting miR-93-5p

Objective:To investigate the effect of long non-coding RNA (lncRNA) HAGLR on the proliferation and migration of gastric cancer cells by inhibiting the expression of microRNA (miRNA, miR)-93-5p.Methods:Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect the expression of HAGLR in gastric cancer cell lines (HS-746T, BGC823, SGC7901, MGC803) and normal gastric mucosal epithelial cells (GES-1). Selected the cell line with the lowest HAGLR expression and transfected with the negative control plasmid (negative control group) or HAGLR-high-expression plasmid (HAGLR group) respectively. The MTS method and the scratch healing test were used to detect the proliferation and migration ability of the cells after transfection. The bioinformatics software miRcode database was used to predict the target gene of HAGLR, and the dual luciferase reporter gene experiment was used to verify the binding of HAGLR to the target gene. qRT-PCR was used to detect the expression of the target gene. Western blot was used to detect the expression of Hippo signaling pathway. The software SPSS 21.0 was used to conduct statistical analysis. The t test was used for comparison between two groups, and the one-way analysis of variance was used for comparison between multiple groups. Results:Compared with GES-1 cells, the expression level of HAGLR in gastric cancer cell lines was lower (all P<0.05), and the cell line with the lowest HAGLR expression was SGC7901 cells ( P<0.01). The HAGLR expression in SGC7901 cells in the HAGLR group and the negative control group were 1.03±0.13 and 9.75±1.10, respectively. The expression level of HAGLR in the negative control group was significantly lower than that in the HAGLR group ( t=7.87, P<0.01). Compared with the negative control group, the absorbance of SGC7901 cells in the HAGLR group was significantly reduced ( P<0.05), and the scratch healing rate was significantly reduced ( P<0.01). The miRcode database showd that HAGLR and miR-93-5p have complementary binding sites. The dual luciferase reporter gene experiment showed that HAGLR can complement miR-93-5p ( P<0.01). Compared with the negative control group, the expression of miR-93-5p in SGC7901 cells in the HAGLR group was significantly reduced ( P<0.01), and the expression of Hippo signaling pathway protein was significantly reduced (all P<0.01). Conclusions:HAGLR is low expressed in gastric cancer cell lines. HAGLR inhibits the proliferation and migration of gastric cancer SGC7901 cells by negatively regulating miR-93-5p.
8.The cranial MRI appearance of cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy in a family

ObjectiveTo recognize the cranial MRI appearance in cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy(CADASIL). MethodsFive patients with CADASIL from two generations in a family underwent routine MRI and MRA examinations.Three patients with CADASIL were confirmed by the Notch3 genetic testing and the vascular pathological Resultsand one was diagnosed on basis of MR and clinical manifestations. The imaging data from 4 patients with CADASIL were analyzed.ResultsFour cases achieved preliminary diagnosis of CADASIL and one was excluded by MRI.In 4 patients with CADASIL,bilateral symmetrical,confluent white matter lesions in the subcortical and Deriventricular regions were seen frequently in the temporal,frontal and parietal lobes,but the occipital lobes were less involved.These lesions appeared as long T1 and long T2 signal.O'Sul]ivan sign was shown in all cases and subcortical lacunar lesions was seen in 2 cases. In the centrum semiovale,well-defined,round or oval cystic infarcts(black holes)were demonstrated in 3 cases and multiple tiny round enlarged perivascular spaces(pepperpot appearance)in all cases.The corpus callosum was involved in all cases,and it was evidently atrophic in 2 cases.The anterior part of internal capsule and external capsule were were present in the basal ganglia and brainstem. Only one case revealed a small infarct in the right cerebellum. Four Datients shared mild or moderate atrophy of brainstem, cerebellum and cerebrum. No marked abnorillalitv of large vessels was seen in all cerebral MRA. ConclusionsThe cranial MRI appearance in CADASIL shows some characteristics.MRI may give some infotznation in the preliminary diagnosis or exclusion of CADASIL.
9.Analysis of the effect that nursing students acting as social volunteers to participate in gerontological nursing practice teaching

Objective To compare the effect of nursing students acting as social volunteers to participate in gerontological nursing practice teaching with traditional practice teaching mode, and promote the reform of teaching mode in nursing practice. Methods Students of two classes from department of nursing, medical college of Wuhan University of Science and Technology were selected as research objects. In class one nursing students acted as social volunteers to participate in gerontological nursing practice teaching (the experimental group), In class two nursing students took practice in geriatric wards according to traditional practice mode(the control group). The practice effect was compared between two groups. Results The average final grade of the experimental group was 86.50, and 78.51 in the control group. Ridit analysis revealed that, setting class one as the experimental group, R=0.796 8,95% confidence interval was 0.715 1-0.878 4, in the control group, R =0.500 0. The investigation showed that the experimental group and the control group were different in moral elevation aspect, teaching level, capability improving aspect and employment promotion aspect, the difference was significant (P<0.01). Conclusions The mode of nursing students acting as social volunteers to participate in gerontological nursing practice teaching has advantage over traditional practice teaching, and it can gain satisfying teaching effect.
10.Influencing factors of survival rate in patients with myocardial infarction treated by intra-aortic balloon pump

Objective To investigate the influencing factors of survival rate in patients with myocardial infarction treated with intra-aortic balloon pump (IABP).Methods A total of 94 myocardial infarction patients with IABP in our hospital were selected,and were divided into survival group and death group according to the survival status.Clinical data,results of coronary angiography,emergency PCI results were compared,and the relevant influencing factors were analyzed by multivariate regression analysis.Results The average age,ratio of Killips Ⅲ/Ⅳ,CK-MB peak showed significant differences between two groups (P < 0.05).Significant differences were seen in the proportion of patients merging LM disease and blood flow with class TIMI3 after PCI in the two groups (P < 0.05).Multivariate regression analysis showed that the age,Killips Ⅲ/Ⅳ,CK-MB peak,blood flow with class TIMI3 after PCI,and merging LM lesions were the risk factors of survival status for patients.Conclusion The survival status of patients with IABP are mainly influenced by age,Killips Ⅲ/Ⅳ,CK-MB peak,blood flow with class TIMI3 after PCI,and merging LM lesions and other factors.
