1.THE EXPRESSION OF SH2-B? IN THE LUNG AND VISCERAL SENSORY AFFERENT SYSTEM OF THE ASTHMATIC MICE

Objective To study the expression of SH2-B? in the lung and visceral sensory afferent system(C-7-T-5 spinal ganglia and the corresponding posterior horn of the spinal cord) of the asthmatic mice,and to explore the role of SH2-B? in asthma.Methods Thirty BALB/c mice were randomly divided into normal control group and asthmatic group on average.The alterations of SH2-B? and nerve growth factor(NGF)immunoreactivity were investigated by immunohistochemical and Western blotting method and analyzed by the Metamoph analysis system.Results Immunohistochemistry demonstrated that the mean optic density(MA)of SH2-B? positive substance in the lungs(0.806?0.023),C-7-T-5 spinal ganglia(0.766?0.014)and the corresponding posterior horn of the spinal cord(0.547?0.014)of the asthmatic mice was much higher than those(0.243?0.018,0.131?0.011,0.215?0.029) in normal control group respectively.Western blotting also demonstrated that SH2-B? and NGF expressions were more increased in the lungs and C-7-T-5 spinal cord of the asthmatic mice than those of normal control group.The mean ratio of SH2-B? MA to ?-actin MA in spinal cord and lung was 0.346?0.017 and 0.512?0.018 in normal control group,0.738?0.021 and 1.526?0.022 in asthmatic group.The mean ratio of NGF MA to ?-actin MA in spinal cord and lung was 0.357?0.021 and 0.734?0.013 in normal control group,1.445?0.015 and 1.265?0.018 in asthmatic group.A positive correlation was found between the levels of SH2-B? and NGF(r=0.651,P

Objective: To study the quality standards for Shenyanghongyao Tablets. Methods: Radix Angelicae Dahuricae was identified by TLC and Ginsenoside R g1 in Radix Notoginseng was determined by TLCS. Results: The linear relationship was at the range of 0.92?g~4.60?g. The average recovery was 97.16%. Conclusion: The methods are available with a good reproducibility and can control the quality of this preparation effectively.

Objective To investigate the application of functional assessment to determine the needs of intermediate care services of elderly,and to analyze the needs of intermediate care services of elderly patients in different grades of hospital,different departments and different age.Method 2481 cases of patients aged over 60 years hospitalized due to acute diseases were collected.The ability of activities of daily living (ADL) scale (Barthel index) and limb function were determined in patients aged over 60 years within 1 week after admission as acute diseases.According to the clinical data and scoring scales,the patients were divided into three groups of self-care group without declined independent function of life with Barthel index > 95 points;intermediate care group in whom the ability of daily activities was decreased due to the acute diseases,the function of the body was decreased,and Barthel index was between 45 ~ 95 points;and long-term care group with long term bed rest,severely declined autonomic function or complete dependence and Barthel index less than 45 points,and no opportunity of further recovery.A total of 382 elderly patients (195 male) were in intermediate care group.Analyzed were the intermediate care needs of elderly patients in different levels of hospitals,different departments (including respiratory,cardiovascular,emergency department,geriatrics) and different age.Results The proportion of needs of intermediate care services was 16.1% (194/1205) in third grade hospitals,24.3% (33/136) in grade 2 hospitals,38.1 % (91/239) in grade 1 hospitals,and 7.1% (64/901) in community hospital.In general hospital the proportion of needs of intermediate care services were 24.5 % (13/53) in emergency department,12.1% (4/33) in cardiology department,14.6% (6/41) in respiratory department,and 10.4% (7/ 67) in geriatric department.The proportion of needs of intermediate care services was 19.0% (22/ 116) in age group of 60~69,42.2% (62/147) in age groupof 70~79,and 28.6% (34/119) in age group of over 80 years.Conclusion The needs of intermediate care services are different in different age groups.Patients aged 70 ~ 79 years have the highest needs of intermediate care services.The needs of intermediate care services are different in different grade levels of hospitals and in different departments.

Neuroendocrine tumors (NETs) is a rare and heterogeneous group of tumors with widely varying morphologies and behaviors. Due to their rarity and heterogeneity, progress in improving its treatment has been slow. Pancreatic neuroendocrine tumors (pNETs) is a subset of NETs, previously known as islet cell tumors, occupies 3% of the primary pancreatic tumors with the annual incidence rate of (1-2)/100 000. In recent years, it is very necessary to improve the diagnosis and treatment of pNETs.

BACKGROUND:There are reports concerning differentiation of adipose-derived mesenchymal stem cells(ADSCs)into chondrocytes using gene transfection technique.However,the transfection of adenovirus and adeno-associated virus into ADSCs is various.It is controversial whether adeno-associated virus(AAV)can transfect ADSCs.OBJECTIVE:To observe the enhanced green fluorescent protein(EGFP)expression following Ad5-EGFP and rAAV2-EGFP transfection into ADSCs,and investigate the cell proliferation ability following transfection.METHODS:ADSCs were isolated from the adipose tissue,which was from 6-month-old New Zealand albino rabbit back and neck by mechanical digestion and enzyme digestion,then ADSCs were cultured and amplified in vitro.ADSCs were infected with Ad5-EGFP and rAAV2-EGFP,and the EGFP expression was observed.A total of 2 μL sodium butyrate(1 mol/L)was added into the medium after rAAV2-EGFP transfection.MTT assay was used to detect the gene transfection effects on reproductive activity of ADSCs.RESULTS AND CONCLUSION:ADSCs isolated and cultured in vitro were flat,long-spindle and amplified stabry.The cell morphology was uniform.Many green fluorescent cells were observed in Ad5-EGFP and rAAV2-EGFP groups.Transfection efficiencies were about 88% and 83%.Adenovirus and adeno-associated virus vector can be transfected with ADSCs,and transfection efficiency is high.Adeno-associated virus needs sodium butyrate to increase its level of gene expression.
10.Analysis for Relevant Clinical Parameters and Biomarkers in Patients of Essential Hypertension Combining Acute Coronary Syndrome

Objective: To analyze the relationship between inflammatory factors and relevant risk factors in patients of essential hypertension (EH) combining acute coronary syndrome (ACS) with its clinical significance. Methods: Our research included 3 groups: EH group, n=79 patients with standard criteria, EH+ACS group, n=85 and Control group, n=48 normal subjects. Blood levels of lipoprotein-associated phospholipase A2 (Lp-PLA2), tryptase (TPS) and relevant clinical, biochemical parameters were measured; risk factors for cardiovascular disease were examined and the relationship between above parameters, risk factors and ACS occurrence in EH patients was studied by Logistic regression analysis. Results: The OR values were all greater than 1 in fibrinogen (Fbg), high-sensitivity C-reactive protein (hs-CRP), TPS, atherosclerotic plaque, Lp-PLA2 and EH grading. Fbg was the most significant independent risk factor (OR=22.242, 95% CI 6.458-76.609, P<0.0001), the standardized partial regression coefficient b'as absolute value (b') was 1.079 which was the highestone in above 6 variables with the strongest impact for ACS occurrence in EH patients. Conclusion: Fbg, hs-CRP, TPS, atherosclerotic plaque and EH grading were the independent risk factors for ACS occurrence in EH patients; Fbg was the highest risk factor for ACS occurrence with the strongest impact, which provided a new direction for ACS prevention and treatment.
