Objective To develop a nested PCR for the detection of early syphilis and genotyping of Treponema pallidum (TP), and to investigate the distribution of genotypes of TP in Guangzhou. Methods Specimens were consecutively collected from genital ulcers of patients with suspected chancre during 2002-2004, and were detected by dark-field microscopy and nested PCR. The acidic repeat protein (arp) gene and the T. pallidum repeat (tpr) gene family were amplified with the positive specimens above. The number of repeats presented in the arp gene and the restriction fragment length polymorphism by Mse I in the tpr gene were analyzed by electrophoresis. The strains were genotyped according to Pillay's criteria. Results Out of 62 patients with suspected chancre, 33 cases (53.2%) were positive by dark-field microscopy and 54 cases (87.1%) by nested PCR. Of 47 TP-positive specimens genotyped by arp gene, 36 (76.6%) were type 14, while of 49 cases genotyped by tpr gene 39 (79.6%) were type d. By combining genotypes of arp and tpr genes, 7 genotypes were found, including 14d (31, 66.0%), 13d (5, 10.6%), 14b (4, 8.5%), 12b (3, 6.4%), 12d (2, 4.3%), 15d(l, 2.2%) and 14i (1, 2.2%). Conclusions Nested PCR shows a high sensitivity in early detection of TP. Genotype 14d seems the predominant type of TP in Guangzhou.

Objective The aim of this study was to explore the inhibitory effect of alcohol extracts of Narcissus bulb on melanogenesis in zebrafish embryos. Methods Zebrafish embryos were exposed to different concentrations of alcohol extracts of Narcissus (0, 50, 100, 200, and 500 μg/mL), and then the formation of melanin was observed.Furthermore, we measured the enzyme activity of tyrosinase (TYR), which plays a pivotal role in melanogenesis.Meanwhile, the spatial and temporal pattern of melanin-specific marker genes were detected.Arbutin (Ar) was used as a positive control in all these experiments. Results The treatment of alcohol extracts of Narcissus bulb inhibited melanogenesis in the zebrafish embryos in a dose-dependent manner.Furthermore, the mRNA expression levels of melanin-related genes such as tyrosinase (TYR), silver (SILV) and Mitfa were significantly reduced after treatment with different concentrations of Narcissus extracts by in-situ and semi-quantitative PCR.Finally, the enzyme activity of tyrosinase was also gradually decreased with increasing concentrations of the alcohol extracts. Conclusions Narcissus alcohol extracts can effectively inhibit the production of melanin in zebrafish embryos. This study provides potential evidence and approaches for the screening of natural whitening compounds using zebrafish models.

ObjectiveTo discuss the effects of targeted nursing intervention on pregnancy outcome and neonatal weight in patients with gestational diabetes mellitus (GDM). MethodsTotally 108 GDM patients treated in our hospital from July 2017 to February 2019 were selected and divided into intervention (n=54) and control (n=54) groups according to the random number table. Patients in the control group received conventional nursing care, while patients in the intervention group received targeted care on this basis. Patient's blood glucose level, pregnancy outcome, neonatal weight and satisfaction with nursing before and after nursing care were compared between the two groups. ResultsThere was no statistically significant difference in blood glucose levels between the two groups before nursing intervention (P>0.05). Fasting blood glucose, 2 h postprandial blood glucose and glycosylated hemoglobin levels were lower after nursing intervention in both groups. The levels of the intervention group were even lower than those of the control group, and there were statistically significant differences between the two groups (P< 0.05). The incidence of malignant pregnancy outcome and neonatal complications of the intervention group were lower than those of the control group after nursing intervention, and patient's satisfaction with nursing was higher in the intervention group than in the control group, and there were statistically significant differences between the two groups (P< 0.05). ConclusionsTargeted nursing intervention can effectively reduce blood glucose levels of GDM patients and the incidence rate of adverse maternal and neonatal outcome, and improve patient's satisfaction, which is worth promoting in clinical practice.
