1.Mechanism of anticarcinogenic function of ellagic acid in gastrointestinal cancers

Recent in vitro and in vivo experiments have revealed that ellagic acid and its metabolites can inhibit the growth of digestive system malignant tumor cells by inhibiting tumor cell proliferation,inducing apoptosis,breaking DNA binding to carcinogens,blocking virus infection,disturbing inflammation,angiogenesis and drug-resistance processes required for tumor metastasis.Ellagic acid and its metabolites are potential chemoprevention and therapeutic drugs against human cancers.
2.TRANSNEURONAL TRANSPORT OF HERPES SIMPLEX VIRUS TYPE 1 IN NEURONAL CONNECTIONS OF THE CEREBELLUM IN THE RAT

he herpes simplex virus type1 (HSV 1) was used for study on tracing neuronal connections of the cerebellum in the rat. The anterior or posterior lobe of the cerebellum in total 23 rats was injected by HSV 1 (HK 2 strain, from Institute of Virology, Chinese Academy of Preventive Medicine) with the titer of 10 -7 in a volume of 10 15?l for each case. Following a postoperative survival period of 1 5 or 10 days, the animals were perfused and their brains and spinal cords were sectioned and stained immunohistochemically by polycolonal antibodies raised in rabbit against HSV 1. Under the LM, the HSV 1 labelled neurons were shown to be a Golgi like appearance, with clearly labelled cell body and dendrites. It showed that the distributions of the labelled neurons in the CNS depended on the postoperative periods and injection sites. 1. After posterior lobe injection, the labelling was limited in the cerebellum, especially in Purkinje cells and the deep nuclei in 1 day of survival time. Two days later, the labelling could be seen in the vestibular, pontive nuclei and inferior olive nucleus. Anterogradely transneuronal labelling in the ventrolateral thalamic nuclus became apparant 3 days after injetions; 2. After anterior lobe injection, the red nucleus, cuneate nucleus, cuniform nucleus and interstitial nucleus of cajal were labelled after 3 days, in addition to the labelling as shown in those cases with injections in the posterior lobe. The results proved that HSV 1 (HK 2 strain) could be transported both retrogradely and anterogradely in CNS, while the transneuronal transport would mainly occur anterogradely. The distances of HSV 1 transport in neuronal pathways would depend upon the postoperative survival times. This indicates that HSV 1 (HK 2 strain) is a powerful tool for demonstrating the chains of synaptically connected neurons in CNS.
3.Experiment study on screening efffective Mcl-1-targeted siRNA squence in gastric cancer cells

Objective To detect the expression of Mcl-1 gene in gastric cancer cell lines SGC7901 and MGC-803, and to screen the most effective Mcl-1-targeted siRNA sequence. Methods Mcl-1 expression was evaluated in human gastric cancer cell lines SGC7901 and MGC-803 by RT-PCR. Four segments of siRNAs (siRNA1, siRNA2, siRNA3 and siRNA4) targeting Mcl-1 mRNA and a no-sense control segment were designed by bioinformatic technology . Mcl-1 specific siRNAs were transfected transiently into SGC7901 and MGC-803 cells by using lipofectamine 2000 . After transfected 24 , 48 and 72 h , quantitative real-time PCR was applied to detect the mRNA expression of Mcl-1 and western-blot analysis was applied to detect the protein expression. Results Mcl-1 gene was expressed in both SGC7901 and MGC-803 cells. Overall, siRNA1 exhibited the best inhibitory effect after being transfected for 48h. The inhibition rate of mRNA level in SGC7901 group and MGC-803 group was 73.8%and 67.6%, and the inhibition rate of protein level in SGC7901 group and MGC-803 group was 79.3%and 96.1%. Conclusion Mcl-1 specific siRNA sequences were successfully designed, and siRNA1 was selected as the most effective sequence, which can simultanandeously inhibit the expression of Mcl-1 in GC7901 and MGC-803 cells.
4.Study Formation of Ammonion-Magnesium Phosphate Crystals in Urine with Bacteria Growing

Objective To study formation of ammonion-magnesium phosphate crystals in urine with bacteria growing and provide guidance for cilinical prevention of urinary calculi.Methods Bacterial culturefluid of Escherichia coli,Proteus mirabilis,Pseudomonas aeruginosa,Klevsiella pneumoniae,Enterococcus in urine was examined directly under the ultrahigh sensitive microscpcope system for ammonion-magnesium phosphate crystasl.The number of ammonion-magnesium phosphate crystasl was measured when the 24th and the 48th hour.Results Ammonion-magnesium phosphate crystasl were observed from the culture fluid without ammonion magnesium phosphate crystasl.The rate of male formation was higher than that of female.Ammonion-magnesium phosphate crystals in culture fluid of Proteus mirabilis was the highest,Pseudomonasaeruginosa was the second,the third was Klebsiella pneumoniae,and there was formed 1 case in 2 ml culturefluid of enterococcus,and 2 cases of formation in 5 ml culturefluid of Escherichia coli.The crystals formed were the most unformed feather crystals,followed by cubic and square cylinders,an d the envelope like crystals were the least.Conclusion Bacteria with urease play a significant role in ammonion-magnesium phosphate crystasl formed,Proteus mirabilis is the main pathlogen.

Objective To analyse the distribution of pathogenic isolates and their durg resistance from inpatient in department of orthopedics from Jan.2008 to Dec.2012 and promote rational use of antibiotics.Methods All the clinical isolates were analyzed ret-rospectivily.Results of 1044 isolates,Gram-negative bacteria accounted for 54.12% (151/279 ),the most common pathogens of which were Escherichia coli,Enterobacter cloacae,and Pseudomonas aeruginosa.Gram-postive bacteria accounted for 41.58%(116/279),Staphylococcus aureus were the most common type.Fungi isolates accounted for 3.94%.The drug sensitive test showed that the resistance of different bacteria to the same antibiotic was different.The same kind of bacteria showed different drug resistance to different antibiotics.The most effective drugs for Enterobactericaeae infection treatment was Imipenem.Pseudomonas aeruginosa was sensitive to all the antibiotics exept for ciprofloxacin,to which the drug resistance was was 80%.The drug-resistance rate of Staphylococcus aureus to erythromycin,SMZ-TMP and clindanmycin were all higher than 50%,while to the rest antibiotic was low.Conclusion The bacteria that caused the infection in patients were widely distributed in the department of orthopedics,antibi-otics should be properly chosen according to the results of microbial culture and antimicrobial susceptibility test.
7.The Effect of Silencing Mcl-1 by siRNA on Biological Behavior of Gastric Cancer MGC-803 Cell

Objective To investigate the effect of inhibiting Mcl-1 gene expression on the biological behavior of gas-tric cancer cell MGC-803 by using a small interference RNA (siRNA). Methods Synthesized siRNA targeting Mcl-1(Mcl-1 siRNA group) was transfected into MGC-803 cells. On the other hand, MGC-803 cells transfected with negative siRNA (Mcl-1siRNA-NC group), MGC-803 cells transfected with Lipofectamine 2000(liposomes control group)and vacant MGC-803 cells(blank control group)were used as controls. Proliferation of MGC-803 cells after transfection of Mcl-1 siRNA was investigated by MTT assay. After 48 h transfection of Mcl-1 siRNA, flow cytometry (FCM) was used to examine the apoptosis cells and cell cycle in all four groups. Polycarbonate membrane transwell chamber was used for evaluating the invasion and migration of the cell line. Results The absorbance of MGC-803 cells decreased greatly after transfected with Mcl-1siRNA for 24、48 and 72 h compared to those in control groups (P<0.05);After transfected 48 h, apoptosis rate in Mcl-1siRNA group was higher than in the blank control group, liposomes control group and Mcl-1siRNA-NC group (%:19.61 ± 1.66 vs 3.69±0.37 vs 3.54±0.47 vs 3.68±0.55,F=12.230, P<0.05),G0/G1 [(41.03±1.86)%] and G2/M phase ratio [(1.80± 0.46)%] in Mcl-1 siRNA group were lower than in the three control groups, S phase ratio [(57.17±1.72)%] in siRNA group was higher than in three control groups (P<0.05). The number of transmembrane cells in Mcl-1 siRNA group in polycarbonate mem-brane transwell chamber experiment (42.00 ± 4.00,76.33 ± 3.51 respectively) were less than in blank control group (79.33 ± 3.51,108.00 ± 3.61 respectively), liposome control group (74.67 ± 2.52,110.67 ± 4.04 respectively) and negative control group (77.33 ± 3.06,109.33 ± 4.51 respectively). However, there was no significant difference in above index among the control groups. Conclusion Inhibitiing Mcl-1 expression can effectively suppress growth, invasion and migration, but promote apoptosis in gastric cancer cells.
8.Determination of individualized PEEP during lung-protective ventilation in patients undergoing general anesthesia: comparison of pulmonary electrical impedance tomography and dynamic lung compliance

Objective:To compare the efficacy of individualized PEEP determined by lung electrical impedance tomography (EIT) and dynamic lung compliance (Cdyn) during lung-protective ventilation strategies in the patients undergoing general anesthesia.Methods:Sixty patients of both sexes, aged 18-64 yr, of American Society of Anesthesiologists physical status Ⅰor Ⅱ, with body mass index of 18.5-28.0 kg/m 2, undergoing elective surgery with general anesthesia and endotracheal intubation in the Second Affiliated Hospital of Soochow University, were selected.Lung-protective ventilation strategy was applied in supine position after general anesthesia.The peak value of PEEP did not exceed 10 cmH 2O, with an increment/decrement of 2 cmH 2O for titration.The corresponding Cdyn value and lung EIT data were collected during titration.The patients were divided into 2 groups ( n=30 each) using a random number table method: titration first increased and then decreased group (group A) and titration first decreased and then increased group (group B). The determination method of individualized PEEP: Cdyn method was the PEEP corresponding to the maximum Cdyn value; EIT method was obtained through PV500 PC software analysis.The level and success rate of individualized PEEP determined by the Cdyn and EIT methods were compared, and the ICC consistency analysis of the determined individualized PEEP was performed. Results:Compared with the Cdyn method, the success rate of individualized PEEP determined by EIT method was significantly increased, and the level of individualized PEEP was decreased in the two group ( P<0.05). In group A, the individualized PEEP titrated by the EIT and Cdyn methods showed good agreement (the ICC value of the increment-Cdyn and increment-EIT methods was 0.761, P<0.05; the ICC value of the decrement-Cdyn and decrement-EIT methods was 0.763, P<0.05). In group B, the individualized PEEP titrated by the EIT and Cdyn methods showed good agreement (the ICC value of the increment-Cdyn and increment-EIT methods was 0.809, P<0.05; the ICC value of the decrement-Cdyn and decrement-EIT methods was 0.797, P<0.05). Conclusion:The agreement between the individualized PEEP determined by lung EIT method and Cdyn method during lung-protective ventilation is good in the patients undergoing general anesthesia, and the success rate of EIT method is higher, and the level of individualized PEEP is lower.

Objective:To summarize the clinical features and experience in diagnosis and treatment of subglottic cysts in children.Methods:The clinical data of 5 children with subglottic cysts admitted in Qilu Children′s Hospital of Shandong University from April 2018 to June 2019 were retrospectively analyzed.Results:Among the 5 patients, 2 cases were premature infants and 3 cases were full term infants.All patients had an endotracheal intubation history.One patient was asymptomatic.Four patients had laryngeal stridor, 2 cases of whom were accompanied by hoarseness and dyspnea.These 2 patients were used to be misdiagnosed with laryngomalacia, laryngitis, etc.Bronchoscopy revealed subglottic gray-white cyst-like lesions, and enhanced CT showed low density without enhancement.All patients were treated with laser and forceps, and bronchoscope was applied in the follow up period for 5-12 weeks.There was no recurrence or subglottic stenosis.Conclusions:Laryngeal stridor in children with a history of endotracheal intubation must be examined by bronchoscopy in time to find out whether there is subglottic cysts.Laser therapy is an effective treatment for subglottic cysts.
10.Four cases of congenital absence of tracheal cartilage ring

The clinical data of 4 children with congenital absence of tracheal cartilage ring in Qilu Children′s Hospital of Shandong University from November 2017 to August 2019 were retrospectively analyzed.The age of the 4 cases was from 2 months to 1 year.One case had no obvious symptoms after birth, and then had repeated wheezing attacks for 3 times.Another 3 cases had symptoms after birth, such as throat ringing, repeated cough and asthma.Bronchoscopy in 4 cases indicated that the local lumen of trachea was round and centripetal stenosis, but the body of bronchoscope could pass smoothly, and no cartilage structure was found in the wall of trachea.Among them, 3 cases of chest enhanced CT examination displayed local and hourglass-like tracheal stenosis, without vascular ring malformation.All the 4 cases were treated by operation, of which 3 cases recovered well and 1 case died of severe infection after operation.
