1.The Clinical value of detecting CD44v6 in plasma for colon and rectum cancer

Objective To explore the clinical value of measuring solubie serum CD44 variant 6sCD44v6 in patients with colorectal cancer. Methods Serum leves of sCD44v6 and CEA in96 patients with colorectal and in 24 healthy indivduals were detected by ELISA andchem-iluninesenle immunoassoly,respectively.Some ehnieal pathological indexes were analyzed.Results Mean sCD44v6 level in patients with colorectal cancer was 275.3+-115.8 ng/ml,which was significantly hirher than that in healthy controls(61.3+-7.8 ng/ml,P<0.05).When using the mean+2s sCD44v6 level of healthy controls as the cut-off value,sCD44v6 level were abnormally elevated in 71 of 96(73.96%)patients with coloreeatal cancer.Conclusion The sCD44v6 leveland positive rate also increased with Dukesstage and tumor metastasis Compared with serum CEA level,the sCD44v6 level might reflect the tumor invasion and metastasis more directly and sensitively.
2.In vitro load test of biological prosthesis replacement after partial resection of femoral head

Objective To investigate effect of the biological prosthesis on load properties of the proximal femur after hemiarthroplasty with partial resection of the femoral head in order to assess stability of the prosthetic implant.Methods Twenty-four young fresh normal femur (with preservation of femoral head) specimens were assigned to prosthesis group (n =8),control group (n =8) and fatigue group (n =8).Specimens in prosthesis group underwent artificial femoral head replacement using self-designed prostheses.Stress at the femoral head with the model standing on one leg was performed.Biomechanical machine was used to detect the difference of load-stress and load-displacement before and after femoral head replacement under the load of 2000 N.Detection points included lateral superior base of femoral neck (A),medial side of lesser trochanter of femur (B),and inferior side of greater trochanter of femur (C).Results At the load of 2 000 N,the load-stress at inferior side of greater trochanter of femur and medial side of lesser trochanter of femur revealed no significance differences between the normal femur and femur with artificial femoral head replacement (P > 0.05),while the difference was significant at lateral superior base of femoral neck (P < 0.05).There were no significant differences of load-stress at A,B,and C points among fatigue group,prosthesis group and control group (P > 0.05).Conclusion After partial femoral head replacement,the biological prosthesis is effective in maintenance of the normal stress transfer in the proximal femur.
3.Evaluation of IVF-ET outcome by measuring the level of NO, VEGF and ET-1 in follicular fluid

Objective To investigate the level of follicular fluid NO, VEGF and ET-1 in assessing IVF-ET outcome. Methods Totally 131 patients undergoing IVF-ET cycles were recruited. The level of follicular fluid NO was measured by chromatometry. The follicular fluid VEGF and ET-1 were measured by ELISA. Transvaginal ultrasound was performed on human chorionic gonadotropin (hCG) injection day to determine ovarian volume and antral follicle count. Results The pregnancy rate was 37.40% (49/131). There were significantly increased level of follicular fluid NO, VEGF and decreased level of follicular fluid ET-1 in the pregnant group than those in the non-pregnant group (P<0.05). Total ovarian volume and antral follicle count on HCG injection day were significantly higher in the pregnant group than those in the non-pregnant group (P<0.05). The levels of follicular fluid NO and VEGF had positive correlations with the total ovarian volume and antral follicle count. However, the level of follicular fluid ET-1 had a negative correlation with the total ovarian volume and antral follicle count. Conclusions The high level of NO, VEGF and low level of ET-1 in follicular fluid are good predictors of ovarian blood flow and ovarian response in IVF-ET.
4.The effects of a stannous-containing sodium fluoride dentifrice and crisscross bristle toothbrush in the management of gingivitis and dental plaque observed by an office-based study

Objective:To investigate the effects of a stannous-containing sodium fluoride dentifrice and crisscross bristle manual tooth-brush in the management of dental plaque and gingivitis.Methods:249 cases with gingivitis were enrolled in an office-based study. The study was unsupervised and single-centered with open-label and self-control.At baseline,gingival health and plaque coverage were assessed by dentists using categorical scales.Participants were given stannous containing sodium fluoride dentifrice and crisscross bristle design manual brush,and were instructed to use the products by manufacturer's usage instructions twice daily for 30 days.At the end of 30 days,plaque and gingivitis were reassessed using the same categorical scales.Results:232 participants(1 78 females and 54 males) completed the study.Gingivitis data of 5 cases and dental plaque data of 3 cases were not judgable.After 30 days of product use,226 cases(99%)showed noticeable improvement in their gingival health;227(96%)cases showed improvement in overnight plaque cover-age.Conclusion:Stannous-containing sodium fluoride dentifrice in combination with crisscross bristle toothbrush is effective in the management of gingivitis and dental plaque.
5.Inhibition of K562 cell growth and tumor angiogenesis in nude mice by antisense VEGF(121) cDNA transfection.

OBJECTIVETo investigate the effect of antisense vascular endothelial growth factor (VEGF)(121) cDNA transfection on the growth of K562 cells in nude mice.
METHODSK562 cells transfected with the antisense (AS) or sense (S) VEGF(121) cDNA, and the vector (V, pcDNA3) alone were transplanted subcutaneously into nude mice and the growth of the transfected cells in vivo was investigated. The effects of transfected K562 cells on human bone marrow endothelial cells (BMEC) were analyzed by MTT assay, the microvessel density (MVD) in tumor mass by vWF immunohistochemistry stain.
RESULTSK562/V tumor grew more slowly [(207.5 +/- 192.9) mm(3) vs (445.0 +/- 150.9) mm(3), P < 0.05] and K562/S tumor more rapidly than K562/V tumor did [(1 174.6 +/- 508.7)/mm(3) vs (445.0 +/- 150.9) mm(3), P < 0.01]. K562/S cell culture supernatant was more strongly in promoting the proliferation of BMEC than K562/V supernatant did, but K562/AS supernatant resulted in a marked decrease of the promoting effect as compared with K562/V's. The MVDs in K562/AS, K562/S, and K562/V tumors were [(11.0 +/- 7.6)/0.72 mm(2) vs (50.8 +/- 11.7)/0.72 mm(2) vs (18.9 +/- 7.0)/0.72 mm(2)], respectively.
CONCLUSIONSAntisense VEGF(121) cDNA transfected K562 cells show growth retardation in transplanted nude mice, decrease of tumor MVD, and decrease of promoting BMEC proliferation capacity.

6.beta1 Integrin Dysfunction in Adult Chronic Myeloid Leukemia Bone Marrow Cells

According to our previous experiments, Ph(+) chronic myeloid leukemia (CML) cell line K562 cells have defects in beta 1 integrin activation. In order to search the same regularity in Ph(+) CML bone marrow cells, bone marrow mononuclear cells (BMMNC) from 12 cases of Ph(+) CML and 10 cases of normal individuals were studied. Their expression rate of 9EG7 epitope on beta1 integrin post treatment by 8A2 or GM- or G-CSF and cell adhesion ability with soluble fibronectin (FN) were evaluated by flow cytometry; in addition, the effects of CGP57148B, a highly specific ABL tyrosine kinase inhibitor, were observed. Our results showed that 9EG7 expression rate and FN binding rate were very low in all the inactivated cells. The parameter increased markedly post 8A2 activation in both NBMMNCs and CMLBMMNCs, but the degree of increase in CMLBMMNCs was significantly lower than that in NBMMNCs; GM-CSF or G-CSF could significantly increase the parameters in NBMMNCs while had no effects on that in CMLBMMNCs. CGP57148B could increase the beta1 integrin activation potential of CMLBMMNCs but had no effects on that of NBMMNCs. The results indicate that decreased activation potential of beta1 integrin in CMLBMMNCs is the major cause of adhesion defects of Ph(+) CML cells; beta1 integrin functional insufficiency in CMLBMMNCs could not be directly reversed by ABL tyrosine kinase inhibitor CGP57148B.
7.Clinical evaluation of the anti-gingivitis efficacy of a novel stabilized stannous-containing sodium fluoride dentifrice

Objective: To evaluate the anti-gingivitis efficacy of a novel stabilized stannous-containing sodium fluoride dentifrice. Methods: A randomized, controlled and double blind clinical study was conducted. 156 adults with gingivitis were enrolled and randomly assigned to experimental group(group of novel stabilized stannous-containing sodium fluoride dentifrice, n = 51), positive control group (group of Yunnan Baiyao dentifrice, n = 54) and negative control group (group of Crest dentifrice, n = 51). Gingival health was assessed using Mazza Modification of the Papillary Bleeding Index(Mazza GI) at Baseline, day 3 and week 4 and pocket depth was evaluated at baseline and week 4, respectively. Results: At day 3 and week 4, the experimental and positive control groups exhibited lower clinical parameters than the negative control group(P< 0. 000 1). At week 4, the mean Mazza GI scores and PD of the experimental group were significantly lower than those of the positive control group(P< 0. 05). Conclusion: The novel stannous-containing sodium fluoride dentifrice has anti-gingivitis efficacy.
8.Experimental analysis of clinical applicability of individualized customized materials

Objective To study the feasibility of clinical application of an individualized customized material. Methods Five batches of individualized customized materials were randomly selected, from which 10 cm × 11 cm samples were intercepted for experimental analysis. Among them, 10 cm × 10 cm materials were selected to perform dosimetric analysis and HU change analysis before and after irradiation with a radiotherapy dose for breast cancer of 50 Gy as the irradiation basis. The center Point 1 on the lower surface of the individualized material and the center Point 2 of the solid water volume were selected for dosimetric analysis before and after the sample is irradiated. After reaching a sufficient amount of irradiation, the 1 cm × 10 cm materials intercepted in the center position and the remaining 1 cm × 10 cm materials after the first sampling were sent to the material science laboratory for analysis of physical properties of density, viscosity, hardness, and tear strength. Results In the comparative analysis of HU values before and after exposure, after receiving 50 Gy dose irradiation, the difference rate of HU value was 5.252%, which was close to the expected 5% difference rate in clinical medicine. In the dosimetric analysis of Point 1 and Point 2, the dose in the irradiated samples was significantly higher than that in the unirradiated samples; the dose in Point 1 increased by 3.742%, and the dose in Point 2 increased by 2.039%. Before and after irradiation, except for the physical density which showed a significant difference, there was no significant difference in viscosity, hardness, and tear strength. Conclusion The individualized customized material can meet the requirements of routine clinical medicine.
