1.Overexpression of FKS1 to improve yeast autolysis-stress.

With the development of high gravity brewing, yeast cells are exposed to multiple brewing-associated stresses, such as increased osmotic pressure, enhanced alcohol concentration and nutritional imbalance. These will speed up yeast autolysis, which seriously influence beer flavor and quality. To increase yeast anti-autolytic ability, FKS1 overexpression strain was constructed by 18S rDNA. The concentration of β-1,3-glucan of overexpression strain was 62% higher than that of wild type strain. Meantime, FKS1 overexpression strain increased anti-stress ability at 8% ethanol, 0.4 mol/L NaCl and starvation stress. Under simulated autolysis, FKS1 showed good anti-autolytic ability by slower autolysis. These results confirms the potential of FKS1 overexpression to tackle yeast autolysis in high-gravity brewing.

3.Association between early functional outcome and serum thyroid hormone level in elderly acute ischemic stroke patients

Objective To study the association between early functional outcome and serum thyroid hormone level in elderly acute ischemic stroke (AIS) patients.Methods Two hundred and twenty-four AIS patients admitted to our hospital from May to December 2016 were divided into good functional outcome group (n=166) and poor functional outcome group (n=58) according to their mRS score.The serum levels of FT3,FT4,TSH were measured.The patients were further divided into FT3≤4.05 pmol/L,FT3 4.06-4.46 pmol/L,and FT3>4.46 pmol/L.The association between early functional outcome and serum levels of FT3,FT4,TSH in elderly AIS patients were analyzed by univariate and multivariate logistic analysis.Results The AIS was severer,the BMI and incidence of AF were higher,the number of WBC was greater,the serum FT3 and CRP levels were higher in poor functional outcome group than in good functional outcome group (P<0.05,P<0.01).The rate of poor functional outcome was significantly higher in FT3≤4.05 pmol/L than in FT3 4.06-4.46 pmol/L and FT3 >4.46 pmol/L (39.5% vs 22.2%,39.5% vs 15.8%,P=0.003).Univariate logistic regression analysis and multivariate logistic regression analysis showed that FT3 was an independent risk factor for early poor functional outcome in AIS patients.Concision The lower the serum FT3 level on admission is,the poorer the early functional outcome is in AIS patients.

Secreted frizzled-related protein (SFRP) can inhibit the expression of Wnt signaling pathway through Frizzled protein.The silencing of SFRP gene promoter methylation is associated with the occurrence and metastasis of many cancers such as colorectal cancer,gastric cancer,liver carcinoma,lung cancer and ovarian cancer.Several studies have found that SFRP gene has latent clinical value,which is expected to become the novel target for the gene diagnosis and treatment of cancer.
5.Immunophenotyping detected by flow cytometry and its clinical significance in acute myeloid leukemia

Objective To investigate the immunophenotypic characteristics of acute myeloid leukemia (AML)and its relationship with chemotherapy response.Methods The immunophenotyping was performrd by flow cytometry in 96 newly diagnosed AML patients.Results In 96 patients with AML,the myeloid antigens were mainly expressed including CD13(94/96,97.92%),CD117(86/96,89.58%),CD33(79/96,82.29%)respectively.The positive expression of stem/progenitor cell differentiation antigens were CD38 (92/96,95.83%),HLA-DR(72/96,75.00%)and CD34(60/96,62.50%).While CD34 and HLA-DR were fewly expressed in M3.The CD14 was only expressed in M4 and M5.The lymphocytic antigens had positive expression in the patients with AML,in which CD4 was 37.5%(36/96),CD7 was 22.92%(22/96)and CD2 was 10.42 %(10/96).In 46 patients for treatment response analysis,total CR rate was 67.39%(31 /46).There was no significant difference of CR rates between the antigens positive group and negative group including CD33,CD117,CD11b,CD14,CD64,CD4,CD34 and HLA-DR.However,the CR rate was significantly lower in CD7 positive cases than CD7 negative ones,with statistically significance difference(P=0.01).Conclusions CD13,CD33and CD117 are the most common antigens in AML.The patients with the positive expression of CD7 have poor response to chemotherapy in AML.
6.Bone Marrow Mesenchymal Stem Cells Differentiated into Hepatocyte-like Cells Induced by Rebirth Liver Tissue Lixivium of Mouse in Vitro

Objective To induce bone marrow mesenchymal stem cells (BMSCs) differentiated into hepatocyte-like cells with rebirth liver tissue lixivium of mouse in vitro.Methods Mouse BMSCs were isolated and directionally induced with rebirth liver tissue lixivium of mouse 36 h after partial hepatectomy (PH). The morphology of cells was observed under an invert microscope, and the characteristics of differentiated cells was identified by immunofluorescence test.Results 7 days after induced by liver lixivium, the spindle shaped BMSCs became round and resembled hepatocyte-like cells. 1~2 weeks later, the differentiated cells expressed albumin, CK8 and CK18, which were known as characteristic markers of the hepatocyte.Conclusion BMSCs of mouse can be differentiated into hepatocyte-like cells under induction of liver tissue lixivium of mouse.
7.Effect of social support as medium in the process of attachment styles and coping styles on criminal teenagers and adolescents with mood disorders

Objective To explore the medium effect of social support in process of attachment styles and coping styles on criminal juveniles and adolescents with mood disorders.Methods The survey used in the research included Experiences of Close Relationships Inventory (ECR),Social Support Revalued Scale(SSRS) and Coping Style Questionnaire(CSQ) conducted by three groups:177 criminal juveniles,80 adolescents with mood disorders and 163 normal teenagers.Results Firstly,there were significant differences in the avoidant attachment,attachment anxiety,objective support,subjective support,utilization of support,solving problem,rationalization,self-accusation and retreat among three groups (F=7.902,8.047,59.009,19.248,5.304,7.596,10.652,6.699,11.009;P<0.05).Secondly,attachment avoidance had significant negative correlation with both utilization of support and help seeking behaviors in the group of mood disorders (r =-0.470,-0.350; P< 0.01).Similarly,avoidant attachment had significant negative correlation with both subjective support and utilization of support respectively (r=-0.232,-0.235 ; P<0.01) in the group of criminal juveniles.The subjective support had significant positive correlation with solving problem (r=0.272,P<0.01) and as well as the utilization of support with both solving problem and help seeking behaviors (r=0.258,0.424; P< 0.01).Attachment anxiety had significant negative correlation with objective support and had significant positive correlation with self-accusation respectively (r=-0.253,0.300;P<0.01).Finally,utilization of support as a medium completely effect on the process of avoidant attachment and help seeking behaviors in the group of mood disorder and criminal juveniles.Moreover,subjective support and utilization of support as medium both affected a part in the process of avoidant attachment and solving problem for the group of criminal juveniles.Conclusion First,both adolescents with mood disorders and criminal juveniles have unsafe attachment mode,which are lacked social support and easy to take negative coping styles.Yet,the level of attachment avoidance and subjective support of criminal juveniles was found different compared to teenagers with mood disorder.Second,the attachment styles for both adolescents with mood disorders and criminal juveniles have impact on coping styles by the means of social support,but there were slightly different way.
8."Outcomes in patients with ""successful"" recanalization in anterior circulation stroke treated with mechanical thrombectomy: TICI 3 versus TICI 2b"

ObjectiveTo investigate the difference in the outcomes in patients with acute anterior circulation stroke having achieved the Thrombolysis in Cerebral Infarction (TICI) 2b and TICI 3 recanalization after mechanical thrombectomy.MethodsThe patients with acute anterior circulation stroke having achieved successful recanalization after mechanical thrombectomy were enrolled retrospectively.They were divided into TICI 2b (almost complete recanalization) group and TICI 3 (complete recanalization) group.The good outcome was defined as the modified Rankin Scale score 0-2 at 3 months after onset.ResultsA total of 83 patients were enrolled in the study, including 38 patients (45.8%) with TICI 2b and 45 (54.2%) with TICI 3;49 (59.0%) had good outcome, and 34 (40.9%) had poor outcome.The good outcome rate in the TICI 3 group was significantly higher than that in the TICI 2b group (68.9% vs.47.4%;χ2=3.946, P=0.047).After adjusting for age, hypertension, diabetes, baseline systolic blood pressure, triglyceride, intravenous thrombolysis, and ASITN/SIR collateral grades, TICI 3 was an independent predictor for good outcome at 3 months after onset (odds ratio [OR] 3.759, 95% confidence interval [CI] 1.098-12.871;P=0.035), while the higher baseline National Institutes of Health Stroke Scale score (OR 0.820, 95% CI 0.715-0.941;P=0.005) and higher fasting glucose (OR 0.610, 95% CI 0.410-0.906;P=0.014) were the independent predictors for poor outcome at 3 months after onset.ConclusionsThere are difference in the outcomes in patients with successful recanalization in anterior circulation stroke treated with mechanical thrombectomy.The outcome is substantially better in TICI 3 than TICI 2b patients.
9.The contrast of epalrestat and mecobalamine in improving diabetic peripheral neuropathy

Objective To compare the curative effect of Epalrestat and mecobalamine .Methods Epalrestat to treat 48 pa-tionts in DPN and mecobalamine to treat 42,measuring blood sugar ,blood pressure, blood fat and body mass index (BMI) prior and post treatment ,and measuring the MCV and SCV of nervus medianus ,nervus peronaeus connunis and nervus tibialis with EMG .Re-sults The symptom of the two sets have all been improved after the treatment ,and the effective power of Epalrestat and mecobalamine is 92.7% and 80.5% respectively.mean while there is improvement in MCV and SCV of nervus medianus ,nervus peronaeus connunis and nervus tibialis,and is more obvious in the set of Epalrestat ( P <0.01).In the whole process of the treat of the two sets ,no one appear to have adverse reactions .Conclusions Epalrestat has significant curative effect with less adverse reactions , and deserves to be spreaded in clinic.
10.Elementary study on construction of engineered blood vessel using chitosan-fibrin gel as scaffold

ObjectiveTo investigate the feasibility of construction of engineered blood vessel using chitosan tube and fibrin gel as scaffold.MethodsVascular endothelial cells and smooth muscle cells were harvested from aortas of a rat, respectively. After expansion in vitro, vascular endothelial cells were seeded onto the inner surface of chitosan tube and smooth muscle cells mixed with fibrin gel seeded onto outer surface of the scaffold to construct engineered blood vessels. Inverted microscope, immunohistochemical staining and scanning electronic microscope were used to evaluate the construct.ResultsVascular endothelial cells formed monolayer and covered the inner surface of chitosan tube. Smooth muscle cells survived in the fibrin gel and grew in a 3-dimensional manner. ConclusionChitosan-fibrin gel may be potentially used as scaffold of engineered blood vessels.
