1.Application of CT-guided self-made localization puncture device in radiofrequency ablation of liver tumors

Objective To evaluate the value of CT-guided self-made positioning puncture device in radiofrequency ablation of liver tumors. Methods A total of 78 tumor foci from 64 patients with primary hepatocellular carcinoma treated with radiofrequency ablation from January 2010 to November 2014 were analyzed. Among them, 13 tumor diameters were 3.5-5.0 cm and 65 < 3.5 cm. The location and puncture of tumor lesion were performed by CT-guided positioning device, and the effect of puncture and lesion was evaluated by using tumor precise radiotherapy planning system. Radiofrequency ablation of tumor lesion was performed by tumor radiofrequency treatment system. Results The puncture error of all tumors in the space was 1-4 mm, the average puncture error was 2.9 mm, the complete ablation rate of tumor lesions was 93.59%(73/78), and there was no serious complication in the course of treatment. After treatment, the alpha-fetoprotein and liver function were significantly improved, and the differences were statistically significant (both P<0.05). At 1 month after surgery, CT found that no tumor and ablation was enhanced, at 3 months after surgery, CT showed that the arterial phase of all the tumor lesions did not strengthen, all of them were low-density lesions, no ablation recurred. The recurrence rates of 1, 2, 3, and 4 years after treatment were 17.9 %, 29.9 %, 45.9 % and 55.0 %. Conclusion CT-guided radiofrequency ablation with self-made puncture device is effective in the diagnosis of primary liver cancer, ablation is complete and safe.
2.Application of gene-knocking out mice in the research of atherosclerosis

These mouse model, the model generated by gene targeting, like IL-1, C-reactive protein, scavenger receptor, IgG Fc,C1q and CD44, play important role in research of inflammatory factor and immune factor on atherosclerosis.
3.New materials and new technology of plant soft capsules

With the popularity of the concept of being ?natural?and ?vegetarian?;plant soft capsules have gained widespread attention in pharmaceutical markets of many countries.Looking for new materials of plant soft capsules and developing the new technologies have become a inevitable requirement due to the traditional gelatin soft capsules could not meet the increasing daily life demand of people.The review illustrates the characteristics of the commonly used plantsoft capsule materials such as modified starch;gelatin or synthetic polymers;as well as the application of advanced technology concerning capsule shell and core material;providing a reference for the development and application of new plant soft capsules.The promising expectation of plant soft capsules are also illustrated.
4. Micro-fracture therapy combined with intra-articular injection of platelet-rich plasma for small sized osteochondral lesion of the talus

Objective: To investigate the effectiveness of micro-fracture therapy combined with intra-articular injection of platelet-rich plasma (PRP) in the treatment of small sized osteochondral lesion of the talus (OLT). Methods: Between September 2014 and October 2017, 43 patients with small sized OLT met the inclusive criteria were admitted and randomly divided into micro-fracture group (21 cases) and combined group (22 cases). Patients in the micro-fracture group were treated with micro-fracture therapy, and patients in the combined group were treated with micro-fracture therapy combined with intra-articular injection of PRP. There was no significant difference in gender, age, disease duration, side of OLT, injured position, lesion area, Mintz classification, and preoperative American Orthopaedic Foot and Ankle Society (AOFAS) ankle-hind foot score and visual analogue scale (VAS) score between the two groups ( P>0.05). After treatment, MRI, VAS score, and AOFAS ankle-hind foot score were used to evaluate the recovery of OLT and the ankle function. Results: All incisions healed by first intention, and no complications such as venous thrombosis and ankle joint infection occurred. All patients were followed up 12-18 months after operation, with an average of 15.6 months. The VAS scores and the AOFAS ankle-hind foot scores were significantly improved at 6 and 12 months after operation in the two groups ( P<0.05), and the scores at 12 months were significantly improved when compared with postoperative scores at 6 months ( P<0.05). Compared with the micro-fracture group, the VAS score and the AOFAS ankle-hind foot score were significantly improved in the combined group at 6 and 12 months after operation ( P<0.05). MRI showed that OLT was well filled in both groups at 12 months after operation. Conclusion: Compared with micro- fracture therapy, micro-fracture therapy combined with intra-articular injection of PRP can effectively reduce pain, improve ankle function, and has a good effectiveness in the treatment of small sized OLT.
5.Expression of minichromosome maintenance 3 from the peripheral blood of fluorosis patients and the liver and renal function

BACKGROUND:Previous studies have shown that minichromosome maintenance 3 is related with fluorosis, but the expression of minichromosome maintenance 3 in fluorosis patients is not clear yet.
OBJECTIVE:To analyze the mRNA expression level of minichromosome maintenance 3 in peripheral blood from patients exposed to fluoride and normal controls.
METHODS:Eleven patients with mild fluorosis by drinking water (exposure group) and 11 cases of control (non-exposure group) were selected for research. SYBRGreen1 real-time quantitative PCR was used to determine the mRNA expression of minichromosome maintenance 3 in peripheral blood mononuclear cel s, and the liver and renal function indicators were detected.
RESULTS AND CONCLUSION:mRNA expressions of minichromosome maintenance 3 in the exposure group and non-exposure group were (0.573 60±0.102 59) and (0.550 0±0.171 81), respectively, and there was no significant difference between two groups (P>0.05). There were no significant differences in the liver and renal function indicators between two groups. The results indicate that mild fluorosis has no significant effect on mRNA expression of minichromosome maintenance 3 in the peripheral blood mononuclear cel s. More indicators are needed to compressively analyze the effect of fluoride on the liver and renal functions.
6.Dynamic observation of EB virus DNA load before and after the treatment in patients with nasopharyngeal carcinoma and its clinical significance.

OBJECTIVE:
Dynamic observation of Epstein-Barr virus (EBV) DNA load before and after the treatment in patients with Nasopharyngeal carcinoma (NPC), predicting the incidence of distant metastasis and offering more personalised choice of therapies.
METHOD:
Fifty-four cases of patients with NPC were taken by fluorescence quantitative PCR assay of EBV DNA load before and after the treatment, all patients were followed up according to plan and carried out the progression-free survival (PFS) and overall survival (OS).
RESULT:
EBV DNA load in plasma of patients with NPC can partly reflect the clinical characteristics of patients; EBV DNA load in some patients with distant metastasis was higher than those patients with continuous remission when they were not started treatment (P < 0.05); For those patients whose EBV DNA copies were lower than 20,000 copies/mI before the treatment, the progression-free survival and overall survival rates were higher than those high expression patients, and the difference were statistically significant (PF < 0.01 and P < 0.05).
CONCLUSION
The EBV DNA load in the plasma of NPC patients can partly predict the occurrence of distant metastases before treatment.

7.Monitoring of blood tacrolimus concentration and lymphocyte subsets in the first uterus transplant recipient in China and its clinical significance

Objective To investigate the changes of peripheral blood tacrolimus concentration and lymphocyte subsets in the uterus transplant recipient,and provide the evidence for monitoring the immune status after uterus transplantation.Methods The peripheral blood tacrolimus concentrations of the uterus transplant recipient during 1 year after transplantation were measured with the microparticle enzyme immunoassay (MEIA).Meanwhile,the whole blood cell counts and lymphocyte subsets were determined by the blood analyzer and flow cytometer,respectively.Results The blood tacrolimus concentrations of the uterus transplant recipient in the first month and second month after transplantation were (13.51 ± 3.92) ng/mL and (15.58 ± 1.19) ng/mL,respectively.The lymphocyte absolute counts were normal before transplantation.At the fifth day after transplantation,the counts of CD3 + T lymphocytes,CD4 + T lymphocytes,CD8 + T lymphocytes and NK cells and the ratio of CD4/CD8 were significantly decreased.One week after transplantation,the counts of CD4 + T lymphocytes were recovered to the normal range and maintained,but its recovery was slower than that of CD8 + T lymphocytes.The ratio of CD4/CD8 ranged from 0.4 to 0.8 during 10 days after transplantation,and increased and maintained between 0.8 and 1.1 after that.The counts of NK cells increased gradually from the 10th day after transplantation,but still did not recover to the level before transplantation even at the 20th day after transplantation.However,the counts and percentages of B lymphocytes did not decrease but increased at the fifth day after transplantation,and recovered to normal gradually from the 10th day after transplantation.There was no significant correlation between the CD3 + T lymphocyte count and blood tacrolimus concentration.Conclusion The dynamic changes of blood lymphocyte subsets and tacrolimus concentration exist in the uterus transplant recipient,which need to be further verified by a large amount of clinical data.
8.Mice cardiovascular imaging using ultrasound biomicroscopy in comparison with magnetic resonance imaging

Objective To establish the method and cardiovascular imaging in mice using ultrasound biomicroscopy(UBM) with anatomical confirmation by magnetic resonance (MR) imaging, and to evaluate the feasibility and value of UBM in mice cardiovascular imaging.Methods The mouse thoracic anatomy was elucidated using Signo 3.0T MR imaging on 2 healthy C57BL/6 adult male mice.The structures and flow of mice heart and great vessels in 16 healthy C57BL/6 adult male mice were showed and analyzed by Visualsonics Vevo 770 UBM through several acoustic windows, including left parasternal, right parasternal, apical regions, and suprasternalis.The systolic and diastolic function of mice heart were also evaluated.ResultsAnatomic characteristic of mice could be displayed by MR imaging.The heart was mostly located in left thoracic cavity.The angle between long axis of the heart and the midline of the chest was about 45 degrees.Left atrium,left ventricle, mitral valve and right ventricle were located on the left side of the midline of the chest.Aortic valve orifice, ascending aorta,right atrium and tricuspid valve were located on the right side of the midline of the chest.Left heart long axis view,left ventricular short axis view,ascending aorta long axis view,long axis view of aortic arch, aortic short axis view, right ventricular inflow tract long axis view, pulmonary artery long axis view and so on were obtained clearly using UBM.The structures of heart and great vessels were displayed and the cardiovascular parameters could be measured accurately,including left atrium,left ventricle,aorta and aortic arch,interventricular septum,right atrium, right ventricle, pulmonary artery, innominate artery, right internal carotid artery and the velocity in each valve orifices from Doppler flow spectrum.No significant difference was found in the measurements between different age groups.Conclusions The cardiovascular structures and hemodynamics could be displayed by UBM in mice when appropriate acoustic windows and sections were selected.This novel imaging protocol provides us a non-invasive way to follow atherosclerosis in genes knockout mice.
9.Assessment of atherosclerosis in ApoE knockout mice feeding with western diet using ultrasound biomicroscopy

Objective To evaluate the value of ultrasound biomicroscopy (UBM) in assessment of atherosclerosis in apolipoprotein E (ApoE) knockout mice feeding with western diet.Methods Sixteen ApoE knockout mice in 8 weeks age were selected,then divided into two groups.One group was fed with west diet as high-fat group,and another group was fed with normal diet as control group.Intima-media thickness (IMT) and plaque area in the aortic root were assessed by UBM in two groups after 8 weeks and 16 weeks.And the measurements of UBM were compared with results of histopathology and blood-fat.ResultsThicken wall and plaque could be find in aortic root in control group and high-fat diet group byUBM.IMT and plaque area in high-fat diet group was significantly higher than those of control group ( P < 0.05).The IMT and plaque area in UBM were good correlation with histopathology ( rwas 0.81 and 0.70 respectively).The triglyceride(TC) and total cholesterol in high-fat diet group was significantly higher than those of control group ( P <0.05),and IMT in UBM were increased with the elevated level of TC,there was a positive correlation between IMT and TC( r =0.528).ConclusionsWestern diet can accelerate the process in formation of atherosclerotic plaque in ApoE knockout mice.UBM can be used to observe this prograss noninvasively in vivo mice.
10.Association between cumulative exposure to blood pressure and new-onset chronic kidney disease

Objective To investigate the association between cumulative exposure blood to pressure (cum BP) and new-onset chronic kidney disease (CKD).Methods In this prospective cohort study,101 510 employees of Kailuan Group receiving annual health examination during 2006 to 2007 were observed.The participants received the second,third,and fourth annual health examinations during 2008 to 2009,2010 to 2011,and 2012 to 2013 year respectively.Their urinary and serum creatinine were tested,and participants with incomplete SBP,DBP data and CKD were excluded.Further excluding those who somehow failed to take annual health examination,with incomplete data,or new-onset CKD 27 809 participants were selected in the analysis.According to cum BP exposure quintile grouping:Q1 < 3.70 scores;Q2:3.70-6.16 scores;Q3:6.17-8.45 scores;Q4:8.46-10.95scores;Q5 ≥ 10.96 scores.Multivariate Logistic regression was used to analyze the association between cum BP level and new-onset CKD by cum BP exposure quintile grouping.Results The rise of cum BP exposure level caused the increased incidence of CKD.The incidences of CKD in the five quintile groups were 2.59%,3.11%,4.19%,5.81%,and 7.73% respectively (P< 0.01).Compared with Q1 group,multivariate logistic regression analysis showed that after the adjustment of age,gender,education,income,smoking,drinking,BMI,FBG,TC,TG,LDL,HDL,UA and CRP,the incidences of CKD gradually increased in the Q2,Q3,Q4,and Q5 cum BP quintile groups,and OR(95%CI) values were 1.08(0.86-1.35),1.26(1.01-1.58),1.57(1.27-1.95),1.78(1.43-2.21) respectively (P for trend <0.01).Similar results were obtained in different genders.For each single point increase of cum BP exposure level,the incidence of CKD increased 6% in the general population (P for trend < 0.01),increased 8% in male (P for trend < 0.01),and 3% in female (P for trend=0.12).Conclusion As the cumulative exposure to blood pressure increases,the risk of CKD incidence rises,especially in men.
