1.The expression of TLR2 and TLR4 in invasive pulmonary aspergillosis mice

Objective To study the roles of TLR2 and TLR4 in the progress of invasive pulmonary aspergillosis(IPA) in experimental mice.Methods The mice were divided into three groups including the group of normal mice,the group of normal mice infected with A.fumigatus and the group of IPA mice.The mice were sacrificed at four time points(8 h,24 h,48 h and 72 h) after infection.The lung tissues from each group were collected for pathological analysis and RT-PcR for detecting the expression level of,TLR2,TLR4 and β-tublin.The ratio of density value of band of each PCR product on electrophoresis to the density value of β-tublin was used to evaluate the expression level of each gene like TLR2.TLR4 and TNF-α.Re-suits The pathological analysis showed the normal structure and no inflananatory reaction in the lungs in the group of normal mice.The infiltration of inflammatory cells,weak injuries and no germination of spore into hypha in the lungs of normal mice infected with A.fumigatus,and serious injuries like destruction of alveolar structure,bleeding,infiltration of inflammatory cells and germination of spore into hypha in the lungs of IPA mice.The expression level of TLR4 at 8 h,24 h,48 h and TNF-α at 24 h and 48 h were lower in IPA mice than that in healthy mice with infection(P<0.05).Conclusion There was low expression of TLR4 and TNF-α in IPA mice lung tissues.Typical pathological injuries in the lungs and germination of spore into hy-pha in IPA mice were observed by the microscope.
2.Recent progress in the studies on taxol

Taxol could induce tumor cell apoptosis throug h activation of Fas/FasL and caspases. bcl-2 also played an important role in t axol-induced cell apoptosis. Furthermore, taxol had the similar action with lip opolysaccharide on inducing the release of bioactive substances including tumor necrosis factor, interleukin and nitric oxide. There were as well some progresse s in taxol-resistance of tumor cells.
3.Problems in China pharmaceutical compulsory licensing system and their solutions

The Doha Declaration has entrusted to the members of WHO the right to use the compulsory licensing for drug patents in the face of global public health crises.Although the basic compulsory licensing system for drug patents has been established in China, the efficiency of its implementation is low, the responsibility of its executive departments is unclear, the related rules are not fully understood by pharmaceutical enterprises, it is thus necessary to improve and perfect the relevant legal provisions and systems in order to effectively improve the accessibility of drugs.
4.Expression of TR6 and survivin gene in the tissue of cardiac cancer and its clinical significance

Objective To explore the clinical significance of expression of DcR3/TR6 gene in the tissue of cardiac cancer and its correlation with survivin gene expression, Methods Two-step immnaohistochemistry technology was adopted to detect the expression of TR6 and survivin gene in 66 cases of cardiac cancer, none of the patients received prior chemotherapy. Results The positive expression rate of TR6 and survivin genes in cardiac cancer was 54.5% and 62.1% , respectively. The expression of TR6 was correlated with histology type,clinical stage and lymph node metastasis of cardiac cancer ( P<0.05 ). The positive expression rate of TR6 in cases that cardiac cancer with survivin expressed was significant higher than those without surviving expressed (χ2=15.145, P<0.01 ). Conclusions The expres-sion of TR6 and survivin can be regarded as valuable indicators for the diagnosis and prognosis of cardiac cancer.
5.Laparoscopic partial gastrectomy and enterectomy for morbid obesity

Objective To explore the short-term result and safety of laparoscopie sleeve gastrectomy and partial enterectomy for the treatment of morbid obesity.Methods Ten patients underwent laparoscopic sleeve gastrectomy,omentectomy,and partial enterectomy as a treatment option for weight reduction between December 2006 and September 2007.The procedure included 70%-75%vertical (sleeve)gastrectomy,omentectomy,and 1/3 to 2/5 enterectomy preserving proximal jejunum and most of the ileum. Results Laparoscopy was completed in all patients,no conversion to open surgery.The operation time averaged at 3.1 hours(range 2.5-3.5 hours),and there was no postoperative complications.The median postoperative hospital stay was 7 days(range 6-8 days).Mean preoperative BMI was 36.1(32.0-40.5)kg/m2.Mean decrease in BMI was 4.1(3.0-4.7)ks/m2,5.6(3.2-9.0)kg/m2,and 7.3(3.2-10.7)kg/m2 respectively,and mean weight loss at postoperative 1,3,and 6 months was 11.7(7-15)kg,17.5(8-25)kg,and 22.0(8-32)kg respectively.Conclusion Laparoscopic sleeve gastrectomy with omentectomy and partial enterectomy is an effective and safe surgical option for the treatment of morbid obesity.
6.Study on in vitro microdialysis recovery of hydroxycamptothecine.

OBJECTIVETo establish the mothod to dectect the microdialysis recovery of HCPT and to investigate the influencing factors, thus to supply experimental basis for in-vivo microdialysis of HCPT.
METHODThe in vitro recovery of HCPT was detected by concentration difference method (increment method and decrement method). The influence of flow rates, medium concentration and temperature on the HCPT recovery and the stability were studied.
RESULTThe recovery detected by increment method was the same as by decrement method. The recovery was independent of HCPT concentrations in the medium. The hydroxycamptothecine recovery had good stability and increased as the temperature rose.
CONCLUSIONMicrodialysis sampling can be used for the pharmacokinetic study of HCPT. Retrodialysis can be used for the determination of the HCPT in vivo recovery.

7.Repair of left ventricular aneurysm secondary to myocardial infarction with cytokine-conjugated collagen scaffolds in rat models

Objective To compare the vascularization of collagen scaffolds with or without growth factors and their efficacy on cardiac function in postinfarcted rats underwent surgical ventricular restoration.Methods Collagen scaffolds were activated with 1-ethyl-3-[3-dimethylaminopropyl]carbodiimide hydrochloride chemistry (EDC) as control or continually covalently immobilized with vascular endothelial growth factor (VEGF) and VEGF + basic fibrohlast growth factor (bFGF) as experimental groups.Adult SD male rats underwent left anterior descending artery (LAD) ligation to generate transmural myocardial infarction(MI).Four weeks later,by echocardiography,rats with moderate scar size(25%-35% akinetic area of freedom wall of left ventricle) were screened out,assigned into 3 groups randomly and received the surgical ventricular restocation (SVR).Then,cardiac function was measured by echocardiography at 1w,2w and 4w after patch implantation.At endpoint of study (4w after patch implantation),the rats were sacrificed and the hearts were harvested.Vascularization of patch were determined by capillary density (evidenced by vWFⅧ staining) or mature vessel density (evidenced by SMA staining) respectively.Results The general mortality of the animal model is 15% (6/40).A significant improvement of cardiac function was observed in all animals at 1 w after patch implantation but that was better preserved in both cytokine-conjugated groups 4w later (control group vs.VEGF group,P < 0.05,control group vs.VEGF + bFGF group,P < 0.01).More capillaries were present in patch with growth factors (P <0.05),while significant functional vessel formation was observed only in VEGF + bFGF group (P <0.01 vs.control or VEGF group).Additionally,we identified a positive correlation between heart function and mature vessel density (P =0.0297,r2 =0.998).Conclusion The mechanical property of collagen scaffold can be effectively improved by EDC,the growth factors immobilized in scaffold were in favor of vascularization of patch,which may facilitate the preservation of cardiac function posterior to SVR.
8.Fetomaternal hemorrhage in RhD-negative pregnant women in Tianjin

Objective To study and monitor the situation of femomaternal hemorrhage (FMH) in RhD-negative pregnant women in Tianjin, obtain the FMH data of such population, and analyze the relationship between FMH and age, blood type, gestational age, hemolytic disease of postpartum neonates, etc. Methods The FMH level was detected by flow cytometry with FITC-anti-HbF monoclonal antibody. The blood type was detected by blood serum method. The irregular antibody was identified by saline method and indirect anti-human ball method. The hemolysis of postpartum neonates was detected by three tests of hemolysis. Results The FMH volume of 86 RhD negative pregnant women was between 0 and 11.48 ml, with an average of 1.82 ml. There were 63.95%of pregnant women showed a volume of FMH<2.0 ml, 23.26%between 2 and 4 ml, 11.63%between 4.0 and 10.0 ml, and 1.16%>10 ml. The proportion of lower FMH in pregnant women≤30 years old was>11.71%higher than that in the pregnant women>30 years old, but the difference was no statistical significant. There was no significant difference in FMH of pregnant women with O, A, B and AB types. The proportion of higher FMH in pregnant women with compatible ABO blood type with her husband was 12.46% lower than that of the heterozygous cases, but the difference was no statistical significant. The proportion of higher FMH in the pregnant women with 28 to 32 weeks gestational age was 14.55% higher than that of ≤28 weeks and was 35.32% higher than that of >32 weeks, and the differences were statistical significant. Three samples in the 86 samples were positive for anti-D antibody, and their three hemolytic test results were strongly positive with the anti-D titer from 1:2 to 1:32 and the FMH volume from 1.50 to 6.93 ml. The proportion of lower FMH in the 10 pregnant women without postpartum hemolysis was 70% higher than that in 5 pregnant women with postpartum hemolysis, but the differences were not statistical significant. Conclusions The results suggest that monitoring FMH content by flow cytometry can reflect FMH in Rh-negative pregnant women. The studies on the relationship between FMH and age, blood type, pregnant time and hemolytic disease of postpartum neonates can provide basically experimental data for standard use of anti-D immunoglobulin in pregnant women.
9.Research progress in research on platelets derived from induced pluripotent stem cells in vitro

Currently, there are insufficient sources of platelets for clinical transfusion, and there are risks of alloimmune reactions and transfusion-transmitted infections (TTI) after transfusion. In recent years, platelets derived from human induced pluripotent stem cells (hiPSCs) have become one of the hottest research topics in the transfusion community, and studies have shown that they have the potential to address the limitations of platelet transfusion and alleviate the conflict between platelet supply and demand in clinical settings. However, the efficiency of hiPSCs in producing functional platelets in vitro is still low, and the yield and quality are still far below clinical transfusion standards. In this review, the basis and applications related to hiPSCs-derived platelets, studies related to human leukocyte antigen (HLA) gene-silenced hiPSC-derived platelets, and challenges faced by hiPSCs-derived platelet products were reviewed, providing references for in-depth research and future clinical applications of hiPSCs-derived platelets.
10.Totally laparoscopic surgical treatment of congenital choledochal cysts in adult patients

Objective To evaluate feasibility of totally laparoscopic surgical treatment of congenital choledochal cysts(CCC) in adult patients.Methods The clinical data of 36 adult CCC patients were analyzed retrospectively.In this study there were 31 type Ⅰ cases and 5 type Ⅳ cases,divided into laparoscopic operation group (n =18) and open operation group (n =18).Results There were no death cases in perioperative period and all patients were followed up.The total operative time of laparoscopy group compared with open group respectively was (212 ± 43) min and (135 ± 20) min (P < 0.05),the volume of blood loss during operation was (75 ± 20) ml and (150 ± 49) ml,(respectively t =6.875,6.000,P <0.05).Postoperative recovery time of intestinal function was (65 ± 15) h vs (94 ± 12) h,celiac drainage tube indwelling time was (72 ± 20) h vs (89 ± 26) h,postoperative hospital stay was (5.8 ± 1.4) d vs (8.9 ± 0.9) d,(respectively t =6.444,2.199,7.908,P < 0.05).One case suffered from reflux cholangitis in the laparoscopic group,1 case of choledochojejunostomy stricture and recurrence of intrahepatic calculi in the open group.Conclusion Laparoscopic cystectomy and cholangiojejunostomy for congenital choledochal cyst in adults is safe and feasible.
