1.Negative regulation of transforming growth factor-β superfamily co-receptor Ⅲ in tumorigenesis

The expression of transforming growth factor(TGF)-β supeffamily co-receptor (Tβ3R) Ⅲ is often lost in many kinds of cancers.Tβ3RⅢ plays a role of negative regulation in tumorigenesis.T3RⅢ could regulate the cellular invasion,migration,proliferation and apoptosis by multiple mechanisms,such as mediating the TGF-β signaling pathway,impacting the mitogen-activated protein kinase(MAPK),nuclear factor-kappa B (NF-κB) and Cdc42 and producing soluble TβR(sTβR) Ⅲ.Defining the mechanisms of absence and physiological functions of TβR Ⅲ in cancers has great significant for the diagnosis,treatment and prognosis of cancer.
2.The relationship between homocysteine, polymorphism of its enzymes and cerebral thrombosis

ObjectiveTo explore the relationship between plasma homocysteine (Hcy), polymorphism in 5,10-methylenetetrahydrofolate reductase (MTHFR) and cystathionine-β-synthase (CBS), and cerebral thrombosis.Methods87 subjects with first-ever acute cerebral thrombosis and 80 controls were studied. The plasma Hcy levels were measured using high-performance liquid chromatography-fluorescence detection (HPLC-FD). The polymorphism in MTHFR was determined by a polymerase chain reaction (PCR) assay and subsequent restriction enzyme digestion and that in CBS was determined by amplification refractory mutation system (ARMS).ResultsThe fast plasma Hcy level in the patient group was (15.28±4.33)μmol/L significantly higher than that ( 11.32 ±3.86)μmol/L in the control group (P<0.001). Different genotype had different influence on the plasma Hcy levels. There were no differences in genotype frequencies or allele frequencies between the patient group and control group (P>0.05).ConclusionCommon mutations in MTHFR, CBS G919A and CBS T833C lead to hyperhomocysteinemia. Hyperhomocysteinemia, but not common mutations in MTHFR and CBS is associated with the increased incidence of cerebral thrombosis.
3.Inhibition of viral protein r on human colorectal cancer cells and the possible mechanisms

Objective To investigate the inhibitive effects of viral protein r (Vpr) of human immunodeficiency virus 1 ( HIV-1 ) on human colorectal cancer cell line HCT-8,and to find the possible mechanisms.Methods The HCT-8 cells were divided into the control group,adv group and adv-Vpr group.HCT-8 cells were not treated in the control group; HCT-8 cells were treated with Adv or Adv-Vpr at different multiplicity of infection (MOI) in the Adv group or Adv-Vpr group,respectively.Cell proliferation was detected by MTT assay.Cell cycle,apoptosis and mitochondrial membrane potential were detected by flow cytometry.The expression of apoptosisrelated proteins was detected by Western blot.All data were analyzed by using the q test,t test and one-way or two-way analysis of variance.Results The proliferation of HCT-8 cells was significantly inhibited by Vpr.The MTT value of HCT- 8 cells in the Adv-Vpr group was 1.03 ± 0.04,which was significantly lower than 2.46 ± 0.15 in the Adv group and 2.51 ± 0.14 in the control group at 72 hours after Adv-Vpr transfection ( MOI =200) ( F =144.6,P < 0.05).The ratio of HCT-8 cells in the G2/M phase was 37.31% ± 5.90% in the Adv-Vpr group,which was significantly higher than 18.30% ± 6.04% in the Adv group and 16.66% ± 3.51% in the control group ( F =10.08,P < 0.05 ).The ratio of HCT-8 cells with decreased mitochondrial membrane potential was 32.07% ±5.64% in the Adv-Vpr group,which was significantly higher than 3.32% ±0.79% in the Adv group and 2.76% ±1.43 % in the control group at 48 hours after Adv-Vpr transfection ( MOI =200) ( F =64.45,P < 0.05).The apoptosis rate of HCT-8 cells was 37.62% ±6.48% in the Adv-Vpr group,which was significantly higher than 3.44% ± 1.11% in the Adv group and 2.93% ± 1.07% in the control group at 72 hours after Adv-Vpr transfection ( MOI =200) ( F =122.4,P < 0.05 ).The results of Western blot showed that Vpr induced cleavage and activation of Caspase-9 and Caspase-3 and phosphorylation of Chk1-S345,while the expression levels of Fas,Fas-L,ERK1,ERK2 remained the same at 48 hours after Adv-Vpr treatment ( MOI =200).Conclusions Vpr inhibits the proliferation of the HCT-8 cells in vitro through G2/M phase arrest and apoptosis.Vpr plays its role by activating DNA damaging pathway and initiating mitochondria apoptotic pathway.Vpr is a potential therapeutic agent for colorectal cancer.
4.Microsurgical treatment of cerebellum hemangioblastoma

Objective To investigate the operative technique and treatment effect for the cerebellum he-mangioblastoma. Methods The clinical data of 18 patients with cerebellum hemangioblastoma who underwent sur-gery were analyzed retrospectively. The clinical data included imaging data,the operative approach and microsurgery method. Results In the 18 cases, 15 undertook complete remove of tumor,and the other 3 cases experienced partial remove. 16 patients were followed up for 1 to 3 years,and 2 cases experienced recurrence. No death occurred. Con-clusion The measures including sufficient preoperative imaging and gentle manipulation during the operation can significantly increase the rate of total removal of the tumors, while radioneurosurgery or γ treatment is applicable after operation.
5.Protective effect of paeonol on human umbilical vein endothelial cells injured by hyperlipidemic serum

OBJECTIVETo investigate protective effect of paeonol (Pae) on human umbilical vein endothelial cells (HUVECs)injured by hyperlipidemic serum and explore its possible mechanism.METHODS The injury model was induced by 20% hyperlipidemic serum incubating HUVECs for 24 h.Pae 124,247 and 495.μmol· L-1 were given followed by administration of hyperlipidemic serum for 24 h.The morphological changes were observed under inverted microscope,cell survival rate was evaluated by MTT method,the nitric oxide (NO) content was measured by nitric acid reductase method and the endothelial nitric oxide synthase (eNOS) mRNA expression was determined by RT-PCR.RESULTSCompared with normal contorl group,most cells in model group split and exfoliated.However,the morphology was tending to normal level after intervention with Pae.Pae significantly improved cell viability(P <0.01).Compared with model group,the survival rate increased from (53.0 ±10.1 ) % to ( 68.4 ± 9.1 ) %,( 84.5 ± 6.7 ) % and (98.1 ± 7.5 ) %,respectively.The NO content and eNOS mRNA expression both increased greatly in Pae groups(P < 0.01 ).Compared with model group,content of NO increased from 54 ± 4 to 79 ± 6,115 ± 5 and ( 136 ± 6) μmol · L- 1,respectively.The expression level of eNOSmRNA improved from 0.215 ± 0.060 to 0.451 ± 0.045,0.563 ± 0.013,0.704 ± 0.068,respectively.CONCLUSIONPae exerts protective effect on HUVECs injured by hyperlipidemic serum by increasing eNOS mRNA expression,which might therefore improve the content of NO.
6.Automatic Oral Drug Dispensing System in Hospital Pharmacy:Application and Experience

OBJECTIVE: To provide references for the application of automatic oral drug dispensing system in hospital pharmacy. METHODS: The chief structure of automatic oral drug dispensing system in our hospital (TR-200FDS system) was introduced; the main working process and employment of this system were briefly introduced,and its advantages and disadvantages were discussed. RESULTS: The application of automatic oral drug dispensing system is characterized by convenience,cleanness,quickness and accuracy,but the relative costs are on the high side. CONCLUSION: The application of automatic oral drug dispensing system in hospital pharmacy can enhance hospital pharmaceutical care level,and it may represent the trend of the development in hospital pharmacy.
7.Thalassemia gene detection and investigate in Dongguan

Objective To investigate the type of gene mutation and its distribution in patients with thalassemia in Dongguan . Methods 7 845 specimens collected from the patients and individuals with physical examination in our hospital from June 2014 to May 2015 were performed according to the results of routine blood and electrophoresis screening with suspected cases .The speci‐mens with phenotype positive were definitely verified the thalassemia type by using Gap‐PCR and reverse dotblot(RDB) .Results Among 7 845 specimens ,suspected cases of 1 132 cases ,662 specimens were finally diagnosed as α‐thalassemia andβ‐thalassemia , with the thalassemia carrying rate of 8 .44% (662/7 845) ,including 412 cases(5 .25% ) of a‐thalassemia .The most common type ofαα/‐‐SEA ,‐α3 .7/ααaccounted for 61 .17% and 17 .48% inα‐thalassemia ,also detected HKαα/‐‐SEA mixed type in 1 case and 250 cases(3 .19% ) ofβ‐thalassemia cases .The most common type ofβCD41‐42/βN ,βIVS‐Ⅱ‐654/βN ,βCD17/βN accounted for 37 .6% , 23 .2% ,16 .0% inβ‐thalassemia .The α‐thalassemia composite β‐thalassemia for 12 cases(0 .18% ) .Conclusion Dongguan city of Guangdong province is a high incidence area of thalassemia .Premarital examination ,genetic counseling should be strengthened ,and reduce the birth rate of the thalassemia children to improve the quality of the population .
8.Immunogenicity, immune mechanisms and immune modulatory effects of mesenchymal stem cells derived from bone marrow of patients with multiple myeloma

Objective To study the immunogenicity,immune modulatory effects and mechanisms of mesenthymal stem ceils (MSCs) derived from the bone marrow of patients with multiple myeloma (MM).Methods The mononuclear cells from the bone marrow of MM patients were obtained and cultured.Immunophenotypes were investigated by using FACS.The levels of cytokines were determined by using enzyme linked immunosorbant assay (ELISA).The endocytosis of monocyte-derived dendritic cells (DCs) was investigated by using FACS.Moreover,the immunoregulatory ability of DCs on proliferation of T lymphocytes was detected by using mixed lymphocyte culture assay.Results MM-derived MSCs had no expression of HLA-DR and costirnulatory molecules (CD40,CD80,CD83 and CD86).MM-derived MSCs could significantly suppress proliferation of T lymphocytes in a dose-dependent manner,which could be reversed by antiTGF-β1 and anti-HGF antibodies.MM-derived MSCs inhibit the endocytosis of DCs.MM-derived MSCs could inhibit the secretion of IL-12 and significantly inhibit the function of DCs on proliferation of T lymphocytes.Conclusion MM-derived MSCs harbored low immunogenicity and immunoregulatory effect in vitro,and this effect was achieved through cytokines.

10.A modified three-vessel occlusion mouse model with ischemic stroke

Objective To develop a reliable and reproducible model of ischemic stroke .Methods Male C57BL/6J mice were randomly divided into three groups:three-vessel occlusion (3VO, 15-min temporary occlusion of the bi-lateral common carotid arteries superimposed on a permanent occlusion of the right distal middle cerebral artery , n=20 ) , suture MCAO control ( n=20 ) , and sham-operated group ( n=10 ) .At 24 h functional outcome was as-sessed using corner test , cerebral ischemic lesion was determined by TTC staining ( n=10 ) , and then the surviral rate was measured till day 7 (n=10).Results The 3VO group got significantly lower neurological deficit score as compared to sham-operated group ( -0.7 vs -0.04 , P<0.01 ) .Infarct volume of mice in 3 VO group was 17.6%±1.6%, while 42.6%±15.0%in MCAO group ( n=10 ) .The 3 VO group got a significantly increased 7-day survival rate compared to suture MCAO group (90%vs 60%, n=10).Conclusions The present three-ves-sel occlusion model is stable and reliable for research on ischemic stroke .
