1.Research progress on the pathogenesis of chronic urticaria

Chronic urticaria is a very common allergic disease in the outpatient.It has complicated pathogenesis and there is no conclusion up to now.The activation of the mast cells and the release of the histamine are the key point of the chronic urticaria onset.In recent years,the studies have demonstrated that the autoimmunity and the chronic infection are the main cause of the disease.There are other factors including dysfunction of the coagulation,deficiency of the vitamin D,pseudoallergen,psychic factor and genetic mechanism.This article will make a review of the pathogenesis of the chronic urticaria from the aspects mentioned above.
2.Effects of Compound Glycyrrhizin on Chemotactic Factors' Expression in Peripheral Blood Mononuclear Cells

OBJECTIVE: To study whether the therapeutic action of compound glycyrrhizin(SNMC) on allergic disorders can be achieved through inhibition on the expression of chemotactic factors in peripheral blood mononuclear cells(PBMC).METHODS: PBMC of 15 healthy subjects were obtained and divided into 4 drug-treated groups: SNMC group,dexamethasone group,mizolastine group,and loratadine group;meanwhile,a positive control group(proinflammatory stimulant was added) and a negative control group(no proinflammatory stimulant was added) were used as control.Fluorescent quantitation polymerase chain reaction was employed to determine the mRNA expression of the chemotatic factors including RANTES,MCP-1,MIP-1? and ELISA was employed to detect their concentrations in supernatant.RESULTS: In the positive control group,both the mRNA expressions and the concentrations of the 3 chemotatic factors increased significantly,and which were significantly lower in the drug-treatment groups than in the positive control group.There were significant differences between the positive control group and dexamethasone-treated group or SNMC-treated group in the expression of MIP-1? mRNA(P

Objective:To analyse the whole DNA methylation status , expression of DNMT1 and DNA methylation status of DNMT1 promoter from patients with systemic lupus erythematosus ( SLE) .To assess the negative feedback effection of DNA methylation on the expression of DNMT1 in the patients with SLE.Methods:The whole DNA methylation status in T cells from 34 SLE patients and 23 healthy controls was assessed by the specific monoclonal antibodies to 5-methylcytosine (5-mc) and the DNA methylation status of DNMT1 promoter was assessed by the Bisulfite Conversion Kit and sequencing.Real time RT-PCR was applied to analyse DNMT 1 mRNA levels in T cells from the patients and controls.Results: SLE patients had siginificantly whole DNA hypomethylation than controls(P<0.05),and the whole DNA methylation was negative correlated with the SLE-DAI.There was no difference in levels of DNA methylation of DNMT1 promoter between patients and controls ,also no correlation between the levels of DNA methylation of DNMT 1 promoter and DNMT1 mRNA.Conclusion:There was no negative feedback effection of DNA methylation on the expression of DNMT 1 in the patients with SLE.The mechanism of lower expression of DNMT 1 in patients with SLE should be studied further.
4.Effect of continuous nursing on the psychological status and quality of life in patients with low rectal cancer after Miles operation

Objective To investigate the effect of continuous nursing on the psychological status and quality of life in patients with low rectal cancer after Miles operation.Methods A total of 84 patients with low rectal carcinoma underwent Miles operation were selected from August 2014 to August 2016 in Henan Rongjun Hospital.The patients were divided into observation group (n =38) and control group (n =46) according to nursing measures.The patients in the control group were given routine nursing measures,while the patients in the observation group were given continuous nursing measures on the basis of routine nursing care.The psychological status and quality of life of the patients were compared between the two groups before and after intervention.Results There was no significant difference in swlf-rating anxiety scale (SAS) and self-rating depression scale(SDS) scores between the two groups before intervention (P > 0.05).After six months intervention,the SAS and SDS scores in the two groups were significantly lower than those before intervention,and the scores of SAS and SDS in the observation group were significantly lower than those in the control group (P <0.05).There was no significant difference in the quality of life between the two groups before intervention (P > 0.05).After six months intervention,the quality of life of patients in the two groups were significantly higher than those before intervention,and the quality of life of patients in the observation group were significantly higher than those in the control group (P < 0.05).Conclusion Continuous nursing can significantly improve the negative emotions of anxiety and depression,and improve the quality of life in patients with low rectal cancer after Miles operation.
5.Detection of lymphocyte function-associated antigen-1 on peripheral blood mononuclear cells from patients with systemic lupus erythematosus

Objective To investigate the expression of lymphocyte function-associated antigen-1 (LFA-1)on peripheral blood mononuclear cells(PBMCs)from patients with SLE and its relation to the occurrence and development of SLE.Methods PBMCS were obtained from 30 patients with SLE and 24 normal human controls.Atier labeled with anti-CD3-FITC and anti-CD11 a-PE,PBMCS were detected for the expression of CD11a by flow cytometric analysis.Results The positivity rate of CD11a was significantly higher in patients with SLE and those with active SLE than in the normal controls (73.74%±7.89%and 77.11%±7.46%vs 68.21%±4.58%.both P<0.05).Furthermore,increased positivity rate of CD11a was observed in patients with active SLE compared with those with stable SLE (77.11%±7.46%vs 69.33%±6.27%,P<0.05).but there was no significant difieFence between the latter group and normal controls(P>0.05).In addition.the expression level of CD11a was positively correlated with SLE disease activity index (r=0.64,P<0.0 1)in patients.Conclusion The overexpression of LFA-1 may take part in the occurrence of SLE and correlate with the activity of SLE.
6.Expressions of miR-17 and miR-20a in CD4+ T cells from patients with systemic lupus erythematosus and their correlations with disease activity

Objective To investigate the expressions of miR-17 and miR-20a in CD4+ T cells from patients with systemic lupus erythematosus (SLE) and their correlations with disease activity.Methods CD4+ T cells were isolated from the venous blood of 30 patients with SLE and 18 healthy human controls.RNA was extracted from the CD4+T cells,and real-time fluorescence-based quantitative PCR (qPCR) was performed to determine the expression levels of miR-17 and miR-20a.The intergroup comparison of miR-17 and miR-20a expression levels was done using t test and Mann-Whitney test,and the correlations of miR-17 and miR-20a expressions with clinical parameters were assessed using Pearson or Spearman correlation coefficients.Results The patients with SLE showed increased expression levels (2-△c~) of miR-17 and miR-20a compared with the healthy controls (0.24 ± 0.08 vs.0.15 ± 0.06 for miR-17,0.19 ± 0.10 vs.0.12 ± 0.09 for miR-20a,both P < 0.05).The miR-17 and miR-20a expression levels were also significantly higher in patients with active SLE than in those with inactive SLE and the healthy controls (0.27 ± 0.07 vs.0.20 ± 0.05 and 0.16 ± 0.08 for miR-17,0.22 ± 0.10 vs.0.15-0.07 and 0.13 ± 0.09 for miR-20a,all P < 0.05),but similar between the patients with inactive SLE and healthy controls (both P > 0.05).The expression levels of both miR-17 and miR-20a were positively correlated with SLE disease activity index (SLEDAI)(r =0.45,0.38,respectively,both P < 0.05) and anti-dsDNA antibody level (r =0.54,0.46,respectively,both P <0.05),but negatively correlated with serum C3 levels (r =-0.43,-0.42,respectively,both P < 0.05).Condusions The expressions of miR-17 and miR-20a are increased in CD4+ T cells from patients with SLE,and correlated with the disease activity in SLE.
7.Preliminary study on efficacy of small doses of furazolidone for duodenal ulcer and clearance of H.pylori

8.Screen of single nucleotide polymorphism of DNA methyltransferase 1 in patients with systemic lupus erythematosus

Objective To screen single nucleotide polymorphism(SNP)of DNA methyltransferase 1 gene in patients with systemic lupus erythematosus(SLE).and to investigate the significance of abnormal expression of DNA methyltransferase in the pathogenesis of SLE.Methods Peripheral blood mononuclear cells(PBMCs)were obtained from 11 patients with SLE and 12 normal human controls.DNA was extracted from the PBMCs.and PCR was performed to amplify some exons(1 to 7)and corresponding introns followed by direct sequencing.Results There was a 21397T/C SNP in the intron 7 between exon 7 and exon 8 of DNA methyltransferase 1 gene in patients with SLE.The frequency of CC homozygote increased in patients with SLE compared with that in normal controls(5/11 vs 1/12,X~2=4.10,P<0.05).Conclusions There is a 21397T/C SNP in DNA methyltransferase 1 gene in patients with SLE.which may be a component of genetic background underlying the abnormal expression Of DNA methyltransferase 1.
9.Effect of audio and video feedback teaching on the rehabilitation of patients with ankylosing spondylitis

Objective To investigate the effect of the use of audio and video feedback on the rehabilitation of patients with ankylosing spondylitis (AS). Methods 60 AS patients were divided into two parts (32 in the experimental group and 28 in the control group) by random number table method, and two groups of patients were treated with the same drug treatment. Patients in the control group were instructed by oral instruction, in the experimental group, the use of audio and video feedback teaching guide patients family exercise, compared with the two groups of patients with four months of follow-up, family exercise compliance, joint function and quality of life, and other indicators. Results Four months after the intervention, the experimental group patients family exercise compliance was better than 84%(27/32), higher than the control group of 36% (10/28), the difference was statistically significant (χ2=9.238, P < 0.01). Patients in the experimental group in four months after the intervention, the health questionnaire to investigate the physiological function, role limitation due to physical problems, body pain, general health, vitality, social function, role limitation due to emotional problems, mental health eight dimension scores respectively were (72.7±8.4), (81.5±9.5), (57.2±8.4), (65.5±8.4), (62.9±7.2), (78.4±7.4), (67.3±6.2), (77.2±7.6) points while in the control group were (60.8±9.1), (72.1±9.6), (50.3±9.4), (60.8±7.0), (58.2±9.1), (74.4±8.7), (60.2±7.2), (74.3±7.4) points, the difference between the two groups had statistical significance (t=1.493-5.266, P < 0.05 or 0.01). Four months after the intervention, the scores of Bath Ankylosing Spondylitis Functional Index (BASFI) of patients with AS in the experimental group were (1.1± 0.8), (1.7±0.2), (1.8±1.3), (1.6±1.0), (3.1±2.1), (1.0±0.6), (1.9±1.6), (3.8±2.5), (2.6±1.7), (2.7±1.6) points, while (3.8±1.6), (2.8±1.5),(2.6±1.5)、(2.8±1.2),(4.4±2.2),(2.2±1.0),(3.3±1.6),(6.5±2.5),(5.6±2.6), (4.3 ± 1.5)control group, the two groups the difference was statistically significant (t=0.282-8.427, P<0.05). Conclusions Application of audio and video feedback teaching in patients with AS rehabilitation guidance, help patients to master the family training methods, improve the joint function of patients, improve the quality of life.
10.Research progress in prognosis of diffuse large B-cell lymphoma

Diffuse large B-cell lymphoma (DLBCL) is the most common type of non-Hodgkin lymphoma with highly heterogeneous both clinically and biologically. So the prediction for prognosis of DLBCL and identifying patients with high risk are coming into the focus of the 58th American Society of Hematology (ASH) Annual Meeting. International prognostic index (IPI) risk model is widely known as the powerful prognostic indicator for DLBCL but in rituximab-era is challenged by a variety of new prognosis systems. Biological markers and mutation and expression of genes play an important role in the pathogenesis and progression of lymphoma, and its prognostic value draw much more attention. The prognostic factors of DLBCL, specific types, such as primary breast DLBCL, intravascular large B-cell lymphoma, the presence of a DLBCL component in a follicular lymphoma and relapsed/refractory DLBCL have been paid more attention in this meeting.
