2.Study of the relationship between thrombopoietin and infection-related reactive thrombocytosis

ObjectiveTo study the mechanism of infection-related reactive thrombocytosis.MethodsSeventy-five infectious disease patients were selected including 42 cases with acute infections phase thrombocytosis (acute infectious phase thrombocytosis group),18 cases with acute infectious phase normal platelet count (acute infectious phase normal platelet count group),15 cases with recovered phase (infectious recovered phase group) and 16 cases with healthy controls(control group).The serum thrombopoietin (TPO),intefleukin-6 (IL-6),white blood cell,platelet was determined and compared among 4 groups.Results The serum TPO in acute infectious phase thrombocytosis group [( 159.1 ± 65.9) ng/L]was higher than that in acute infectious phase normal platelet count group,infectious recovered phase group,and control group [(43.5 ± 14.4),(40.3 ± 15.2),(41.8 ± 18.9) ng/L](P< 0.05).There was no significant difference in the serum IL-6 between acute infectious phase thrombocytosis group [(542.7 ± 247.0) ng/L]and acute infectious phase normal platelet count group [(598.5 ± 250.4) ng/L] (P > 0.05 ),but which was higher than that in infectious recovered phase group [(43.5 ± 20.7 ) ng/L] and control group [( 38.3 ± 17.6 )ng/L] respectively (P < 0.05 ).The serum IL-6,TPO was positively correlated with platelet,white blood cell.The serum TPO was positively correlated with IL-6.ConclusionElevated TPO leads to the thrombocytosis,which is the possible mechanism of infection-related reactive thrombocytosis.

Pullulan is a linear glucosic polysaccharide produced by the polymorphic fungus Aureobasidium Pullulans, which has long been applied for various applications in medical and food industry due to its security, stability and low adhesive ability.At present, the two problems in restricting pullulan industrial production are the low polysaccharide production and melanin secreted which is hard to erase completely, giving the following process some problem.As a starting point, this review article collects and analyzes the progress on the breeding of pullulan high-yield strain without melanin in recent years, in order to find more efficient strains breeding methods, laying a foundation for further breeding of pullulan high-yield strain without melanin.

The first research on the NS1 protein as the non-structural protein of the type A influenza virus was emphasized on the considering of its depressant effect on the composition of the protein of host cell.And now,with deep research,the evolution of its genes and the antigenicity of its protein have been explained.The NS1 protein of influenza virus has the association with the apoptosis induced by the influenza virus,the regulating function of apoptosis has the direct correlation with possibility of producing interference and the cell line infected by influenza virus.The NS1 protein restrained to producing the interferon by infected cell.The NS1 protein plays an important role on the host anti-viral cytokine responses,it possesses nagitive regulation for interferon's antiviral activity,most observation indicated nagitive regulation might associate with the virulence of influenza virus.Furthermore NS1 protein as an inspection antigen to differentiate and diagnose the poultry which was immunized or naturally infected has a very wide prospect,because the traditional vaccine was used extensively.
5.Effects of miR-200b on proliferation and migration of colon cancer cells

Objective To study the effects of miR-200b on proliferation and migration of sw620 colon cancer cells,and its regulation effect on E-cadherin expression.Methods The expressions of miR-200b in sw620 cells at 24 h and 72 h after pEGP-miR-200b transfection were detected by real-time PCR (RT-PCR).The change of the expression level of E-cadherin after miR-200b transfection was detected using the methods of RT-PCR and Western blot.The proliferation and migration abilities were measured by MTT and scratch test after miR-200b transfection.Results The expressions of miR-200b in sw620 cells at 24 h and 72 h after pEGP-miR-200b transfection raised significantly compared to the control group (t =11.579,P < 0.01 ; t =11.579,P <0.01).MiR-200b transfection inhibited the proliferation abilities of sw620 cells.It is the most significant of the inhibitory effect on the third day and the inhibition rate was 55.34%.MiR-200b transfection markedly inhibited the migration abilities of sw620 cells.The two groups had significant difference in the migration distance of 24,48,72 h (t =11.579,P <0.01 ; t =10.419,P <0.01 ; t =6.955,P <0.01).The mRNA and protein expressions of E-cadherin gene increased significantly by transfecting miR-200b gene in sw620 cells (t =10.432,P < 0.01 ; t =8.325,P < 0.O1).Conclusion Up-regulated expression of miR-200b could inhibite the proliferation and migration abilities of sw620 colon cancer cells.The involved molecular mechanism is probably related to the change of E-cadherin expression.
6.A study on the relationship between extent of hepatic fibrosis and serum thyroid hormone levels of patients with liver cirrhosis

Objective To investigate the relationship between extent of hepatic fibrosis and serum thyroid hormone levels in pa-tients with liver cirrhosis .Methods Chemiluminescence immunoassay technology was adopted to detect serum hyaluronic acid (HA),laminin(LN),collagen type Ⅳ (CIV),procollagen type Ⅲ (PC Ⅲ ),thyroid stimulating hormone(TSH),triiodothyronine (T3) ,thyroxine(T4) ,free thyroxin 3(FT3) and FT4 of 240 patients with liver fibrosis (liver fibrosis group) and 80 healthy people (control group) .Results In the control group ,serum HA ,LN ,CIV ,PCⅢ levels of healthy people in ≥45-year group were signifi-cantly higher than those in <45-year group ,and those in male group were obviously higher than female(P<0 .05) .Serum TSH , T3 ,T4 ,FT3 ,FT4 levels of healthy people in male group were significantly lower than female (P<0 .05) .With the increase of grade in Child-Pugh classification ,serum levels of hepatic fibrosis indexes of patients with liver fibrosis increased markedly (P<0 .05) , while their thyroid hormone levels significantly decreased (P<0 .05) ,especially in T3 and FT3 .Serum HA ,LN ,CIV ,PCⅢ levels of patients with A ,B or C grade were markedly higher than those in control group(P<0 .05) ,and serum T3 ,T4 ,FT3 ,FT4 levels of patients with B or C grade were obviously lower than those in control group (P<0 .05) .Conclusion The extent of liver fibrosis is correlated to serum thyroid hormone levels .
7.Analysis of the clinical risk factors of type 2 diabetes complicated with cerebral infarction

Objective To analyze the clinical risk factors of type 2 diabetes complicated with cerebral infarction .Methods 508 patients with type 2 diabetes were selected and divided into two groups :group A (complicated with cerebral infarction ,n=240) and group B (without cerebral infarction ,n=268) .The basic clinical information of two groups were recorded .The coagulation indica‐tors and biochemical indicators (including blood glucose ,blood lipid ,HbA1c ,blood β2 microglobulin ,urineβ2 microglobulin ,urine microalbuminuria ,fasting insulin ,fasting C peptide etc) were detected respectively .Results There were significant differences in age ,age at onset of diabetes ,duration of hypertension and systolic blood pressure between group A and group B (P<0 .05) .The in‐cidence of hypertension and coronary heart disease in group A were higher than group B (P<0 .05) .Comparing with group B ,the levels of HbA1c ,ApoB ,fasting insulin ,fasting C‐peptide ,lipoprotein (a) ,Hcy ,blood β2 microglobulin ,urineβ2 microglobulin ,and urine microalbuminuria of group A were significantly different (P<0 .05) .The ATⅢ level of group A was significantly lower than that of group B (P<0 .05) .Conclusion The risk factors of type 2 diabetes complicated with cerebral infarction include age ,hyper‐tension ,HbA1c ,ApoB ,fasting insulin ,fasting C‐peptide ,lipoprotein (a) ,Hcy ,etc make diabetics be more prone to cerebral infarc‐tion ect .
8.The expression of miR-200b and CEA and the correlation between the two factors in colon cancer

Objective To study the expression of transcription factor miR‐200b and CEA and the correlation between the two transcription factors in the process of occurrence and development of colorectal cancer .Methods Expression miR‐200b and CEA mRNA by real‐time PCR in 60 colon cancer tissues and corresponding normal tissues were detected and the results were compared with the clinical features and pathological characters .The relationship between the mRNA expression of miR‐200b and CEA in 60 colon cancer tissues was determined .Results The expression rate of CEA mRNA was detectable to highly expressed rates in colon cancer tissues than that in matched normal tissues (P<0 .01) ,whereas miR‐200b mRNA in the colon cancer tissues was signifi‐cantly lower than that in the matched normal tissues(P<0 .01) .There was no significant correlation between miR‐200b ,CEA mR‐NA expression and age ,sex ,tumor location(P>0 .05) .Loss expression or down regulation expression of miR‐200b mRNA was de‐tected ,whereas CEA mRNA was detectable to highly expressed in the different histological grade and Dukes stages .The expression of miR‐200b mRNA and CEA mRNA were positively correlated with the histological grade in colon cancer .A significant correlation was found between the expression of miR‐200b mRNA and CEA mRNA (r= -0 .790 ,P<0 .001) .Conclusion Loss or down regu‐lation expression of miR‐200b mRNA ,whereas CEA is detectable to highly expressed in colon cancer .Negative correlation occurred in miR‐200b mRNA and CEA mRNA indicates that miR‐200b and CEA provide the new clues of genetic diagnosis and treatment .

Objective: To study the effects of transcription factor activator protein-2?(AP-2?)on invasive growth and estrogen receptor-?(ER-?) expression in human colon cancer SW620 cells,and to probe into the involved molecular mechanism.Methods: Plasmid pcDNA3.1(+)-AP-2? and pcDNA3.1(+) were transfected into SW620 cells by liposome-mediated transfection.The adhesion,invasion and migration abilities of SW620 cells were measured by metrical gel adhesion assay and modified Boyden chamber(Transwell assay).The gene and protein expression levels of AP-2? and ER-? in SW620 cells were examined by Real-time PCR,Western blotting and immunofluorescence cytochemistry.The interaction between AP-2?
