Diffuse large B-cell lymphoma (DLBCL) is the most common type of non-Hodgkin lymphoma with highly heterogeneous both clinically and biologically. So the prediction for prognosis of DLBCL and identifying patients with high risk are coming into the focus of the 58th American Society of Hematology (ASH) Annual Meeting. International prognostic index (IPI) risk model is widely known as the powerful prognostic indicator for DLBCL but in rituximab-era is challenged by a variety of new prognosis systems. Biological markers and mutation and expression of genes play an important role in the pathogenesis and progression of lymphoma, and its prognostic value draw much more attention. The prognostic factors of DLBCL, specific types, such as primary breast DLBCL, intravascular large B-cell lymphoma, the presence of a DLBCL component in a follicular lymphoma and relapsed/refractory DLBCL have been paid more attention in this meeting.

Objective To investigate DNA methylation status as well as signal transducer and activator of transcription 3 (STAT3) gene expression in peripheral blood T-lymphocytes from patients with systemic lupus erythematosus (SLE),and to explore their relationship.Methods Peripheral blood samples were obtained from 34 patients with SLE and 23 healthy controls followed by the isolation of T-lymphocytes.Flow cytometry was performed to evaluate DNA methylation status using antibodies against 5-methylcytosine (5-me),reverse transcription (RT)-PCR to detect the mRNA expression of STAT3 in T-lymphocytes.Results Compared with healthy controls,17 patients with active SLE showed significantly decreased DNA methylation levels (8.50 ± 1.42 vs.11.31 ± 1.34,P ＜ 0.01),but increased STAT3 mRNA expression levels (1.34 ± 0.32 vs.1.02 ± 0.28,P ＜ 0.01).However,there were no significant differences between 17 patients with stable SLE and healthy controls in DNA methylation levels (11.30 ± 1.34 vs.11.31 ±1.34,P ＞ 0.05) or STAT3 mRNA expression levels (1.01 ± 0.27 vs.1.02 ± 0.28,P ＞ 0.05).Patients with active SLE also showed significantly reduced DNA methylation levels,but enhanced STAT3 mRNA expression compared with those with stable SLE (both P ＜ 0.01).As Pearson correlation analysis showed,SLE disease activity index (SLE-DAI) was negatively correlated with DNA methylation levels in patients with SLE (r =-0.78,P ＜ 0.01),but positively correlated with STAT3 mRNA expression (r =0.50,P ＜ 0.01),and no significant correlation was observed between STAT3 mRNA expression and DNA methylation levels (r =-0.13,P ＞ 0.05).Conclusion The aberrant expression of STAT3 in peripheral blood T-lymphocytes may be related to disease activity in SLE,but unrelated to DNA methylation levels.

Objective To investigate the effects of 5-azacytidine and estradiol on the methylation of CpG motifs and expression of DNA methyltransferasel (DNMT1) mRNA in patients with systemic lupus erythematosus (SLE) and normal controls.Methods Peripheral blood lymphocytes isolated from 12 patients with SLE and 11 normal human controls were stimulated with phytohaemagglutinin for one day followed by additional 3-day culture with or without the presence of 5-azacytidine of 1 μmol/L or estradiol of 30μg/L respectively.Then.the methylation of CpG motifs was detected by flow cytometry using anti-5-methylcytosine antibody,and DNMT1 mRNA expression by real time reverse transcription-PCR Results After treatment with 5-azacytidine,a decrease wag observed in the methylation of CpG motifs, but not in the expression of DNMT1 mRNA in peripheral lymphocytes from patients with SLE (1=18.60,P<0.01;t=1.56.P>0.05) and in those from the normal controls (t=5.63,P<0.01;t=2.17,P>0.05) compared with untreated lymphocytes.Nevertheless,there were no significant changes in the methylation of CpG motifs or expression of DNMT1 mRNA in lymphocytes from patients with SLE (t=1.53,0.93,respectively,both P>0.05) and normal controls (t=1.93,0.11,respectively,both P>0.05) after the treatment with estradiol.Conclusions The methylation of CpG motifs is suppressed efficiently by 5-azacytidine,and the suppression is unlikely to be associated with the decrease of DNMT1 mRNA.Estradiol has no significant impact on the methylation of CpG motifs and expression of DNMT1 mRNA in lymphocytes.

AIM: To investigate the changes of cytosolic free calcium concentration ([Ca 2+ ] i) and expression of Bcl-2 in HL-60 cells treated by 6F isolated from Pteris semipinnata L. (PSL), and to discuss the relations between calcium ion and cytotoxicity and DNA fragment induction effects of 6F. METHODS: HL-60 cells were used as in vitro model. [Ca 2+ ] i was measured on fluorescent spectrophotometry using Fura-2/AM as Ca 2+ indicator. Bcl-2 expressing level was measured by flow cytometry. Tetrazolium salt (MTT) and diphenylamine staining methods were applied for cytotoxicity assay and DNA fragmentation detection, respectively. RESULTS: [Ca 2+ ] i increased obviously in a dose and time dependent manner after treated HL-60 cells with 6F. 6F decreased the expressing level of Bcl-2. Adding 2 mmol/L Ca 2+ to the medium, or 1 mmol/L EDTA to chelate Ca 2+ , or 4 ?mol/L calcium ionophore A 23187 to increase the concentration of cytosolic Ca 2+ , the DNA fragment induction by 6F was not affected, whereas the cytotoxicity of 6F was enhanced. 250 ?mol/L Zn 2+ attenuated the DNA fragment induction, and the cytotoxicity of 6F against HL-60 cells was enhanced significantly. CONCLUSION: It was speculated that the decreased expressing of Bcl-2 by compound 6F was related to increased [Ca 2+ ] i in HL-60 cells, and DNA fragment induction was possibly catalyzed by Ca 2+ - independent DNase.

ABSTRACT AIM To investigate the effect of ch-itosan on protein kinase C (PKC) activitives and diacyl glycerol(DAG)concentration in murine peritoneal macrophages. METHODS A new ion-pair reversed-phase high liquid chromatography and ra-dioimmunoassay were used to determinate the activity of PKC and the concentration of DAG respectively. RESULTS ( 1) Chitosan induced activation and translocation of PKC in murine peritoneal macrophages. The peak time was 25 min and the activity of PKC came back the basic level at 1. 5 h. (2)Chitosan increased the production of DAG in murine peritoneal macrophages. The peak time was 30 s and the concentration of DAG came back the basic level at 3 min. CONCLUSION The im-munopoteniating effect of chitosan may be associated with the channel of DAG /PKC.

Objective: To investigate the knowledge, attitudes and practice of AIDS control knowledge among students in a university in Hangzhou City, so as to provide the evidence for evaluating the effectiveness of AIDS control in universities. Methods: The knowledge, attitudes and practice towards AIDS control were investigated among students in a university in Hangzhou City using self-designed questionnaires in 2020, and were descriptively analyzed. Results: Totally 6 300 questionnaires were allocated, and 6 164 valid ones were recovered, with an effective recovery rate of 97.84%. The respondents included 2 837 male students ( 46.03% ) and 3 327 female students ( 53.97% ), and had a mean age of ( 18.20±0.76 ) years. The overall awareness of AIDS control knowledge was 73.49%. The awareness rates of “The rights of marriage, employment and school admission are protected among HIV-infected individuals by Chinese laws” ( 61.11% ) and “Homosexual contact in men is the current main transmission pattern among young students in China” ( 66.37% ) were relatively low. There were 768 university students with a history of sexual behaviors ( 12.46% ), and regular sex partners were the predominant sex partners at the first sexual contact ( 679 students, 90.78% ) and at the sexual behaviors during the past one year ( 402 students, 71.91% ). There were 515 students (67.14%) using condoms during the first sexual contact, and there were 300 ( 63.83% ) and 16 students ( 40.00% ) using condoms with regular and casual sex partners during the past one year, respectively. Among 20 men who had sex with men, only 5 students ( 25.00% ) used condoms at each sexual contact. Of students with sexual behaviors, there were 355 (46.22%) and 354 ( 46.09% ) students that considered to be infected with AIDS and other sex-transmitted diseases, and among the university students with sexual behaviors, there were 614 ( 79.95% ) students that were willing to receive HIV testing, and 409 ( 53.26% ) and 305 ( 39.71% ) students that were willing to receive HIV testing in hospitals and disease control and prevention institutions, respectively. Conclusions The awareness of AIDS control knowledge is low among students in the university in Hangzhou City, which does not reach the national goal of supervision and assessment for AIDS control in China. There are high-risk sexual behaviors and the rate of condom use is low.

OBJECTIVE: To provide reference of the ideal entry point for antegrade femoral nailing according to analysis correlation between highest point of greater trochanter and the middle line of the medullary cavity in adults by three-dimensional images. METHODS: From January 2016 to January 2017, 107 adults who underwent continuous computed tomography (CT) scans were ultimately enrolled, including 64 males and 43 females with an average age of (51.7±16.4) years old;54 patients on the left side and 53 patients on the right side. Three-dimensional images were built by using image-processing software (Volume Viewer) to reconstruct geometry of cortex and medullary canal. All people weregrouped according to different femoral greater trochanter morphology, such as anterior apex (AA), posterior apex (PA), middle apex (MA) and none apex (NA). Forwards inclination was adjusted to apparent neck shaft angle (ANSA) and true neck shaft angle (TNSA) on the coronal and saggittal view, recorded as C-ANSA, C-TNSA, S-ANSA, S-TNSA respectively, vertical distance from the middle line of femur medullary cavity to the highest point of greater trochanter of femur on the 4 positions were measured and performed statistical analysis, multiple linear regression was applied to analysis relationship between clinical data and VD value. RESULTS: (1)Comparison of VD value among 4 groups on the 4 positions, there were no difference in VD value between AA and MA groups on the S-ANSA position;and no differences in VD value among AA, MA and NA groups on the C-ANSA and C-TNSA position. (2)There were differences in VD value among AA, MA and NA groups on the sagittal plane;while had difference in VD value between PA and NA group on the coronal plane. (3)Prediction equation of VD value on S-ANSA and S-TNSA position by multiple linear regression showed R=0.343, F=3.409, =0.012 on the S-ANSA position;R=0.317, F=2.846, =0.028 on the S-TNSA position; neck shaft angle and sex were risk factors of VD value on the sagittal plane, while no difference in VD value on the coronal position. CONCLUSION (1)When indentify insertion point in adult femoral nail according to the highest point of greater trochanter as anatomic landmark, the morphology of greater trochanter of femur should be distinguished to certain observation position, then evaluate migration before and after on the sagttial plane and lateral offset on the coronal plane. (2)Migration before and after on the sagttial plane is increase with increase of neck shaft angle, and the degree of migration of female before and after is less than that of male.

Objective Toinvestigateserumalanineaminotransferase(ALT)andtotalbilirubin(TB)concentrationsofchildrenin Panyu district of Guangzhou ,and establish the reference ranges for ALT and TB tests for healthy children of different gender and different age .Methods 4 065 children were chosen randomly ,and their serum ALT and TB were determined .After excluding the outliers ,the results were analyzed statistically .Results After excluding outliers ,a total of 4 000 children were enrolled in the stud-y ,including 2 241 males ,1 759 females ,aged 2- <9 years old .According to statistical analysis ,reference ranges were established . For male the ALT reference ranges were as follows ,2- <3 years old :5 .0-36 .0 U/L ,3- <4 years old :7 .0-21 .0 U/L ,4- <8 years old :6 .0-21 .0 U/L ,8- <9 years old :5 .0-24 .0 U/L ;TB reference ranges of male were as follows ,2- <3 years old :3-12μmol/L ,3- <7 years old :4-15 μmol/L ,7- <8 years old :4-16 μmol/L ,8- <9 years old :7-12 μmol/L .ALT reference range of female were as follows ,2- <3 years old :6 .0-30 .0 U/L ,3- <4 years old:7 .0-22 .0 U/L ,4- <9 years old :6 .0-19 .0 U/L ;female TB reference range ,2- <3 years old :3 .0-12 .0 μmol/L ,3- <4 years old :3 .0-13 .0 μmol/L ,4- <9 years old:4 .0-16 . 0 μmol/L .Conclusion Established the reference ranges of ALT and TB test for healthy children in Panyu district .

OBJECTIVE: To explore the effect of metabolic syndrome on 15 days postoperative adverse events of femoral intertrochanteric fractures with internal fixation. METHODS: From January 2011 to June 2019, 986 elderly patients with intertrochanteric fracture were treated with internal fixation, including 312 males and 674 females, with an average age of(77.71± 7.58) years old. And there were 97 patients with metabolic syndrome and 889 patients without metabolic syndrome. Through the electronic medical record system, the patient's age, gender, fracture type, cause of trauma, body mass index, smoking history, preoperative comorbidities, preoperative bloodtransfusion, operation timing, ASA classification, anesthesia method, internal fixation type, operation duration, and 15 days postoperative adverse events, which include surgical site infection, acute heart failure, acute respiratory failure, pulmonary infection, acute renal failure, DVT, embolism in important organs, urinary tract infection, death, and blood transfusion after surgery were collected. The differences of preoperative and intraoperative baseline datas and 15 days postoperative adverse events between the two groups were compared and analyzed by t text or univariate RESULTS: There were statistical differences in age, body mass index, history of cardiac insufficiency, history of COPD, history of renal insufficiency, operation timing, ASA classification, operation duration, surgical site infection, acute heart failure, DVT, urinary tract infection and blood transfusion between two groups ( CONCLUSION Elderly patients with intertrochanteric fracture with metabolic syndrome had higher postoperative surgical site infection rate, DVT incidence rate, urinary tract infection rate, and postoperative blood transfusion rate. Therefore, the orthopedic treatment team should give more attentionand optimize the treatment plan during the perioperative period with the cooperation of internal physician and anesthesiologist.

AIM　To investigate the poteintation of vincristine-induecd apoptosis by tetrandrine, neferine and dauricine isolated from Chinese medicinal plants in the human mammary MCF-7 multidrug resistant cells. METHODS　The apoptotic cells were detected by fluorescent staining of a combination of Hoechst 33342 and propidium iodide (PI), flow cytometry and agarose electrophoresis. RESULTS　The apoptotic cells induced by vincristine alone accounted for about 10% of all the cancer cells, while the percentage of apoptotic cells induced by a combination of vincristine with tetrandrine, neferine, or dauricine was found to be significantly higher than that by vincristine alone, and their reversal effects were positively correlated with the drug concentration and the exposure time. In addition, tetrandrine was shown to be the most potent in the reversal efficacy among the three compounds to be tested for apoptosis in vitro. CONCLUSION　Tetrandrine, neferine and dauricine showed obvious potenitiation of vincristine-induced apoptosis in the human mammary MCF-7 multidrug-resistant cells.
