1.Relationship between insomnia and non-suicidal self-injury behavior among senior high school students with suicide ideation: the pathway of anxiety

BackgroundSuicidal ideation serves as a sensitive predictive indicator for non-suicidal self-injury (NSSI) behavior, and NSSI behavior brings profound effects to both mental and physical health of senior high school students. Existing studies have made exploration on the relationship among insomnia, anxiety and NSSI behavior in the general population, but the internal mechanism of NSSI behavior in senior high school students with suicidal ideation requires further study. ObjectiveTo explore the pathway of anxiety between insomnia and NSSI behavior in senior high school students, so as to provide references for the intervention on NSSI behavior in such demographic. MethodsFrom December 10 to 15, 2021, cluster sampling was adopted to select 2 572 senior high school students across three senior high schools in Hubei Province as the research objects. Assessment was conducted by using Insomnia Severity Index (ISI), Generalized Anxiety Disorder Scale-7 item (GAD-7) and Adolescents Self-Harm Scale. Spearman correlation analysis was conducted to examine the correlation among scores of scales finished by senior high school students with suicidal ideation. Model 4 from the SPSS 20.0 Process 4.2 was employed to test the mediating path of anxiety between insomnia and NSSI behavior. ResultsA total of 2 421 valid questionnaires (94.13%) were recovered, which revealed 554 cases (22.88%) with suicidal ideation and 408 cases (16.85%) with NSSI behavior. The scores of ISI, GAD-7 and Adolescents Self-Harm Scale of senior high school students with suicidal ideation were higher than those of students without suicidal ideation (Z=17.124, 20.611, 21.314, P<0.01). As correlation analysis showed, the ISI score of senior high school students with suicidal ideation was positively correlated with scores of GAD-7 and Adolescents Self-Harm Scale (r=0.646, 0.354, P<0.01), and GAD-7 score was positively correlated with the score of Adolescents Self-Harm Scale (r=0.375, P<0.01). Among senior high school students with suicide ideation, anxiety acted in a mediating role between insomnia and NSSI behavior, with effect value of 0.111 (95% CI: 0.053~0.185), accounting for 53.88% of the total effect. ConclusionInsomnia of senior high school students with suicidal ideation can not only directly affect NSSI behavior, but also indirectly affect NSSI behavior through anxiety. 2.Clinical analysis of 176 cases of fungal keratitis in Jingzhou

AIM: To analyze the infection characteristics and clinical treatment outcome of patients with fungal keratitis(FK)in Jingzhou area, so as to provide basis for clinical diagnosis and treatment of FK. METHODS: The data of 176 patients with FK diagnosed by etiology in Jingzhou Central Hospital from January 2015 to July 2020 were collected, and the epidemiological characteristics, identification results of fungal strains, drug sensitivity and clinical treatment outcome of the patients were analyzed. RESULTS:Among the 176 patients with FK, 118 were male and 58 were female, and the ratio of male to female is 2.03:1. The age of onset was the largest number of patients in 50-59 years old and 60-69 years old. The peak seasons of onset were April to June and October to December. Risk factors include trauma and possible history of trauma, ocular surface diseases, post ophthalmic surgery, wearing corneal contact lenses and long-term local drug use. The main pathogens were Fusarium(39.8%), aspergillus(30.7%), alternaria(15.9%)and candida(5.1%). The results of drug sensitivity showed that voriconazole and amphotericin B were sensitive, while the drug resistance rate of fluconazole was very high. There were 125 eyes improved or cured after conservative treatment, 12 eyes were injected into corneal stroma, 15 eyes were covered with conjunctival flap or amniotic membrane transplantation, and 21 eyes were treated with corneal transplantation. Enucleation of ocular contents was performed in 8 eyes, including 5 eyes with severe endophthalmitis after conjunctival flap covering. CONCLUSION:The incidence of FK in Jingzhou area is mostly middle-aged and elderly men, mostly caused by corneal trauma in the process of agricultural labor. The main pathogens were fusarium and aspergillus. It is very important to select reasonable antifungal drugs and treatment methods according to the drug sensitivity results.
3.Study on the effect of chlorine disinfectants on the destruction of New coronavirus nucleic acid

Objective To observe the destructive effects of chlorine disinfectants on the nucleic acid of novel coronavirus, and to assess the feasibility of real-time fluorescent quantitative PCR technology for the evaluation of the disinfection effect of novel coronavirus. Methods A suspension quantitative inactivation test was used to observe the inactivation effects of different concentrations of chlorine containing disinfectants and different action times on the novel coronavirus. RT-PCR method was used to detect novel coronavirus nucleic acid for the disinfection effect evaluation. Results The chlorine disinfectants of 1 000 mg/L and 2 000 mg/L could destroy the nucleic acid of ew coronavirusafter application for over 30s. The chlorine disinfectant of 750 mg/L could destroy the nucleic acid of new coronavirusafter application for over 15min. Chlorine disinfectant of 500 mg/L could not completely destroy the nucleic acid of new coronavirusafter 30 min action. Conclusion The disinfection method stipulated in the national prevention and control plan using 1 000mg / L effective chlorine for the epidemic sites of new coronavirus for 30 minutes could completely destroy the nucleic acid of new coronavirus. The Real-time quantitative PCR can be used to evaluate the disinfection effect of new coronavirus.
4.Nanopore DNA single molecule sensing method and synthetic nanopore fabrication

Nanopore single molecule sequencing(SMS) is considered as one of the most promising third generation DNA sequencing method by virtue of fast sequencing and low cost. As the most significant part of the sequencing system, the synthetic solid nanopore has recently been the research highlight in the field of nanopore DNA sequencing because of its fine-controlled size, high reliability and wide applicability. Also, its fabrication method is currently one of the main challenges in this field. In this article, the principles of the nanopore SMS are introduced followed by the review of the difficulties and challenges in the current research stage with emphasis on the nanopore fabrication methods.
5.Analysis of influenza surveillance results in Jingzhou, Hubei Province from 2016 to 2021

ObjectiveTo understand the epidemiological characteristics of influenza in Jingzhou from 2016 to 2021, so as to provide scientific evidence for the formulation of influenza prevention and control policies in this region, and effectively protect people's health. MethodsData of influenza-like illness (ILI) and pathogen surveillance in Jingzhou during 2016‒2021 were collected and statistically analyzed. ResultsA total of 46 272 ILI cases were reported from two hospitals in Jingzhou City from 2016 to 2021. The difference in the constituent ratio of ILI was statistically significant among different age groups (P<0.05). A total of 12 812 specimens were collected from two hospitals for influenza surveillance. A total of 1 513 cases were RNA positive,and the positive rate of influenza virus nucleic acid detection was 11.81%. The RNA positive specimens were mainly B (Victoria), accounting for 39.33%. There were statistically significant differences in the positive rate of influenza virus nucleic acids and different types of influenza virus nucleic acids among different years (P<0.05). ConclusionThe influenza epidemic in Jingzhou peaks in winter and spring, and the new A (H1),A (H3), B (Victoria) and B (Yamagata) types alternate and mixed epidemics dominate.
6.Clinical efficacy of Matrine and Sodium Chloride Injection in treatment of 40 cases of COVID-19.

In this paper, we analyzed medical records of 40 patients with coronavirus disease 2019(COVID-19), in order to explore the clinical efficacy of Matrine and Sodium Chloride Injection in the treatment of COVID-19. The investigation was based on the results of a previous animal test, which was aimed to investigate and confirme the clinical efficacy of Matrine and Sodium Chloride Injection in the treatment of COVID-19. The animal test demonstrated that Matrine and Sodium Chloride Injection has a significant therapeutic effect on the human coronavirus pneumonia for the model mice. The lung inhibition index reached up to 86.86%. The evaluation was conducted on 40 confirmed cases of COVID-19 treated at Jingzhou Hospital of Infectious Disease(Chest Hospital) of Hubei Pro-vince from January 30~(th) to March 21~(th), 2020. In these cases, patients were treated with other integrated Chinese and Western medicines regimens in the recommended Matrine and Sodium Chloride Injection diagnosis and treatment regimen. The clinical manifestations, laboratory data, nucleic acid clearance time, and imaging data were compared and analyzed before and after treatment. After administration with Matrine and Sodium Chloride Injection, the clinical symptoms of 40 cases were alleviated markedly, and their blood analysis and biochemical indexes returned to normal. The lung CT showed more than 50% of lesion absorption rate, and the viral nucleic acid test showed the average clearance time of patients was 16.6 days, and the average length of hospital stay was 25.9 days. After administration with Matrine and Sodium Chloride Injection, the symptoms of cough and fatigue were alleviated significantly, and the appetite was significantly improved compared with before, especially for patients with gastrointestinal symptoms. Additionally, laboratory indicators, especially absolute value and ratio of lymphocytes and CRP were significantly alleviated. According to the chest CT for short-term review, the absorption of lung lesions was faster than before, especially for grid-like and fibrotic lesions. Compared with antiviral drugs, such as Abidol and Kriging, the nucleic acid clearance time was significantly shorter than the cases treated with Matrine and Sodium Chloride Injection. The clinical effective rate of 40 cases was 100.0%. We believed that Matrine and Sodium Chloride Injection have a good clinical effect in the treatment of COVID-19, and suggested increasing the clinical application and further conducting large-sample-size cli-nical verification.

7.Effects of Epigallocatechingallate on Lipid Metabolism Related Gene and Long Non-coding RNA Expression Profile in HepG2 Cells

Objective: To investigate the effects of epigallocatechingallate (EGCG) on lipid metabolism related gene and long non-coding RNA (lncRNA) expression proifle by biochip technology, and to explore the possible relationship between the two elements.
Methods: HepG2 cell was cultured with EGCG at 25μmol/L for 24 hours, the total RNA was extracted and hybridized into the biochip of Human Transcriptome Array 2.0 for mRNA and lncRNA expression profile analysis. Bioinformatics technology was used to establish the possible relationship between lncRNA and the predicted target genes;the data obtained from biochip microarray was conifrmed by real time RT-PCR examination.
Results: The microarray revealed that EGCG treated HepG2 cell expressed 27 differential lipid metabolism genes and 11 of them involved in cholesterol metabolism. In addition, there 285 lncRNA expressions were up-or down-regulated. Bioinformatics technology indicated that the predicted target genes for lipid metabolism might be cis-or trans-regulated by lncRNA;the data from real-time RT-PCR was consistent with the data from biochip microarray.
Conclusion: Tea polyphenols improves lipid metabolism and lncRNA might be involved in the regulation of lipid metabolism related gene.
8.Experimental study on expression of TNF-α and ICAM-1 in cerebral ischemia-reperfusion injury

Objective To detect the expression of TNF α and ICAM 1 in cerebral ischemia reperfusion injury. To explore their probable modulation mechanism.Methods Immunohistochemistry was used to examine the expression of TNF α and ICAM 1.The MPO activity was determined by spectrophotometry.Result The expression of TNF α and ICAM 1 were obviously rised after cerebral ischemia reperfusion (P< 0.05).Conclusion The up regulation of TNF α and ICAM 1 after cerebral ischemia reperfusion induced the accumulation of leukocytes, which involved in ischemia reperfusion injury mechanism. TNF α play an important role in expression of ICAM 1.
9.Sonographic characteristics of high-frequency ultrasound and elastography of thyroid microcarcinoma and the analysis of misdiagnosis

Objective To summarize the sonographic characteristics of high frequency ultrasound and elastography of thyroid microcarcinoma(TMC),and to analyze the causes of misdiagnosis.Methods The preoperative ultrasonic data of 245 suspicious TMCs in 202 patients,as confirmed by operation pathology,were retrospectively analyzed.Results Preoperative ultrasonography accurately diagnosed 221 TMCs,the diagnosis rate was 90.2%,and the misdiagnosis rate was 9.8%.Among the missed diagnosed lesions,18 lesions were nodular goiter,others were 3 nodular goiter with focal papillary hyperplasia of follicular epithelium,2 nodular goiter with adenomatous hyperplasia,1 focal lymphocytic thyroiditis,respectively.The thyroid lesions≤0.5 cm in diameter were more easily to misdiagnose.According to the importance of the ultrasonographic features of TMC,the order were aspect ratio (A/T) ≥ 1,irregular-shape,microcalcifications,low or very-low echo.Based on the above corresponding characteristics and considering other features together,the diagnostic accuracy rate were 94.1 %,93.9%,92.4% and 90.5%,respectively.Less blood supply and ill-defined boundary were the secondary sonographic signs of TMC.The elastographic scores of TMC were most showed 4 to 5 points.Diagnosis of TMC relied on elastography alone is less effective,but when elastograph diagnosis based on high frequency ultrasound,the diagnostic accuracy is much higher,especially when there is no calcification in the lesions.Conclusions High-frequency ultrasound has a very important value in the diagnosis of TMC,while elastography has certain assistant value on the basis of high-frequency ultrasonic diagnosis.
10.Changes of somatostatin and acetylcholine contents in vascular dementia rats

Objective To explore the changes and significance of somatostatin (SS) and acetylcholine (Ach) contents in vascular dementia rats. Methods Model of vascular dementia rats was established. Active avoidance responase (AAR) ratio was used as the index for the judgement of the learning and memory abilities of vascular dementia rats. The SS and Ach contents in different cerebral sections of rats with and without intervention of huperzine A were determined by radioimmunoassay. Results A remarkable decrease in SS and Ach contents was found in vascular dementia rats, which could be reversed by intervention of huperzine A. Conclusion The decreased SS content in cerebrum after ischemia is related with vascular dementia. SS may be involved in the pathological process of vascular dementia through the mutual regulation with Ach.
