1.Determination of depressor constituent sanggenon of Cortex Mori in different sources

Objective:To study the contents and determinate method of Sanggenon C of Cortex Mori in different sources.Methods:Sanggenon C was determined by HPLC. The mobile phase was methanol water(75∶25). Detection wavelength was at 280nm.Results:The average recovery was 96.94% with RSD = 1.72% and the linear range was 0.32~4.80?g. The highest content of Sanggenon C was 0.55% and the lowest content was 0.02% from different producing area. Conclusion: The method is accurate and reliable with good reproducibility.
2.Studies on TLC identification of root bark of Morus alba

Object To study specific TLC identification of the root bark of Morus alba L. (RBMA)Methods The ethanol extracts of RBMA and other confusable species were subjected to TLC in different development system. The TLC plats were examined under shortwavelength UV light or colored by FeCl 3 solution.Results A mixture of chloroform and methanol (5∶1) is used for development and ferric chloride solution is used to color, a specific purple spot of the certified RBMA can be found.Conclusion The specific spot showed with TLC can be regarded as the basis of identification of RMBA and it was separated and identified as sanggenon C.
3.Cognitive analysis on nurses' multiple-sites practice from nurse groups

Objective To investigate the cognition of nurse groups toward nurses' dual practice,and to analyze nurses' tendency to pursuc dual practice and its influencing factors.Methods A questionnaire survey was conducted,and 1010 registered nurses were recruited using stratified sampling method.Results Most (76.4%) nurses agreed with nurses' dual practice.The willingness about nurses' dual practice was significantly different(P=0.022,P=0.008) due to different age and length of service.The top three benefits of nurses' dual practice were to increase reasonable salary of nurses,improve quality of nursing care in community institutions for the aged and increase the utilization of health care resource.As for the disadvantages,disrupting the medical and nursing order,increasing difficulties of management and government's supervision,increasing medical risks.Nurses believed that the main obstacles for nurses' dual practice were the unclear medical risks and the unprotected nurses' interests.Conclusion The nurse groups had a positive and optimistic attitude towards nurses' dual practice.In the case that the government implements relevant laws and regulations,and the interests of nurses are well-guaranteed,nurses' dual practice is a good measure that can improve nurses' work motivation.
4.Determination of Chlorogenic Acid in Fuyanqing Perfusate by HPLC

Objective To establish a method for the determination of chlorogenic acid in Fuyanqing perfusate (FP). Methods The sample was extracted with methyl and the chromatographic condition were as follows: C18 chromatographic column (250 mm? 4.6 mm,5 ? m),a mobile phase of methyl alcohol and water and methyl acid(30 ∶ 70 ∶ 1),the detection wavelength at 327 nm and the flow rate being 0.5 mL/min.Results A linearity of chlorogenic acid in FP was obtained in the range of 0.16 ? g~ 0.58 ? g,r=0.9996(n=6).The average recovery was 98 % and RSD =1.0% (n=6).Conclusion This method is easy,sensitive and accurate for the determination of chlorogenic acid in FP.
5.GC fingerprint of the constituents in the volatile oil of Eucalyptus globulus Labill

AIM:To establish a method for GC fingerprint determination of the chemical constituents in the volatile oil of Eucalyptus globulus Labill.in Yunnan Province,China. METHODS: GC was used to optimize the fingerprint determination method,and to identify the main peaks in the GC fingerprint. RESULTS: 16 mutual peaks were gained on the base of 10 batches of samples. CONCLUSION: Stable and reproducible GC fingerprint is helpful to further analyze the volatile oil constituent of Eucalyptus globulus Labill.
6.Efficacy and safety of tirofiban treatment combined with percutaneous coronary intervention in the elderly with acute ST segment elevation myocardial infarction

Objective To analyze the efficacy and safety of tirofiban treatment combined with percutaneous coronary intervention (PCI) in the elderly with acute ST segment elevation myocardial infarction prospectively. Methods From May 2008 to May 2010, 106 patients who presented with acute STEMI within 12 hours from onset and received successful primary PCI were enrolled into the study. All patients had angiographic evidence of initial total occlusion of infarct-related artery and finally restored toTIMI3 flow after PCI. All patients were divided into tirofiban group (n = 54) and control group (n = 52) according to whether tirofiban was used or not. Assessment of myocardial perfusion included Myocardial Blush Grades (MBG), and the resolution of the sum of ST-segment elevation (sumSTR) at 90 minutes after the procedure. Left ventricular ejection fraction (EF) was measured one week later. Major adverse cardiac events in hospital and bleeding complications were also assessed. Results Baseline clinical and angiographic characteristics of the two groups were similar. Significant higher rates of MBG 3 were observed in the tirofiban group (88. 9% vs57. 7%, P < 0.05). Patients received tirofiban were more likely to achieve higher sumSTR (70. 3% vs 42. 3%, P <0. 05). Ejection fraction was also markedly increased in tirofiban group than control group (56. 2 ± 7.6 vs 46. 7 ± 8. 5, P < 0. 05). In-hospital major adverse cardiac events, it was not different between the two groups(P >0. 05). There were slightly more minor bleeding complications in tirofiban group compared with control(11.1% vs 6. 0%, P >0. 05). No patient had major bleeding or thrombocytopenia.Conclusions Tirofiban can further ameliorate microvascular perfusion and it is safe and feasible for patients with STEMI undergoing primary PCI.
7.Analysis of 17 175 cases with results of physical examination crowd EB virus antibody positive

Objective To study the relationship between nasopharyngeal carcinoma and positive IgA antibodies of EB virus nuclear antigen 1 (NA),EB virus capsular antigen (VCA) and EB virus Zta protein.Methods The serum EB virus VCA-IgA,NA1-IgA and Zta-IgA antibody in 17 175 cases were detected by ELISA.782 cases of EB virus antibody-positive subjects were further given nasopharyngeal CT imaging,electronic nasopharyngoscopy.Finally confirmed by biopsy and immunohistochemistry.Comparison of serology results EB antibody-positive individual,while the two antibody-positive and antibody while three positive nasopharyngeal carcinoma detection rate,combined with evaluation of EB virus antibody detection in nasopharyngeal carcinoma screening value of high-risk groups.Results 17 175 cases of medical groups,the EB virus antibodies were detected in 782 cases,with the most positive individual,accounting for 535 cases (68.41%),nasopharyngeal cancer diagnosed in 15 cases (2.80%);two antibody positive while 213 cases (27.24 %),diagnosed 49 cases of nasopharyngeal carcinoma (23.00%);least three antibody positive while only 34 cases (4.35%),nasopharyngeal cancer diagnosed 18 patients (52.94%);by physical examination,in 16,393 cases of EB virus antibody negative population by nasopharyngoscopy and throat CT and other medical examination,the final diagnosis of nasopharyngeal carcinoma in 6 cases (0.04%).Conclusion Three EB virus antibody combined detection greatly improves the rate of nasopharyngeal cancer diagnosis;we must also do other physical examination to prevent EB virus antibody-negative nasopharyngeal crowd missed.
8.Negatively regulatory effects of bone marrow-derived mesenchymal stem cells supernatant on allogeneic lymphocytes proliferation

BACKGROUND: Preliminary study has proven that adult human bone marrow-derived mesenchymal stem cells (MSCs) suppress peripheral blood lymphocytes proliferation.But the mechanism was still to be investigated.OBJECTIVE: To study the negatively regulatory effect of adult human MSCs on allogeneic lymphocyte proliferation by cell-free condition.DESIGN,TIME AND SETTING: Cytological observation in vitro,which was performed in the Lanzhou General Hospital,Lanzhou Military Area Command of Chinese PLA between October 2005 and December 2007.MATERIALS: The bone marrow sample was provided by the allo-transplantation donor.The peripheral blood lymphocytes were provided by the healthy volunteer.METHODS: Adult human MSCs were separated with Percoll + adherence method.Allogeneic peripheral blood lymphocytes were obtained from healthy donors with Ficoll solution and the cell concentration was adjusted as 2×109/L for use.100μ L MSCs culture supernatant was taken out in 96-well plates.The groups were following: A superuatant + 3-day MSCs culture media (100 μ L/well); B superuatant + phytohemagglutini (PHA; 1 g/L,5 μ L); C medium + LG-DMEM culture media containing 10% fetal bovine serum (100 μ L); D medium + PHA (1 g/L,5 μ L).The cells were incubated at 37 ℃ with 5% CO2 in a fully humidified atmosphere for three days.MAIN OUTCOME MEASURES: Effect of MSCs supematant on proliferation and transformation of variant lymphocytes.RESULTS: Peripheral blood lymphocytes proliferation was suppressed as compared with the blank control group and PHA group after MSCs culture,and the inhibition ratio was 9.00% (P < 0.05).When lymphocytes were stimulated by PHA,the suppression effects were even stronger and the inhibition ratio was 20.91% (P < 0.01).CONCLUSION: Adult human MSCs supernatant can suppress peripheral blood lymphocytes proliferation and transformation; furthermore,PHA can enhance the inhibitory effect,suggesting the negative regulation is at least in part due to indirectly inhibiting lymphocytes via soluble cytokines.

A new molecular method for simultaneously rapid detection and differentiation of Mycobacterium tuberculosis, Mycobacterium bovis, Mycobacterium avium and Mycobacterium paratuberculosis was established by using denaturing high-performance liquid chromatography (DHPLC) combined with multiplex nucleic acid amplification. These 4 important pathogenic mycobacteria were identified by separation of 4 specific PCR-amplified target fragments by DHPLC analysis. A total of 51 Mycobacterium strains and 22 other bacterial species were tested to confirm the specificity of the multiplex PCR-DHPLC assay. The sensitivity of the assay was as low as 10~2-10~3 gene copies. This method rapidly identify the positive clinical samples from human and bovine with higher detection ratio than traditional culture method and was able to identify simultaneously four pathogenic Mycobacterium, which provided a new molecular tool for rapid detection of tuberculosis and paratuberculosis in human and animals.
10.Deep Learning-Based Segmentation of Extra-Pelvic Organs and Metastases in Advanced Prostate Cancer Based on MET-RADS-P

Purpose To explore the feasibility of the deep learning-based segmentation of extra-pelvic region and metastases in advanced prostate cancer based on metastasis reporting and data system for prostate cancer(MET-RADS-P).Materials and Methods Four datasets(68,91,57 and 263 patients with head,neck,chest and abdomen metastases,respectively)from Jan 2017 to Jan 2022 in Peking University First Hospital were retrospectively collected for the development of the classification model of scanning range and segmentation model of different regions and metastases according to the scanning sites(head,neck,chest and abdomen).In addition,90 patients with prostate cancer confirmed by pathology and underwent whole-body MRI were collected for external validation of the developed model.The manual annotation of the regions and metastases were used as the"reference standard"for the model evaluation.The evaluation indexes included dice similarity coefficient(DSC)and volumetric similarity(VS).Results In the external validation set,the classification accuracy of head,neck,chest and abdomen were 100%(90/90),98.89%(89/90),96.67%(87/90)and 94.44%(85/90),respectively.The range of DSC,VS values of the segmentation model for organs in different regions were(0.86±0.10)-(0.99±0.01),(0.89±0.10)-(0.99±0.01),respectively.The range of DSC,VS values of the segmentation model for metastases in different regions were(0.65±0.07)-(0.72±0.13),(0.74±0.04)-(0.82±0.13),respectively.Conclusion The 3D U-Net model based on deep learning may achieve the segmentation of extra-pelvic region and metastasis in advanced prostate cancer.
