1.Effect of Buxin Huoluo Capsule for Coronary Heart Disease and Its Anti-lipid-peroxidation Mechanism

[Objective] To evaluate the therapeutic effect of Buxin Huoluo Capsule (BHC) for coronary heart disease (CHD) and to explore its anti-lipid-peroxidation mechanism. [Methods] One hundred and seventy-five cases of CHD were treated with BHC and 121 cases with isosorbide dinitrate (ID) . Effects of BHC on angina pectoris, electrocardiograph, superoxide dismutase (SOD) activity and lipid peroxides (LPO) level were observed. [Results] In BHC group, the total effective rate in relieving angina pectoris was 88.0 % and that in improving electrocardiogram was 80.0 % , the difference being not significant as compared with ID group. As for the reduction of frequency of angina pectoris and the decrease of dosage of nitroglycerin, BHC were superior to ID. 2.Effects of superoxide dismutase on the periapical periodontitis in rat.

The purpose of this investigation was to elucidate the effects of superoxide dismutase (SOD) on the induced periapical periodontitis in rat, and to shed light on the possible mechanism involved in root canal therapy. 48 Wistar rats with induced periapical periodontitis were randomly divided into 3 groups; their root canals of molar teeth were prepared; and the root canals were filled with SOD, CH, and nothing, respectively. At 1, 2, 3 and 5 weeks after operation, 4 animals were killed respectively. Periapical roentgenogram and periapical pathological slices were made. The results were analyzed statistically by F test and t test with SPSS12.0 software package. After root canal therapy, mild inflammatory infiltration was observed in SOD and CH groups at 1 week. At 2 weeks, periapical periodontitis was improved. After 3 weeks, there was no inflammatory cell observed in SOD/CH group. At 3 and 5 weeks, no significant difference in periapical radiographic findings was noted between the experimental groups and normal group (P > 0.05). The periapical radiographic lesion in control group was larger than that in SOD and CH groups (P < 0.01). The results showed that SOD could control periapical inflammation and induce the proliferation of dentin and cementum. Therefore, SOD might be an ideal biocompatible material for root canal therapy.

3.Application of laparoscopic contrast enhanced ultrasound in laparoscopic surgery for hepatocellular carcinoma combined with cirrhosis

Objective:To explore the application value of laparoscopic contrast enhanced ultrasound in laparoscopic surgery for patients with hepatocellular carcinoma combined with cirrhosis.Methods:The clinical data of 71 patients with hepatocellular carcinoma combined cirrhosis from February 2018 to February 2020 in Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology were retrospectively analyzed. The patients underwent preoperative enhanced CT and multi-parameter MRI examination, followed by laparoscopic partial hepatectomy, and intraoperative laparoscopic contrast enhanced ultrasound examination. Based on histological examination and follow-up results, the diagnostic efficacy of preoperative imaging and preoperative imaging combined with intraoperative laparoscopic contrast enhanced ultrasound in patients with hepatocellular carcinoma combined with cirrhosis was compared.Results:Among the 71 patients, 69 completed laparoscopic surgery and 2 converted to open surgery. One hundred and ten HCC lesions were diagnosed by preoperative imaging examination, 105 lesions were detected by intraoperative ultrasound among them, of which 98 lesions were diagnosed as HCC by intraoperative laparoscopic contrast enhanced ultrasound. There were no statistically significant difference in sensitivity, specificity, accuracy, positive predictive value and negative predictive value between preoperative imaging and preoperative imaging combined with intraoperative laparoscopic contrast enhanced ultrasound in the diagnosis of malignant liver lesions: 94.4% (102/108) vs. 99.1% (107/108), 81.0% (34/42) vs. 66.7% (28/42), 90.6% (136/150) vs. 90.0% (135/150), 92.7% (102/110) vs. 88.4% (107/121) and 85.0% (34/40) vs. 96.6% (28/29), P>0.05. Laparoscopic contrast enhanced ultrasound revealed an additional 11 suspected malignant lesions, of which 5 lesions were histologically confirmed as HCC. Seven patients underwent surgical strategy changes. Conclusions:Laparoscopic contrast enhanced ultrasound in patients with HCC combined with cirrhosis during laparoscopic surgery can be used to detect, identify, accurately locate of the lesions and modify the surgical plan.
4.Expression and antibody preparation of recombinant truncated glycoprotein of Guertu virus

Objective:To express truncated glycoprotein (Gn, Gn1, Gn2, Gn3, Gc1 and Gc2) of Guertu virus (GTV) in Escherichia coli ( E. coli) cells, and prepare polyclonal antibodies against recombinant proteins Gn-His, Gc1-His and Gc2-His after purification. Methods:Gene fragments encoding Gn, Gn1, Gn2, Gn3, Gc1 and Gc2 of GTV DXM strain were amplified by RT-PCR, and cloned into the prokaryotic expression vector pET-32a (+ ) to construct recombinant expression plasmids. The transformed E. coli BL21(DE3) strains carrying expression plasmids were induced by IPTG to express target proteins, which were identified by SDS-PAGE. Recombinant proteins Gn-His, Gc1-His and Gc2-His were purified by nickel affinity chromatography and detected by Western blot using GTV-positive sheep serum for analysis of their antigenicity. New Zealand white rabbits were immunized with the purified recombinant proteins. The titers and specificity of serum antibodies were analyzed by ELISA. Meanwhile, eukaryotic expression vectors pcDNA3.1-Gn, pcDNA3.1-Gc1/Gc2 were constructed and transfected into mammalian Vero cells to evaluate the binding activity of rabbit polyclonal antibodies by indirect immunofluorescence method. The specific reactivity of serum antibodies to recombinant proteins was detected by Western blot. Results:Restriction enzyme analysis and DNA sequencing confirmed that the recombinant expression vectors of pET-32a-Gn, pET-32a-Gn1/Gn2/Gn3, pET-32a-Gc1/Gc2, pcDNA3.1-Gn and pcDNA3.1-Gc1/Gc2 were constructed successfully. The relative molecular mass ( Mr) of the expressed recombinant proteins Gn-His, Gn1/Gn2/Gn3-His, Gc1/Gc2-His were approximately 63.4×10 3, 37.1×10 3, 31.9×10 3, 30.8×10 3, 40×10 3 and 54.4×10 3, respectively. The recombinant proteins could be recognized by GTV-positive sheep serum. The titers of polyclonal antibodies against GTV Gn, Gc1 and Gc2 were 1∶409 600, 1∶204 800 and 1∶6 400, respectively. Indirect immunofluorescence assay and Western blot showed that the prepared rabbit polyclonal antibodies could specifically react with the proteins expressed in eukaryotic cells and the recombinant proteins. Conclusions:The recombinant GTV glycoproteins Gn-His and Gc1/Gc2-His were efficiently expressed and purified and characterized with good immunity. The prepared polyclonal antibodies had high titers and good specificity. This study provided reference for further studying the biological function and detection methods of GTV glycoproteins and research on vaccines.
