1.Effect of acetyl-L-carnitine preconditioning on PC12 cell apoptosis induced by oxygen-glucose deprivation

Objective To investigate the effect of acetyl-L-carnitine (ALC) preconditioning on the PC12 cell apoptosis induced by oxygen-glucose deprivation.Methods PC12 cells were seeded in 96-well plates and randomly divided into 5 groups ( n =6 each):control group (group C),cell injury group (group Ⅰ) and preconditioning with different concentrations of ALC groups (groups A1-3 ).In group C,the cells were incubated with DMEM liquid culture medium containing glucose 0.5 g/L for 3 h.In groups Ⅰ and A1-3 the cells were incubated with DMEM liquid culture medium containing sodium hydrosulfite (Na2S2O4) 3 mmol/L and glucose 0.5 g/L for 3 h,and in addition the cells were pre-incubated with ALC 0.2,0.4 and 0.6 mmol/L for 24 h in groups A1-3 respectively.Cell viability was evaluated by MTF assay,while the apoptosis in cells was detected using TUNEL.The activities of ATPase and SOD and MDA content were also detected.Results Oxygen-glucose deprivation significantly increased the number of apoptotic cells and the content of MDA,and decreased the cell viability and activities of SOD and ATPase in group Ⅰ compared with group C ( P < 0.05).Preconditioning with ALC significantly increased the cell viability and the activities of SOD and ATPaes,and decreased the number of apoptotic cells and the content of MDA in groups A1-3 compared with group Ⅰ ( P < 0.05).Conclusion ALC preconditioning can attenuate PC12 cell injury induced by oxygen-glucose deprivation through inhibition of apoptosis in cells.
2.Expression of serum miR-145 and miR-195 in OSCC patients and their relationship with cervical lymph node metastasis

Objective To analyze the expression of serum microRNA(miR)-145 and miR-195 in patients with oral squamous cell carcinoma(OSCC)and their relationship with cervical lymph node metastasis.Methods A total of 102 patients diagnosed with OSCC in the hospital from January 2020 to June 2022 were selected as OSCC group.Clinicopathological parameters of OSCC patients were collected and sorted.According to whether cervical lymph node metastasis occurred,the patients were divided into a cervical lymph node me-tastasis group(43 cases)and a non-cervical lymph node metastasis group(59 cases).In the same period,79 patients with precancerous lesion were selected as the precancerous lesion group,and 86 healthy subjects with-out oral disease were selected as the healthy control group.Real-time fluorescence quantitative polymerase chain reaction was used to detect the relative expression levels of serum miR-145 and miR-195.The evaluation value of relative expression levels of serum miR-145 and miR-195 in cervical lymph node metastasis in OSCC patients was examined by receiver operating characteristic curve analysis.Results The relative expression lev-els of serum miR-145 and miR-195 in OSCC group were lower than those in precancerous lesion group and healthy control group,and the difference was statistically significant(P<0.05).The relative expression levels of serum miR-145 and miR-195 in precancerous lesion group were lower than those in healthy control group,and the difference was statistically significant(P<0.05).The relative expression levels of serum miR-145 and miR-195 in OSCC patients with low tumor differentiation and TNM stage Ⅲ+Ⅳ were lower than those in OSCC patients with medium/high tumor differentiation and TNM stage Ⅰ+Ⅱ,with statistical significance(P<0.05).The relative expression levels of serum miR-145 and miR-195 in cervical lymph node metastasis group were lower than those in non-cervical lymph node metastasis group,and the difference was statistically significant(P<0.05).The relative expression levels of serum miR-145 and miR-195 were 0.840(95%CI:0.765-0.915),0.832(95%CI:0.754-0.910)and 0.898(95%CI:0.754-0.910)of cervical lymph node metastasis in OSCC patients respectively.The specificity was 79.1%,88.4%and 77.5%,and the sensitivity was 74.6%,67.8%and 87.4%,respectively.Conclusion Serum miR-145 and miR-195 are low expressed in OSCC patients,and are related to cervical lymph node metastasis,tumor differentiation,and TNM stage in OSCC patients,which can be used as an important indicator to evaluate cervical lymph node metastasis in OS-CC patients.
3.The value of contrast-enhanced CT and MR DWI in differentiating between autoimmune pancreatitis and pancreatic carcinoma

Objective To investigate the diagnostic value of contrast-enhanced CT and MR diffusion-weighted imaging (DWI) in differentiating autoimmune pancreatitis (AIP) from pancreatic carcinoma (PC).Methods Twelve patients with AIP (AIP groups) and 15 patients with PC (PC groups) were included in this study.Contrast-enhanced CT and DWI were performed in all patients.The manifestations of the pancreatic lesions,pancreatic duct and bile duct were observed.The CT attenuation values and the apparent diffusion coefficient (ADC) values of the lesions were calculated.Frequencies of the CT manifestations,and the mean CT attenuation values,and the ADC values were compared between the two groups.Results Most of the AIP showed diffuse enlargement,sausage shape,peripancreatic capsule,penetrating sign of pancreatic duct and thickness of the bile duct.The frequencies of the findings mentioned above were significantly different between AIP and PC (P<0.05).The CT attenuation values of the lesions,and the rate of lesions' CT attenuation values to livers' in 3 phases of contrast enhanced CT were significantly different between AIP and PC (P<0.05).The ADC values of AIP were significantly lower than that of PC (P<0.05).Conclusion The changes of morphology of pancreas,pancreatic and bile duct may facilitate in differentiating AIP from PC.Quantitative analysis of CT attenuation values and ADC values measured on MR images may be helpful in differentiating AIP from PC.
4.The curative effects of twinlight laser treatment on chronic periodontitis

Objective:To observe the effects of Er ∶YAG(erbium:yttrium-aluminum-garnet) laser combined with diode laser in the treatment of chronic periodontitis.Methods:30 patients with chronic periodontitis patients were treated by mechanical scaling and root planning(SRP),pure Er ∶ YAG laser(ERT),diode + Er ∶ YAG + diode laser therapy(TPT) respectively.In the treatment VAS pain score was recorded.Probing deepth (PD),clinical attachment level (CAL) and bleeding on probing (BOP) were examined,IL-1 β,TNF-α and b-FGF in GCF,Porphyromonas gingivalis(Pg) in subgingiva plaqure were measured before and after treatment.Data were analyzed by SPSS 21.0 software.Results:6 weeks,12 weeks after treatment PD,BOP and CAL in TPT group were lower than those in ERT and SRP groups(P <0.05),TGF-β and b-FGF in TPT group were higher,IL-1β and TNF-α were lower than those in ERT and SRP groups (P < 0.05).The content of Pg relative to total bacteria in TPT group were lower than that in ERT and SRP groups (P < 0.05),VAS value in twinlight laser group was significantly lower than hand instruments group(P < 0.05).Conclusion:The twinlight laser is effective in the treatment of periodontitis.
