1.Comparison of statistical methods of antimicrobial resistance based on clinically isolated bacteria and infection-related non-repetitive bacteria

Objective To compare the differences between two statistical methods for evaluating non-sensitivity of pathogenic bacteria to antimicrobial agents,and explore effect of non-consideration of clinical background on evalua-ting extent of bacterial resistance.Methods Data of Staphylococcus aureus and Acinetobacter spp .in a hospital in the first half year of 2008,2010 and 2013 were collected and conducted statistical analysis with two methods (me-thod 1 :based on all clinically isolated bacteria;method 2 :based on infection-related non-repetitive bacteria),two methods for evaluating bacterial non-sensitive rates to antimicrobial agents were compared.Results The non-sensi-tive rates of Acinetobacter spp .to various antimicrobial agents :statistical results by using method 1 were generally higher than those using method 2,absolute difference between two statistical methods was 10.46%-33.77%;the non-sensitive rates of Staphylococcus aureus to various antimicrobial agents :except compound sulfamethoxazole in 2010 and 2013(difference were 6.17% and 10.21 % respectively),penicillin G (difference was 3.86%),erythromy-cin (difference was 2.71 %),and azithromycin in 2013 (difference was 2.43%),statistical results by using method 1 were generally higher than those using method 2,absolute difference between two statistical methods was 0-18.04%.Conclusion There are deviation in the non-sensitive rates of bacterial strains to antimicrobial agents by using two different statistical methods,deviation is larger in Acinetobacter spp ..The resistance level might be incorrectly higher when evaluating the resistance status without considering clinical background of bacteria.
2.Prevalence and factors associated with frailty among institutional older adults

Objective To investigate the prevalence and influencing factors associated with frailty among institutional older adults and to provide evidence for intervention study.Methods A total of 237 older adults living in four welfare homes from Chengdu in Sichuan Province were recruited by convenience sampling.A cross-sectional survey was conducted,and frailty was assessed by the frailty phenotype;general information was collected by selfmade general information questionnaire;functional capacity was assessed by Barthel questionnaire;depressive status was assessed by GDS-15;cognitive function was assessed by clock drawing test and nutritional status was assessed by short form mini nutritional scale questionnaire (MNA-SF).Results The reported rate of frailty was 55.69% among institutional older adults,and 44.31% reported no frailty;grip weakness (207,87.3%),slow walking speed (172,72.6%) and low physical activity(131,55.3%) were the main frailty problems.Univariate analysis showed that the elderly'frailty conditions were different in the pre-retirement occupations,smoking,alcohol consumption,physical exercise,chronic diseases,medication,acute events (last year),self-reported health,using assisted walking devices,ability of daily life,depression,cognitive ability and nutritional status,and the differences were statistically significant(P<0.05).Conclusion The status of frailty is not optimistic among institutional older adults and many factors are associated with frailty.Healthcare providers should pay more attention to the frailty issue and take timely intervention strategies to prevent or delay the frailty.
3.Preparation of Legumain enzyme and mitochondrial double-stage targeted harmine liposome and evaluation of its in vitro characterization

OBJECTIVE To prep are Legumain enzyme and mitochondrial double-stage targeted harmine (HM) liposome (KA@HM-LPS)and preliminary evaluate its pharmaceutical properties ,in vitro antitumor effect and biocompatibility. METHODS Firstly,the preparation and homogenization methods of KA@HM-LPS was screened ,and prepared liposomes were characterized. Secondly,the serum stability ,in vitro release rate ,hemolysis percentage of KA@HM-LPS and cell survival rate under KA@BLPS were determines respectively. Finally ,the cell surivival rate ,mitochondrial targeting and inhibitory effects on cell migration and invasion of KA@HM-LPS were determined. RESULTS KA@HM-LPS was prepared by the thin-film dispersion method ,with encapsulation efficiency of (90.50 ± 0.62)% . The extrusion moulding method was selected as homogenization method of KA@HM-LPS. The particle size ,polydispersity index ,and Zeta potential of KA@HM-LPS were (211.40±11.67)nm,0.316± 0.014 and(-14.20±0.49)mV,respectively. In 37 ℃,10% FBS,the particle size of KA@HM-LPS kept stable after 12 h. In vitro release curve of KA@HM-LPS in 20% plasma conformed to Weibull distribution and had the property of sustained release. When HM concentration was 160 μg/mL,the hemolysis percentage of KA@HM-LPS was (4.23±0.19)%,which was much lower than that of free HM ,with safety. When the mass concentration of KA@BLPS reaches 400 μg/mL,the survival rate of LO 2 cells was (94.40 ± 6.12)% ,and the biocompatibility was good. Cell test results in vitro showed that ,inhibitory effect of KA@HM-LPS on liver cancer cells with overexpression of Legumain enzyme (LGMN -SK-Hep-1) was significantly higher than that of normal liver cancer cells SK-Hep- 1; compared with SK-Hep-1,LGMN +-SK-Hep-1 cells had a higher uptake efficiency of the liposome ;KA@HM-LPS could significantly inhibit the migration and invasion of LGMN +- SK-Hep-1 cells. CONCLUSIONS KA@HM-LPS is prepared successfully ,which can effectively inhibit the migration and invasion of liver cancer cells with Legumain enzyme overexpression ,and improve the blood compatibility of HM.
