Chinese Journal of Tissue Engineering Research 2005;9(3):250-251
OBJECTIVE:To retrospect the experiments and clinical research on tropomyosin(TM) and make sure the 4 genes of TM in mammals and the relationship between its gene mutation and weakness of skeletal muscle as well as hypertrophic cardiomyopathy in order to provide support for preventative measures and rehabilitation interventions. DATA SOURCES:Conducting computerized search on experiments and clinical reports from January 1987 to Augsut 2004 regarding TM to Medline and Wanfang Database. The key searching words of " tropomyosin, gene, mutation" both in English and Chinese were used with the restricted language of English and Chinese respectively. DATA SELECTION:The fiirst trial was conducted to the found literatures while the articles with " single case, experience" in title and non-research ones were excluded. Then full text of the rest reports was searched.Randomized control clinical trial(RCT) was used as the selective standard. DATA EXTRACTION:There were 30 articles found regarding the randomized and non-randomized trials on the research and advance of molecular biology of TM with 21 of them met the selective standard and 9 articles were excluded. DATA SYNTHESIS:The study showed that TM was an important regulatory protein for muscle contraction, which existed widely in various kinds of eukaryotes in the way of isomers. Four TM genes of mammals had been confirmed by which 20 TM isomers could be expressed.Different TM gene could induce different diseases respectively. CONCLUSION:At present, research of TM is not just limited to the process of muscle contraction, study on its effects on the conversion of vascular smooth muscle cell has been attached much attention. The application of gene engineering will essentially diagnose and cure the disease therefore medical behaviors and quality of life will be consequently changed.

This study was aimed to explore the clinical effect of San-Jiao (SJ) acupuncture combined with Chinese medicine and Cisapride tablets in the treatment of senile constipation. A total of 118 patients were selected according to the inclusion criteria. According to the visiting sequence, patients were randomly divided into the acupuncture with Chinese medicine group of 60 cases and drug group of 58 cases. In the acupuncture with Chinese medicine group, SJ acupuncture was combined with Huang-Di (HD) powder in the treatment. In the drug group, Cisapride tablets were applied. Both groups were treated for 20 times. The scale was used in the comprehensive evaluation of integral effect evaluation of main symptoms, psychological evaluation before and after the treatment. The results showed that there was significant difference on the integral before and after treatment in both groups (P< 0.01). The posttreatment integral showed that the acupuncture with Chinese medicine group was better than the drug group (P<0.01). There was no significant difference on total effective rate between two groups. However, comparison on good improvement rate showed that the acupuncture with Chinese medicine group was better than the drug group (χ2 =28.25, P < 0.01). It was concluded that the acupuncture with Chinese medicine group had better effect on the im-provement of clinical symptom integral, psychological integral, and comprehensive effect evaluation. SJ acupuncture combined with HD powder was a safe treatment method with good effect.

Object To study the effects of WUZI SIWU GUASHITANG (WT) against multi glycosides of Tripterygium wilfordii Hook. f. (GTW) induced genital system toxicity in male rats and its mechanism. Methods Male rats in treatment groups were treated respectively with a 80 days oral administration of large , middle and small doses of WT plus a fixed dose of GTW. The rats as control were treated with GTW alone or saline. Thereafter, the changes in testis organ index, the number of spermatozoa, the rate of movable spermatozoa and the structure of spermatogenic cell epithlium were observed and the serum testosterone level was determined by IRMA. Then the comparison was made between treatment and control groups as to above variables. Results As compared with GTW group, large or middle dose of WT significantly increased the weight of testis, the number of spermatozoa and the rate of movable spermatozoa. Large dose of WT could relieve the damage of GTW to the spermatogenic cell epithelium completely and keep the serum testosterone at normal level. Conclusion The combined use of GTW with WT is very useful in relief of GTW induced adverse reactions of genital system in male rats. The mechanism by which WT exerts the counteractive effects is related to protecting the spermatogenic cell epithelium from being damaged by GTW, and keeping the serum testosterone at normal level.

AIM: To study the effect of genistein on activation and proliferation of T cells, and explore the molecular mechanism of genistein. METHODS: Fluorescence conjugated monoclonal antibodies and flow cytometry were used to detect the express of CD69 and CD25 by activated T cells in vitro in response to Concanavalin (ConA )and Phorbol 12,13-dibutyrate(PDB) or T cell proliferation stained by CFSE in response to PDB / Ionomycin or ConA. RESULTS: Genistein inhibited the expression of CD69 and CD25 in activated T cells in response to Con A in a concentration-dependent manner and in response to PDB in a high concentration. Genistein inhibited proliferation of T cells in both groups in a concentration-dependent manner. CONCLUSION: Genistein inhibited activation and proliferation of T cells in vitro in response to polyclonal stimulus, and it may hold potential as a new immunosuppressant.

Aim To investigate the neuroprotective effects of osthole(Ost)on the primary cultured cortical neurons transfected with APP595/596 gene and its underlying mechanism. Methods Neonatal mouse cortical neurons were transfected with APP595/596 gene to establish AD cell models for the further study. Then, the cell viability was detected by CCK-8 assay, and the leakage of lactate dehydrogenase(LDH)was assayed by LDH kit to evaluate the injury degree.Transferase-mediated nick end labeling(TUNEL)was used to evaluate the cell apoptosis. The expression of β-amyloid peptide(Aβ)and β-site APP cleaving enzyme 1(BACE1)was measured by immunofluorescence, while the miRNA-107 was measured by RT-PCR.Results Compared to model group,Ost could significantly improve the neurons viability, decrease the LDH release and prevent the apoptosis.Ost also inhibited the expression of Aβ and BACE1 at protein level, while enhanced the expression of miRNA-107 at gene level. Conclusion Ost plays a neuroprotective role in neurons transfected with APP595/596 gene in part through up-regulating miRNA-107.

Objective To investigate the effects of osthole on neural stem cells ( NSCs) differentiation and explore the potential mechanism. Methods Brain-derived NSCs from newborn mice were isolated and cultured in vitro and determined by immunofluorescence. The P5 generations of NSCs were placed in culture solution with osthole at concentrations of (0,10,50, 100 μmol·L-1 ) . The neuron, astrocyte and oligodendroglia cell differentiation were determined by immunofluorescence. The mRNA expression of Notch 1 and its target genes Mash 1 and Neurogenin 2 were assessed by RT-PCR. Results The neurosphere displayed Nestin and Sox 2-postive by immunofluorescence, suggesting that the cultured cells were NSCs. Osthole promoted NSCs differentiating into more neuron(P<0. 01) and oligodendrocyte(P<0. 05), but not astrocyte. Meanwhile, osthole significantly reduced the mRNA expression of Notch 1(P<0. 01) and increased Ngn 2(P<0. 01)at the dose of 100 μmol·L-1. Conclusion Osthole enhances NSCs differentiating into more neuron and oligodendrocyte via probablly inhibiting Notch signal pathway.
