1.Expressions of MMP-2,MMP-3,MMP-9 and CD147 in human ovarian cancer and their clinicopathological significance

Objective:To investigate the expressions of MMP-2,MMP-3,MMP-9 and CD147 in human ovarian cancer and their clinicopathological significance.Methods:Immunohisto chemical S-P method was performed to detect the expressions of MMP-2,MMP-3,MMP-9,CD147 in 40 malignant and 9 benign ovarian tumors.Result:The expression rates of MMP-2,MMP-3,MMP-9 and CD147 in ovarian cancer were 92.5%,100%,100% and 95%,respectively.The expressions of MMP-2,MMP-3,MMP-9 and CD147 in ovarian cancer were related with clinical stage and pathological garade(P0.05).There were positive correlations between the expressions of CD147 and MMP-2,MMP-3,MMP-9(r=0.802,0.786,0.914)in ovarian cancer.Conclusion:Expressions of CD147,MMP-2,MMP-3 and MMP-9 have relevance to clinical stages and pathological grades in ovarian cancer,and overexpression of CD147 may induce the secretion of MMP-2,MMP-3,MMP-9.

3.NQO1 C609T gene polymorphism associated with an increased risk of post operative cognitive dysfunction

Objective To investigate the association between the C609T polymorphism of NAD (P)H:quinoneoxidoreductase (NQO1) gene and post operative cognitive dysfunction (POCD).Methods 90 ASA Ⅰ-Ⅱ patients of 59 to 78 years old, undergoing elective hip replacement with epidural anesthesia were enrolled.All patients were given a battery of 5 neuropsychological tests before operation and seven days after operation.Patients were divided into POCD group and control group according to test results (45 patients in each group).The single nucleotide polymorphism C609T of NQO1 gene was detected using real-time PCR by Taqman probes and subjected to odd ratio assessment.Results 5 samples in control group couldn' t be used in the real-time PCR analysis due to quality control.The frequency of C/C genotype in POCD control was lower than that of control group ( 30.0% vs 11.1% ) with statistical significance ( OR = 0.292,95 % CI 0.092 ~ 0.92 1, P < 0.05 ).The C/T +T/T genotype frequency was significantly higher in group POCD than in the control group(88.8% vs 70% ).Patients presented with C/T + T/T genotype showed an evidently increased risk of POCD ( OR =3.42,95% CI 1.08 ~ 10.82,P < 0.05 ).The frequency of C allele of NQO1 gene in group control was 56.2%, as compared with 40% in group POCD with significance ( OR = 0.519,95% CI 0.282 ~ 0.955, P < 0.05 ).The frequency of T allele of NQOI gene in control group was 43.7% ,as compared with 60.0% in POCD group( OR = 1.93,95% CI 1.047 ~3.552,P<O.05).Conclusion The NQO1 gene single nucleotide polymorphism C609T is evidently associated with the increased risk of POCD.
4.In vitro susceptibility of Geotrichum silvicola to five antifungal agents and electron microscopic observation of terbinafine-treated Geotrichum silvicola

Objective To evaluate in vitro antifungal activities of five clinically common antifungal agents against Geotrichum silvicola,and to observe morphological changes of Geotrichum silvicola after treatment with terbinafine.Methods Five antifungal agents commonly used in clinic,including terbinafine (TER),amphotericin B (AmB),flucytosine (FC),fluconazol (FCZ) and itraconazole (ICZ),were tested in this study.According to the National Committee for Clinical Laboratory Standards (NCCLS) M27-A2 document and relevant literatures,a susceptibility test was performed to determine the minimal inhibitory concentration (MIC) values of the five antifungal agents against G eotrichum silvicola.Scanning electron microscopy (SEM) was used to observe morphological changes of Geotrichum silvicola treated with different concentrations of TER.Results The MIC values against Geotrichum silvicola were as follows:TER,0.01 μg/ml;AmB,0.4 μg/ml;FC,2 μg/ml;FCZ,2.69 μg/ml;ICZ,0.25 μg/ml.Geotrichum silvicola was sensitive to TER,AmB,FC and FCZ,and sensitive to ICZ in a dose-dependent manner.SEM revealed that after TER treatment,the surface of Geotrichum silvicola became rough and shrinking with irregular defects and holes,and even gave a fragment-like appearance.Conclusions Among the five tested antifungal agents,TER shows the strongest activity against Geotrichum silvicola.Within a certain range,the higher the concentration of TER,the more serious the damage to Geotrichum silvicola.
5.Influence of all-trans retinoic acid on the methylation of the RARa gene in ovarian cancer cell line COC2

Objective: All-trans retinoic acid(ATRA) is a classic drug that can induce tumor differentiation,and its influence on the aberrant methylation of cancer cells has been studied insufficiently.The objective of this study was to observe the influence of ATRA on the methylation of the RARa gene promoter in ovarian cancer cell line COC2 and its relationship with the RARa expression.Methods: The ovarian cancer cell line COC2 was treated with different concentrations of ATRA for different times.Methylation specific PCR(MSP) and bisulfate sequencing methods were used to detect the changes in the methylation of the RARa promoter after ATRA treatment.RT-PCR was employed to observe the changes in the expression level of RARa mRNA.Results: Aberrant methylation of the RARa gene promoter was found in the ovarian cancer cell line COC2.ATRA treatment decreased the number of RARa promoter methylation sites in COC2 within a certain scope in a concentrationand time-dependent manner,and increased the expression of RARa mRNA.Conclusion: Aberrantly high methylation of the RARa promoter exists in the ovarian cancer cell line COC2;ATRA can partially reverse the methylation and increase the RARa mRNA expression.
6.Regulation of MHC class Ⅱ transactivator on MHC class Ⅱ expression of pancreatic cancer cell line CaPanC-2 and PanC-02 in vitro

Objective To investigate the effect of MHC class Ⅱ transactivator( MHC Ⅱ TA ) on MHC Ⅱ expression of human and rat pancreatic cancer cell line CaPanC-2 and PanC-02. Methods The recombinant adenovirus (Ad-C Ⅱ TA) was transfected into the CaPanC-2 cell and the PanC-02 cell, then it was cultured for 24, 48, 72 h. The C Ⅱ TA mRNA expressions were assayed by Real Time PCR and CⅡ TA protein expressions were determined by Western blotting. The percentage of positive expression cells of MHC Ⅱ were measured by flow cytometry. Results The C Ⅱ TA mRNA expressions of CaPanC-2 cell at 24, 48, 72 h were 16769 ± 6455, 261568 ±348850 and 834816 ±97783 folds higher than that of control group (P <0.05). The CⅡTA mRNA expressions of PanC-02 was 548546 ± 87755, 1242684 ± 624888 and 1401647 ± 726145 folds higher than that of control group ( P <0.05). CⅡ TA protein was not expressed in CaPanC-2 cell and PanC-02 cell. After transfection for 48 h, the CⅡ TA protein expressions of CaPanC-2 and PanC-02 were 0.746 ± 0.499 and 0.631 ±0.244. The percentage of positive expression cells of MHC Ⅱ in CaPanC-2 cells after 24, 48, 72 h were (8.1±0.3)%, (18.9±0.3)%, (78.5±0.90%, which were significantly higher than that in control group [ ( 7.0 ± 0.1)% , P < 0.01 . The percentage of positive expression cells of MHC Ⅱ in PanC - 0 2 were (5.1 ±0.2)% , (37.3 ±2.0)% , (68.8 ±2.2)%, which were significantly higher than that in control group [ (2.2 ±0.2)% , P <0.01). Conclusions Transfection of Ad-C Ⅱ TA could increase the expression of CⅡTA and MHC Ⅱ molecules of pancreatic tumor cells in vitro.
7.EFFECT OF FORTIFIED FOOD ON THE NUTRITIONAL STATUS OF PRESCHOOL CHILDREN

Three kinds of fortified biscuits and bread were given as breakfast for 5 months to 106 preschool children 4-6 years old lodged in a kindergarten, with a control group taking common biscuits and bread without fortification. 6 g protein and 160 kcal were supplied by 50 g of fortifed foods in which 0.25 g lysine, 0.25 g lysine and 0.8 mg riboflavin, and cod liver oil and calcium in addition to lysine and riboflavin were fortified respectively. The results showed that a diet with intake of 2.1 g protein and 71 kcal/kg body weight fortified on the average by 228 mg lysine or 228 mg lysine and 1.1 mg riboflavin increased the monthly increment of body weights, heights, and urinary excretions of creatinine per day. There was also a tendency of improvement, in nitrogen retention. The urinary excretion of riboflavin in load test was increased in the group taking the second kind of fortified foods.

Objective: To observe the influence of all-trans retinoic acid(ATRA) on the expressions of E-cadherin,heparanase and VEGF in epithelial ovarian carcinoma cell line COC2,and to investigate the anti-metastatic potential and possible action mechanism of ATRA.Methods: We used flow cytometry to examine the expressions of E-cadherin,heparanase and VEGF proteins in the epithelial ovarian carcinoma cell line COC2 treated with different concentrations of ATRA.Results: ATRA significantly increased the expression of E-cadherin and decrease that of VEGF and hepareanse in a dose-dependent manner.Conclusion: ATRA can inhibit cell proliferation,improve cell-cell adhesion and downregulate the expressions of VEGF and heparanse proteins,suggestive of an anti-angiogenic and anti-metastatic potential.
9.Effect of oxymatrine on mouse allergic contact dermatitis induced by DNFB and lymphocyte proliferation stimulated by Con A

AIM: To explore the inhibitory effect of oxymatrine (OMT) on the allergic contact dermatitis (ACD) stimulated by dinitrofluorobenzene (DNFB) and its effects on the proliferation of the lymphocytes. METHODS: ① An ACD mouse model was established by stimulation with DNFB, and then the mice were injected intraperitoneally with different dosages of OMT, PBS and hydrocortisone (HCT) respectively, the swelling degree of their auricles was examined. ② Carboxyfluorescein diacetate, succinimidyl ester (CFDA-SE) dye and flow cytometer were used to examine the fluorescence intensity changes of lymphocytes stimulated by polyclonal stimulator ConA and OMT. RESULTS: ① compared with PBS group, OMT possessed the strong inhibitory effect on the ACD caused by DNFB in a dose-dependent manner, and its inhibitory effect was equivalent to the HCT of the same dosage with fewer side effects. ② In vitro experiments proved that OMT (500, 125 and 31 mg/L) had the ability to restrain the proliferation of lymphocytes of mouse. CONCLUSION: OMT possesses an inhibitory effect on the ACD induced by DNFB, and OMT is a kind of immunosuppressor.
10.Effects of One Finger Massage on Nerve Morphology and Function of Sciatic Nerve Injury Rats

Objective To study the effects of one finger massage on sciatic nerve injury rats. Methods The sciatic nerves were exposed, and the sciatic nerve was held by micro needles to make the sciatic nerve injury model. SD rats were randomly divided into sham-operation group, model group and massage group. In the sham-operation group, the sciatic nerves were exposed but not held. 7 d after the establishment of modeling, rats in massage group received one finger massage for 30 d. After 30 d, SFI and BBB of sciatic nerves were detected. HE staining, transmission electron microscope and immunochemistry assay were used to measure the changes of sciatic nerves. Results Compared with model group, massage group could speed up the recovery of SFI and increase BBB, promote the recovery of sciatic nerve morphology, increased protein level of S-100β, and enhance ultrastructure of newborn nerve growth and recovery. Conclusion One finger massage can effectively promote neurological and functional recovery after sciatic nerve injury in rats.
