China Pharmacy 2015;(18):2578-2580
OBJECTIVE:To provide reference for improving the practical application of fluorescence quantitative reverse tran-scription-polymerase chain reaction (RT-PCR) for the live-attenuated mumps vaccine manufacturing. METHODS:A regression equation of standard titer-ct value was established;micro cytopathic effect(micro-CPE)and fluorescence quantitative RT-PCR were respectively adopted for the titer of 10 batches of single harvest liquid of live-attenuated mumps vaccine and end products, and the results were compared. RESULTS:According to the t-test analysis, the P were respectively 0.743 and 0.868,and the average val-ues were in the range of (-0.174,0.094) and (-0.113,0.153),with no significant differences. They were similar. CONCLU-SIONS:Fluorescence quantitative RT-PCR can be used for the titer detection of single harvest liquid and end products in the pro-cess of live-attenuated mumps vaccine manufacturing. And it is expected to further upgrade to Chinese Pharmacopoeia method.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2014;28(9):627-630
OBJECTIVE:
To investigate the bacteria distribution, drug bacterial sensitivity characteristics of the rural chronic rhinosinusitis (CRS). And to explore the effect of antibiotic on pathogenic bacteria culture.
METHOD:
Choose nasal sinus secretions from 115 CRS patients living in rural areas. Aerobic bacteria culture, anaerobic bacteria culture and drug sensitive test were procedured for each sample. At the same time the use of antibiotics nearly 2 months and nearly 2 weeks were collected.
RESULT:
Among one hundred and fifteen specimens, 17 kinds of germs were detected in 37 cases, the positive rate of aerobic bacteria was 32.17%. Staphylococcus aureus and epidermis staphylococcus aureus the most common type of aerobe in CRS patients at rural areas. There was negative result in the anaerobic bacteria culture of 17 maxillary sinus specimen. The cases of using antibiotics nearly 2 months was up to 90, accounting for 78.26%. Nearly 2 weeks, 73 cases, accounting for 63.48%. The chi-square analysis showed high bacterial culture rate, in chronic rhinosinusitis with nasal polyps (CRSwNP group), which revealed correlation between bacterial infection factors and nasal polyps formation. For CRS patients with positive result of bacterial culture, they were sensitive to ofloxacin, cefotaxime, organism, ciprofloxacin, magnitude cephalosporin, and were drug fast to penicillin G, ampicillin, erythromycin.
CONCLUSION
No specific differences was found in the bacteria distribution of rural CRS. antibiotics abusage in rural CRS patients and the anaerobic bacteria culture techniques is the main factor resulting in low culture rate. Rational use of antimicrobial agents should be established on the basis of the bacterial culture and drug sensitive test.

Cancer Research and Clinic 2011;23(12):807-809,816
ObjectiveTo investigate the expression and the clinical significance of Clusterin protein in invasive ductal breast cancer.MethodsThe expression of Clusterin protein in 70 cases of invasive ductal breast cancer,20 cases of breast hyperplasia, and 10 cases of adjacent normal breast tissue was examined with SP immunohistochemistry (IHC),and their correlation with some clinicopathological parameters were studied.ResultsClusterin expression was detected in 71.4%of breast cancer tissues, which was significantly higher than that in benign lesions (20.0 %) and adjacent normal tissues (0) (x2 =17.143,P < 0.05; x2 =19.048,P < 0.05).Clusterin expression in breast cancer was relevant to the histological grade (x2 =5.233,P < 0.05),lymph node metastasis (x2 =4.677,P < 0.05),and the progression in the clinical stage (x2 =4.667,P < 0.05),but irrelevant to the age (x2 =0.024,P > 0.05) and tumor size (x2 =0.406,P > 0.05).Clusterin expression was negatively correlated to the expression of ER and PR in breast cancer tissues (r =-0.362,P < 0.05 and r =-0.290,P < 0.05,respectively),but uncorrelated with C-erbB-2 expression(r =0.129,P > 0.05).Conclusion Clusterin overexpressed in breast cancer tissues suggests an important role of clusterin in invasive ductal breast cancer carcinogenesis and development through inhibition of cell apoptosis. It may become new clinical diagnosis marker for breast cancer and novel target for breast cancer therapy.

Basic & Clinical Medicine 2006;0(10):-
Caveolin-1(Cav-1) is a scaffold protein of caveolae that acts as a tumor modulator by interacting with cell adhesion molecules and signaling receptors.Evidence from a variety of studies indicate that caveolin-1 exhibits the heterogeneity of Cav-1 expression in different tumors.Current research has clearly established a role for Cav-1 as a a novel prognostic marker,Future studies will undoubtedly offer novel exciting opportunities to develop anti-cancer therapies.

Objective:To explore the influence on inflammation cytokines for anti-IL-1βand TNF-αIgY intranasal treatment in guinea pigs with allergic rhinitis. Methods:The allergic rhinitis model in guinea pigs was established using ovalbumin(OVA).Hartley guinea pigs were randomly divided into the control group(group C,n=17),the allergic rhinitis model group(group M,n=27),the 0.1%anti-IL-1βand TNF-αIgY treatment group(group Z1,n=21)and the fluticasone propionate treatment group(group Z2,n=21). At 2 h,4 h and 8 h after the last treatment, blood was got by heart puncture,as well as nose was lavaged using 0.9% saline and the nasal lavage fluid( NLF) was collected. The level of cytokines was examined using ELISA kits. Results: In the peripheral blood, the levels of IL-1β,IL-5,IL-9,IL-13,IL-18,IL-33 and TGF-β1 from 2 h to 8 h;TNF-αand OVA-specific IgE from 2 h to 4 h;and IL-22 from 4 h to 8 h were significantly decreased in the 0.1%anti-IL-1βand TNF-αIgY treatment group compared with the allergic rhinitis model group(P<0.05).In the NLF,the levels of IL-1β,IL-5,IL-9,IL-13,IL-22,IL-33,TNF-α,TGF-β1 and OVA-specific IgE from 2 h to 8 h;and IL-18 at 2 h were significantly decreased in the 0.1% anti-IL-1βand TNF-αIgY treatment group compared with the allergic rhinitis model group ( P<0.05 ) .Conclusion: Anti-IL-1βand TNF-αIgY intranasal treatment can significantly reduce inflammation cytokine levels in allergic rhinitis guinea pigs.

Progress in Biochemistry and Biophysics 2001;28(1):81-85
With redox-sensitive fluorescene probes DCFH-DA and DHR123, the formation of cytosolic and intramitochondrial reactive oxygen species (ROS) inside immature rat cerebellar granule cells during the apoptosis induced by nitric oxide donor S-nitroso-N-acetyl-pennicillamine (SNAP) was monitored by laser confocal scanning microscopy. The cytosolic and intramitochondrial ROS increase significantly after 0.5 mmol/L SNAP treatment for 1 h. Pre-treatment with the nitric oxide scavenger hemoglobin can effectively inhibit the formation of cytosolic and intrarnitochondrial ROS and protect neurons from apoptosis. Adding glutathione can also protect neurons from apoptosis, and the cytotoxity of nitric oxide increases significantly while the synthesis of glutathione is inhibited. The results indicated that ROS might be involved in NO-induced apoptosis in neural cells and glutathione might be the endogenesis antioxidant to protect neurons from oxidative injury.
