1.Overview of Fuwai hospital research platforms and their operating results

Fuwai Hospital of Chinese Academy of Medical Sciences constructed three research plat-forms by integration of resources in 2004 which included central laboratory, pathophysioiogy center and ani-mal center. It mainly aimed to provide the use of equipment, technical training, guidance and experimental animals, etc. For hospital researchers and graduate students in this paper, we reviewed the three research platforms and analyzed the positive effect of research platforms on personnel training and promoting the re-search and development of the hospital.
2.THE EXPERIMENTAL STUDY OF A RAT C6 BRAIN TUMOR MODEL

Rat C6 glioma cells were cultured in vitro,and 10?L C6 cell suspension containing 10g/L agarose and 1?10 6 C6 cells was injected into the right caudate nucleus of rat brains to establish a rat brain tumor model by directional implantation method.General observation and MRI scan were conducted after implantation.Trans aorta paraformaldehyde perfusion for 5 group rats having received implantation was carried out at 10,15,20,25,days and before natural death.The tumor containing samples were prepared histologically by hematoxylin and eosin stains.MRI scan showed that intracerebral growth occurred in 50 implanted rats,with a distant metastasis of 4%.The results indicated that tumor globular intracerebral growth occurred and extracranial growth extension was scarce after implantation.The model was very stable and the implanted rat survival duration is easily determined. The experiment lays the foundation for chemotherapy,radiotherapy and gene therapy of the glioma.
3.Research progress on the relationship between sleep disorders and Alzheimer's disease

Sleep disorders are common diseases with various dys-function during sleep-wake process, including difficulty falling or staying asleep, falling asleep at inappropriate times, excessive total sleep time, or abnormal behaviors associated with sleep. Sleep disorders can lead to the deposition of amyloid beta protein ( Aβ) by affecting the normal metabolism of amyloid beta protein in the brain. Patients with Alzheimer’s disease ( AD) often suffer from sleep disorders, and its pathology always results in more se-vere sleep disorders, which leads to a risk of cognitive impair-ment and hypofunction. Sleep disorders could interact closely with AD, forming a positive feedback loop, which causes serious damage to the body health. This review summarized the current research about sleep disorder in the onset of AD and the current status of medication.
4.Analysis of 7 578 malignant tumor cases in Kashga, Xinjiang

Objective:To investigate the incidence and distribution of malignant tumor in Uygur People and provide a theoretical basis for epidemiological surveys. Methods:A retrospective study was conducted from November 2007 to October 2012 on inpatients with malignant tumors at The First People′s Hospital of Kashgar Prefecture. The top 10 kinds of malignant tumor were found in Uygur People and Han People. Results:A total of 7578 patients were registered, including 6840 (about 90.26%) Uygur People, 628 (nearly 8.29%) Han People, and 110 (about 1.45%) from other minority groups. The top 10 kinds of malignant tumor for Uygur People were gastric cancer, esophageal cancer, leukemia, cervical cancer, malignant lymphoma, primary hepatic carcinoma, breast cancer, head and neck cancer, lung cancer, and colorectal cancers. Meanwhile, the order for Han People were lung cancer, colorectal cancer, breast, gastric, primary hepatic, head and neck, leukemia, cervical, esophageal cancer, and pancreatic cancer. Conclusion:A different epidemic character of malignant tumor existed between Uygur People and Han People in Kashga, Xinjiang.
5.Clinical application of continuity care model in stroke patients

ObjectiveTo probe into the clinical application effect of continuity care model in stroke patients. Methods58 patients with stroke in our hospital during June 2008 to June 2011 were selected.According to block randomization method,the selected patients were divided into the study group and the control group with 29 patients in each group.The control group received routine care interventions,while patients in the study group was given routine care interventions plus continuity care model.Activities of daily living,compliance behavior,quality of life,nursing satisfaction degree and other related indicators of patients in two groups were comparatively analyzed. ResultsCompared with the control group,the activities of daily living of the study group at discharge did not show significant differences,while 1 month,3 months,6 months after discharge,the activities of daily living was significantly increased.Regular medication,exercise time,timely return visit,proper diet and other indicators of compliance behaviors were significantly improved.The quality of life score did not show significant differences at discharge,while 3 months after discharge,the quality of life score were significantly higher in the study group than that of the control group.The satisfaction degree of patients was significantly higher in the study group than that of the control group. ConclusionsApplication of continuity care model in stroke patients can improve activities of daily living,compliance behavior,quality of life,satisfaction degree with nurses.It's worthy of further clinical application and promotion.
6.Study on the isoniazid induced cellular damage and the expressions of Fas/Fas ligand of HepG2

Objective To establish a model of isoniazid induced necrosis and apoptosis in HepG2 cell and to observe the expressions of Fas/Fas ligand (FasL) in this model.Methods HepG2 cells were treated with different dosages of isoniazid (0,1,2,4,6 and 8 mg/mL) or blank control for 24 hours.Flow cytometer was used to observe the cellular morphology of the HepG2 cell.Annexin V/propidium iodide double staining and flow cytometry were employed to detect the necrosis and apoptosis of HepG2 cells.The expressions of Fas/FasL on the cells were also determined by flow cytometry.The data were analyzed by one-way ANOVA.The comparisons between the drug groups and the control group were performed by using Dunnett t test. Results The higher the dose of isoniazid (4,6,8 mg/mL) was,the more necrosis and apoptosis were observed.In the 4,6 and 8mg/mL isoniazid arms,the total mortality rates were all higher than the control group [(32.1 ±7.5)%,(34.9±8.1)%,(38.2±9.4)% vs (7.2±1.5)% respectively](t=4.62,5.14 and 5.75,respectively; all P<0.01 ).The expression levels of Fas increased along with the dose of isoniazid increasing [(8.7±2.2)%,(11.5±2.8)%,(12.3±3.0)% and (10.6±2.9)% in isoniazid 2,4,6and 8 mg/mL arms,respectively],which were all higher than that in control arm [(3.1 ±0.8) %](t=2.97,P<0.05; t=4.46,P<0.01; t=4.88,P<0.01; t=3.98,P<0.05).Furthermore,the expressions of FasL increased as well when the dose of isoniazid increased.The expression levels of FasL were (16.2±3.5)%,(21.7±4.8)% and (18.7±4.9)%,respectively in isoniazid 4,6 and 8 mg/mL arms,which were all higher than that in the control group [(7.4±1.4)%](t=3.11,P<0.01; t=5.06,P<0.01; t=3.99,P<0.05).HepG2 cell necrosis increased with isoniazid of 8 mg/mL.However,the increase of apoptosis was not observed.Conclusion Isoniazid can induce HepG2 cell necrosis and apoptosis,and the apoptosis may be related with the increased expressions of the Fas/FasL on the cells.
7.Study on the Preventive Effect and Mechanism of Butylphthalide on Myocardial Ischemia-reperfusion Inju-ry in Rats

OBJECTIVE:To study the preventive effect and mechanism of butylphthalide on myocardial ischemia-reperfusion injury in rats. METHODS:SD rats were randomly divided into sham operation group,model group,butylphthalide low-dose,me-dium-dose,high-dose groups (50,75,100 mg/lg),10 in each group. All rats were intragastrically administrated related drugs once a day,sham operation group and model group were intragastrically administrated equal amount of normal saline,for 3 d. Af-ter last administration,except for sham operation group,120 min of repertusion was conducted. Ratio of myocardial infarction ar-ea, apoptosis rate of myocardial cells, expressions of apoptosis-related protein [proapoptotic proteins (Caspase-3, Fas, Cas-pase-9),apoptotic protein (Bcl-2)],left ventricular ejection fraction (LVEF),and content of vascular endothelial growth factor (VEGF)in serum of rats in other groups were detected. RESULTS:Compared with sham operation group,myocardial infarction ar-ea and apoptosis rate of myocardial cells in model group were obviously increased;expressions of Caspase-3,Fas,Caspase-9 pro-tein in myocardial cells were obviously enhanced,Bcl-2 protein expression was obviously weakened;LVEF and serum VEGF con-tent were obviously reduced (P<0.01). Compared with model group,except that LVEF in butylphthalide low-dose group didn't changed obviously (P>0.05),above indexes in other administration groups improved obviously (P<0.05). Compared with bu-tylphthalide low-dose group,myocardial infarction area and apoptosis rate of myocardial cells in butylphthalide high-dose group were obviously decreased(P<0.05);expressions of Caspase-3,Fas,Caspase-9 protein in myocardial cells in butylphthalide medi-um-dose,high-dose groups were obviously weakened,and Bcl-2 protein expression was obviously enhanced(P<0.05). CONCLU-SIONS:Butylphthalide can effectively reduce myocardial ischemia-reperfusion injury in rats,protect cardiac functions,and its mechanism may be related with up-regulating VEGF.

9.Pharmacodynamic Research of Tongmai Oral Liquid on Preventing and CuringⅡPeriod Diabetic Nephropathy

Objective To investigate the effect of Tongmai oral liquid on the early diabetic nephropathy. Methods Diabetic nephropathy rats were induced by excision of the right kidney and a single intraperitoneal injection of streptozotocin (STZ). Forty male Wistar rats were divided randomly into 3 groups:Tongmai group, nephrotic model group, and normal group. At the end of the 8th week, the rats general states and some parameters such as kidney hypertrophy index, 24 h urine volume, blood glucose, 24 h urinary microalbumin (24 h UMA), renal function [serum creatinine (Cr), blood urea nitrogen (BUN)], serum lipid, and the change of nephritic pathologic lesion were measured. 10.A STUDY OF THE MECHANISM OF MORPHOLOGICAL CHANGES IN AXONS AFTER SEVERE BRAIN INJURY

To explore the molecular pathological mechanism of severe brain injury, the brain diffuse axon injury (DAI) model and Mamarou free drop model were produced in rats. Sagittal sections of the brain were processed by immunohistochemical ABC method using the mouse serum against NF68 subunit and HSP70. The medulla oblongata was observed under the microscope and electron microscope. Left parietal lobe of the free drop model was examined with HE and HSP70 immunohistochemistry. At 30 min post injury, the axons in medulla oblongata were seen to be crooked, swollen,and deranged. The myelin sheath became slightly separated, and the NFs in axoplasma were abnormal . At 2~24 h post injury,obvious axonal swelling, disconnection and formation of axonal retraction balls were seen. Obvious separation of myelin sheaths, local disconnection, vacuolization,peripheral aggregation of mitochondria and partial dissolution of axoplasma were seen. The NF68 positive axons increased gradually in staining intensity. HSP 70 positive cells of the two groups were detected at 3h after brain injury, reached the peak at 24h, and decreased at 72h. The HSP expression of the two groups were in accord. The research indicated that DAI could lead to a derangement in structure of NFs. Ischemia and anoxia may aggravate the brain injury.
