1.A comparison of surface electromyographic signals recorded from the paraspinal muscles of adolescents with idiopathic scoliosis during three loading tests

Objective Using surface electromyography (sEMG) techniques to investigate the patterns of frequency domain indexes of sEMC signals recorded from adolescent idiopathic scoliosis (AIS) patients' paraspinal muscles. Methods Twenty-five AIS patients were enrolled. All the patients undertook the Biering Sorensen test ( BST) ,the object-lifting test and a semi-bridge test, the ME3000P sEMG instrument was applied to record the electric activities of paraspinal muscles (convex/concave) of all subjects, and the time domain indexes such as averaged EMG(AEMG)and spectrum area(SPA) were analyzed, values in three tests were compared with one way ANOVA. Results The differences of AEMGs and SPAs on both convex and concave sides among the three tests showed no statistical significances (P = 0. 10,0.23, 0. 24 and 0. 45, respectively). Conclusions Any of the Biering Sorensen test( BST) , the object-lifting test or the semi-bridge test can be used to test the electric activities of paraspinal muscles (convex/concave) of AIS patients.
2.Expression of KLK10 in endometrioid adenocarcinoma and its relationship with ER and PR

Objective To evaluate the expression of human kallikrein10 (KLK10) in different endometrioid tissues and analyse the relationship of KLK10 with ER and PR in endometrioid adenoearcinoma.Methods The expression of KLK10 protein in 12 normal endometria,19 endometrial hyperplasia and 34 endometrial carcinoma were detected by immunohistochemistry.The correlations of the expression of KLK10 protein,ER and PR were analyzed.Results The expression of KLK10 in stage Ⅰ,Ⅱ and Ⅲ were 64.3 %(9/14),30.0 % (3/10),10.0 % (1/10),and the difference was statistically significant (P < 0.05).The expression of KLK10 in endometrial carcinoma,normal endometria,endometrial hyperplasia were 66.7 % (8/12),33.3 %(4/12),10.0 % (1/10),and the difference was statistically significant (P < 0.05).The expression of KLK10 in G1,G2,G3 were 66.7 %,33.3 %,10.0 %,and the difference was statistically significant (P < 0.05).The positive rate of KLK10 expression was 38.2 % and the positive rates of ER and PR expression were 67.6 %and 55.9 %,respectively,in 34 endometrial carcinoma.The expression of KLK10 was positively correlated with ER and PR expression (x2 =0.448,P < 0.01).Conclusion Absence or down-regulated expression of KLK10 may play an important role in the formation and development of endometrioid adenocarcinoma.The low expression of KLK10 is correlated with low expression of ER and PR in endometrioid adenocarcinoma.The positive expression of KLK00,ER and PR predicts a better prognosis.
3.Anti-intestinal cancer effect and mechanism of Wnt/β-catenin signaling pathway in Wuzhishan CallicarpanudifloraHook.etArn in Hainan Province

Objective:To investigate the anti-intestinal effect and mechanism of Wnt/β-catenin signaling pathway in Wuzhishan CallicarpanudifloraHook.etArn in Hainan Province.Methods:The colony formation assay was used to detect the effect of Wuzhishan CallicarpanudifloraHook.etArn on the proliferation of intestinal cancer cells in Hainan Province. The effect of Wuzhishan CallicarpanudifloraHook. etArn on the colony growth of intestinal cancer cells was detected by HE staining. The effect of Wuzhishan CallicarpanudifloraHook.etArn on transmembrane protein of intestinal cancer cell line IFITM1 was detected by laser confocal microscopy. Immunocytochemistry was used to detect the effect of Wuzhishan CallicarpanudifloraHook.etArn on the tumor suppressor protein of intestinal cancer cells. Western Blot was used to detect the expression of Wnt/β-catenin signaling pathway-related proteins.Results:With the increase of Wuzhishan CallicarpanudifloraHook.etArn in Hainan Province concentration, the colony number of SW480 and SW620 cells was down-regulated and the inhibition rate was up-regulated in a dose-dependent manner. Compared with that in control group(con group), the colony size of SW480 and SW620 cells in group Wuzhishan CallicarpanudifloraHook.etArn in Hainan Province group (Lhzz group) decreased, the cell morphology was blurred and the number of colony cells decreased significantly. Compared with that in con group, the green fluorescence intensity of IFITM1 transmembrane protein expression in SW480 and SW620 cells in Lhzz group decreased, the number of colony formation decreased, the volume decreased, and the nucleus showed pyknosis and irregular shape. The expression of tumor suppressor protein DCC in SW480 cells in Lhzz group was significantly higher than that in con group (2.13 ± 0.33 vs. 1.05 ± 0.16, t = 3.542, P < 0.05). The expression level of tumor suppressor protein DCC in SW620 cells in con group was significantly higher than that in con group (1.89 ± 0.26 vs. 0.96 ± 0.12, t = 3.466, P < 0.05). The expression level of oncoprotein DCC in SW620 cells in con group was significantly higher than that in con group (0.64 ± 0.11 vs. 0.38 ± 0.05, t = 3.154, P < 0.05), and the expression level of tumor suppressor protein in SW620 cells in con group was significantly higher than that in con group (0.52 ± 0.09 vs. 0.24 ± 0.03, t = 3.143, P < 0.05), and the expression level of β-catenin in SW480 and SW620 cells in Lhzz group was significantly higher than that in con group ( P < 0.05). Conclusions:Wuzhishan CallicarpanudifloraHook.etArn in Hainan Province can inhibit the proliferation of intestinal cancer cells by activating Wnt/β-catenin signaling pathway.
4.Role of protein kinase C in reduction of mitochondrial injury during myocardial ischemia-reperfusion by ischemic preconditioning in rats

Objective To evaluate the role of protein kinase C (PKC ) in reduction of mitochondrial injury during myocardial ischemia-reperfusion (I/R) by ischemic preconditioning in rats .Methods Forty male Sprague-Dawley rats ,aged 12-13 weeks ,weighing 280-320 g ,were randomly divided into 4 groups ( n=10 each) using a random number table:sham operation group (S group) ,I/R group ,ischemic preconditioning group (IP group) and PKC inhibitor chelerythrine group (C group) .Myocardial I/R was produced by 35 min occlusion of left anterior descending branch of coronary artery followed by 120 min reperfusion .Ischemic preconditioning was induced by 3 episodes of 5 min occlusion of left anterior descending branch at 5 min intervals before myocardial ischemia . Chelerythrine 1 mg/kg was injected intravenously via the caudal vein before ischemic preconditioning in group C . At 120 min of reperfusion ,the animals were sacrificed and the hearts were immediately removed .Mitochondrial suspension was prepared for determination of activities of succinate dehydrogenase (SDH ) , xanthine oxidase (XOD ) , glutathione peroxidase (GSH-Px ) and Ca2+-ATPase , content of Ca2+ , myocardial mitochonerial permeability transition pore (mPTP) opening and membrane potential (Δψm ) .Results Compared with S group , the activities of XOD and Ca2+-ATPase ,content of Ca2+ and mPTP opening were significantly increased ,and the activities of SDH and GSH-Px and Δψm were decreased in I/R group ( P<0.05) .Compared with I/R group ,the activities of XOD and Ca2+-ATPase , content of Ca2+ and mPTP opening were significantly decreased , and the activities of SDH and GSH-Px and Δψm were increased in IP group ( P<0.05) .Compared with IP group ,the activities of XOD and Ca2+-ATPase , content of Ca2+ and mPTP opening were significantly increased , and the activities of SDH and GSH-Px and Δψm were decreased in C group ( P<0.05) .Conclusion PKC is involved in reduction of mitochondrial injury during myocardial I/R by ischemic preconditioning in rats .
5.Role of ROS/PKC/p38 MAPK pathway in protective effects of polysac-charide from Fructus corni on rat cardiomyocytes against hypoxia-reoxy-genation injury

[ ABSTRACT] AIM:To investigate the effect of polysaccharide from Fructus corni ( PFC) on cardiomyocytes against hypoxia/reoxygenation ( H/R) injury and its possible relationship with ROS/PKC/p38 MAPK pathway.METHODS:Prima-ry cardiomyocytes were isolated from neonatal SD rats and randomly divided into normal group, H/R group, PFC (20 mg/L, 100 mg/L and 200 mg/L) preconditioning+H/R groups, chelerythrine+PFC (100 mg/L)+H/R group and SB203580+PFC (100 mg/L)+H/R group.The cell viability was measured by inverted microscopic observation.Apoptosis in the car-diomyocytes was detected by Hoechst 33258 staining and fluorescence microscopy.The levels of lactate dehydrogenase ( LDH) and superoxide dismutase ( SOD) in the cell culture supernatants, and the reactive oxygen species ( ROS) in the cells were also measured by microplate reader.The protein levels of PKC, p-p38 MAPK and HSP70 in the cells were detec-ted by Western blotting.RESULTS:Compared with normal group, the cell viability and beating frequency were decreased in H/R group.LDH and ROS contents, apoptotic rate and p-p38 MAPK level increased significantly (P<0.01).Compared with H/R group, PFC preconditioning increased beating frequency, SOD activity and the protein level of PKC and HSP70, and decreased ROS production, the protein level of p-p38 MAPK and cell apoptotic rate.However, the effect of PFC was in-hibited by chelerythrine or SB203580.CONCLUSION:PFC may protect cardiomyocytes from hypoxia/reoxygenation injury. Its mechanism is possibly involved in the inhibition of ROS via increasing the activity of SOD and the activation of PKC, and suppression of excessive activation of p38 MAPK.
6.Application of SAT-TB in the evaluation of curative effect of initial treatment of smear-positive pulmonary tuberculosis patients

Objective To evaluate the application value of simultaneous amplification and testing for Myco-bacterium tuberculosis(SAT-TB)in evaluating the curative effect of initial treatment of smear-positive pul-monary tuberculosis patients.Methods A total of 62 newly treated smear positive pulmonary tuberculosis pa-tients from June 2022 to June 2023 in Guangzhou Panyu District Chronic Disease Control Station were selected as the study objects,and the curative effect was evaluated by liquid-based sandwich cup method,Roche culture method and SAT-TB detection method.All patients received the standard anti-tuberculosis treatment regimen of 2HRZE/4HR standard regimen,and sputum samples were detected by liquid-based sandwich cup method,Roche culture method,and SAT-TB detection method at the 2nd,4th,and 8th week of treatment,respectively.Results Among 62 patients,54 cases were positive and 8 cases were negative using Roche culture method,47 cases were positive and 15 cases were negative using SAT-TB detection method.60 cases were positive and 2 cases were negative by Mycobacterium tuberculosis(MTB)-DNA test.The positive rates of the three methods were 87.10%(54/62),75.81%(47/62)and 96.77%(60/62),respectively.Taking Roche culture method re-sults as the standard,the sensitivity of SAT-TB detection method and MTB-DNA was 97.87%(46/47)and 90.00%(54/60),and the specificity was 46.67%(7/15)and 100.00%(2/2),respectively.There were signifi-cant differences between Roche culture method and SAT-TB detection method and MTB-DNA test(x2=20.070,P<0.05,x2=13.985,P<0.05),the difference between the results of SAT-TB detection method and MTB-DNA test was also statistically significant(x2=8.365,P<0.05).The negative conversion rates of MTB in sputum samples were 69.35%(43/62),29.03%(18/62)and 41.94%(26/62)at the 2nd,4 th,and 8 th weeks,respectively.77.42%(48/62),59.68%(37/62),58.06%(36/62),82.26%(51/62),79.03%(49/62),75.81%(47/62).There were significant differences in the negative conversion rates of MTB in sputum sam-ples between SAT-TB and liquid-based sandwich cup method at the 2nd and 4th weeks(x2=8.365,P<0.05,x2=4.465,P<0.05),while there was no significant difference between the results of Roche culture method at the 2nd,4th and 8th weeks(x2=1.726,P>0.05,x2=0.000,P>0.05,x2=0.046,P>0.05).Conclusion The use of SAT-TB detection method in clinical practice to evaluate the efficacy of smear positive pulmonary tuberculo-sis patients could accurately and quickly assess the negative conversion rate and treatment effect of patients,and provide a reliable basis for guiding clinical treatment.It could be considered as an effective auxiliary diag-nosis and evaluation method of curative effect,worthy of promotion and practical application.

8.Single-cell transcriptomic sequencing coupled with Mendelian randomization analysis elucidates the pivotal role of CTSC in chronic rhinosinusitis

Objective:To investigate the molecular mechanisms of chronic rhinosinusitis (CRS), to identify key cell subgroups and genes, to construct effective diagnostic models, and to screen for potential therapeutic drugs.Methods:Key cell subgroups in CRS were identified through single-cell transcriptomic sequencing data. Essential genes associated with CRS were selected and diagnostic models were constructed by hdWGCNA (high dimensional weighted gene co-expression network analysis) and various machine learning algorithms. Causal inference analysis was performed using Mendelian randomization and colocalization analysis. Potential therapeutic drugs were identified using molecular docking technology, and the results of bioinformatics analysis were validated by immunofluorescence staining. Graphpad Prism, R, Python, and Adobe Illustrator software were used for data and image processing.Results:An increased proportion of basal and suprabasal cells was observed in CRS, especially in eosinophilic CRS with nasal polyps (ECRSwNP), with P=0.001. hdWGCNA revealed that the "yellow module" was closely related to basal and suprabasal cells in CRS. Univariate logistic regression and LASSO algorithm selected 13 key genes ( CTSC, LAMB3, CYP2S1, TRPV4, ARHGAP21, PTHLH, CDH26, MRPS6, TENM4, FAM110C, NCKAP5, SAMD3, and PTCHD4). Based on these 13 genes, an effective CRS diagnostic model was developed using various machine learning algorithms (AUC=0.958). Mendelian randomization analysis indicated a causal relationship between CTSC and CRS (inverse variance weighted: OR=1.06, P=0.006), and colocalization analysis confirmed shared genetic variants between CTSC and CRS (PPH4/PPH3>2). Molecular docking results showed that acetaminophen binded well with CTSC (binding energy:-5.638 kcal/mol). Immunofluorescence staining experiments indicated an increase in CTSC +cells in CRS. Conclusion:This study integrates various bioinformatics methods to identify key cell types and genes in CRS, constructs an effective diagnostic model, underscores the critical role of the CTSC gene in CRS pathogenesis, and provides new targets for the treatment of CRS.
9.Analysis on the atypical clinical accidents of acupuncture based on the individual factors in patients.

The hidden risk of acupuncture has become the factor of the high incidence of adverse reaction of acupuncture in clinical practice. The retrospective analysis and the typical cases analysis are especially important for the prevention from the hidden risk of acupuncture. In the paper, the relevant literatures were reviewed, the basic diseases were listed such as diabetes, hypertension and digestive gastric ulcer that were neglected by the physicians, and the adverse reactions induced by the therapeutic history and the body constitutions were analyzed. It was stated in the paper that because of the individual factors of patient, the risks of the atypical clinical accidents of acupuncture were extremely serious and easily neglected by the clinical acupuncture physicians. It is very necessary to remind the clinical acupuncture physicians to understand the basic diseases, basic treatment as well as the peculiarity of body constitution of the patients besides their chief complaints. Moreover, acupuncture should be applied very carefully after identifying the pulse condition and the contraindication so as to ensure the patient's safety and obtain the optimal efficacy.

10.Resting-state electroencephalogram classification of patients with schizophrenia or depression.

The clinical manifestations of patients with schizophrenia and patients with depression not only have a certain similarity, but also change with the patient's mood, and thus lead to misdiagnosis in clinical diagnosis. Electroencephalogram (EEG) analysis provides an important reference and objective basis for accurate differentiation and diagnosis between patients with schizophrenia and patients with depression. In order to solve the problem of misdiagnosis between patients with schizophrenia and patients with depression, and to improve the accuracy of the classification and diagnosis of these two diseases, in this study we extracted the resting-state EEG features from 100 patients with depression and 100 patients with schizophrenia, including information entropy, sample entropy and approximate entropy, statistical properties feature and relative power spectral density (rPSD) of each EEG rhythm (δ, θ, α, β). Then feature vectors were formed to classify these two types of patients using the support vector machine (SVM) and the naive Bayes (NB) classifier. Experimental results indicate that: ① The rPSD feature vector performs the best in classification, achieving an average accuracy of 84.2% and a highest accuracy of 86.3%; ② The accuracy of SVM is obviously better than that of NB; ③ For the rPSD of each rhythm, the β rhythm performs the best with the highest accuracy of 76%; ④ Electrodes with large feature weight are mainly concentrated in the frontal lobe and parietal lobe. The results of this study indicate that the rPSD feature vector in conjunction with SVM can effectively distinguish depression and schizophrenia, and can also play an auxiliary role in the relevant clinical diagnosis.
