1.Research in influence of comprehensive relaxation training on perioperative psychological stress in patients with coronary intervention treatment

Objective The aim of this study is to provide guidance on rehabilitation nursing via observing the effect of comprehensive relaxation training on perioperative psychological stress in patients with coronary intervention treatment. Methods In this study, eighty patients ready to receive coronary intervention were randomly divided into the experimental group and the control group (40 patients in each). The control group were treated with routine percutaneous coronary intervention care, and the experimental group received both comprehensive relaxation training and routine care. The anxiety state and subjective feelings on comprehensive relaxation training in both groups were dynamically investigated via State-Trait Anxiety Inventory questionnaire and subjective feeling questionnaire from pre-operation to 7 days after operation. Results Anxiety levels between two groups showed the similar trend, and there was statistically significance between two groupsin the state anxiety level. The subjective feeling in the experimental group improved. Conclusions The psychological stress in patients receiving coronary intervention surgery maintains at high level form pre-operation to 24h post operation, and comprehensive relaxation training can significantly reduce the level of state anxiety. These patients consider that this simple training shows obvious relaxing effect.
2.THE EXPERIMENTAL STUDY OF A RAT C6 BRAIN TUMOR MODEL

Rat C6 glioma cells were cultured in vitro,and 10?L C6 cell suspension containing 10g/L agarose and 1?10 6 C6 cells was injected into the right caudate nucleus of rat brains to establish a rat brain tumor model by directional implantation method.General observation and MRI scan were conducted after implantation.Trans aorta paraformaldehyde perfusion for 5 group rats having received implantation was carried out at 10,15,20,25,days and before natural death.The tumor containing samples were prepared histologically by hematoxylin and eosin stains.MRI scan showed that intracerebral growth occurred in 50 implanted rats,with a distant metastasis of 4%.The results indicated that tumor globular intracerebral growth occurred and extracranial growth extension was scarce after implantation.The model was very stable and the implanted rat survival duration is easily determined. The experiment lays the foundation for chemotherapy,radiotherapy and gene therapy of the glioma.

4.Safty research of intraoperative intraperitoneal chemotherapy with raltitrexed for advanced colorectal cancer

Objective To investigate the intraoperative intraperitoneal chemotherapy security with raltitrexed in advanced colorectal cancer surgical operation.Methods Sixty patients with colorectal cancer undergone surgery were randomly divided into trial group (n =30) and control group (n =30) according to the random number table method.The trial group was given surgical operation plus with intraperitoneal chemotherapy with raltitrexed.The control group was given surgical operation plus with intraperitoneal saline perfusion.Theroutine blood test,liver and kidney functions,toxic side effects and complications in two groups before and after surgery were investigated.Results The white blood cells in trial group before and after surgery was (6.36 ± 2.63) × 109/L vs.(8.20 ± 2.08) × 109/L,with statistically significant difference (t =3.06,P <0.05).The ratio of absolute neutrophil count in trial group before and after surgery was 65.17% ± 10.36% vs.72.21% ± 10.53% (t =3.22,P < 0.05).The platelets in trial group before and after surgery was (261.03 ±84.74) × 109/L vs.(228.47 ± 58.69) × 109/L (t =2.07,P < 0.05).The white blood cells,the ratio of absolute neutrophil count and the platelets after surgery had no statistically significant differeuces between the two groups (P >0.05).The trial group had higher 1,2 level vomiting (60.00% vs.23.33%;x2 =8.30,P < 0.05),and nausea (30.00% vs.6.67%;x2 =5.46,P < 0.05) incidence rates,but there was no statistically significant difference in other toxic side effects (P > 0.05).The major complications post operation included intestinal obstruction,incision infection,abdominal cavity bleeding,and anastomotic fistula.There were equivalent complications in two groups (6.67% vs.3.33%,x2 =0.35,P >0.05;10.00% vs.6.67%,x2 =0.22,P>0.05;0 vs.0;3.33% vs.0,P>0.05).Conclusion For patients with advanced colorectal cancer,intraoperative intraperitoneal chemotherapy with raltitrexed is safe and feasible,and the adverse reactions can be tolerated without increasing postoperative complications.
5.The relationship between the changes of peripheral blood natural killer cells and HLA-Cw alleles in liver cirrhotic patients with chronic HBV infection

Objective To investigate the relationship between the frequency of peripheral blood natural killer cells (NK) and HLA-Cw alleles in liver cirrhotic patients with chronic HBV infection and a-cute hepatitis B patients. Methods Thirty liver cirrhotic patients and 30 patients with acute hepatitis B were included in our study, and 41 healthy individuals were enrolled as controls. The numbers of circulating NK cells and activated NK ceils were analyzed by flow cytometry. HLA-Cw genotyping was conducted with polymerase chain reaction-sequence specific oligonucleotide (PCR-SSO). Results The numbers of circu-lating NK cells and activated NK cells in liver cirrhotic patients were 13.22% ± 4.61% and 45.68% ± 14.64%, which was lower than that in healthy subjects (P < 0.05). The numbers of circulating NK cells and activated NK cells in acute hepatitis B patients were 22.62% ± 3.70% and 65.28%± 14.45%, which was higher than that in healthy subjects(P < 0. 05). There were statistically significant differences between the two groups(P < 0.01). The allele frequency of HLA-Cw * 15 in the patients with cirrhosis was signifi-cantly higher than that in the healthy (P < 0.05), and there was a significant negative correlation between the frequency of HLA-Cw * 15 and the numbers of activated NK cells in liver cirrhosis(r =4). 862, P < 0.05). No statistically significance was found between the group of acute hepatitis B and healthy subjects a- bout HLA-Cw(P > 0. 05). Conclusion The function of NK cells in liver cirrhotic patients is low, HLA-Cw * 15 gene may be one of the causes of effecting the antiviral function of NK ceils to induce the persistence of hepatitis B virus(HBV) infection.
6.Change of lactic acid concentration in patients receiving extracorpOreal membrane oxygenation

BACKGROUND:Extracorporeal membrane oxygenation(ECMO)adopts heparin-coated oxygenator as key membrane oxygenation,which is a covalent bonding of artificial material surface with mucopolysaccharide on one side of heparin.Thas artificial materials can stimulate human vascular endothelial function partly,leading to decrease production of lactic acid,increase biocompatibility of the matedals,prevent thrombogenesis and relieve inflammatory reaction after operation.OBJECTIVE:To discuss the relationship between the plasma lactic acid level and the prognosis of patients after ECMO applied with heparin-coated oxygenator.DESIGN:Self-controlled clinical study,grouping by age and survival prognosis.SETTING:Department of Cardiopuimonary Bypass,Fuwai Cardiovascular Hospital,Chinese Academy of Medical Sciences.PARTICIPANTS:Forty patients received ECMO in Fuwai Cardiovascular Hospital,Chinese Academy of Medical Sciences were selected from December 2004 to September 2006.They suffered from failing weaning of cardiopulmonary bypass postcardioctomy,cardiogenic shock based on dilated cardiomyopathy or cardiogenic shock based on coronary heart disease.All patients were divided into four groups according to age and survival prognosis:adult survival group,adult death group,infant survival group and infant death group.METHODS:All the patients received venoarterial ECMO support treatment.ECMO perfusion system consisted of centrifugal pump,water tank,heparin-coated membrane oxygenator(Jorstra Novalung,Germany)and corollary circuits (Bioline Coating,Germany,batch number:SFDA-I-20063450801).MAIN OUTCOME MEASURES:The concentrations of lactic acid were measured at the ECMO institution,6 hours later,the median time at ECMO support,6 hours before weaning off and at the time of weaning.RESULTS:①In 26 patients of adult groups,20 of them(76.9%)weaned from ECMO,16 of them(61.5%)survived and 10 of them died.In 14 patients of infant groups,7 of them weaned from ECMO(50%),5 of them(35.7%)survived and 9 of them died.②The concentrations of lactic acid in two survival groups were significantly lower than those in death groups(P<0.05 or 0.001).The concentration decreased significantly at the time points of median time,6 hours before weaning off and at the weaning time,compared with the level at the time of ECMO institution(P<0.01),indicating a decreasing concentration of lactic acid.CONCLUSION:①The concentration of lactic acid in patients after ECMO decreascs obviously.②Patients with high lactate level at the weaning time show unfavourable prognosis.
7.Study on antibacterial ability of silver nanoparticles loaded titanium nanotubes

Objective To detect the inhibitory effect of siliver nanoparticles loaded titanium nanotubes on staphylococcus aureus, and provide a theoretical basis for implant local application. Methods Orderly arrangement of titania nanotubes produced by anodic oxidation, loaded silver nanoparticals by situ replacement. Scanning electron microscopy (SEM) and transmission electron microscopy(TEM) were used to detect the morphology topology of silver nanoparticals, titanium nanotubes and siliver particals loaded titanium nanotubes. The minimum inhibitory concentration of silver nanoparticles was calculated. The antibacterial of planktonic bacteria was detected 1 day, 3 days and 5 days after culturing staphylococcus aureus on siliver particals loaded titanium nanotubes. The inhibitory bacterial adhesion properties were detected by scanning electron microscopy. Results The uniform and orderly diameter of 80~120 nm TiO2 nanotubes were prepared under 18 V voltage, loaded diameter of 20 nm silver nanoparticals, which effectively inhibited adhesion and proliferation of staphylococcus aureus. Conclusion Titanium nanotubes produced by 18 V have a stronger drug loading capacity. The 100 mmol/L silver nanopartical solution loaded nanotubes can effectively inhibit staphylococcus aureus adhesion and proliferation within three days.
8.Circ_0075960 targets the miR-202-5p/CTNND1 axis to promote the growth and migration of endometrial carcinoma cells via regulating Wnt/β-catenin signaling activity

Background:
Endometrial carcinoma (EC) is one of the most common malignant tumors of the female reproductive tract, involving multiple molecular alterations. Circular RNA (circRNA) dysregulation is frequently observed in EC tissues, suggesting the involvement of circRNA in EC development. We aimed to investigate the role of circ_0075960 in EC.
Methods:
Real-time quantitative polymerase chain reaction (RT-qPCR) and western blot assays were applied for expression analysis. CCK-8, EdU, colony formation, flow cytometry and wound healing assays were employed for functional analysis. The predicted binding relationship between miR-202-5p and circ_0075960 or CTNND1 was validated by dual-luciferase reporter experiment. In vivo animal models were constructed in nude mice to verify the role of circ_0075960 in tumor growth.
Results:
Circ_0075960 and CTNND1 were upregulated, while miR-202-5p was downregulated in EC. Knockdown of circ_0075960 induced EC cell apoptosis, suppressed cell proliferation and migration, and repressed tumor growth in animal models. MiR-202-5p was targeted by circ_0075960 and it directly bound to CTNND1 3’UTR. The inhibition of circ_0075960 knockdown or miR-202-5p enrichment on EC cell proliferation and migration was reversed by miR-202-5p depletion or CTNND1 overexpression, respectively. Circ_0075960 targeted miR-202-5p to positively regulate CTNND1 expression. Moreover, circ_0075960 knockdown weakened the activity of Wnt/β-catenin signaling via targeting the miR-202-5p/CTNND1 axis.
Conclusion
Circ_0075960 targets the miR-202-5p/CTNND1 axis to modulate Wnt/β-catenin signaling activity, thus contributing to the malignant development of EC.
9.Effects of nano-lead exposure on learning and memory as well as iron homeostasis in brain of offspring rats.

OBJECTIVETo investigate the effects of nano-lead exposure on learning and memory and iron homeostasis in the brain of the offspring rats on postnatal day 21 (PND21) and postnatal day 42 (PND42).
METHODSTwenty adult pregnant female Sprague-Dawley rats were randomly divided into control group and nano-lead group. Rats in the nano-lead group were orally administrated 10 mg/kg nano-lead, while rats in the control group were administrated an equal volume of normal saline until PND21. On PND21, the offspring rats were weaned and given the same treatment as the pregnant rats until 42 days after birth. The learning and memory ability of offspring rats on PND21 and PND42 was evaluated by Morris water maze test. The hippocampus and cortex s amples of offspring rats on PND21 and PND42 were collected to determine iron and lead levels in the hippocampus and cortex by inductively coupled plasma-mass spectrometry. The distributions of iron in the hippocampus and cortex were observed by Perl's iron staining. The expression levels of ferritin, ferroportin 1 (FPN1), hephaestin (HP), and ceruloplasmin (CP) were measured by enzyme-linked immunosorbent assay.
RESULTSAfter nano-lead exposure, the iron content in the cortex of offspring rats on PND21 and PND42 in the nano-lead group was significantly higher than those in the control group (32.63 ± 6.03 µg/g vs 27.04 ± 5.82 µg/g, P<0.05; 46.20 ±10.60 µg/g vs 36.61 ± 10.2µg/g, P<0.05). The iron content in the hippocampus of offspring rats on PND42 in the nano-lead group was significantly higher than that in the control group (56.9 ± 4.37µg/g vs 37.71 ± 6.92µg/g, P<0.05). The Perl's staining showed massive iron deposition in the cortex and hippocampus in the nano-lead group. FPNl level in the cotfex of offspring rats on PND21 in the nano-lead group was significantly lower than that in the control group (3.64 ± 0.23 ng/g vs 4.99 ± 0.95 ng/g, P<0.05). FPN1 level in the hippocampus of offspring rats on PND42 in the nano-lead group was significantly lower than that in the control group (2.28 ± 0.51 ng/g vs 3.69 ± 0.69 ng/g, P<0.05). The escape latencies of offspring rats on PND21 and PND42 in the nano-lead group were longer than those in the control group (15.54 ± 2.89 s vs 9.01 ± 4.66 s; 6.16 ± 1.42 s vs 4.26 ± 1.51 s). The numbers of platform crossings of offspring rats on PND21 and PND42 in the nano- lead group were significantly lower than those in the control group (7.77 ± 2.16 times vs 11.2 ± 1.61 times, P<0.05; 8.12 ± 1.51 times vs 13.0 ± 2.21 times, P<0.05).
ONCLUSIONn Nano-lead exposure can result in iron homeostasis disorders in the hippocampus and cortex of offspring rats and affect their learning and memory ability.

10.Establishment of interventional access via the sheathing of the caudal artery in rats

Objective Digital silhouette angiography in animal models is limited by the lack of methods that can perform continuous intra-arterial manipulations.We aimed to investigate the way that allowed to repeat interventional maneuvers through a single sheath access in the rat caudal artery.Methods Six adult male Sprague Dawley rats (650~700 g) were anesthetized,the caudal arteries were exposed and a fish-mouth-shaped incision was made with a microscissor.After the access was established,whole brain angiography and aortic dilatation with balloon and stent catheters were performed via the tail artery sheath access to test the feasibility and effectiveness of the tail artery sheathing method as an interventional access.Results The diameter of caudal artery approximately (1.19±0.14) mm,and the 18G sheath was successfully implanted with guidewire.The sheathing time of the caudal artery (11.00±1.90 min,P<0.01) was significantly reduced compared with the carotid artery (24.50±4.50) min and the femoral artery (18.17±3.43) min.A balloon catheter and a stent catheter could be passed through the caudal artery sheath and released in the aorta,and the catheter could be withdrawn smoothly after the release.Each rat can be used for multiple tail artery sheathing operations,and repeatable angiographic and interventional procedures can be performed through a single sheath channel.Conclusions The method of establishing interventional access using a sheath introduced through the rat tail artery is suitable for intravascular imaging,treatment,and the construction of rat animal models.This method further enhances the scope of application of rats as human disease models.
