1.Observation of effects of zinc on the injury induced by oxygen free radical in cultured heart cells

In present study, the injury induced by oxygen free radical in cultured cardiac cells was observed by adding xanthine and xanthine oxidase into the medium The effects of zinc on the injury of cells were also observed, taking the electro physiological changes and threshold concentration of BaCl_2 which induces cell stop beating as index. The results showed that the electric parameters of action potential (the amplitude, overshoot, maximum diastolic potential, threshold, maximum rate of depolarization and duration of action potential ) were decreased in XOD group, compared with the control. The threshold concentration of BaCl_2 was decreased too. However, there were no significant differences between Zn group and control group except the TP value. The electric parameters of action potential was increased notab(?)y in Zn group, compared with XOD group. Furthermore, the threshold concentration of BaCl_2 in Zn group was increased too. It is suggested that Zn possessed the protective effects on cultured cardiac cells from the injury induced by oxygen free radical.
2.HSV_1-tk/GCV GENE THERAPY IN GLIOMA ANIMAL MODEL

Objective To evaluate the therapeutic effect and the mechanism of herpes simplex virus thymidine kinase Ⅰ type (HSV 1\|tk) and Ganciclovir(GCV) in animal model of human glioma. Methods We demonstrated that tk gene was stably integrated into the genome of retrovirus vector producing cell line pN 2A\|tk/VPC(VPC) by Southern blot hybridization; then we established the nude mice model of glioma by inoculating the VPC and the human glioma cells DBTRG\|05MG(05MG) at 1∶1,1∶4 ratios respectively(only inoculated 05MG cells as control group), then GCV treatment was administrated i.p.,30mg/(kg\5d), twice daily for 14 days. A week later, the nude mice were sacrificed and the pathological sections of tumor tissues were observed by light microscopy. 3.Effects of Ca~(2+)/ calmodulin dependent protein kinase Ⅱ signal pathway in DPDPE long term treatment NG108-15 cells

AIM To observe the change of Ca 2+ /calmodulin dependent protein kinaseⅡ (CaMKⅡ) signal pathway in opioids dependent NG108-15 cells and the relationships between Ca 2+ /calmodulin dependent protein kinaseⅡ (CaMKⅡ) signal pathway and cAMP accumulation. METHODS NG108-15 cells were used as an in vitro model system. Competitive protein binding assay and radioimmunoassay were used to examine the intracellular cAMP accumulation. Calmodulin activity was assayed by PDE method. CaMKⅡ activity was assayed by ?- 32 P incorporation of syntide-2. mRNA expression of mDOR was measured by RT-PCR. RESULTS DPDPE long-term treatment also increased cAMP accumulation, and resulted in opioids dependence in NG108-15 cells; DPDPE long-term treatment increased calmodulin activity and CaMKⅡ activity in NG108-15 cells. Specific calmodulin antagonist W-7 was found to inhibit significantly the elevation of calmodulin and CaMKⅡ activity which resulted from DPDPE long-term treatment, and CaMKⅡ inhibitor KN-62 also inhibited elevation of CaMKⅡ activity by DPDPE long-term treatment. DPDPE long-term treatment increased cAMP accumulation, when W-7 or KN-62 was added at the same time, cAMP accumulation decreased. When naloxone was added to NG108-15 cells which were long-term treated by DPDPE, calmodulin and CaMKⅡactivity increased more. mRNA level of mDOR did not change significantly in opioids dependent NG108-15 cells. CONCLUSION Ca 2+ /CaMKⅡ signal pathway was involved in the mechanisms of opioids dependence through regulating cAMP level when DPDPE is long-term administered to NG108-15 cells.
4.Determination and Comparison of Ion Residues in Different Autosampler Plastic Vials by Ion Chromatography

Objective The present paper compared the ion residues including cations residues and anions residues in autosampler plastic vials from 4 different ion chromatography companies and discussed the relationship between soaking duration time and ion releasing. Methods The separation and determination were carried out by IonPac CS12A cation exchange column, MSA isocratic eluent together with suppressed conductivity detector; and IonPac AS15 anion exchange column, NaOH isocratic eluent together with suppressed conductivity detector. 4 plastic vials from 4 companies were soaked in DI water with a particular volume followed by injecting the liquid samples to IC system in order to measure residue ions with low level(?g/L). Results The detection limits were 0.79-5.28 ?g/L.The linear range were 1-20 000 ?g/L for Cl-, NO2-, NO3-, SO42-, Na+, K+, Mg2+, Ca2+. The linear coefficients were 0.998 3-0.999 9. RSDs were below 3%.The recovery rates were 83.3%-112%. Conclusion This method satisfies the requirement of practical examination, and the suitable methods for repairing the plastic vials of autosampler is proposed.
5.The role of antimicrobial peptides in immune system

Many species of animals contain antimicrobial peptides as components of their immune systems. Many antimicrobial peptides have a broad spectrum of activity not only against gram negative and gram positive bacteria but also against antibiotic resistant bacteria, fungi, viruses, and parasites. Such peptides can act in synergy with other host molecules to kill pathogens. It has been found that certain peptides have a high affinity for bacterial products, allowing them to modulate the host inflammatory response to sepsis. They have also been found that they have the ability to recruit inflammatory cells. These properties as well as some other functions of antimicrobial peptides enable them to have an important impact in innate immunity.
6.Observation of the effed of tiotropium bromide combined with ambroxol in of the treatment of chronic ob-structive pulmonary disease

Objective To explore the effect of tiotropium bromide combined with ambroxol hydrochloride oral liquid on chronic obstructive pulmonary disease (COPD) at remission.Methods According to the digital table, 38 patients with COPD at remission were randomly divided into treatment group and control group .The control group was given with conventional treatment ,and anti-infection and symptomatic treatment when acute attacking ,while the treatment group was treated with inhaled tiotropium bromide and ambroxol Hydrochloride oral liquid base on the con -trol group.Both groups were treated and observed for 6 months.Times of expectoration and acute attack of COPD were observed,and scores of COPD Assessment Test ( CAT) were noted everyday during therapy .Results Compared with the control group ,the total times and means of expectoration in treatment group were much less ,with very significant statistical difference(t=5.19,P<0.05);the cases,times and means of acute attack of COPD were obviously less than those in control group,with very significant statistical difference (t=2.92,P<0.05);the total scores and means of CAT in treatment group were much lower,with significant statistical difference(t=2.65,P<0.05).Conclusion With long-term treatment ,inhaled tiotropium bromide combined with ambroxol hydrochloride oral liquid can effectively decrease the times of expectoration and acute attack in patients with COPD at remission,and improve their qualities of life.

Objective To analyze the association HLA DQ gene polymorphism to MG susceptibility.Methods HLA DQA1 and DQB1 were genetyped using PCR RFLP Results Comparing the frequency of HLA DQA1 alleles between MG patients with hyperplasia or MG males with disease onset less than 30 years and the healthy controls It was found that the frequency of DQA1*0301 was significantly increased Furthermore,in comparison with the frequency of HLA DQB1 alleles between MG patients (included subgroups)and the healthy controls,the frequency of DQB1*0303 was significantly increased,while the frequencies of DQB1*0601 and DQB1*0602 were significantly decreased And these differences were statistically significant. Conclusions HLA DQA1*0301 contributes to the genetic susceptibility of MG with hyperplasia and MG males with disease onset ≤30 years old DQB1*0303 was susceptible to MG DQB1*0601 and DQB1*0602 were protective genes

Pro-rich antimicrobial peptides are a group of linear peptides isolated from animals. These peptides have antibacterial activities and play an important role in innate immunity. Pro-rich antimicrobial peptides are devided into two classes:Pro-rich antimicrobial peptides from mammals and Pro-rich antimicrobial peptides from insects and other invertebrates.Members of Pro-rich antimicrobial peptides are all characterised by a high content of proline residues and predominantly active against Gram-negative bacterial species which they kill by a non-lytic mechanism,at variance with the majority of the known antimicrobial peptides. Evidence is accumulating that the Pro-rich peptides enter the cells without membrane lysis and,once in the cytoplasm,bind to,and inhibit the activity of DnaK protein that is essential to bacterial growth,thereby causing responsive bacteria death. This mode of action makes these peptides suitable for drug development efforts. As one of the best characterized Pro-rich peptides,PR-39 plays an important role in a number of biological processes. It has been found that PR-39 can induce the syndecan expression in mesenchymal cells, inhibit the NADPH oxidase activity of neutrophils and block the degradation of 1?B? and HIF-1?. These findings show that PR-39 can provide novel means in the control of inflammation, wound repair, ischemia-reperfusion injury,and angiogenesis for therapeutic purposes.

As for the ? opioid receptor, there are some disputes about the domains involved in the selective recognition of ligands, the relative spatial orientation of the amino acids within the TM affect the affinities of selective ligands. Regulation of opioid receptor activities does not appear to involve in their ability to promote the association of GTP onto the G proteins and the subsequent dissociation of heterotrimers. It is not very clear if phosphorylation correlates with agonist induced receptor desensitization. The celluar processing of ? opioid receptors requires the formation of multiple protein complexes, it is clear that interactions of ubiquitous transcriptional factors determine gene transcription.
