1.EFFECTS OF INTERLEUKIN-2 AND POLYSACCHARIDE OF ASTRAGALUS ON MURINE NK CELL: CYTOCHEMISTRY AND ULTRASTRUCTURE

In this paper, murine natural killer (NK) cells were studied with cytochemistry and electron-microscopy after treated with interleukin-2 (IL-2) and polysaccharide of astragalus (PA). The results showed that acid phosphatase (AcP), acid naphthylacetate esterase (ANAE) and naphthol-AS-D chloroacetate esterase (NCAE) were positive in NK cells, and the activity of AcP and ANAE was enhanced by IL-2 and PA. Significant changes of NK cells in ultrastructure took place after preculturing with IL-2.

lymphocyte,and especially the small lymphocytes were moreheavily labeled than the larger ones,and the Con A receptor of NK cell exhibitedpolarization along the cell surface.These results suggest that:1.the extent of ConA receptors was related to the functional activities of the ceils,the more active thecell is,the more the Con A receptors will be;2.the receptors of Con A wereprobably related to the cell recognition,for instance,on the NK cell Con A rece-ptors showed polar distribution;3.the number of receptors of Con A was dependenton the cell differentiation,the more mature the cell is,the more the Con Areceptors are.

The effects of the Biological Suppressor of Cancer(BSG)on human normal hone marrow cells and leukaemic cells were studied.There are some evidences that the BSC possesses a biological activities which apparently kill leukaemic cells and inhibit their DNA synthesis,but it has no such effects on erythroblast.The BSC extracted from ascites of ascitic tumors shows no cancer lines specificity,nor tissue specificity.
4.Analysis of Piper puberulum (Benth.) Maxim.extracts preparation and the preliminary qualitative chemical composition

Objective To investigate optimal extraction process of Piper puberulum ( Benth.) Maxim.and qualitative analyze the chemical component of the extracts.Methods Method of solvent heating reflux was used for extraction.On the basis of single factor experiment, L9 (34 ) orthogonal experiment was designed with the variants of extraction frequency, time, material-liquid ratio, and immersion time.Extraction rate as index, extraction processes were optimized to achieve best extraction.The extracts, including total extract, water elution, and ethanol elution, were physiochemically analysed to achieve an initial qualitative result.Results The optimal extraction process was: extractions 3 times for 2 hours, with an 1︰30 material -liquid ratio and 2 hours of immersion, Initial qualitative analyzed the total extracts containing amino acids, polypeptides, proteins, alkaloids, steroids or triterpenes, flavones, saponins, polysaccharides, reducing sugars or glucosides, cumarins, terpene lactones, phenols, and tannins.The water elution containing: amino acids, polypeptides, proteins, saponins, polysaccharides, reducing sugars or glucosides, cumarins, and terpene lactones.The ethanol elution containing: amino acids, polypeptides, proteins, alkaloids, steroids or triterpenes, flavones, polysaccharides, reducing sugars or glucosides, phenols, and tanins.Conclusion The experiments show that optimal extraction process can achieve high extraction yield, stable and practical.
5.EFFECTS OF WEHI-3 SUPERNATANT ON GROWTH AND PROLIFERATION OF MURINE BONE MARROW STROMAL CELLS IN VITRO

In order to observe the effects of the supernatant of WEHI-3 cell culture which contains IL-3 (interleukin 3), we have Performed some experiments by adding WEHI-3 supernatant to murine bone marrow cells culture. Cell culture of six days shows that the supernatant of WEHI-3 can promote adherence and proliferation of the stromal cells. There are 1.334?10~6 stromal cells/L in the culture which contained 20% WEHI-3 supernatant. Four kinds of stromal cells can be identified under both light microscope and SEM. Fibroblast-like cells which tend to be spindle-shaped with many filament-like and microvilli-like dendrites on the cell surface; Riticular-like cells which are irregular with some folds on the dendrites surface occasionally; Macrophage-like cells which are round with many phagocytic granules in the cytoplasm, and are characterized by positive ANAE staining, and many lamina-like folds and long filament-like dendrites on the cell sufrace; Fat cells are few in number, the cell body is ovoid with lipid droplet, and show strong cytoplasmic staining for Sudan black B, the cell surface is smooth. At the same time we have also observed the close contact between the hematopoietic cells and the stromal cells. In the culture without WEHI-3 supernatant, we have found few small adhered cells with less cytoplasm and weak enzyme activaties under light microscope, and with atrophied cell body and flat surface under SEM. The cell count of the stromal cells in normal culture is 58.83/mm~2. The results showed that WEHI-3 supernatant can promote the growth and proliferation of the stromal cells of murine bone marrow with some histochemical changes.
6.A primary study of the relationship between apparent diffusion coefficient value of rectal adenocarcinoma on DWI and its pathological grading

Objective To investigate the relationship between apparent diffusion coefficient (ADC)value of rectal adenocarcinoma on DWI and its pathological grading.Methods The ADC values of 46 rectal adenocarcinomas were measured and compared with their histopathological grades.Results The 46 rectal adenocarcinomas included well differentiated adenocarcinomas in 14,moderate-ly differentiated ones in 20,and poorly differentiated ones in 12.The ADC values of well,moderately and poorly differentiated ade-nocarcinomas were (1.125±0.103)×10 -3 mm2/s,(1.030±0.098)×10 -3 mm2/s and (0.922±0.091)×10 -3 mm2/s,respective-ly,exhibiting a statistical difference (χ2 =1 7.35 1,P =0.000).Mann-Whitney U test showed that difference in ADC value between different histopathological grades was statistically significant.Conclusion ADC value of rectal adenocarcinoma can be used as a bio-marker for cell grading to guide treatment decision and prognosis assessment.
7.Synthesis and phosphodiesterase type-5 inhibitory activity of sildenafil analogues possessing a guanidine group in the phenyl ring

In this study,a series of new sildenafil analogues 11-27 possessing a guanidine group were synthesized to investigate their PDE5 inhibitory activity and selectivity using[~3H]cGMP SPA kit in vitro and efficacy in the rat model of erection.Most of the compounds showed potent activity against PDE5,and more importantly,several compounds exhibited higher PDE5 selectivity over PDE6 than that of sildenafil.Structure-activity relationship of these sildenafil analogues was also discussed.Within this series of compounds,compound 15(IC_(50) =1.7 nmol/L)not only exhibited more potent PDE5 inhibitory activity than that of sildenafil (IC_(50) = 6.5 nmol/L),but also showed functional efficacy in the rat model of erection.

AIM:To study the role of prostaglandin F2?(PGF2?) in cardiac hypertrophy and its relation with calcineurin(CaN) signal transduction pathway in vitro.METHODS:The cultured neonatal rat cardiomyocyte was used to observe the hypertrophic effect of PGF2?,and the hypertrophic response was assayed by measuring the cell diameter,protein content and atrial natriuretic factor(ANF) mRNA expression.For mechanism studies,the intracellular free calcium concentration([Ca2+]i) in cultured cardiomyocytes was measured by using Fura-2/AM as a fluorescent indicator.ANF and CaN mRNA expressions,and the expressions of CaN and its downstream effectors,NFAT3 and GATA4 proteins were assayed by RT-PCR and Western blotting,respectively.RESULTS:In cultured cardiomyocytes,PGF2? induced profound hypertrophic morphology change and the significant increase in cell diameter,and protein content in a concentration-dependent manner compared with those in vehicle control(P
9.Distrubution and excretion of protopine in rats by RP-HPLC method

AIM To study the distrubution and excretion of protopine in rats. METHODS Reversed phase high performance liquid chromatographic method (RP HPLC) was developed for determining the level of protopine in rats. The analytical column were packed with 5 ?m C 18 . The mobile phase was a mixture of methanol, water and 10% acetic acid (80∶20∶2), in which the pH was modulated to 5 6 with 15% ammonia. Protopine biological samples were isolated well, in which two extraction with ether under basical condition and an extraction with 0 02 mol?L -1 sulfuric acid were performed, respectively. The content of protopine in the biological sample was measured by an UV detector at 285 nm. The distrubution and excetion of protopine have been investigated in rats after intravenous administration 10 mg?kg -1 . RESULTS Protopine distrubuted in many tissues after iv a dose of 10 mg?kg -1 . The higher level of protopine was found in lung, kidney, spleen and brain, and the highest was observed in lung at 5, 15 minutes after administration. However the top level tissue was testicle at 3 h, which may be due to small blood circulation. The excretion of the parent compound in urine was 36 87% of dose, but the excretion of the parent compound in feces and bile was less than 1% of dose. Plasma protein binding was less than 5%. CONCLUSION The distrubution of protopine is extensive and the parent compoud was mainly excreted by urine and plasma protein binding was low.
10.Role of high resolution CT in diffuse pulmonary nodules.

OBJECTIVETo evaluate high resolution CT (HRCT) in the diagnosis of diffuse pulmonary nodules.
METHODSFifty normal chest radiographs, conventional CT and HRCT were used to evaluate the visualization of pulmonary lobule. The configuration, distribution and intrinsic structure of lesion in 38 patients with diffuse pulmonary nodules were analyzed by HRCT.
RESULTSThe chest radiographs were not able to show the structure of pulmonary lobule. The visualization rates of pulmonary lobule were 20% by conventional CT and 50% by HRCT (P < 0.05). Of 38 patients with diffuse pulmonary nodules, 19 had interstitial nodules which were located in the pulmonary intestities, the lobular septa and under the pleura. HRCT could clearly show their para-bronchial distribution with clear cut margin. Four had airspace nodules, chiefly shown as solidification of the air spaces. There was no nodule beneath the pleura or in the lobular septa. HRCT revealed even density and hazzy margins. Fifteen had randomly distributed nodules, with the nodules scattered at random. HRCT showed nodules with high density, sizes varying greatly but the margin was clear.
CONCLUSIONHigh resolution CT is able to show the pattern of distribution, intranodular structures and background of the diffuse pulmonary nodules, which is valuable in the diagnosis and differential diagnosis of this disease.
