1.Analysis of the risk factors for recurrence of gastrointestinal stromal tumors after resection

ObjectiveTo investigate the postoperative risk factors for gastrointestinal stromal tumors.MethodsFrom December 2005 to December 2010,sixty-one gastrointestinal stromal tumors patients of People' s Hospital of Tongling city gastrointestinal surgery department were analyzed retrospectively.Using the logistic regression model to calculate the postoperative recurrence risk factors. Results Sixty-one patients underwent operation,including gastrectomy (40 cases ),partial intestina parva resection (7 cases),partial colon resection ( 3 cases ),Dixon' s operation ( 2 cases ) and tumorectomy ( 9 cases ).Fifteen patients were given postoperative imatinib Mesylate Capsules (Glivec) treatment.Fifty-seven patients were followed-up more than 1 year and 7 cases were found palindromia ( 12% ).Logistic model shows that the malignant degree of gastrointestinal stromal tumors is the only risk factor of postoperative palindromia.ConclusionsMalignant degree of gastrointestinal stromal tumors is the main risk factor for postoperative palindromia.Imatinib mesylate capsules treatment may be beneficial to the greater stage patients.
2.The effects of tetrahydroxystilbene glucoside on mouse liver cytochrome P 450

Objective To investigate the effects of tetrahydroxystilbene glucoside (TSG) on cytochrome P450(CYP) in mouse livers .Methods Kunming male mice were divided into the blank ,low dose and the high dose of TSG groups .3 ,5 and 7 after intra-gastrical administration of TSG ,mice were sacrificed and the mRNA expressions of CYP isoenzymes in mouse livers were measured by real time reverse transcription-polymerase chain reaction(RT-PCR) ,respectively .Results TSG significantly inhibited CYP1A2 and CYP 3A4 mRNA expressions at 3th ,5th and 7th day after treatment .TSG time-dependently increased CYP2E1 mRNA expres-sion .TSG inhibited CYP4A14 mRNA expression at 7th day after treatment .Moreover ,TSG had no significant effects on CYP2B10 , CYP3A11 and CYP3A25 mRNA expressions .Conclusion TSG has significant effects on CYP1A2 ,CYP2E1 ,CYP3A4 and CYP4A14 mRNA expressions but no significant effects on CYP2B10 ,CYP3A11 and CYP3A25 mRNA expressions .

4.Inhibitory effect and mechanism of cobra venom metalloproteinase atrase A on platelet aggregation

Aim To investigate the effect and mechanism of cobra venom metalloproteinase atrase A on inhibiting platelet aggregation.Methods Platelet aggregation induced by collagen,ADP,PAF,AA,ristocetin and thrombin,respectively,was measured turbimetrically after platelet incubated with atrase A.Western blot was used to detect the effect of atrase A on cleavage of platelet membrane glycoprotein and von Willebrand Factor.Results Atrase A significantly inhibited platelet aggregation induced by ristocetin and thrombin in a dose-and time-dependent manner.Meanwhile,atrase A just showed slight inhibitory effect on platelet aggregation induced by PAF,AA,collagen,and ADP after incubated with PRP for 5 min.After incubation time was prolonged to 30 min,significant inhibition was shown on platelet aggregation.Western blot revealed that atrase A cleaved platelet membrane glycoprotein GPIb.Conclusions Cobra venom metalloproteinase atrase A significantly inhibits platelet aggregation induced by ristocetin and thrombin due to the cleavage of platelet membrane glycoprotein Ib.And atrase A also has inhibitory effect on platelet aggregation induced by ADP,PAF,AA,and collagen.
5.Inhibitory effect of anticomplementary protein atrase B from cobra venom on platelet aggregation induced by activated complement

Aim To investigate the inhibitory effect of atrase B on human platelet aggregation induced by activated complement.Methods By employing CVF to activate complement,the effect of atrase B on gel filtered platelet aggregation induced by activated complement was measured by turbidimetry and the expression of P-selectin and GPⅡb/Ⅲa on platelet membrane were detected by flow cytometry.Results Atrase B inhibited platelet aggregation and the expression of P-selectin and GPⅡb/Ⅲa on platelet membrane induced by activated complement.Conclusion Anticomplementary protein atrase B from Naja atra venom can significantly inhibit platelet activation and aggregation induced by activated complement.
6.Synthesis and phosphodiesterase type-5 inhibitory activity of sildenafil analogues possessing a guanidine group in the phenyl ring

In this study,a series of new sildenafil analogues 11-27 possessing a guanidine group were synthesized to investigate their PDE5 inhibitory activity and selectivity using[~3H]cGMP SPA kit in vitro and efficacy in the rat model of erection.Most of the compounds showed potent activity against PDE5,and more importantly,several compounds exhibited higher PDE5 selectivity over PDE6 than that of sildenafil.Structure-activity relationship of these sildenafil analogues was also discussed.Within this series of compounds,compound 15(IC_(50) =1.7 nmol/L)not only exhibited more potent PDE5 inhibitory activity than that of sildenafil (IC_(50) = 6.5 nmol/L),but also showed functional efficacy in the rat model of erection.

8.Roscovitine inhibits inflammatory hyperplasia of carotid artery intima in rats via suppressing nuclear factor-κB activation

AIM:To study the effect of roscovitine on the inflammatory hyperplasia of carotid artery intima in rats and the related mechanisms .METHODS: SD rats ( n=60 ) were randomly divided into 3 groups including control group, model group and treatment group .The rat model was established by trypsin digestion injury .The rats in control group were given sham operation .The rats in treatment group were administered with 0.5 mL roscovitine (2 g/L) slow-re-leasing gelatin.The rats in each group were fed normally for 4 weeks, then killed to take out carotid arteries for further ob-servations .The effects of roscovitine on the inflammatory hyperplasia of carotid artery intima and the related mechanism via nuclear factor-κB ( NF-κB) in the rats were detected by Western blot .RESUITS:Roscovitine inhibited the activation of NF-κB and the expression of inflammatory factors cyclooxygenase-2 (COX-2), vascular cell adhesion molecule-1 (VCAM-1),TNF-αand IL-6 via blocking the phosphorylation activation of NF-κB and inhibiting the degradation of IκB-α.CON-CLUSION:Roscovitine inhibits inflammatory hyperplasia of carotid artery intima in the rats via suppressing NF-κB activa-tion.
9.Effect of Pneumatically Ballistic Extracorporeal Shockwave on Chronic Nonspecific Low Back Pain

Objective To observe the clinical effect of pneumatically ballistic extracorporeal shockwave on chronic nonspecific low back pain. Methods 66 cases with chronic nonspecific low back pain were randomly divided into treatment group (n=33) and control group (n=33). There were 1 case drop-out in the treatment group and 2 in the control group. The treatment group accepted pneumatically ballistic extracorporeal shockwave, and the control group accepted hot magnet for 2 weeks. They were assessed with Visual Analogue Scale (VAS) and Finger-floor Distance (FFD) before and after treatment. Results The scores of VAS and FFD improved in both groups after treatment (P<0.05), and the difference before and after treatment were more in the treatment group than in the control group (P<0.05). Conclusion The pneumatically ballistic extracorporeal shockwave is more effective on chronic nonspecific low back pain than hot magnet.
10.Robot-assisted Laparoscopic Partial Nephrectomy (Report of 6 Cases)

Objective:To summarize our clinical experience of robot-assisted laparoscopic partial nephrectomy (RALPN), and to discuss its efficacy and safety. Methods:Between December 2007 and October 2008, 6 patients with small exophytie renal masses underwent intraperitoneal robotic partial nephrectomy utilizing the Da Vinci sur-gical system. The perioperative data were collected, and were compared with those performed the same operation in abroad and those performed laparoseopic partial nephrectomy(LPN) with the same team in internal. Results: All the operations were accomplished successfully except the one which required conversion to open nephron-sparing surgery (NSS) due to bleeding occured after the renal artery had been clamped. The mean lesion diameter was 3.2 (2.2-3.6)cm; the mean operative time (not including preoperative set-up time of the Da Vinci surgical system) was 130 (110-160) minutes; the mean warm ischemia time were 40(33-50)minutes; the mean estimated blood loss was 188 (100-380) ml. The patients were ambulant in the 7th postoperative days, and tubes were removaled in 3 days, and mean hospital stay was 9 (8-12)days. Renal function of all patients was in the normal range. Pathology revealed renal cell carcinoma in five, papillary renal cell carcinoma in one. All resection margins were negative. Follow-up ranged from 4 to t5 months, no local residual lesions, local recurrence, incision implantation and dis-tant metastasis were found in all patients. Conclusions.. Robot-assisted laparoscopic partial nephrectomy can be safe-ly performed in selected patients, and it is a feasible approach and a minimally invasive operation for small renal tumors.
