1.Design experiments in the teaching of higher vocational microbiology test

Objectve To study director-leading teaching method,raise the students' main body consciousness and innovation spirit and improve the teaching effect of microbiology test.Method 158 students majored in laboratory medicine of Grade 2013 were divided in two groups.80 from class 1 were in test group,and the rest from class 2 were served as control group.The clinical microbiological specimens were collected and identified;the experimental class was designed with the experimental teaching mode,while the control group used the traditional experimental teaching mode.Through the experimental evaluation and questionnaire survey,the difference between new teaching model and the traditional teaching mode were evaluated.SPSS 21.0 software was used in the collected data for processing.Result The theoretical results of the experimental class were (88.65 ± 7.64) and the practical skills were (89.72 ± 7.69) and the control group were (79.88 ± 10.64) and (80.55 ± 9.06).The scores of the experimental class were significantly higher than those of the control class,and there and statistically significant difference between the scores of two classes.T values were 6.851 and 5.938 and P values were 0.000 and 0.000 respectively.Survey results show that:90% of the students agree with the new teaching methods.Conclusion The teaching result of the new teaching mode is superior to the traditional teaching mode.
2.Up-regulation of Keap-1 inhibits the nuclear import of Nrf-2 and decreases cisplatin resistance in ovarian cancer Skov3/DDP cells

Objective To test the status of Keap-1/Nrf-2 pathway in cisplatin-resistant Skov3/DDP cells,and the contribution of these molecular in cisplatin (cis-pt) resistance.Methods Skov3/DDP was divided into four groups:blank,cis-pt,Keap-1 up-regulated and Keap-1 plus cis-pt.Keap-1 was up-regulated in Skov3/DDP cells by trans-faction with pFlag-CMV-Keap-1 plasmid which was validated by expression of flag and increase of Keap-1 level using Western blot.The effect of Keap-1 up-regulation on Nrf-2 re-distribution was analyzed by Western blot.FCM was employed to detect apoptosis.ROS level was measured by DHE staining.Results The expression of Keap-1 was decreased obviously in Skov3/DDP cells compared with Skov3 cells (P<0.05),Nrf-2 in Skov3/DDP cells detected mainly in nucleus.Then,the Keap-1 was successfully up-regulated (3.5 fold) by exogenous expression of pFlag-CMV-Keap-1 plasmid,as a result,nucleus portion of Nrf-2 was significantly reduced (0.2 fold).Next,the FCM results showed significant increase of apoptosis in Keap-1 up-regulated Skov3/DDP cells after cis-pt administration (P<0.05).Finally,ROS level was significantly raised in Skov3/DDP cells treated with cis-pt plus Keap-1 up-regulation.Conclusions Up-regulation of Keap-1 reduced the nucleus portion of Nrf-2 and reverses cisplatin resistance in Skov3/DDP cells.
3.ALT abnormality among workers from food industry and related people from public places and influencing factors

Objective To explore the influencing factors of ALT abnormality among workers from food industry and related people from public places in Shenzhen and influencing factors.Methods 2,411 workers from food industry and people from public places who had physical examinations in our department from May to October in 2013 were involved in the investigation.ALT abnormality rate and the influencing factors were analyzed.Results The incidence of ALT abnormalities among workers from food industry and related people from public places in Shenzhen was 9.37%.The ALT abnormalities were correlated with censue register,gender,age,marital status,work duration in Shenzhen,education level and monthly income respectively(all P<0.05). Conclusion The ALT abnormality has a higher rate among people from cities,of male gender and old age,with longer work duration in Shenzhen and relatively high level of education and higher monthly income.Therefore,for the population,the health education should be strengthened and the healthy lifestyles should be advocated to effectively reduce the ALT abnormality rate.
4.Discussion on teaching reform of Microbiological Examination under mixed teaching mode

Objective:To reconstruct the curriculum based on the systematization of working process, and discuss the teaching effect of task-driven teaching method in microbiology examination teaching reform under the "student-centered and teacher-guided" mixed teaching mode.Methods:A total of 120 medical laboratory students were selected as research objects, and they were divided into experimental group and control group. The experimental group implemented the reconstructed curriculum content using task-driven teaching method under the mixed teaching mode, and the control group used traditional teaching method. At the end of the course, the questionnaire survey and final examination were combined to evaluate the teaching effect. SPSS 21.0 was used for statistical analysis to evaluate the differences of the teaching methods.Results:The questionnaire survey showed a generally higher evaluation on the teaching effect of the students after the reform, and the performance of the experimental group were significantly higher than that of the control group, with significant differences ( P<0.005). Conclusion:The task-driven teaching under the mixed teaching mode to carry out the teaching form combining theory with practice aimed at strengthening skills training can greatly promote the improvement of students' professional knowledge and skills as well as the cultivation of their professional qualities, so that the course of microbiological examination technology can better meet the requirements of higher vocational medical examination training objectives.
5.Expressions of survivin, COX-2, VEGF and their correlations with angiogenesis in condyloma acuminatum

Objective To investigate the expressions of survivin, cyclooxyenase-2 (COX-2) and vascular endothelial growth factor (VEGF) and their relationship with angiogenesis in condyloma acuminatum (CA) tissues. Methods Immunohistochemistry using PowerVision staining kit was performed to detect the expression of survivin, COX-2 and VEGF protein in 60 CA tissue samples from patients and 21 normal skin samples from the foreskin of human controls. At the same time, the microvessel density was determined in CA tissues by staining blood vessel endothelium with anti-CD105 monoclonal antibody. Results The positivity rate of survivin and COX-2 expression was 56.67% and 63.33%, in CA tissues, 9.52% and 0 in normal skin tissues, respectively. There was a significant difference between the two groups of tissue samples in the positivity rate and intensity of survivin and COX-2 expression (all P < 0.05). VEGF was expressed in all of the CA tissues and normal skin tissues, while the intensity of VEGF expression was statistically different between the two groups of tissue samples (P < 0.05). The MVD was 16.38 ± 5.46 and 0.62 ± 0.44 in CA tissues and normal skin tissues, respectively (P < 0.05). There was a significant positive correlation between the expressions of survivin, COX-2 and VEGF, as well as between MVD and the expressions of survivin and COX-2 in CA tissues. Conclusion The expression levels of survivin, COX-2 and VEGF are significantly higher in CA tissues than in normal skin tissues.
6.Preliminary study on WPBL teaching method in microbiological test teaching

Objective To explore the effect of WPBL teaching method in microbiologicla test teach-ing. Method A total of 120 students majoring in clinical laboratory medicine were divided into the experi-mental group (WPBL teaching) and the control group (the traditional teaching) with 60 in each group in 2015. Results Questionnaire survey showed that in the experimental group, 92.6%of the students believe that WPBL teaching method can stimulate learning interest and promote student self-directed learning, 97.3% believe that WPBL teaching method can improve the ability to analyze and solve problems, 94.7%believe that WPBL teaching method can strengthen teamwork and promote learning exchanges, and 99.6%hold that WPBL teaching method can help them recognize the importance of microbiological tests in the di-agnosis of clinical infectious diseases. The final scores and the usual performance of the experimental group were (75.23 ±11.21) and (24.15 ±1.71), respectively, while those of the control group were (63.42 ±8.21) and (20.26±1.48), respectively. The differences were statistically significant (P<0.005). Conclusions PBL teaching method can fully integrate teaching resources, stimulate students' enthusiasm and initiative, improve students' comprehensive qualities, including the ability to analyze and solve problems, teamwork ability and critical thinking ability, and improve the quality and effect of microbiological test teaching.
7.Change of serum zinc-α2-glycoprotein level before and after methimazol treatment in hyperthyroidism patients with Graves disease

Objective To explore the roles and possible significance of zinc-α2-glycoprotein(ZAG) in Graves disease(GD) by detecting the change of serum ZAG level before and after methimazol(MMI) treatment in the patients with initial onset of Graves disease.Methods The enzyme linked immunosorbent assay(ELISA) was adopted to detect serum ZAG level in healthy population,patients with newly diagnosed GD and GD patients treated by 12-week MMI treatment.The serum thyroid hormones and thyroid related autoantibodies were determined by adopting the electrochemiluminescence immunoassay.The blood lipid levels were examined by enzymatic colorimetry.Furthermore the relationship between serum ZAG level with thyroid function,thyroid related autoantibodies and blood lipid in the patients with initial onset of GD was analyzed.Results The levels of ZAG,FT3,FT4,TT3,TT4,TGAb,TPO and TRAb in the patients with newly diagnosed GD were much higher than those in the age-and gender-matched normal healthy population,while the TSH level was significantly lower than that in the normal control group(P<0.05);after 12-week MMI treatment in the patients with initial onset of GD,the levels of serum ZAG,FT3,FT4,TT3,TT4 and TRAb were markedly decreased,whereas the TSH level was significantly enhanced(P<0.05 or P<0.01).The correlation study showed that fasting serum ZAG level in the GD patients was positively correlated with FT3,FT4,TT3 and TT4(P<0.05);and negatively correlated with TSH,FFA,TC and TG(P<0.05).With fasting serum ZAG in the GD patients as the dependent variable,the multiple linear regression analysis showed that TSH,TC and TG were the independent related factors of plasma ZAG level.Conclnsion The serum ZAG level is closely correlated with the occurrence and development of GD,and might play an important role in the blood lipids metabolism of GD patients.
8.Value of transvaginal three-dimensional ultrasound in evaluating the curative effect of Yangmo decoction in the treatment of uterine adhesion.

OBJECTIVES:
Intrauterine adhesions (IUA) is the damage of the basal layer of the endometrium caused by various reasons, resulting in adhesion of the uterine muscle walls to each other, which is manifested as clinical symptoms such as spanomenorrhea, amenorrhea, and infertility. Hysteroscopic adhesiolysis (HA) is the main treatment, for patients with moderate or severe adhesion or angular adhesion, the incidence of postoperative adhesion is high. Traditional Chinese medicine "Yangmo decoction" can promote endometrial growth. Three-dimensional transvaginal ultrasonography (3D-TVUS) can judge IUA and evaluate uterine receptivity through three-dimensional imaging. This study aims to investigate the value of 3D-TVUS in judging the efficacy of Yangmo decoction in the treatment of intrauterine adhesions.
METHODS:
The clinical data of patients who underwent HA at two different centers in department of Gynecology of Third Xiangya Hospital of Central South University and Changsha Jiangwan Hospital from January 2021 to August 2021 were retrospectively collected. A total of 275 eligible patients were included. According to the postoperative management measures, the selected patients were divided into two groups. Yangmo decoction group (n=138): the use of Yangmo decoction and uterine-shaped silicon stent post HA; Hormone group (n=137): the use of estrogen, progesterone and uterine-shaped silicon stent post HA. The preoperative general data, preoperative and postoperative 3D-TVUS parameters of the two groups were analyzed.
RESULTS:
The endometrial thickness of Yangmo decoction group was thicker than that of hormone group (P<0.001), the intercornual distance was wider (P=0.016), the endometrial echo was more homogeneous (P=0.018), the percentage of bilaterally visible tubal opening was higher (P<0.001), the endometrial morphology was better (P=0.012), and endometrial blood flow, endometrial motility and uterine motion were better in Yangmo decoction group than that in the hormone group (all P<0.001).
CONCLUSIONS
The endometrial thickness, echo, blood flow, and peristalsis, the number of visible tubal opening, uterine motion, and the intercornual distance obtained by 3D-TVUS examination are important factors to evaluate the prognosis of postoperative drug treatment for IUA. 3D-TVUS is of great significance in evaluating the efficacy of Yangmo decoction in the treatment of IUA.
