1.Three years retrospective clinical evaluation of endodontically treated premolars restored by cast ceramic onlays.

OBJECTIVEThis study aimed to evaluate the success and survival rate of endodontically treated premolars restored by ceramic onlays by comparing restored by quartz fiber posts and metal ceramic crowns.
METHODSNinety-four patients with 126 endodontically treated premolars were enrolled in this study and divided into 4 groups according to the remaining axial walls and restorative methods. The observation time was 36 months. The success and survival rates of the restorations, as well as causes of failures, were analyzed.
RESULTSAt the final follow-up, the success and survival rates of the mild defect endodontically treated premolars were restored by quartz fiber posts and the crowns were at 96.3% and 98.1%; the success and survival rates of the severe defect premolars restored by quartz fiber posts and crowns were at 88.5% and 96.2%. The success and survival rates of the mild defect premolars restored by cast ceramic onlays were both at 96.6%, the success and survival rates of the severe defect premolars restored by cast ceramic onlays were at 94.1% and 100%, respectively. The success and survival rates of the different groups were no significant difference (P>0.05).
CONCLUSIONBased on the results and within the limits of this study, cast ceramic onlays is a very reliable method to restore endodontically treated premolars.

2.Determination of Content and Encapsulation Efficiency of Genistein-loaded Eudragit Nanoparticles by HPLC

Objective:To establish an HPLC method for the determination of genistein in genistein-loaded Eudragit nanoparticles. Methods:The HPLC analysis was carried out on a DiamonsilTM C18(250 mm ×4.6 mm,5μm)column with the mobile phase of meth-anol-water (85∶15) at the flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 261nm and the column temperature was 25℃. Results: A good separation was displayed between the drug peak and the excipient peaks. An excellent linearity range for genistein was obtained from 1.0 μg·ml-1 to 100.0 μg·ml-1(r =0.999 8). The recovery of genistein was 99.85%(RSD =0. 65%,n=9). Conclusion:The method is simple, rapid, accurate and specific, which is suitable for the determination of content and encapsulation efficiency of genistein-loaded nanoparticles.
3.Effect of different ages on sufentanil pharmacokinetics in patients undergoing cardiac valve replacement

Objective To assess the effect of different ages on sufentanil pharmacokinetics in patients undergoing cardiac valve replacement.Methods Sixteen NYHA Ⅱ or Ⅲ patients undergoing selective cardiac valve replacement were randomly divided into two groups:elderly group (aged 65-69 years,group Ⅰ,n =8) and young adult group (aged 36-45 years,group Ⅱ,n =8).Intravenous and intra-arterial cannulae were placed.Sufentanil 5 μg/kg injected intravenously during anesthesia induction.Blood samples from the radial artery were obtained at 1,3,5,10,20,30,60,120,180,240 and 360 min after sufentanil injection (3 ml each).Then 1 ml plasma was immediately separated from the 3 ml blood sample and stored at - 80 ℃ until being assayed.Plasma sufentanil concentration was determined with liquid chromatography mass spectrometry and pharmacokinetic parameters were calculated with 3P97 pharmacologicl program.Results Plasma sufentanil concentration versus the time decay curve in patients undergoing cardiac valve replacement before cardiopulmonary bypass (CPB) was fitted to a twocompartment model and could be expressed by bi-exponential equations:Cp (t) =27.4 e-0.41t + 3.2 e-0.029t in group Ⅰ and Cp (t) =14.4 e-0.51t +3.4 e-0.032t in group Ⅱ,respectively.There were significant differences in t 1/2 α,t1/2 β and CL between the two groups ( P < 0.05 or 0.0l ).During CPB,plasma sufentanil concentration versus the time decay curve in patients undergoing cardiac valve replacement was fitted to a three-compartment model and could be expressed by tri-exponential equations:Cp(t) =22 e-0.51t + 3.5 e-0.045t + 0.21 e-0.0029 t in group Ⅰand Cp(t) =15 e-0.52t + 3.9 e-0.048t + 0.32 e-0.004t in group Ⅱ,respectively.There were no significant differences in pharmacokinetic parameters between the two groups ( P > 0.05).Conclusion There are significant differences in sufentanil phannacokinetic characteristics before CPB but different ages have no obvious influence on sufentanil pharmaco-kinetics during CPB.
4.Transgenic mouse fetus generated from embryonic stem cells by tetraploid embryo complementation

Objective To use tetraploid embryo complementation combined with gene transfer to produce genetically modified embryonic stem cells (EsCs) clones. Methods In this study, EGFP was introduced into ESCs by electroporation, and transfected positive cells were selected by G418 resistance. The tetraploid embryos were obtained from diploid blastomere electrofusion which preformed at 2-cell stage. Afterwards, 19-21 EGFP-ESCs were inserted into each tetraploid blastocyst cavity by piezo drilled microinjection,then the injected blastocysts were transferred into the uterus of pseudo-pregnancy at 2.5-day or the oviduct of 0.5-day female mice. Results The transfected ESCs maintained normal karyotype even after long-term passage (2n=40). The rate of fusion was 95.07%, and the developmental rate of tetraploid blastocyst was 95%.Totally 410 injected blastocysts were obtained. Unfortunately, we have not got any vital offsprings, except 151 implantation sites (pseudo-pregnancy 2.5 days:29.41%;the oviduct of half one day:64.37%). Furthermore, scattered EGFP expressions in transgenic fetus were observed under invert fluorescent microscope. Conclusion The transfected ESCs were observed in transgenic fetus, and the implantation rate in oviduct was higher than that in uterine.
5.Clinical Observation of Exenatide Combined with Clomiphene Citrate in the Treatment of Polycystic Ova-ry Syndrome with Insulin Resistance

OBJECTIVE:To observe clinical efficacy and safety of exenatide combined with clomiphene citrate in the treatment of polycystic ovary syndrome with insulin resistance. METHODS:98 patients with polycystic ovary syndrome complicated with in-sulin resistance were randomly divided into control group (49 cases) and observation group (49 cases). Control group was given Clomiphene citrate capsule 50 mg orally,once a day,for 5 d+Metformin enteric-coated tablet with initial dose of 0.25 g orally, twice a day,adjusted to 0.50-0.75 g orally,twice a day,for 3 menstrual cycles. Observation group was given Clomiphene citrate capsule(usage and dosage same as control group)+Exenatide injection 5 μg subcutaneously,twice a day,adjusted to 10 μg subcu-taneously,twice a day,for 2 months. Clinical efficacies of 2 groups were observed as well as the levels of LH,FSH,LH/FSH and IR before and after treatment,ovulation and pregnancy of infertility patients after treatment. The occurrence of ADR was record-ed. RESULTS:Total response rate,ovulation rate and pregnancy rate of observation group were significantly higher than that of con-trol group,with statistical significance(P<0.05). Before treatment,there was no statistical significance in the levels of LH,FSH, LH/FSH and IR between 2 groups (P>0.05). After treatment,the levels of LH,LH/FSH and IR in 2 groups were significantly lower than before,and the observation group was significantly lower than the control group,the levels of FSH in 2 groups was sig-nificantly higher than before,and the observation group was significantly higher than the control group,with statistical significance (P<0.05). There was no statistical significance in incidence of ADR between 2 groups(P>0.05). CONCUSIONS:Exenatide com-bined with clomiphene citrate shows significant therapeutic efficacy for polycystic ovary syndrome complicated with insulin resis-tance and can increase ovulation rate and pregnancy rate through improving insulin resistance,but doesn't increase the occurrence of ADR.

7.The clinical study on the use of diode laser irradiation in the treatment of periodontal-endodontic combined lesions.

OBJECTIVETo evaluate the adjunctive effect of diode laser irradiation for the treatment of periodontal-endodontic combined lesions.
METHODS30 cases with periodontal-endodontic combined lesions were selected. All cases were randomly divided into experimental group and control group (n=15). In the control group, patients were treated with root canal therapy and conservative periodontal treatment (scaling and root planning), and patients in the experimental group were treated with root canal therapy and conservative periodontal treatment followed by diode laser irradiation. Clinical measurements including pocket probing depth (PPD), clinical attachment level (CAL), modified bleeding index (mBI), periapical index (PAI) were taken before treatment (baseline). Three and six months after the treatment, the adjunctive effects of diode laser in the treatment of periodontal-endodontic combined lesions were evaluated.
RESULTSThree months after the treatment, the average PPD, CAL and mBI levels were reduced significantly in both groups. But the average PPD reduction in the experimental group was significantly greater (P<0.05) than in control group. However the average CAL and mBI reduction were not significantly different between two groups. Six months after the treatment, the average PPD, CAL and mBI levels were further reduced in the experiment group, but not in the control group. The average PAI levels were reduced significantly in both groups, but the PAI reduction was not significantly different between two groups.
CONCLUSIONThe use of diode laser as an adjunct procedure in the treatment of periodontal-endodontic combined lesions can aid in achieving success.

8. Research of CircRNAs in the diagnosis and treatment of common orthopedic diseases

Circular RNA is a special kind of endogenous non-coding RNA which is widely existing in eukaryotic cells. More studies confirmed that most circRNAs were produced in the process of RNA shearing and played a vital role in the occurrence and development of many diseases. Though there was not much research on circRNAs related to orthopaedic diseases in recent years, study found that circRNAs played a great significant role in the pathogenesis, diagnosis, target therapy of degenerative diseases(osteoarthritis, lumbar degenerative disease), metabolic diseases(osteoporosis) and motor system tumor(osteosarcoma). This study summarizes the research progress of circRNAs in orthopaedic diseases.
9.Porphyromonas gingivalis FimA genotype distribution among periodontitis patients with type 2 diabetes

Objective To investigate the distribution differences of Porphyromonas gingivalis(Pg) FimA genotypes between periodontitis patients with and without type 2 diabetes for a better understanding of the relationship between diabetes and periodontitis.Methods Questionnaires and detailed periodontal examinations(6 sites per tooth) were conducted in 80 subjects with moderate-severe chronic periodontitis in the Department of Periodontology, Peking University School and Hospital of Stomatology.There were 40 type 2 diabetic patients and 40 systemicly healthy patients enrolled respectively.The periodontal parameters including plaque index(PLI), probing depth(PD), bleeding index(BI), attachment loss(AL) by 6 sites per tooth and numbers of missing teeth were also recorded.Pooled subgingival plaque samples using pocket method with Whatman No3 filterpaper were collectedat each of the 6 sites from one incisor and one molar.Pg and its FimA genotype distributions were investigated using DNA extrctedfrom plaque samples by PCR.Results Diabetic patients had a significantly higher score of PLI[2.35(0.58) vs 1.64(0.76), P<0.05] , while rest of periodontal indexes observed(PD, BI and AL) did not differ significantly between diabetic patients and systemicly healthy controls(P>0.05).The detection rate of Pg did not show statistically significant difference between the two groups(50% vs 60%, P>0.05).However, the proportion of FimA Ⅱ was significantly higher in diabetic group than systemicly healthy group(80% vs 42%, P<0.05).Conclusions Type 2 diabetic patients were prone to be infected by highly virulent strains of Pg: FimA Ⅱ Pg.

Objective:
The therapeutic effect of acid fibroblast growth factor 1(FGF1) on rats with diabetic cardiomyopathy (DCM) was evaluated by using nano-liposomes combined with ultrasound-targeted microbubble destruction technique (UTMD).
Methods:
The FGF1-loaded nano-liposomes were prepared by water-in-water emulsion method combined with lyophilization technique.TypeⅠdiabetes model was induced by intraperitoneal injection of streptozotocin (STZ, 70 mg/kg) in 60 male SD rats.Sixteen weeks later, diabetic rats were randomly divided into: placebo group (saline treatment), FGF1 group, FGF1-loaded nano-liposomes group, and FGF1-loaded nano-liposomes plus UTMD group (
