1.Effects of PDCD5 on hypoxia/reoxygenation-induced autophagy and apoptosis of cardiomyocytes

AIM:To investigate the influence of programmed cell death 5 (PDCD5) on apoptosis and autoph-agy in the cardiomyocytes exposed to hypoxia/reoxygenation ( H/R) and its potential mechanism .METHODS:H9c2 cells were exposed to H/R.PDCD5 was downregulated by RNA interference .The cell viability was measured by MTT assay . TUNEL assay was used to detect cell apoptosis .The mRNA and protein levels were determined by RT-qPCR and Western blot.RESULTS:The expression of PDCD5 was upregulated in the cardiomyocytes after H/R injury.Furthermore, H/R injury obviously reduced the cell viability and enhanced the apoptosis and autophagy of the cardiomyocytes .However, knockdown of PDCD5 increased the cell viability , and attenuated H/R-induced apoptosis , accompany with reduction of Bax and augment of Bcl-2 expression .Additionally , silencing PDCD5 markedly inhibited H/R-induced autophagy by regulating the expression of LC3-II/LC3-I and beclin-1.Moreover, downregulation of PDCD5 suppressed NF-κB signaling by redu-cing the protein level of p-P65.CONCLUSION: Silencing PDCD5 suppresses H/R-induced H9c2 cells apoptosis and autophagy by blocking NF-κB signaling pathway .The result indicates a new strategy for the prevention and treatment of myocardial I/R injury.
2.Multidimensional spatial triangular area as an index for the evaluation of the release-absorption correlation of multiple component traditional Chinese medicines

The paper is aimed to provide a novel index, named as multidimensional spatial triangular area, for the evaluation of the release-absorption correlation of multiple component traditional Chinese medicines (TCMs). The applicability of the method was demonstrated by the example data. The method and standard practice for evaluation of the release-absorption correlation for western medicines with single compound could not be applied to TCMs with multiple components. The release percentage or absorption percentage of the multiple components for TCMs at the sampling time was a point in the multidimensional space. The area of the triangle formed byt the sequential three points rrepresented the changing characteristics of the components' release and absorption kinetics. The side lengths of the triangle could be calculated from the spatial distances between each two of the sequential three points. Then the triangle area could be obtained by the side lengths. The in vitro release-in vivo absorption correlation of the multiple components could be represented by the correlation between the integrating values of the release triangle areas and that of the absorption triangle areas. The results of the examples indicated that the multidimensional spatial triangular area method could treat the multiple components in a holistic way, in line with the holism the hi he TCMs. Therefore, the multidimensional spatial triangular area method provided new methodology for the release-absorption correlation of the TCMs with multiple components.
3.The comparative study of the clinical effect for different surgical approach to treat children with supracondylar fractures

Objective To investigate the clinical effect of tongue-shaped flap cut into the triceps and triceps both sides of the road approach road into the way of two surgical treatments of supracondylar fractures in children.Methods 90 patients with pediatric supracondylar fractures were chosen.50 cases of tongue-shaped flap to triceps road cut into were set as A group,40 patients taking brachial three muscles on both sides of the first approach were set as the B group.The clinical effects of the two groups were compared.Results The operative time,blood loss,cast immobilization time in B group was superior to group A ( P < 0.05 ),there was statistically different.The pain score improvement in group B was significantly better than group A before and after treatment(P <0.05),the difference was statistically significant.After treatment,A group of patients with excellent results was in 22 cases,good in 24 cases,the fine rate was 92%.B group with excellent results was in 23 cases,good was in 16 cases,the good rate was 97.5%.The superior quality of the treatment effect in B group was better than A group( P < 0.05),the difference was statistically significant.Conclusion The operative time,blood loss,time cast immobilization,pain improvement,the treatment effects for triceps both sides of the approach for the treatment of supracondylar fractures in children are better than the triceps tongue-shaped flap cut into the road,and there is high clinical value.
4.Percutaneous minimally invasive techniques in the treatment of cervical disc herniation

ObjectiveTo compare the therapeutic effects of percutaneous cervical discectomy (PCD group),percutaneous cervical disc nucleoplasty(PCN) and the association of them (PCDN) for the treatment of cervical intervertebral disk displacement and instability of cervical vertebral column.Methods From February 2003 to April 2011,171 consecutive patients with cervical disc herniation have presented at the authors' hospital and were retrospectively studied.The average age of patients was 47.8 years(ranged,21-74).Ninety-seven cases were treated with PCD,50 cases with PCN,and the other 24 cases with PCDN.Clinical result and the stability of cervical vertebral column after operation were evaluated and compared among the 3 groups.ResultsAll cases had been followed up for a median of 4.1 years.There was significant difference in the pre- and post-operation the Japanese Orthopaedic Association(JOA) scoring system on within 3groups (PCD:t=21.85,P<0.05; PCN:t=14.50,P<0.05; PCDN:t=8.56,P<0.05).All cases had been successfully operated.There was no significant difference between groups among the 3 groups in terms of the clinical outcomes(The recovery rate of JOA standard evaluation,F=2.19,P=0.12).According to Odom criteria,the excellent and good rate are as follows:81.35% in PCD,82.44% in PCN,83.19% in PCDN,respectively.There was no significant difference between groups among the 3 groups in terms of the clinical success rate (P>0.05).There was no instability of cervical vertebral column cases in 3 groups after operation(P>0.05),and no significant difference was found in terms of cervical vertebral column stability in pre- and post-operation in each group.ConclusionAll the three operations including PCN,PCD and PCDN are safe,minimally invasive spine surgery for the treatment of cervical intervertebral disk displacement; they achieve good clinical outcomes and there are no difference on the stability of cervical vertebral column between preoperation and postoperation.
5.Clinical Analysis of 63 Cases of Myocardial Bridge

Objective To evaluate the significance of myocardial bridge and find a reasonable diagnosis and treatment strategy.Methods Sixty-three myocardial bridge patients and sixty-three patients with negative results of coronary artery angiography were reviewed.The clinical data of symptoms,electrocardiogram,exercise tests,coronary artery angiography,therapeutics and the serum levels of C-reactive protein(CRP)were analyzed.Results The symptoms of chest distress and chest pain were found in myocardial bridge patients.Myocardial consumption of oxygen augmentation causes the symptoms of aggravation.Positive results of electrocardiogram and exercise tests in many of myocardial bridge patients were examined.There were no relationship with severity of myocardial bridge artery stenosis.Most of myocardial bridge were discovered in anterior descending branch.At present,the main treatment of myocardial bridge was drug therapeutics.After treatment,the serum levels of CRP was significantly decreased.Conclusion Myocardial bridge was anatomy abnormality with important clinical significance.The serum levels of CRP can be used to evalue the therapeutic efficacy of myocardial bridge.
6.Preventive effect of Polygonum Multiflorum on deteriorated micro-structure and biomechanical properties induced by prednisone

Aim To investigate the preventive effect of Polygonum Multiflorum (PM)on the deteriorated mi-cro-structure and biomechanical properties induced by prednisone.Methods Ninety 6-month-old male Sprague-Dawley rats were randomly divided into nine groups,which were control,prednisone,CAL,30%ethanol eluent of the PM(H,M,L),PM(H,M,L). Prednisone was gavaged to rat for 21 weeks as model group of osteoporosis.Meanwhile,tested herbal ab-stract were orally administrated to the modeled rats in-duced by prednisone.At the end of the experiment, the right femur was collected for micro-CT scanning, three-dimensional reconstruction and biomechanical test.Results Compared with the control group,mod-el group showed destruction of bone microarchitecture, BV /TV fell 28.6%(P <0.05),bone biomechanical parameters decreases,and stiffness fell 29.7%(P <0.01 ). Compared with the model group, positive group had significantly improved effect on bone micro-architecture,and biomechanical parameters,BV /TV increased 46.7%(P <0.01 ),and stiffness increased 25.9%(P <0.01 ).30% ethanol eluent of the PM (M,L)dose may improve bone microstructure by in-creasing BV /TV 46.7% (P <0.01 ),40.0% (P <0.05)respectively,PM(H)may improve the biome-chanical parameters by increasing stiffness 24.7%(P<0.05),and 30% ethanol eluent of the PM(H)and PMhigh-dose may improve the biomechanical parame-ters,but not as positive group.Conclusions Predni-sone reduces biomechanical properties of rat femur and deteriorates femoral microstructure.30% ethanol eluent of the PM(M,L)and PM(H)plays a preventive role in the changes of micro-structural and biomechanical properties by prednisone,and increases BMD,whereas other groups have no significant preventive effect.

8.Association between oxidative stress and pancreatic cancer

Pancreatic cancer (PC) is often asymptomatic in the early stage and most patients have progressed to the advance stage at the time of diagnosis.At present,chemotherapy is mainly used in patients with advanced PC,but PC patients have poor response to chemotherapy,which brings great challenges to the treatment of PC.Oxidative stress-induced reactive oxygen species (ROS) can cause damages in DNA,proteins,and lipids and produce the toxic and mutagenic metabolites that alter the biological behavior of tumor and transform the tumor into a malignant phenotype.Antioxidants have an antitumor effect and provide a basis for the design of anti-oxidative stress drugs for the treatment of PC.This article summarizes the research advances in the molecular pathways associated with oxidative stress and PC,in order to explore new ROS-targeted methods for the treatment of PC.

10.Discussion on the peacetime and wartime management of emergency medical materials in public health emergencies

As an important part of the construction of national emergency management system, emergency material support is the material basis and important guarantee for the smooth implementation of emergency work. In the face of the outbreak and spread of COVID-19, China′s public health emergency management system and emergency management system have achieved certain results in epidemic prevention and control, but also exposed some shortcomings. The authors summarized the management experience and existing problems of China′s emergency medical materials in storage, production, transportation and deployment during the COVID-19 epidemic, and discussed the management path and method of emergency medical materials in peacetime and wartime, which should be jointly built by the government, society, enterprises and the public. The following suggestions are put forward: multi agent participation in expanding the scope and scale of emergency medical supplies reserve, refining and adjusting the inventory and structure of emergency medical supplies reserve; establishing a supply chain list of emergency medical supplies to ensure the " wartime" production capacity; establishing a comprehensive management information platform for medical materials with " peacetime and wartime integration" ; and improving the awareness and ability of community and family′s " peacetime" medical material reserve.
