1.ORIGINS OF THE NADPH-d POSITIVE TERMINALSIN THE MEDIAL AMYGDALOID NUCLEUS OF THE RATS

Objective To estimate the effect of the NO on the medial amygdaloid nucleus(Me), we studied the origins of the NOS positive terminals in the Me. Methods Noergic afferent projection to Me was identified by a combined NADPH-d histochemical staning and retrograde CTb immunocytochemical method after microinjecting CTb into Me. Results The double labeled of neurons (NOS and CTb) were located in dorsal raphe nucleus, locus ceruleus, basolateral amygdaloid nucleus, parabrachial nucleus, ventrolateral part of periaqueductal gray.Conclution The NADPH-d positive terminals in the Me originates from the aforementioned nucleus, and may relate to the function of the Me.
2.Effect of play therapy-based group counseling on behavior disorders and mental coping of school-age nephrotic syndrome children

Objective:To investigate the effect of play therapy-based group counseling on behavior disorders and mental coping of school-age nephrotic syndrome children.Methods:A total of 140 school-age nephrotic syndrome children were randomly divided into experiment group ( n=70) and control group ( n=70). Children in the control group recieved routine care, while experiment group carried out play therapy-based group counseling based on the routine care. With the Conners Parent Symptom Questionnaire (PSQ) and Coping with a Disease (CODI), the overall curative effect was assessed before and after intervention by their scores. Results:After intervention, the incidence rate of behavior disorder of nephrotic syndrome children in the experiment group was 18.0% (11/63), lower than in the control group 33.8% (23/68), the difference was statistically significant ( χ2 value was 4.131, P=0.042). The conduct, psychosomatic disorder and hyperactivity index scores in the experiment group were (1.12±0.23), (0.24±0.08), (1.22±0.32) pionts, lower than (1.25±0.31), (0.29±0.09), (1.39±0.37) pionts in the control group, the difference was statistically significant ( t values were 2.625, 2.950, 2.825, P<0.05); the scores of acception, escape, negative feelings and total CODI scores were (12.87±3.19), (22.51±5.24), (25.83±4.51), (101.23±11.48) pionts, higher than (11.27±3.77), (20.41±4.00), (24.13±4.38), (95.38±8.33) pionts in the control group, the difference was statistically significant ( t values were 2.191-3.361, P<0.05). Conclusion:Play therapy-based group counseling can effectively alleviate behavior disorders and improve mental coping of school-age nephrotic syndrome children.
3.Relationship between acceptance of disability and posttraumatic stress response in patients with brachial plexus injury

Objective To determine the relationship between acceptance of disability and posttraumatic stress response in patients with brachial plexus injury. Methods Total 160 patients with brachial plexus injury were recruited and investigated with the Impact of Event Scale-Revised (IES-R) and the Acceptance of Disability Scale (ADS). Results The total score of ADS was (79.07±11.99) which showed medium level of acceptance of disability. The total score of IES-R was 4-66 (33.51±14.41), which showed that most of the patients suffered from posttranmatic stress response. Significantly negative correlation was found between acceptance of disability and posttraumatie stress response(r=-0.480, P<0.001). Conclusions Nurses should pay more attention to the acceptance of disability and posttranmatic stress response in patients with brachial plexus injury, and provide appropriate health education and effective psychological intervention to improve the patients' mental health and quality of llfe.
4.Improvement of Content Determination for Polygala XanthoneⅢin Polygalae Radix

OBJECTIVE:To improve the content determination for polygala xanthone Ⅲ in Polygalae Radix contained in Chi-nese Pharmacopoeia (2010 edition). METHODS:HPLC was performed on the column of Hypersil BDS C18 with mobile phase of acetonitrile-0.05% phosphoric acid(gradient elution)at a flow rate of 1.0 ml/min;detection wavelength was 320 nm,column tem-perature was 25℃,and the injection volume was 10μl. RESULTS:The linear range of polygala xanthoneⅢwas 0.029-0.928 μg/ml (r=0.999 1);RSDs of precision,stability and reproducibility tests were lower than 2.0%;recovery was 94.66%-100.90%(RSD=2.46%,n=6). CONCLUSIONS:The method is simple,stable and reproducible. Although the determination time is prolonged,it has improved the accuracy and it is more suitable for the content determination of polygala xanthoneⅢin Polygalae Radix.
5.Role of occult hepatitis B virus infection in the pathogenesis of mul-tiple myeloma

Objective:To explore the relationship between hepatitis B virus (HBV) infection and the pathogenesis of multiple my-eloma (MM), in order to provide an epidemiological evidence for the prevention and treatment of MM. Methods:Clinical and epidemi-ological data of 185 MM patients and 182 non-tumorous patients were collected. Subjects were randomly selected from in-patients who were homeochronously admitted to the same five grade-III A hospitals, including Xi'an Central Hospital, Shaanxi People's Hospital, Xi'an Jiaotong University Xibei Hospital, and so on. MM patients were selected in terms of age and gender. Peripheral blood HBsAg was assayed by ELISA. If HBsAg was negative, the S and C-gene fragments of HBV DNA were tested using nested PCR . Fisher's ex-act test orχ2 test (SPSS statistical software) was used to compare the differences between the groups. Logistic regression was employed to examine the association between the pathogenesis of MM and HBV infection. Results:In MM patients, the HBsAg positive rate was 8.11% (15/185), the occult HBV infection (OBI) positive rate was 3.53% (6/170), and the total HBV infection rate was 11.35% (21/185). For the control group, the HBsAg positive rate was 4.40%(8/182), the OBI positive rate was 0.57%(1/174), and the total HBV in-fection rate was 4.95%(9/182). No statistical difference in HBsAg or OBI positive rate was found between the two groups (P>0.05). However, MM patients showed significantly higher total HBV infection rate than that of the controls [χ2=5.02, P<0.05;OR was 2.46 (95%CI:1.10-5.53, P<0.05)]. Additionally, the proportion of ISS stage III was significantly higher in MM patients with HBV infection than in uninfected MM patients (85.71%vs. 60.37%,χ2=5.15, P<0.05). Patients with HBV infection showed reduced albumin level (χ2=5.60, P<0.05) and aκ/λlight chain ratio (P<0.05) compared with uninfected patients. Conclusion:The risk of MM pathogenesis after HBV infection is significantly increased as OBI is included in the analyses. Furthermore, MM patients with HBV infection will develop more severe liver damage, indicating that OBI in MM patients with negative HBsAg should be screened before chemotherapy to pre-vent HBV reactivation.

OBJECTIVETo identify the mutation in the keratin 6A(K6A) gene in a pedigree with pachyonychia congenita type I (PC-I).
METHODSBlood samples were obtained from 2 affected, 3 unaffected members in this family, and 100 unrelated healthy individuals. Mutation detection was carried out by PCR amplification of the K6A gene and direct DNA sequencing.
RESULTSA heterozygous mutation of T--> C transition at position 521 in exon 1 of the K6A gene was found in the 2 affected, but not in the unaffected members and 100 unrelated healthy individuals.
CONCLUSIONThe mutation of 521T--> C in the K6A gene is the disease causing mutation in this PC-I family.

8.In vivo and in vitro MR imaging of glioma targeted block copolymer micelles

Objective To explore the in vivo/in vitro MR imaging effect of Angiopep2 modified glioma targeted block copolymer micelles(ANPs/CPT). Methods Angiopep2 modified DOTA-Gd conjugated polymeric micelles based on beta-cyclodextrin(β-CD) were successfully synthesized.Water proton longitudinal relaxation rate (1/T1) were calculated to compare the r1of DOTA-Gd and ANPs/CPT. Different concentration of DOTA-Gd, NP/CPT, ANP/CPT were co-cultured with C6 cells, the intracellular content of Gd3+was analyzed by ICP-AES;The signal intensity(SI) and contrast to noise ratio(CNR) were measured by 1.5 T MR;The CNR of ANPs/CPT was evaluated on different organs of normal rats. The enhancement feature of ANP/CPT in glioma as well as T/N and CNR between tumor and normal brain tissue were also investigated.Several comparisons were made between the 3 groups by one-way ANOVA,and the difference were significant when P<0.05.Results The r1of ANPs/CPT(13.900 s-1·mmol/L-1)was 3 times as much as DOTA-Gd(4.927 s-1·mmol/L-1).C6 cells uptake different concentrations of Gd3 +had significant difference among groups(F=362.502,1636.136,386.880,918.173, P<0.001).The difference of SI and CNR among groups were significant(F=55.240,155.419,P<0.001).The CNR of the brain tissue had significant difference with other organs at all times after tail vein injection of ANPs/CPT(F=9.417,21.808, 21.383,107.318,178.762,P<0.01).The ANPs/CPT showed excellent brain-targeting efficiency and theT/N ratio(30 min after),CNR in different time point had significant difference among groups (F=5.349,6.594,24.078,18.508 and 6.840,6.780,5.895,12.620,37.139,368.893, P<0.05).Conclusions ANPs/CPT shows nice agnetic relaxation performance,extended blood circulation duration,excellent brain-targeting efficieney, which will render the theranosticnano-carriers with important reference for the construction of new generation multifunctional nanomedicine.

Objective:To explore the clinical effect of moderate exercise therapy and fracture liaison service in osteoporotic hip fracture.Methods:From June 2018 to June 2020, 120 patients with osteoporotic hip fracture admitted to Nanjing Hospital Affiliated to Nanjing Medical University were selected. The patients were divided into study group and control group by random number table method, 60 cases in each group. The control group was treated with routine nursing and moderate exercise therapy, and the control group was treated with fracture liaison service, clinical observation and comparison of treatment compliance, bone mineral density (BMD), Numerical Rating Scale (NRS) score and nursing satisfaction before and after intervention.Results:The BMD and NRS scores in the study group were (0.90±0.25) g/cm 2, (1.0±0.3) points, higher than those in the control group (0.80±0.18) g/cm 2, (1.0±0.25) points. The difference was statistically significant ( t values were 2.514, 2.547, P<0.05). The total compliance rate and nursing satisfaction of the study group were 93.30% (56/60) and 95.00% (57/60) respectively, which were significantly higher than 80.00% (48/60) and 83.33% (50/60) of the control group. The difference was statistically significant ( χ 2 values were 4.615, 4.227, P<0.05). Conclusion:Moderate exercise therapy and fracture contact service intervention can effectively improve bone mineral density and total compliance rate, relieve pain and improve nursing satisfaction.
