3.Relationship between transforming growth factor-β1 and interleukin-10 single nucleotide polymorphism and susceptibility of recurrent aphthous ulcer.

OBJECTIVETo explore the possible relationship between recurrent aphthous ulcer (RAU) and single nucleotide polymorphism (SNP) of transforming growth factor-β1 (TGF-β1)-509T/C and interleukin-10 (IL-10)-1082A/G sites.
METHODSA total of 138 RAU patients were recruited for this study. The control group consisted of 124 subjects. TGF-β1-509T/C and IL-10-1082A/G sites were detected by restriction fragment length polymorphism-polymerase chain reaction (RFLP-PCR) and sequence specific primer-polymerase chain reaction (SSP-PCR). Relative risk ratios were estimated by odds ratios (OR) and 95% confidence interval (95%CI).
RESULTSSignificant differences were found in the genotype frequencies or allele frequencies of TGF-β1-509T/C and IL-10-1082A/G sites between the RAU patients and controls (P < 0.05). CT genotype (OR = 1.231, 95% CI = 0.702-2.160), TT genotype (OR = 2.482, 95% CI = 1.250-4.927), and T allele (OR = 1.465, 95% CI = 1.036-2.074) at the TGF-β1-509 site exhibited high risks. AG genotype (OR = 1.391, 95% CI = 0.808-2.396), GG genotype (OR = 4.165, 95% CI = 1.944-8.924), and G allele (OR = 2.134, 95% CI = 1.474-3.089) at the IL-10-1082A/G site also showed high risks.
CONCLUSIONTGF-β1-509T/C and IL-10-1082A/G sites are associated with the risk of RAU. The TGF-β1 gene-509T allele and IL-10 gene-1082G allele may serve as genetic determinants for RAU.

4.International Experience of Chronic Disease Management and Enlightenment

OBJECTIVE:To provide reference for comprehensive intervention and management of chronic disease in China.METHODS:The global chronic disease trends and disease burden were summarized;theoretical framework,practice and experience of international chronic disease management were summarized and analyzed as well as enlightenment on domestic chronic disease management.RESULTS&CONCLUSIONS:Worldwide prepresentative chronic disease theory model mainly involved USA chronic disease nursing model and WHO innovation care for chronic conditions.Main experience of international chronic disease management is that managing based on community,confirming preferential intervened disease types,adopting standardized clinical diagnosis and treatment pathway,designing rational transfer treatment system,providing patient self-management support.At pres ent,chronic disease management have been improved in China,but is still poor in community management.It is necessary to strengthen community medical staff training about chronic disease prevention and treatment and health education for social group.
5.Relationship between IFN-γ+874 A/T site and IL-2-330 T/G site single nucleotide polymorphism with the susceptibility of recurrent aphthous ulcer

Objective:To explore the possible relationship between recurrent aphthous ulcer( RAU) and the single nucleotide polymor-phism(SNP)of IFN-γ+874A/T site and IL-2-330T/G site. Methods:138 RAU patients and the 124 health control subjects were in-cluded. IFN-γand IL-2 serum concentration were detected by ELISA. IFN-γ+874A/T site and IL-2-330T/G site of the whole blood were detected by RFLP-PCR and SSP-PCR. Results:IFN-γand IL-2 levels in serum of the patients were significantly lower than those of the controls(P<0. 05). There was significant difference in the genotype frequencies and allele frequencies of IFN-γ+874A/T site between the 2 groups(P<0. 05). The AA genotype(OR=9. 964)and the A allele (OR=3. 801) at the IFN-γ+874A/T site was with high-risk of RAU. Carry A allele in patients with risk of RAU was to carry the T allele of 3. 801 times. There was no significant differ-ence in the genotype frequencies or allele frequencies of IL-2-330T/G site between the 2 groups(P>0. 05). Conclusion: IFN-γ+874A/T site was associated with the risk of RAU. But,IL-2-330T/G site was not.
6.The research of lymphocyte proliferation using CFSE and FCM

Aim To quantitatively analyze the lymphocyte proliferation using the time-series data of carboxyfluorescein diacetate succinimidyl ester(CFSE).Methods The model of immune suppression in mice was created by cyclophosphamide and CFSE dye was used for staining the lymphocyte of both control group and treatment group animals. The data were analyzed through mathematical model-fitting.Results The first generation of cell proliferation of the control group and treatment group were 27.17 h and 22.88 h; cell death rates in each division were respectively 20% and 40%; the half-life of cells before the proliferation was respectively 31.53 h and 43.32 h.Conclusions The mathematical fitting of CFSE data can quantitatively analyze the mechanism of the drugs attecting the proliferation of lymphocyte.In this experiment the mechanism of cyclophosphamide inhibiting lymphocyte proliferation may be due to the increase of cell death rate of each division caused by cyclophosphamide.

8.Chemical constituents in butanol extraction from stems and leaves of Arachis hypogaea

Objective To investigate the chemical constituents in the stems and leaves of Arachis hypogaea.Methods The isolated compounds Ⅰ—Ⅳ were obtained by the combination of silica gel,ODS-18,and Sephadex LH-20 column chromatographies.Structural elucidation was conducted by the moedrn spectral method.Results Seventeen compounds were isolated and identified from the water extraction,and there were four norsesquiterpene compounds from butanol extraction.They are arachiside A(Ⅰ),cucubalactone(Ⅱ),roseoside(Ⅲ),and citroside A(Ⅳ).Conclusion Compound Ⅰ is a new compound named arachiside A and compounds Ⅱ—Ⅳ are isolated from the plants of Arachis Linn.for the first time.
9.Chemical constituents in leaves of Piper laetispicum

Objective To study the chemical constituents in the leaves of Piper laetispicum.Methods Compounds were separated and purified by silica gel column and macroporous resin.Their structures were identified on the basis of spectral methods.Results Sixteen compounds were isolated and identified from the leaves of P.laetispicum:(2E,4E)-N-isobutyl-11-phenylundecadienamide(Ⅰ),(2E,4E)-N-isobutyl-15-phenylpentadecadienamide(Ⅱ),laetispicine(Ⅲ),(2E,4E)-N-isobutyl-7-(3',4'-methylene-dioxyphenyl)heptadienamide(Ⅳ),brachystamide A(Ⅴ),dihydropipercide(Ⅵ),pipermacramide A [(2E,4E)-N-isobutyleicosadienamide,Ⅶ],(+)-spathulenol(Ⅷ),d-sesamin(Ⅸ),piperitol(Ⅹ),vitexin(Ⅺ),vitexin-2″-O-?-D-glucoside(ⅩⅡ),N-p-coumaroyltyramine(ⅩⅢ),?-sitosterol(ⅩⅣ),stigmasterol(ⅩⅤ),and 1-dotriacontanol(ⅩⅥ).Conclusion Compounds Ⅰ and Ⅱ are isolated for the first time as natural substance,compounds Ⅴ,Ⅵ,and Ⅷ are first isolated from P.laetispicum,compounds Ⅺ,ⅩⅡ,and ⅩⅥ are obtained from the plants of Piper L.for the first time.
10.Effect of DCN gene on the anti-tumor and immunological functions of mice bearing S180 sarcoma

Objective To investigate the anti-tumorigenesis function of DCN gene and its effect on the immunological funotions of mice bearing S180.Methods Twenty four mice bearing S180 were divided into three groups at random(eight mice every group),and then the normal saline group(negative control),pcDNA3.1(+)vector group and pcDNA3.1(+)-DCN group were set up for the experiment.They all started to inject next day by armpit.The volume of tumors were measured at the 5th day,7th day,9th day,11th day.The transformation rate of T lymphocytes were detected by means of MTT,the mRNA level of TGF-β by RT-PCR and the level of VEGF in serum by ELISA.Results The average volume of tumor in pcDNA3.1 (+)-DCN group were deflated to(2.219±0.105)cm3 comparing with the others(P<0.001);the transformation rate of T lymphocytes were increased significantly to(67.21±6.32)%(P<0.01,P<0.05);meanwhile it can decrease the mRNA level of TGF-β in serum,and depress the expression of VEGF markedly,whose absorbance were2.6175±0.099(P<0,01,P<0.05).Conclusion DCN gene could significantly inhibit the growth of tumor,and enhance the immunological function of mice bearing S180 in vivo.
