2.Internal medical management of extranodal nasal type NK/T cell lymphoma
Journal of International Oncology 2015;42(1):71-73
Extranodal nasal type NK/T cell lymphoma (ENKTL) is a highly aggressive non-Hodgkin lymphoma.Due to its low occurrence even in prevalent areas, there has been no large sample randomized controlled clinical trials. Therefore,no standard therapeutic strategy is currently identified in this disease.Tumor cells are insensitive to conventional anthracyclines-containing chemotherapy because of high expression of multi-drug resistant gene 1.Regimens that incorporate the use of L-asparaginase or gemcitabine result in substantial improvements in overall response rate and are promising treatment for ENKTL.Targeted therapy, immunomodulatory therapy and hematopoietic stem cell transplantation are still under research.
3.Association between γ-glutamyl transpeptidase-to-platelet ratio and hepatic fibrosis in patients with chronic hepatitis B
Journal of Clinical Hepatology 2017;33(3):548-552
Early and accurate diagnosis of hepatic fibrosis and application of antiviral therapy are the key to improving the prognosis of patients with chronic hepatitis B (CHB).Liver biopsy and transient elastography cannot be widely used for the diagnosis of hepatic fibrosis in clinical practice, and therefore, the serological diagnostic model has become a hot research topic in recent years. This article introduces a new serological diagnostic model,γ-glutamyl transpeptidase-to-platelet ratio (GPR),which has a high value in the diagnosis of hepatic fibrosis in CHB patients;however, the accuracy of GPR varies between different populations and different areas.GPR is also an excellent predictor for the prognosis of hepatitis B-associated liver cancer. It is pointed out that GPR has a promising future in the diagnosis of hepatic fibrosis in CHB patients, but due to a lack of clinical research data on GPR,further studies are needed to support its application in China.

Journal of Acupuncture and Tuina Science 2006;4(5):274-278
Objective: To research the clinical curative effect and explore the mechanism of penetrating of scalp-point on acute hypertensive intracerebral hemorrhage (ICH). Methods: 66 patients were randomly divided into two groups: one is the scalp-point therapeutic group, the other is the western medicine group. The two groups were all treated by dehydration, etc. In addition, there are 20 normal people to be set as the healthy group. To select the nervous functional deficit mark, activity of daily living (ADL) form, the amount S-100B protein as indexes and analysis the results. Results: the recordings indicate the patients' Serum S-100B protein are related with nervous functional deficiency mark. After 28 days, the recovery of patients' nervous function of scalp acupuncture group was obviously superior to that of control group (P<0.05). Conclusions: The results suggested that the patients' Serum S-100B had positive correlation with nervous functional deficit; The penetrating of scalp-point had the better regulatory effect on the level of Serum S-100B, and could improve the nervous functional deficit and ADL, so as to improve the coordination and compensation function and the general nervous function between cortical functional areas

Journal of Chinese Physician 2017;19(7):1014-1017
Objective To explore the expressions of glutathione reductase in gastric cancer,to investigate the relationship between glutathione reductase (GSR) and clinical pathological characteristics of gastric cancers,to identify the role of GSR in evaluation of the prognosis of gastric cancer patients, and to investigate the role of GSR in the development of gastric cancer. Methods The gastric cancer datasets were searched and downloaded from The Cancer Genome Atlas (TCGA),and chip data were analyzed with clinical information.Gene set enrichment analysis (GSEA) was conducted to explore the gene sets enriched in samples with high GSR expression. Results The expression of GSR was down-regulated in high grade tumors (P < 0.01).No significant difference was found between different age,Shortest tumor diameter,American Joint Committee on Cancer (AJCC) M stage,Barrett's esophagus, family history of gastric cancer, and Helicobacter pylori (H.pylori) infection.Higher expression of GSR indicated poor prognosis in gastric cancer.GSEA indicated that GSR regulates gene sets associated with oxidative phosphorylation, metabolism of nucleotides, mitochondrial protein import, and mitotic G1 S phases. Conclusions GSR can be used as an indicator to predict the prognosis of gastric cancer patients and a target for the treatment of gastric cancer.

Background The application of mesenchymal stem cells to transfer specific genes is under investigation in various diseases.Using this strategy may provide a more effective method to supply exogenous neurotrophic factors to the cental nervous system, including retina. Objective This study was to construct ciliary neurotrophic factor(CNTF)-overexpressing bone marrow mesenchymal stem cells (BMSCs) using lentiviral vectors. Methods Rat secreted-CNTF gene cDNA was synthesized and subcloned into a lentiviral vector plasmid pHⅣ-dTomato to construct recombinant vector CNTF-dTomato.CNTF-dTomato/pH Ⅳ-dTomato plasmid were co-transfected into 293T packaging cell line with packaging plasmid psPAX2 and enveloped plasmid pMD.2G to produce recombinant lentivirus CNTF-lenti and control-lenti.Rat BMSCs were infected with CNTF-lenti/control-lenti to produce CNTF-BMSCs and control-BMSCs.Expression of dTomato and efficiency of infection was evaluated under the fluorescence microscope.Uninfected BMSCs(pure BMSCs) served as the blank control.CNTF protein level in the supernate was detected by enzyme-linked immunosorbent assay (ELISA) and compared among the blank-BMSCs group,control-BMSCs group and CNTF-BMSCs group.Adipogenic and osteogenic differentiation of CNTF-BMSCs were induced using adipogenic-inducible medium and osteogenic-inducible medium and identified using oil-red O staining and alizarin red S (ARS) staining. Results After CNTF-dTomato plasmid was transfected into Stbl3 competent cells, the colony PCR product was 1 033 bp.The inserted sequence in the pHⅣ-dTomato plasmid was coincident with the expected one.The results of DNA sequencing showed that CNTF-dTomato plasmid was successfully constructed. The infection rate of CNTF-lenti was approximately 95%.ELISA showed that on the post-infected day 2,3,7,the CNTF protein levels in the supernate were significantly higher in the CNTF-BMSCs group than those in the blankBMSCs group and control-BMSCs group (all at P=0.000).In the CNTF-BMSCs group, the CNTF protein levels in the supernate were significantly increased on post-infected day 2,3,7 compared with day 1 (P =0.013,0.004,0.042).The adipogenic-induced cells showed the red staining to oil red O,and osteogenic-induced cells presented the orange staining to ARS.Conclusions BMSC line with stable expression of CNTF is successfully established by lentiviral vectors.CNTF-BMSCs have the potential to differentiate towards adipocytes and osteoblasts.
