2.Alpha-lipoic acid pretreatments provide protection against hypoxia/reoxygenation injury in human umbilical vein endothelial cell

Objective To observe the protective effect of α-lipoic acid (LA) pretreatment on hypoxia/reoxyg-enation injury in vascular endothelial cells and preliminary explore its mechanism. Methods HUVEC were cultured intro and devided into control group,hypoxia/reoxygenation model group and LA group. Cell morphology was observed under inverted phase contrast microscope,cell viability was detected by MTT,total antioxidant capacity (TAOC),nitric oxide(NO),malondialdehyde (MDA),superoxide dismutase (SOD) activity or level were measured by various commercial kits respectively. Apoptosis of HUVEC in each group were detected by flow cytometry , cleaved caspase-3 level was measured by western blot. Results As compared to control group, the survival rate, TAOC, SOD, NO activity were significantly lower(P<0.05), and MDA level, apoptosis rate and apoptosis protein levels were significantly higher (P < 0.05) in model group. However,LA can increase the survival rate,TAOC, SOD,NO activity (P < 0.05),reduce MDA level and cell apoptosis (P < 0.05)in a dose dependent manner. Conclusion LA pretreatments provide protection against hypoxia/reoxygenation injury in HUVEC. LA exerts protective effects through inhibition of HUVEC apoptosis.
3.Pancreatic acinar cell carcinoma: a clinicopathological and immunohistochemical analysis of 8 cases

ObjectiveTo investigate the clinicopathological features,immunohistochemical phenotype of pancreatic acinar cell carcinoma.MethodsEight cases of pancreatic acinar cell carcinoma admitted to our hospital from January 2001to January 2011were retrospectively analyzed. The clinicopathological characteristics,and immunohistochemical staining for phenotype were analyzed,then the follow-up data were summarized.ResultsAll 8 patients with pancreatic acinar cell carcinoma was male,with a median age of 47 years old.Tumors were located in the pancreatic head in 4 patients,pancreatic body and tail in 4 patients.The average tumor size was 4.5 cm × 4.0 cm × 3.2 cm,the section appeared as gray or gray-red and presented as solid or cystic lesions.Larger tumors were often accompanied by hemorrhage and necrosis.Microscopically,the tumor cells arranged in acinic,cord,trabecular or solid nests.The cytoplasm was abundant and eosinophilic.The nuclear was round,oval,slightly atypia.lmmunohistochemical staining showed diffusely positive for CAM5.2,α-AT,α-ACT and focally positive for CA19-9,CEA,E-cad,β-cat and MUC-1 and only occasionally positive for AFP,NSE,Syn and CgA.Follow-up data showed there was one case of postoperative death due to postoperative pancreatic leakage with abdominal infection.Liver metastasis occurred in 4 cases,among whom,2 cases died.ConclusionsPancreatic acinar cell carcinoma is a rare epithelial malignant tumor of pancreas,with distinct phenotype characteristics.
4.Effects of acupuncture on NO and NOS in restoration environment of interstitial cells of Cajal after colonic anastomosis.

OBJECTIVETo explore the mechanism of acupuncture on promoting the restoration of interstitial cells of Cajal (ICCs).
METHODSThirty SD rats were randomly divided into a blank group, a model group and an acupuncture group, ten rats in each one. The rats in the model group and acupuncture group were treated with colonic anastomosis to establish the model. After successful establishment of the model, the rats in the acupuncture group were treated with acupuncture at bilateral "Zusanli" (ST 36), "Sanyinjiao" (SP 6) and "Taichong" (LR 3) for 15 min, once a day for 10 days. Rats in the model group and blank group were put into the fixator for 15 min at the same time daily. The propulsive rate of small intestine was measured in each group. Colonic tissues were collected to detect c-kit expression by using immunohistochemistry. The nitricoxide (NO) content was measured by nitrate reductase method and nitric oxide synthase (NOS) activity was measured by method of L-arginine.
RESULTSCompared with the blank group, the propulsive rate of small intestine in the model group was decreased; NO content was increased; iNOS activity was elevated; cNOS activity was declined; total NOS (tNOS) activity was increased and the counting of c-kit positive ICCs was decreased (all P < 0.05). Compared with the model group, the propulsive rate of small intestine in the acupuncture group was increased; NO content was decreased; iNOS activity was reduced; cNOS activity was elevated; NOS activity was decreased and the counting of c-kit positive ICCs was increased (all P < 0.05).
CONCLUSIONAcupuncture can regulate NO content and NOS activity in postoperative restoration environment of ICCs, which may participate in the process of acupuncture promoting the restoration of ICCs.

5.Expression and significance of SP-A in nasal mucosa of allergic rhinitis and nasal polyp

Objective: Surfactant protein A (SP-A) is protein that appears to play an important role in mammalian first-line host defense. The objective of this study was to immunolocalize SP-A in human sinonasal tissue. Method:Eleven cases of allergic rhinitis, fifteen cases of polyp and seven cases of normal middle turbinate were studied with immunohistochemistry and immunofluorescence method to detect the expression of SP-A. Result:The expression of SP-A in allergic rhinitis and polyp were dramatically higher than that in controls(P<0.05), and there was no remarkable difference in the expression of SP-A between allergic rhinitis and polyp(P>0.05). The result demonstrated that SP-A was positivly correlated with eosinophils within the basement membrane of epitheli-um(R=0.81,0.55). In the result of immunofluorescence, there was significantly higher expression SP-A in nasal mucosa of allergic rhinitis and nasal polyp than that in control group(P<0.05). Conclusion:SP-A is likely to play key roles in the inflammatory reaction process of allergic rhinitis and polyp. Its secretion in the upper airway indicates that future studies may allow manipulation of this protein and development of novel treatments for sinonasal pathology.
6.Relationship between podocyte molecule’s distribution and expression,foot process morphology and proteinuria

Objective: To investigate the association between nephrin, podocin and ? actinin of the glomerular podocyte molecules, the morphometric change of podocyte foot process and the development of proteinuria. Methods: Puromycin aminonucleoside (PAN) nephrosis was established. Immunofluorescence staining, image analysis and real time quantitative PCR were employed to study the distribution and quantitation of glomerular expression of nephrin, podocin and ? actinin. Morphometric methods were applied to evaluate the morphology change of podocyte foot processes under electron microscopy. Results: (1) Before the onset of proteinuria, 2 days after PAN injection, the podocyte foot process became swollen;nephrin and podocin staining were changed into discontinuous pattern accompanied by the decrease of podocin staining intensity. The foot process became more swollen on day 5,and podocin intensity continued to decrease. Meanwhile, nephrin decreased significantly both in protein intensity and at mRNA level. (2) When heavy proteinuria [(130.8?30.7) mg/d, P =0.02]occurred, complete effacement of podocyte foot processes was revealed; both podocin and nephrin staining intensity decreased dramatically( P
7.Effect of Air Sterilization with Nanometer Photocatalysis Air Sterilizer in Hospital

OBJECTIVE To observe the sterilization effect of nanometer photocatalysis air sterilizer mounted in hospital.METHODS We carried out a dynamic survey on air average numbers of colony through field experiment and practical operation.RESULTS The field experiment displayed that the air bacterium dissolution rate was 55.64-75.94% and the bacterium content degraded as time extended,after nanometer photocatalysis air sterilizer been used 1-2 h;the result of practical operation in four seasons of the year revealed that the average bacterium colony numbers in each ward region was lower than before sterilizer mounted,the air bacterium dissolution rate was 73-82.3% in the third ward region and 44.5-76.3% in the fourth ward region.CONCLUSIONS It is effective to use the nanometer photocatalysis air sterilizer to sterilize the air in hospital under the condition of inspection.The airflow of the central air conditioning can practice the air sterilization function more validly.
8.The effect of calcium channel blockade on matrix metalloproteinases and fibronectin in the rat myocardial remodeling

Aim To investigate the contribution of cardiac L-and L/T-type Ca 2+ channels blockers on matrix metalloproteinases (MMPs) protein expression and fibronectin (FN) degradation following myocardial infarction(MI). Methods Rat MI model was established by permanent ligation of left coronary artery. Infarcted rats were orally treated with placebo, amlodipine (L-channel blocker, 4 mg?kg -1?d -1) or mibefradil (L/T-Channel blocker, 10 mg?kg -1?d -1) for 7 days before induction of myocardial infarction. Protein levels of MMP-2,3,9 and FN distribution were assayed by immunoflorescence at 1, 3, 7 days post coronary occlusion in left ventricular free wall (LVFW) and myocardium interventricular septum (IS). Results Pathological changes of myocardial tissues in IS and LVFW showed typical myocardial remodeling. The hypertrophy was dominant in IS, but in LVFW the characteristics including disordered alignament, hypertrophy of the myocytes, the discontinuity, dissolving of carediomyofibrills, and the hyperplasia of interstitial tissue were found 7 days after MI. The protein levels of MMP-2,3,9 increased in IS 1,3,7 days post MI gradually, whereas the protein levels of FN decreased gradually, the protein levels of MMP-2,3,9 increased significantly in LVFW 7 days post MI, and the protein levels of FN decreased significantly compared to control group, respectively(P
9.Formula Screening and in vitro Drug Release of Tegafur Thermo-sensitive Gel for Intratumor Injection

Objective:To screen the best formula of tegafur temperature-sensitive gel for intratumor injection and investigate the in vitro drug release behavior. Methods:The drug dose was determined by cytotoxicity experiment. The thermo-sensitive gel was prepared with PLGA-PEG-PLGA and HPMC as the matrix. With the in vitro release as the index, the effects of PLGA-PEG-PLGA and HPMC at different concentrations on gel were investigated. The gelation temperature, viscosity and pH were detected. Results:The best formula was as follows:25% PLGA-PEG-PLGA, 1% HPMC, and tegafur dose of 1 mg·ml-1 . The average gelation temperature was 36. 7℃, the average viscosity was 7550 mPa·s, and the average pH was 7. 2. Conclusion:Tegafur thermo-sensitive gel for intratumor in-jection shows temperature sensitivity and obvious sustained-release property, which provides experimental basis for the further clinical research.
10.Diffusion kurtosis imaging for demonstrating the microstructure of brain gray and white matter in patients withchronic mountain sickness

Objective To explore the differences between patients withchronic mountain sickness (CMS) and healthy people in the microstructure of brain gray and white matter by using diffusion kurtosis imaging (DKI). Methods 21 CMS patients were recruited to a study group and 20 healthy volunteers were assigned to a control group. Both groups received conventional MRI and DKI sequence scans. The mean kurtosis (MK) values ,radial kurtosis (RK) values and kurtosis anisotropy (KA) values in each region of interest (ROI) in the cerebral gray and white matter were measured and the same part of the left and right side ROI parameters was counted. The data satisfied the normal distribution and the paired samples t?test was used;the ROI parameters between the two groups in the same parts of the same side followed the normal distribution and the two independent samples t?test was used. Results The KA values in both sides of the anterior limb of internal capsule ,the MK values and RK values in bilateral caudate nucleus head and thalamus had side difference in CMS group. In both sides of the anterior limb of the internal capsule′s MK values,KA values and thalamus′s MK values,RK values existed side difference in the control group. The KA values of the right anterior limb in internal capsule were lower than those in the left in both groups;the right thalamus′s MK values and RK values were higher than the other side. The RK value in genu of corpus callosum differed significantly ,and it was lower the CMS group than in the control group. The difference of the KA values in corpus callosum ,the MK value in the left anterior limb of internal capsule ,the RK values in the left posterior limb of internal capsule ,the MK values in the right caudate nucleus head ,the MK values in bilateral lenticular nucleus and the MK values and KA values in the right thalamus were significant ,and they were higher inthe CMS group than in the control group. Conclusions The left and right cerebral hemispheric micro?structure is not exactly symmetrical between patients with CMS and healthy people living in high altitude areas , there are differences in some brain areas. The sensitivity and reactivity of brain tissue to high altitude hypoxia are different in patients with CMS ,there are various degrees of difference between the CMS and the normal in the corpus callosum,basal ganglia region and thalamus.
