Objective To investigate the effect of Huangqikeli on immune function of mice. Methods 30 mice were randomly divided into 3 groups. The mice in A and B group were administrated with distilled water,C group were administrated with Huangqikeli. On the tenth day,all groups except A group were injected with cyclophosphamide to induce the model. The effects on the phagocytic function of Huangqikeli to macrophage in abdominal cavity of mice,delayed hypersensitivity induced by DNCB,and production of hemolysin antibody in mice that CRBC sensitized were observed. 3.Effects of Suoquan Wan on Endocrine and Immune Function of Polyuria Rats With Kidney-yang Deficiency

Objective To observe the effects of Suoquan Wan(SQW) on the endocrine and immune function of the polyuria rats with kidney-yang deficiency. Methods The model rats were induced by gastric infusion of adenine(250 mg/kg) for 4 weeks, then treated respectively with SQW (at low, middle and high dose respectively), Shenqi Wan and desmopressin for another 4 weeks. The bodyweight and organ index were recorded, and serum cortisone concentration were detected. T cell subsets in peripheral blood were determined by flow cytometry. Results The bodyweight of rats in model group was lower than that in the normal control group. And the bodyweight of rats in desmopressin group, middle-and high-dose SQW groups differed from that in the model group (P < 0. 05 or P < 0. 01). The decreased organ indexes of pituitary, adrenal and thymic glands were found in the model group (P < 0. 05 or P < 0. 01 compared with the normal group). The thymus index was elevated in all the medication groups except the low-dose SQW group (P < 0. 05). The elevated organ indexes of pituitary and adrenal glands were only found in Shenqi Wan group and high-dose SQW group (P < 0. 05). In the model group serum cortisone concentration was decreased (P < 0. 01). In Shengqi Wan group and middle-and high-dose SQW groups, the cortisone concentration was significantly increased (P < 0. 05 or P < 0. 01, compared with the model group). In the model group, the percentages of CD3+ and CD3+/CD4+ T cell subsets in pe-ripheral blood were decreased, and the percentage of CD3+/CD8+ T cell subset was increased (P < 0. 05 or P < 0. 01 compared with the normal group). Compared to the model group, the percentages of CD3+ and CD3+/CD4+ T cell sub-sets were increased and the percentage of CD3+/CD8+ T cell subset was decreased in SQW group and Shenqi Wan group. But the differences of T lymphocyte subsets were insignificant between desmopressin group and the model group. Conclu-sion SQW can regulate the endocrine and immune function of the polyuria ratwith kidney-yang deficiency.
4.Expression and clinical significance of Pin1 and cyclin D1 in pancreatic carcinoma

Objective To investigate the expression of Pin1 and cyclin D1 in human pancreatic carcinoma and to discuss its role in oncogenesis of pancreatic cancer.Methods The mRNA expression of Pin1 and cyclinD1 in pancreatic tumor tissues and corresponding adjacent nontumor tissues 27 patients was detected by the real-time quantitative reverse transcription-polymerase chain reaction(RQ-RT-PCR).The results of RQ-RT-PCR were analyzed by one-way ANOVA and Fisher's exact probabilities test.Results Cyclin D1 and Pin1 were overexpressed at mRNA level in pancreatic carcinoma tissues compared with their adjacent nontumor tissues.Cyclin D1 overexpression were found in 14 of 20 pancreatic carcinoma tissue specimens and Pin1 overexpression in 13 of 20 carcinoma tissue specimens.The expression of cyclin D1 and Pin1 in pancreatic cystadenoma tissues was not different than that of corresponding adjacent nontumor pancreatic tissue.Pin1 overexpression positively correlated with an increase in cyclin D1 levels as shown by Fisher's exact probabilities test.However,Pin1 and cyclin D1 expression was not correlated with clinical stage and pathological parameters.Conclusions The overexpression of Pin1 in pancreatic carcinoma tissues could promote cyclin D1 expression,which might be a critical event in oncogenesis of pancreatic carcinoma.Pin1 may play a key role in pancreatic carcinoma.

6.Clinical significance of anti-β_2 glycoprotein 1 antibody and matrix metalioproteinase-9 and their association with Kawasaki disease

Objective To explore the levels and clinical significance of anti-beta 2-glycoprotein 1 antibodies (β_2GP1), matrix metalloproteinase 9 (M M P-9) in the plasma of children with Kawasaki diseases (KD). Methods Serum level of anti-β_2GP1 antibody and MMP-9 was measured in 47 children with KD by ELISA, and the data was analyzed using SPSS11.5 software. Thirty age matched children with infectious diseases(sepsis or pneumonia), exclusive of heart, liver, kidney, blood diseases and autoimmune diseases such as rheumatoid were chosen in the fever control group. Results Coronary artery lesions (CAL)were found in 17 children of KD group (17/47) by Doppler ultrasound examination. Significant differences (P < 0.05) of serum level of anti-β_2GP1 antibody was showed between KD group ((7.46 ± 2.13) U/ml)and the control group ((4.38 ± 0.43) U/ml) ; serum level of MMP-9 was (886.62 ± 92.72) ng/ml and (460.06 ± 179.59) ng/ml in KD group and the control group respectively, with significant difference between the two groups(P < 0.05). In KD group, levels of anti-β_2GP1 were (8.83 ± 0.89) U/ml among children with CAL and (6.18 ± 1.42) U/ml among children without CAL, serum level of MMP-9 was (948.62 ± 81.76) ng/ml and (872.00 ± 34.74) ug/ml respectively, with significant differences(beth P < 0.05). In children with KD, the serum levels of anti-β2GP1 antibody and MMP-9 were significantly correlated (correlation coefficient r = 0.665). Conclusions Serum levels of anti-β_2GP1 antibodies and MMP-9 increased in the acute phase of KD, and were significantly higher in those KD children with CAL.Anti-β_2GP1 antibodies and MMP-9 may play a role in the pathogenesis of KD, and can be used as an important serological indicator of KD with CAL.
7.The inhibitive effects of resveratrol on adhesion of neutrophils to HUVECs induced by fMLP

Aim To study the effects of resveratrol(Res)on the release of soluble adhesion molecules from human umbilical vein endothelial cells(HUVECs)induced by N-formyl-methionyl-leucyl-phenylal-anine(fMLP)and the adhesion between neutrophils and HUVECs.Methods The effects of Res on neutrophils adhesion to human umbilical endothelial cell(HUVECs)triggered by fMLP were examined.The soluble intercellular cell adhesive molecule-1(ICAM-1),the soluble vascular cell adhesive molecule-1(VCAM-1)and E-selectin release from fMLP(10 ?mol?L-1)stimulated HUVECs were determined by ELISA kits.The neutrophils isolated from human vein blood were loaded with Fluo-3,a fluorescent indicator,to detect intracellular free calcium concentration([Ca2+]i),and CLA was used as a chemiluminescent indicator to determine superoxide production in neutrophils.Results Res(1~50 ?mol?L-1)significantly inhibited neutrophil adhesion to fMLP-stimulated HUVECs and also obviously downregulated the levels of ICAM-1,VCAM-1 and E-selectin in supernatant of HUVECs culture stimulated by fMLP in the dose-dependent pattern.Res also suppressed fMLP-activated superoxide generation and[Ca2+]i increase in neutrophils.Conclusions Res involved in neutrophil adhesion to HUVECs intermediated by cell adhesive molecules expression trigged by[Ca2+]i and superoxide production in neutrophils,which means a lot to prevent inflammatory diseases.
8.Study on the effects of mM-CSF in the functional polarization of macrophages

Objective To investigate the anti-inflammatory/immune modulatory effects of high-expressing membrane bound M-CSF-hematopoietic malignant cells on macrophages. Methods After coculturing RAW264.7 and murine macrophage cell line with Namalwa-M, a cell line stably expressing mM-CSF, and companing with Nainalwa-V, a cell line stably transfected with the empty vector as the control, flow cytometry was used to detect the expression of the marker of alternatively activated macrophages, CD206, and intracellular expression of IL-10, IL-12, IL-6 and TNF-α to study the immunophenotype of RAW264.7; phagocytic assays to investigate their functional activity in vitro. Results RAW264.7 cocultured with Namalwa-M consistently showed high-level expression of CD206, which indicated activities of these macrophage cells were increased. Furthermore, these RAW264.7 expressing high levels of IL-10, TNF-a and low levels of IL-12, IL-6, as determined by intracellular staining were suggested that the phagocytic activity was increased. Functionally, RAW264.7 cocultured with Namalwa-M showed a higher level of phagocytic activity. Conclusion Macrophage generated in vitro after cocultured with mM-CSF-expressing hematopoietic malignant cell line could be transformed into abnormal macrophage with an immunophenotype defined as IL-10-high, IL-12-low, IL-6-low and TNF-α-high.
9.Ultrastructure of neurovascular unit in chronic compressive cervical myelopathy rat model

Objective To observe the ultrastructure of the neurovascular Unit (NVU) in chronic compressive cervical myelopathy rat model at different stages.Methods From March, 2014 to March, 2015, 32 rats were divided into two groups: sham control group (n =8) and compressive spinal cord injury group (n =24).The model was established by inserting the compression sheet made of polyurethane at the level of C6.BBB and somatosensory evoked potentials (SEP) were used to evaluate the spinal cord function status of model rat.Transmission Electron Microscopy (TEM) examination of compressive cervical spinal cords was performed separately at the 14th, 21st, 28th and 42nd day after modeling.Results At the 14th, 21st, 28th and 42nd, the BBB score were 17.571 ± 0.870, 15.952 ± 0.870, 15.476 ± 0.602 and 16.190 ± 0.632 were significantly lower than those in the control group (the BBB score of 4 points were 19.600 ± 0.516, 19.500 ± 0.527, 19.600 ± 0.699 and 19.800 ± 0.6232 respectively) (P < 0.05).Latency prolongation and amplitude reduction of somatosensory evoked potentials (SEP) were presented in the compressive spinal cord injury group.At the 14th day, edema around the capillaries was observed, the morphological structure of endothelial cells and basement membrane was normal, the tight junction between endothelial cell was intact, the mitochondria in the axons, oligodendrocytes and astrocyte foot processes were edematous.At the 21st day, extensive edema, even partial necrosis around the capillaries were found, the surrounding structure were arranged loosely, partial loss of endothelial cells and basement membrane, cavitation occurrence in endothelial cells, basal membrane density was significantly lower, mitochondria vacuoles and shrinkage in the cytoplasm, axon myelin loose or broken;at the 28th day, the edematous range surrounding capillaries narrowed, low density of basement membrane and endothelial cells, vacuoles in endothelial cells, loose axon myelin, while some mitochondrias backed to normal.At the 42nd day, capillary integrity, no abnormalities were found in endothelial cells, basement membrane, tight junction and mitochondria, double layers of endothelial cells and basement membrane could be seen, local broken and loose structure were presented in part of the axons;The TEM of the sham control group showed normal ultrastructure of NVU.Conclusion The ultrastructure of NVU in chronic cervical spinal cord compression presented various in the different periods, NVU disruption were found in the early stages (14th-28th days), and compensatory and repair process were developed incompletely later.
10.Comparative study on drug resistance of pseudomonas aeruginosa isolated from different sites of patients

Objective To analyze the infection sites of pseudomonas aeruginosa (PAE) isolated from patients in hospital ,and in‐vestigate their drug resistance situation ,in order to provide reference information for clinical use of antibiotics rationally .Methods The sample distribution of PAE between January 2012 and December 2013 were retrospectively analyzed .And the resistance rates of PAE to antibacterial drugs from different sites of patients were statistically compared .Results The isolation rate of PAE in re‐spiratory tract was the highest ,accounting for 74 .1% ,closely followed by isolation rate in urine and wound secretion .The resist‐ance rate of PAE to antibacterial drugs in these three kinds of specimen is statistically different (P<0 .05) .The resistance rate of PAE is high in respiratory tract ,and low in wound secretion .Conclusion The pseudomonas aeruginosa infection is mostly common‐ly found in respiratory tract ,and has the highest drug resistance rate .The choice of antibacterial drug should be made according to the infection sites of patients ,because the resistance rate of PAE in different sites of patients is significantly different .
