Objective To understand the serological detection of suspected measles cases in Xicheng District of Beijing, and provide basis for monitoring and prevention of measles. Methods Detection results of 200 blood speci-mens and age distribution of suspected measles cases in Xicheng District of Beijing in 2011 -2014 were analyzed. Results The positive rate of serological detection of suspected measles cases in 2011-2014 was 32.50%(65/200);positive rates in each year were 12.00%,4.35%,33.90%,and 44.09% respectively;there was significant differ-ence in the positive rate of serological tests in different years(P <0.001).Of 200 cases,65 were <5 years old,and 97 were >24 years old, positive rates of serological detection were 35.38% and 34.02% respectively;31 cases (15.50%)received vaccination of measles;vaccine coverage rate in IgM antibody positive cases was lower than IgM antibody negative cases (3.08% vs 21 .48%,P <0.001 ).Conclusion Positive rate of serological detection de-creased in 2012,then increased year by year, for children under 5 years, measles are controlled by immunization with measles vaccine,in order to achieve the goal of eliminating measles effectively, control and prevention of adult measles is of great significance.

OBJECTIVE To discuss the management methods of sputum sample retained clinically based on the praxiology, and aim at improving the quality of the sputum retained. METHODS Through investigating the quality of sputum sample retained routinely from the department of internal medicine in a general hospital at the duration from Sep 2004 to Feb 2005,the intervention effect was evaluated by carrying out the measure of praxiology intervention. RESULTS The qualification rate was 23.7% for the sputum sample retained routinely from the hospital at the duration from Sep 2004 to Feb 2005,after carrying out the measure of praxiology intervention, the qualification rate was 30.1% at the duration from Apr 2005 to Aug 2005.Statistic data showed that there was no significant difference. CONCLUSIONS The quality of the sputum sample retained routinely is poor, and the measure of praxiology intervention failed to improve the quality of the sputum sample.Microscopy is the most important measure to ensure the quality of the sputum sample for the cytology test before cultivating the sample, and should be a routine test in hospital. The study on praxiology intervention will be further carried out in the future.

Objective To investigate the expression of TLR2 and HIF-1α in pancreatic cancer and explore the relationship between TLR2 or HIF-1α protein and the clinical or pathological changes in pancreatic cancer. Methods The mRNA of TLR2 and HIF-1α in 30 cases of pancreatic cancer and its adjacent tissues were detected with real-time PCR and with immunohistochemical methods in 65 cases of pancreatic cancer and 38 cases of corresponding adjacent tissues. The relationship between TLR2 or HIF-1α and pathologic features of pancreatic cancer was analyzed. The correlation between TLR2 and HIF-1α in pancreatic cancer was also assessed. Kaplan-Meier method was used to assess the impact of TLR2 or HIF1αexpression on survival. Results The relative quantification of TLR2 mRNA and HIF-1α mRNA in pancreatic cancer tissues was 0.84±0.17 and 0.87±0.11, respectively, which was significantly higher than that in adjacent tissues 0.70±0.13 and 0.68±0.13 respectively,P<0.05. The protein expression of TLR2 and HIF-1α in pancreatic cancer tissues was 63. 10% and 70.8%, respectively, significantly higher than that in adjacent tissues (34.20% and 36.8%, respectively). There was no significant correlation between the expression of TLR2 or HIF-1α protein and the age, gender, tumor location, the degree of differentiation in patients with pancreatic cancer (P>0.05). However, there was significant correlation between the expression of TLR2 or HIF-1α protein and tumor size, lymph node metastasis, venous invasion and clinical staging. TLR2 and HIF-1α had a significant impact on survival. Conclusion TLR2 and HIF-1α overexpressed in pancreatic cancer and TLR2 signaling pathway may promote development of the pancreatic cancer with HIF-1α together.

Objective To construct the eukaryotic plasmid expression vector mediated short hairpin RNA(shRNA) interference targeting TLR4 gene, and transfect it into pancreatic adenocarcinoma cell line PANC1,then screen stably transfected clonal cell line. Methods Three shRNA interference expression plasmid vectors targeting the TLR4 gene were constructed, named TLR4-1,TLR4-2,TLR4-3.The shRNA plasmid with highest inhibitory efficiency was selected and transfected into PANC1 cells with liposome. The silencing efficiency and transfection efficiency of TLR4-shRNA was assayed with real-time quantitative PCR and flow cytometry analysis.Monoclonal cell with stable transfection of TLR4-shRNA were selected by geneticin 418 (C418) and limiting dilution analysis. Results Transient transfection efficiency of PANC1 was (46.72 ±5.06) %.TLR4 mRNA expressions were 0.025 ± 0.004,0.027 ± 0.003,0.019 ± 0.006in cells transfected with TLR4-1,TLR4-2,TLR4-3,respectively,which were significantly lower than that in untransfected group (0.061 ±0.018) and negative control group (0.057 ±0.015,P <0.05).The transfection efficiency of TLR4-3 vector in stably transfected clones [(82.79 ±8.16)%] was significantly higher than that of transient transfection (P =0.001 ).The expression of TLR4 mRNA was decreased to 0.010 ± 0.002,which was significantly lower than that of transient transfection ( P =0.001 ).The expression of TLR4 protein was (0.54±0.32) %,which was significantly lower than that of untransfected cells [( 87.42 ± 5.00 ) %] and that of negative control [(82.9±5.00)%,P =0.000].Conclusions Stable transfection PANC1 cell lines with TLR4 gene silencing are successfully identified.

Objective To explore the effect of neuromuscular electrical stimulation(NMES) on hyolaryngeal structural movement in post-stroke dysphagia. Methods Thirty patients with post-stroke dysphagia were randomly divided into 3 groups (n =10) namely group A,group B and group C.On the basis of routine swallowing training, medication and rehabiliation given to group C,group A was provided with VitalStim therapy on both suprahyoid and infrahyoid muscles, while group B was only on suprahyoid muscles lasting 2 weeks. Before and after treatment, the Video Fluoroscopy Swallowing Study (VFSS) was carried out and the upward and forward displacement distances of the hyoid and thyroid cartilage when swallowing pap were measured. Moreover, water swallow test, dysphagia severity scale and DOSS scores were used to assess the swallowing function. Results After 2 weeks' treatment, the water drinking test score, dysphagia severity scale and DOSS scores increased significantly in all groups than before treatment. The water drinking test score of group A (2.40 ± 1.26) and group B (2.10 ± 0.99),dysphagia severity scale of group A (5.30 ± 1.89) and group B(5.20 ± 1.69),as well as the DOSS level of group A (5.20 ± 1.40) and group B (5.10 ± 1.45) were significantly better than the control group. However,no significant difference was observed in all measurements between group A and group B.The group B revealed a significantly increase in anterior hyoid excursion distance of (12.15 ± 7.59) mm,much bigger than that before treatment and that of group A and B after treatment. Conclusion NMES on the basis of traditional swallowing training can improve the swallowing function of post-stroke dysphagia patients, and NMES on suprahyoid muscles can further induce an increase in anterior hyoid excursion.
