1.Determination of Total Flavonoid in Fructus Rosae Laevigatae by Dual Wavelength TLC-Scanning

OBJECTIVE:To determine the content of the total flavonoid in Fructus Rosae Laevigatae.METHODS:Dual wavelength TLC scanner was selected with rutin as standard substance.The developing solvent was acetic ether-formic acid-water(8∶1∶1);?s=266 nm,and ?R=230 nm.RESULTS:The linear range of Fructus Rosae Laevigatae was 1~6 ?g,and the average recovery was 98.9% 2.397%(RSD=1.39%,n=6).CONCLUSION:The method is simple and accurate,and it is applicable for the quality control of Fructus Rosae Laevigatae.
2.Experimental Study of Ruzengping Granule on Hyperplasia of Mammary Glands in Model Rats

Objective To observe the effect of Ruzengping granule on mammary gland of rat with mastoplasia. Methods After benzoate estradiol and progestogen had been injected into rats for 35 days, the mastoplasia model was made. The model rats were treated with Ruzengping granule for 30 days, to measure the mammary papilla diameter of the rats in all groups, to observe morphologic changes of mammary papilla and the changes of mammary gland under microscope, to determine the content of estradiol in blood serum and blood rheology by radio-immunity. Results Ruzengping granule could significantly inhibit the mammary papilla's diameter of the mastoplasia rats, lessen mammary papilla’s symptoms in morphology, reduce the quantity of lobule of mammary gland, lobule-alveolus in mammary gland and the diameter of acinar lumina. Ruzengping granule could also significantly improve blood rheological nature and degrade content of estradiol in blood serum. Conclusion Ruzengping granule can healing efficaciously rat’s mastoplasia induced by benzoate estradiol.
3.Efficacy of quantitative CT in assessment of fracture healing

Objective To investigate the efficacy and accuracy of quantitative computed tomography (QCT) in assessment of fracture healing. Methods Twenty-four healthy New Zealand rabbits were enrolled in the study and randomly divided into two groups, ie, Croup A (union model group, transverse fracture open created on mid-shaft of tibia and inter-fixed by kirschner wire) and Group B (non-union model group, transverse fracture with 5 mm defect on mid-shaft of tibia open created, then sealed with bone wax on fracture gap and medulla cavity, inter-fixed by kirschner wire). At 2, 4, 8 and 12 weeks, Kirschner wire was withdrawn and plain X-ray and QCT scanning were performed on the bilateral tibia. Then, rabbits were scarified and its bilateral tibia were desected and histologically examined. The result of X-ray and histological analysis was used as the "golden standard" for evaluation of fracture healing. Receiver operating characteristic curve (ROC) was used to analyze the evaluation performance of QCT. The corresponding segments of the contralateral healthy tibia were used as control to investigate the change of QCT parameters. Results In Group A, X-ray and histological analysis verified clear fracture line filled with irregular callus at 2 and 4 weeks but proved vague or vanishing fracture line and continuous and intact cortex of irregular callus at 8 and 16 weeks. In Group B, X-ray and histology analysis found clear fracture line with inactive ossification at 2 and 4 weeks but vague fracture line with scarce osteocyte and bone trabecula at 8 and 16 weeks. ROC analysis of QCT results showed the following results; (1) the areas under curve (accuracy) of material parameters including bone mineral density (BMD) and bone mineral content (BMC) were 0.781 and 0.750 respectively; (2) structure parameter-cross-sectional area (CSA) and the area under curve of cross-sectional moment of inertia (CSMI) were 0.781 and 0.469 respectively (P <0.05); (3) the areas under curve of the extending parameters bone strength indices (BSICSA) and CSMI bone strength indices ( BSICSMI) were 0. 913 and 0. 813 respectively (P < 0.05); (4) the area under curve (accuracy) of BSICSA, CSA and BMD were 0.905, 0.921 and 0.905 respectively (P<0.05). Conclusions QCT has potential in distinguishing fracture union and nonunion models in measurement of local fracture pattern. The screening parameters with more accuracy are BSICSA, CSA, BMD, which have advantages of accuracy and specialty in assessing fracture healing.
4.Bacteriological examination of digital X-ray imaging device before and after disinfection

The digital X-ray imaging device was disinfected with trichloro-isocyanuric acid solution.The samples were taken with cotton swab pouring method before and 1,2,3 and 6 h after disinfection and the bacteriological examination was performed.Before disinfection the colony count of samples was all above the standard (> 10 cfu/cm2) with bacilli and gram-positive cocci predominantly.The highest count was detected in the X-ray photographic bed (58 cfu/cm2) and the lowest was in the exposure button(19 cfu/cm2).After disinfection the colony count in radiation suits still exceeded the standard,the colony count in diagnosis bed 2 h after disinfection,that in all parts of device 3 h after disinfection was off levels.The results indicate that the chlorine disinfectants can only achieve short-term effect in disinfection of digital X-ray imaging device.
5.Pseudolaric acid B induced apoptosis and mitotic arrest circumventing Fas receptor pathway in MCF-7 cells

Aim To study the mechanisms of Pseudolaric acid B(PAB)-induced MCF-7 cell apoptosis and mitotic arrest.Methods MTT assay was performed to assess the cell growth inhibition,contrast phase microscope was used to observe cellular morphologic alteration,and the change of DNA was detected by fluorescent microscopy.The distribution of cell cycle was determined by flow cytometric analysis of propidium iodide staining,and the protein expression was examined by Western blot analysis.Results PAB inhibited MCF-7 cell growth in a dose-and time-dependent manner.4 ?mol?L-1 PAB induced DNA condensation at 24 h.PAB cleaved PARP in a time-dependent manner.At 36 h,PAB up-regulated the expression of cdc 2 and nuclear cyclin B1.Fas antagonistic antibody UB2 had no effect on apoptosis,but agonistic antibody CH11 enhanced the apoptosis induced by PAB.UB2 exerted no effect on cell cycle arrest,and CH11 had the same action as UB2 except for reducing the mitotic arrest through enhancing apoptotic subdiploid peak.Conclusion PAB inhibited MCF-7 cell growth through mitotic arrest and apoptosis.Apoptosis and mitotic arrest were independent of Fas pathway.

7.Determination of ferulic acid in Jianpibuxue tablets by HPLC

Objective To establish a determination method of ferulic acid in Jianpibuxue tablets.(Methods)HPLC was used with Lichrospher C_(18)(4.6mm?250mm,5?m) as the column.The mobile phase was a mixture of acetonitrile-0.378mol/L acetum(30∶70) and in a flow rate of 0.8mL/min;The detective wavelength was 321nm;Column temperature was room temperature and the injection quantity was 10?L.(Results)Ferulic acid in samples was well separated.The linear range was 1.0-10.0mg/L(r=0.9996).The average recovery was 99.8% and RSD was 0.26 %(n=6).Conclusion This method can be used for quality control of ferulic acid in Jianpibuxue tablets.

Objective Based on the traditional Chinese medical knowledge service and sharing system, to explore the application of famous doctor Yu Ying'ao's regulating the liver method. Methods The medical records of Yu Ying'ao from the outpatient department of Chinese Academy of Sciences were used as the data source. The traditional Chinese medical knowledge service and information sharing system(V1.1.5) was applied to construct database. The regulating liver core combination of drugs, drug application mode, and combined treatment of diseases and syndromes were determined by analysis methods of frequency and association rules. Results The use of regulating liver method was often used in the treatment of irregular menstruation, cirrhosis, pain and other diseases. Differentiation of liver qi stagnation, blood stasis, liver stagnation and spleen deficiency, liver and kidney deficiency were the main TCM syndromes. Treatment included Tongluo, nourishing Yin treatment. The core drugs were Bupleurum and Rhizoma Cyperi. Conclusions The use of traditional Chinese medical service and knowledge sharing system could explore the clinical diagnosis and medication rules of regulating liver methods.

Objective To assess the impact of intervention with Insulin therapy management unit program in the poorly controlled type 2 diabetic patients in a community of Shanghai.Methods There were 55 patients with type 2 diabetes with poorly controlled hyperglycemia (HbA1C ≥ 8%) enrolled in this study.They were divided at random into 2 groups:intensive care group and standard care group.The subjects in intensive care group were provided with a glucose meter and required to monitor their blood glucose levels at least 2 or 3 times per day.Community health provider acquired information of blood glucose level,episodes of hypoglycemia,and dosage of insulin every week by cell phone.Standard care patients received diabetes care from the same provider in outpatient clinic every month.Results By the end of 6 months of intervention,the intensive care group showed a significant difference in HbA1C as compared to the standard care group (7.40% ±0.91% vs 8.65 % ± 1.28%,P<0.01).The frequency of self-monitoring of blood glucose (SMBG) was 4 times per month in both groups at baseline.After intervention,the frequency of SMBG in intensive care group was greatly increased compared to standard care group (50times per month vs 5 times per month,P<0.01).The frequency of self-reported hypoglycemia in intensive care group was increased compared to standard care group (3 times per month vs once per month,P<0.01).The average daily dose of insulin in intensive care group was increased 6 units by the end of the present study(P>0.05).Conclusion After 6 months of intervention,the glycemic control was obviously improved in type 2 diabetic patients treated with insulin and the daily dose of insulin was not increased significantly.TheInsulin therapy management unit is effective and safe.
