AIM To study whether chlorpromazine(CPZ) and verapamil (Ver) have protective effects on the nephrotoxicity of cadmium (Cd). METHODS Thirtytwo Wistar rats were divided randomly into four groups. Each agent was injected 5 times per week for 6 weeks. The rats in Cd-treated group were sc injected with CdCl2 7μmol·kg-1. The rats of CPZ- and Ver- pretreated group were ip injected with CPZ 5 mg· kg- 1, Ver 4 mg· kg- 1,respectively, 1 h later sc injected with CdCl2 7 μ mol·kg- 1. The control group was sc injected with saline 2 mL·kg-1 at corresponding time. Twenty-four hours after the last injection, the 24-h urine samples were collected. The renal cortex was also excised. Lactate dehydrogenase (LDH) activity, protein and Cd concentration in urine were determined. The activities of protein kinase C (PKC), Na + -K + -ATPase, Ca2 + -ATPase and Cd concentration of renal cortex were also measured. RESULTS Cd concentrations of renal cortex and urine in rats from Cd-treated group were significantly higher than those of control group.Cd concentrations in urine of rats from CPZ- and Ver-pretreated groups were significantly lower than those of Cd-treated group, but there was no significant change in renal cortex. As compared with control group, LDH activity, protein content in urine and the activities of PKC, Na+ -K+ -ATPase and Ca2+ -ATPase in the rats of Cd-treated group increased significantly. LDH activity, protein content in urine and activities of PKC,Na+ -K+ -ATPase and Ca2+ -ATPase in rats of CPZ- and Ver-pretreated groups were significantly lower than those of Cd-treated group. CONCLUSIONCd could activate the activities of PKC,Na+-K+-ATPase and Ca2+-ATPase. Moreover, pretreatment of CPZ and Ver could reduce nephrotoxicity of Cd.

Objective To study the effect of the mouse angiostatin cDNA gene transfecting into human liver cancer cell line HCC7721 on cell growth, cell cycle phase distribution, cell morphology in vitro and tumorigenesis in vivo , and the mechanisms. Methods The gene fragment of mouse angiostatin cDNA was directly cloned into an eukaryotic expression plasmid pcDNA3.1(+) of the promoter CMV between the multicloning sites HindⅢ and XbaⅠ and confirmed the correct recombinant plasmid pcDNA3.1(+) angio through enzymatic digestion and gene sequencing. Then it was transfected into human liver cancer cell line HCC7721 with liposome, pcDNA3.1(+) as vecter control and liposome as mock control. After 30 day selection by neomycin G418, angiostatin expression at the levels of mRNA and protein in vitro were tested by RNA dot blot and FACS respectively. Cell morphology under was observed light microscope, made growth curves were made, and cell cycle distribution checked in FACS. Animal model was set up to study tumorigenesis in vivo of those cells transplanted subcutaneously in the right hind legs of nude mice BALBc. Microvessel density (MVD) was analyzed and angiostatin expression in the tumor tissues by in situ immunohistochemistry. Results The recombinant eukaryotic expression plasmid pcDNA3.1(+) angio was constructed Auccessfully. After selection of G418 for about 4 weeks there were macroscopic cell clones in the experimental group and vector control group, but no survival cells in the mock control. In vitro angiostatin were expressed in the experimental group, but not in the vector control. There weren't significant changes in cell morphology, cell growth curves and cell cycle distribution between the experimental and the control group. However, the nude mice experiment showed that tumorigenic capability of the experimental cells had been reduced greatly. Immunochemistry study showed that there were much less microvessels and conversely stronger angiostatin positive staining in the tumor tissues than in the control group. Conclusions Mouse angiostatin has no direct inhibition on human liver cancer cell line HCC7721 in vitro , but it inhibits effectively the tumorigenesis of HCC7721 in vivo , which is probably due to its inhibition on tumor angiogenesis in a paracrine path way.

Objective To search for and investigate the relationship between the differently expressed gene and pulmonary thromboembolism. Methods Peripheral blood mononuclear cells were isolated from pulmonary thromboembolism patients who were diagnosed and sex,age matched healthy controls. To screen differently expressed mRNA fragment which was related to pulmonary thromboembolism.Analyze one of the new ESTs.Simultaneity and homology comparison were made. Results First,fifty-five candidate mRNA fragments were chosen.Analysis of ten candidate mRNA fragments among them(up regulation or down regulation more than 3 times)were made by using Homology search, which confirmed that g44 and g48 were new sequence and were registered to GenBank.Making g44 be probe, northern blot was used to analyze one of the new ESTs.Signal of cross was found in controls and patients with pulmonary thromboembolism. It was significantly strong in the patient group compared with the control group. Conclusion These results reveal that new fragments have a strong expression in pulmonary thromboembolism group than control group. It suggests that these two new gene fragments may play an important role in the process of pulmonary thromoembolism as pathogenic gene.

Objective To observe the effects of lanthanum on learning and memory, and explore the corresponding mechanism on the basis of the levels of excitatory amino acids and calcium homeostasis of cerebral cortex in rats. Methods Forty female Wistar rats were randomly divided into control,low-,moderate-and high-dose lanthanum groups(ten rats in each group). The offspring of lanthanum exposure groups were administrated with lanthanum from weaning for one month. The capability of learning and memory was measured by neuroethology testing. The levels of excitatory amino acids, intracellular calcium ion and Nissl body expression of cerebral cortex were also detected. Results The capabilities of learning and memory in lanthanum exposure groups were significantly lower compared with the control, with a dose-dependent manner. The levels of glutamate and intracellular calcium ion of cerebral cortex in low-and moderate-dose group were significantly higher than those in controls, and the levels of glutamate, aspartate and intracellular calcium ion of cerebral cortex in high-dose group were increased significantly as compared with the control group, low dose group and moderate dose lanthanum group.Besides,Nissl body expression of cerebral cortex in low-dose group was lower than that in control group, and the levels of Nissl body expression of cerebral cortex in middle dose lanthanum group and high dose groups decreased further. Conclusion The increased levels of excitatory amino acids, calcium homeostasis unbalance and cellular injuries in cerebral cortex may be involved in lower capacity of learning and memory in the rats exposed to lanthanum.

Objective To investigate the effect of high glucose and losartan on cell proliferation and cyelooxygenase-2 (COX2) expression in normal human mesangial cells (NHMCs), and to examine the effect of losartan on COX2 and transforming growth factor-betal (TGF-β1) expression in a model of diabetic nephropathy (DN). Methods NHMCs were cultured in vitro in high glucose media with or without losartan. NHMCs proliferation and COX2 expression were determined by WST-1, Western blot, and RT-PCR. The rat model of DN was produced by injections of streptozocin (STZ). After the treatment with losartan for 4 weeks, glomerular hypertrophy, urinary thromboxane B2(TXB2) and 24 h urinary pro-tein counts were measured, and COX2 and TGF-β1 expressions were investigated using immunohistochem-ical techniques and RT-PCR. Results Losartan dose-dependently inhibited the proliferation of NHMCs in response to high glucose. Losartan also decreased COX2 expression in NHMCs at high or low glucose concentrations. In vivo experiments found kidney weight/body weight (KW/BW), urinary TXB2 and 24 hurinary protein counts increased significantly in the DN group. Losartan reduced KW/BW, urinary TXB2,and 24 h urinary protein counts and significantly suppressed the over-expression of COX2 and TGF-β1.Conclusion Losartan reduces COX2 expression in NHMCs, especially at high glucose concentrations.Losartan could suppress the expression of COX2 and TGF-β1 in the kidney of DN rats and attenuate the renal lesions caused by DN.
