1.Effect ofJianpi-Huoxue decoction combined with conventional therapy of western medicine on adrenocorticotropic hormone levels and vascular endothelial growth factor of patients with chronic nephritis

Objective To observe the effect ofJianpi-Huoxue decoction on the adrenocorticotropic hormone levels and vascular endothelial growth factor of patients with chronic nephritis.Methods 120 patients with chronic nephritis from January 2012 to December 2013 in TCM Hospital of Yanggu county were selected and recruited into a control group and a treatment group, with 40 patients in each group, according to random number table method. The control group was given oral indomethacin, 1 tablets each time, 3 times/d; intramuscular injection of methotrexate, 1 piece/time, 1 times/week. The treatment group was additionally treated by Jianpi-Huoxue decoction on the basis of the control group. Both groups were treated for 3 months. The serum adrenocorticotropic hormone and vascular endothelial growth factor levels were measured before and after the treatment,and efficacy and safety were also evaluated.Results After the treatment, the total effective rate was 72.5% (29/40) in the control group and 97.5% (39/40) in the treatment group, with significant differences (χ2=7.941,P<0.05). The ACTH levels after treatment was (13.82 ± 5.94 ng/Lvs. 8.01 ± 4.16 ng/L) in the treatment group and (18.73 ± 6.84 ng/Lvs. 8.07 ± 4.08 ng/L) in the control group, showing significant increase compared with before treatment (P<0.01). The level of VEGF was (61.92 ± 26.37 ng/Lvs. 83.72 ± 30.63 ng/L) in the treatment group and (49.36 ± 23.93 ng/Lvs.84.34 ± 33.49 ng/L) in the control group, showing significant decrease than before treatment (P<0.01). After the treatment, the increase of ACTH and the decrease of VEGF in the treatment group were better than those in the control group (t=3.427, 2.231,P<0.05).ConclusionsJianpi-Huoxue decoction combined with conventional western therapy can increase the level of ACTH in patients with nephritis, reduce VEGF, and improve clinical curative effects.
2.Association between serum glial fibrillary acidic protein and hypertensive intracerebral hemorrhage

Objective To investigate the association between serum glial fibrillary acidic protein (GFAP) and hypertensive intracerebral hemorrhage(HICH). Methods The level of serum GFAP was detected in 68 patients with HICH by enzyme linked immunosorbent assay(ELISA) and its relationship with bleeding volume,bleeding part,neural deficit score(NDS) and prognosis was analysed, also compared those with the normal control (NC) group. Results Compared with NC group, the levels of serum GFAP in HICH group were increased significantly in 1~14 d after onset (all P20 ng/ml had worse prognosis.Conclusions The level of serum GFAP in patients with HICH is increased significantly, and it is correlate with bleeding volume, condition and prognosis.
3.Hepatitis B surface antigen terminates codon bias selection

Objective:To investigate the existence condition of hepatitis B surface antigen(HBsAg) termina-tion codon bias.Methods: A total of 174 reference sequences of all kinds of Hepatitis B virus(HBV) genotypes were chosen from GenBank,and compared by BioEdit.Then secondary structure of RNA was constructed and analyzed together.Results:(1) There were two types of HBsAg termination codon: TAA and TGA in 174 reference sequences.TAA was in 124 cases(71.26%);and TGA in 50 cases(28.74%).(2) There was codon bias selection in HBsAg termination codon,and it could affect the secondary structure of RNA and amino acid sequence encoding protein.Conclusion: HBsAg termination codon bias exists and may be related to RNA structure and the conservatiom of protein function in the evolutionary progress.
4.Clinical value of expression of CEA and CK-20 in peritoneal washes on predicting peritoneal micrometastasis and prognosis of gastric carcinama

Objective To explore the clinical value of expression of CEA and CK-20 detected by flow cytometry (FCM) in peritoneal washes on predicting peritoneal metastasis and prognosis of gastric carcinama.Methods Clinicopathological characteristics and follow up data of 105 patients with gastric carcinoma who were underwent D2 radical resection (R0) were collected.Peritoneal washes was collected.Peritoneal lavage cytology examination (PLC) was used to find intraperitoneal free cancer cells (IFCC).FCM was used to determine the CEA and CK-20 expressions.Meanwhile,13 patients with benign lesion on the stomach and the gastric carcinama cell line SGC-7901 were served as the negative and positive control,respectively.Results Positive expression of CEA was in 48 (45.7 %) patients,of CK-20 was found in 67 (63.8 %) patients,and of CEA and CK-20 was in 85 (81.0 %) patients by FCM.However,positive expression of IFCCs was found in 31 patients(29.5 %) by PLC.The expression of CEA and CK-20 were related to serosa invasion,lymphnode metastasis and pTNM stage (P < 0.05).The median survival of patients with negative expression of both CEA and CK-20 (n =20) was significantly longer those with positive expression (n =30)(52 vs 18 months,P < 0.05).Conclusions Combined Detection of CEA and CK-20 in peritoneal washes by FCM can be used to predict peritoneal micrometastasis and may predict the prognosis of gastric carcinoma.
5.Effect of HPV E6 siRNA on the proliferation and chemotherapy sensitivity of cervical cancer HeLa cells

Objective To elucidates the effects of HPV18 E6 siRNA targeting at human papillomavirus(HPV)18 E6 gene on the proliferative activity of HeLa cells and chemotherapy sensitivity.Methods HPV18 E6 expression of HeLa cells was inhibited by siRNA interference,the change of P53 and P21 proteins expression level was measured by Western blot.MTT assay was used to detected proliferative activity and sensitivity to paclitaxel liposome of HeLa cells.Results After inhibition of E6 expression,P53 and P21 proteins increased and the growth of HeLa cells was decreased(P <0.01).The inhibition rate of HeLa was markedly increased after transfection of HPV18 E6 siRNA and paclitaxel liposome.Conclusion HPV18 E6 siRNA can effectively silence gene expression of E6 and inhibit proliferation of HeLa cells.HeLa cells are more sensitive to combine HPV18 E6 siRNA with paclitaxel liposome than that of control groups.

Systems biology, which includes genomics, proteomics, transcriptomics and metabolomics, is an integration of system theory and biology. It studies the life phenomena with thinking method of systems theory. And it provides an important methodological basis and technical conditions for recognizing syndrome of traditional Chinese medicine (TCM) and mechanism of therapeutic effect from multiple disciplines and perspectives. Systems biology is similar to the core idea of TCM system and holism concept. Therefore, systems biology can be used as a platform and pivot of non-alcoholic fatty liver disease (NAFLD) in modern TCM study. Systems biology will accelerate the modernization research of TCM in the prevention and treatment of NAFLD. It will also provide new ideas and methods for TCM study on NAFLD.
7.To explore learning burnout and professional commitment of nursing students studying at the traditional Chinese medicine college

Objective To explore learning burnout psychology and professional identity of nursing students studying at the traditional Chinese medicine college.Methods 200 students were surveyed by learning burnout and professional commitment scale.Results Learning burnout level of nursing students was higher,depression scores were the highest in four dimensions.Professional commitment kept within medium level.The professional identity was low.The inheritance commitment score was the lowest in each dimension analysis.Conclusions Schools and teachers should take corresponding measures to improve professional commitment level,so as to reduce learning burnout level and ensure long-term stable development of nursing profession.
8.Study on Isolation and Purification Technology of Active Components of Senecio cannabifolius with Macro-porous Adsorption Resin

OBJECTIVE:To optimize the isolation and purification technology of active components of Senecio cannabifolius with macroporous adsorption resin. METHODS:The type of macroporous adsorption resin was selected with static adsorption using adsorption capacity and resolution rate of chlorogenic acid and hyperoside as index. The isolation and purification condition was op-timized with dynamic adsorption using the elution volume of chlorogenic acid and hyperoside as index,such as maximal sample size,rinse water quantity,volume fraction and collective volume of eluant ethanol. RESULTS:Among 7 kinds of resin,HPD100 had the best purification property;the optimal purification technology was as follows as mass concentration of sample 6 mg/ml,the speed of sample loading 2 ml/min,maximal sample size 3 times of column volume(BV), rinse water quantity of 2.5 BV,60%ethanol as eluting reagent. The contents of chlorogenic acid and hyperoside were increased from 0.90,0.18 mg/g to 7.26,1.04 mg/g after purification. RSD of each index were all ≤3.0%(n=3) in validation test. CONCLUSIONS:The isolation and purification technology of active components of S. cannabifolius with HPD100 macroporous adsorption resin is stable and effective.
9.Study of the expression of osteopontin in epithelial ovarian cancer tissue and serum

Objective To study the expression and signficance of the osteopontin (OPN) in epithelial ovarian cancer tissue and serum. Methods Immunohistochemistry method and ELISA were used to detect the expression of OPN in 64 cases of epithelial ovarian cancer tissue and serum, 20 cases of ovarian benign tumors and 10 cases of ovarian nomal tissues. Results The OPN expression was associated with the clinical staging and histological grading of epithelial ovarian cancer tissue and serum (P < 0.01). The level of OPN in epithelial ovarian cancer tissue was significantly higher than that in benign ovarian tumor and normal control groups (P < 0.01). Conclusion OPN is remarkably correlated with the carcinogenesis and the development of epithelial ovarian cancer.
10.Neo-adjuvant chemotherapy sensitivity and associated gene expressions in cervical cancer

Objective To study the associations between neo-adjuvant chemotherapy (NACT)sensitivity and expression of relative proliferation and apoptosis genes in cervical cancer tissues. Furthermore,the potential roles of relative genes expression as monitor in the chemotherapy against cervical cancer tissues were studied. Methods Fifteen pathologically proved Ⅱ A stage cervical cancer patients with HPV 16 positive were divided into effective group and non-effective group according to the clinical response to NACT, the changes of HPV16-E6, p53 genes and proteins expressions were measured by real-time quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blot. At the same time, MTT assay was used to detected proliferative activity to paclitaxel liposome and carboplatin (paraplatin) in HeLa cells. Results In effective group, the overall response rate was 73.3 % (11/15) and the HPV16-E6 mRNA and protein level after NACT was significantly decreased than that before NACT (t=3.359, t=3.614, P<0.05), while p53mRNA and protein level increased significantly (t =5.852, t=2.838, P <0.05). However, there have no significant different between pre-NACT and Post-NACT of the two genes in the non-effective group. After using chemotherapy HeLa cells growth decreased. Conclusion HPV16-E6 and p53 expreesion level are useful paraneters in evaluating NACT response in cervical cancer tissues. The detection of the two genes expression is a method for predicting efficacy in NACT.
